1.Sinicization and measurement invariance testing of the Health Behavior Scale for Cancer Patients

Objective To translate the Health Behavior Scale for Cancer Patients(HBSCP)into Chinese and conduct reliability and validity tests.The measurement invariance of the Chinese version of the scale among differ-ent cancer patients was measured to form a health behavior scale suitable for assessing cancer patients in China.Methods The original scale was translated and translated back according to the Brislin translation model.The Chinese version of the HBSCP was formed through cross-cultural adaptation and pre-test.A questionnaire survey was conducted on 600 cancer patients from 2 tertiary A hospitals in Tianjin and Nanchang from February to July 2024 by convenience sampling method to test the reliability,validity,and measurement invariance of the Chinese version of the HBSCP.Results 567 valid questionnaires were collected.The Chinese version of the HBSCP in-cludes 9 items with 2 factors.Confirmatory factor analysis supported the 2-factor model,and all indexes meet the ideal fitting standard.The factor load of each item on the scale ranged from 0.697 to 0.815.The composite relia-bility of the 2 dimensions was 0.815 and 0.874.Average variance extracted was 0.656 and 0.526.The Chinese version of the HBSCP was positively correlated with Health Promoting Lifestyle Profile-Ⅱ(r=0.760,P<0.001).The con-tent validity index of items ranges of the scales from 0.930 to 1.000,and the content validity index/average of the scales was 0.993.The total Cronbach's alpha coefficient of the scale was 0.900,and the two-factor Cronbach's alpha coefficient was 0.849 and 0.873,respectively.The scale has measurement invariance in the equivalence test for 3 clusters of lung,gastrointestinal,and gynecologic cancers.Conclusion The Chinese version of the HBSCP has good reliability and validity and measurement invariance across cancer types,and can be used to measure the health behavior level of cancer patients.
2.Construction of p97 mutant of Mesomycoplasma hyopneumoniae based on the homologous recombination system

The aim of this study is to establish an gene editing method of Mesomycoplasma hyo-pneumoniae(Mhp)based on the homologous recombination principle.The restriction enzyme di-gestion and ligation method combined with gene synthesis were used to construct a shuttle plasmid to achieve replication in both Mhp and Escherichia coli(E.coli).The pGEM?-T vector was used as the skeleton.The oriC sequence of Mhp which can achieve the replication of the plasmid in Mhp was inserted into the vector.Sequences of the Spiroplasma promoter and puromycin resistance gene were then inserted into the above constructed plasmid to screen recombinant clones.The up-stream and downstream homologous arms of p97 were constructed to initiate homologous recombination.The recA gene of E.coli is inserted to improve the efficiency of homologous recom-bination.The obtained shuttle plasmid was then delivered into Mhp by electro-transformation or chemical transformation.A shuttle plasmid,pGEM?-Mhp-oriC-p 97,which can replicate in both Mhp and E.coli was constructed.With the transformation of this plasmid,the carried puromycin gene and recA gene can be expressed,the p97 gene can be edited.Finally,the genetically unstable p97 gene mutant was initially obtained.In this study,a tool for Mhp gene editing based on the principle of homologous recombination was established,which laid a foundation for the develop-ment of tools for studying the pathogenesis of Mhp.

5.Effects of gabapentin on plasma β-endorphin level in the patients with painful diabetic neuropathy

Objective To investigate the effects of gabapentin(GBP) on plasma β-endorphin(β-EP) level in the patients with painful diabetic neuropathy(PDN). Methods We detected the plasma β-EP level in 24 PDN patients in the treatment with GBP, 18 PDN patients without GBP treatment, 20 diabetic mellitus patients without PDN and 24 healthy control subjects. Results (1)The level of β-EP in diabetic mellitus patients was lower than in the healthy control(P<0.01). (2)The patients who received GBP had a significant change of β-EP level after treatment(P<0. 01).. Conclusions Gabapentin is effective for the treatment of PDN and its adverse effects are mild. It can lower the plasma β-EP level in the patients with PDN.
6.Preparation and characterization of an extracellular matrix of artificial tendon tissue from natural macromolecules.

Collagen and chitosan are well natrual polymers to be used as extracellular matrix on tissue engineering because of their biocompatibility, certain mechanical strength and biodegradability. But there are some disadvantages when they are used to construct extracellular matrix respectively. This experiment utilized their complementary performances to prepare a composite extracellular matrix of artificial tendon tissue that had adequacy mechanics strength and good biocompatibility, cell affinity, biodegradability. Collagen and chitosan were covalently crosslinked using EDC and NHS to obtain a porous scaffold material that the porous was oriented under an external force. Then RGD peptide was covalently attached to scaffold material surface to improve its affinity with cells. The microstructure of scaffold material was observed under microscope and scanning electron microscope. Simultaneously, the physical performance, hydrophilicity, ecto-degradation rate and cell compatibility of scaffold material were measured in the experiments. The results showed that this scaffold material was soft and stretchy. Its tensile strength was 15.0 MPa, corresponding shape extension was 7.33%, and its porosity was 79.4%. Its water absorption rate and water retention rate were 772% and 206% respectively. Its degradation rate in RPM 1640 culture mediun with 10% fetal bovine serum and in human serum were 4.13% and 37.2% respectively after three weeks. These degradation rates are suitable for the rehab course of injured tendon. Moreover the degradation rates can be controlled by adjusting technological conditions and degree of cross linking. Significantly higher affinity with 3T3-Ll cell was detected on the scaffold material modified by RGD peptide. The various physical performances of this complex scaffold material are appropriate for constructing extracellular matrix of artificial tendon tissue or artificial skin. Moreover, it could be used as soft tissue slurry for plastic surgery.
