1.Improvement effect of arbutin on myocardial fibrosis model rats and its mechanism

OBJECTIVE To study the improvement effects of arbutin on myocardial fibrosis (MF) model rats and its mechanism. METHODS The network pharmacology was used to predict the potential target of arbutin in improving MF and molecular docking was used to validated. Totally 50 SD rats were given isoprenaline subcutaneously (5 mg/kg, once a day, for 14 consecutive days) to induce the MF model. Modeled rats were randomly divided into model group, captopril group (9 mg/kg), arbutin low-dose, medium-dose and high-dose groups (50, 100, 200 mg/kg), with 10 rats in each group. Another 10 healthy rats were included as normal group. Each group was given the corresponding drugs, once a day, for 28 consecutive days. Twenty-four hours after the final administration, electrocardiograms and heart-related indexes [heart weight index (HWI), left ventricular weight index (LVWI)] of rats were detected; the levels of creatine kinase (CK), lactate dehydrogenase (LDH), N-terminal pro-brain natriuretic peptide (NT-proBNP) and type Ⅰ collagen (Col Ⅰ) and Col Ⅲ were detected in myocardial tissue of rats; the pathological changes of myocardial tissue were observed, and protein and mRNA expressions of adenosine deaminase (ADA) and adenosine kinase (ADK) were detected in the myocardial tissue of rats. RESULTS The results of network pharmacology showed that the main targets of arbutin improving MF were ADA and ADK. The results of molecular docking showed that arbutin bind stably with ADA and ADK. The results of experimental verification showed that compared with model group, the amplitude of ST and T waves in electrocardiogram were improved in administration groups, and the symptoms of atrial flutter were alleviated; HWI (except for arbutin medium-dose group), LVWI, the levels of CK, LDH, NT-proBNP, Col Ⅰ and Col Ⅲ in the myocardial tissue of rats were decreased significantly (P<0.05); the degree of myocardial fibrosis in rats decreased; protein and mRNA expressions of ADA and ADK in the myocardial tissue were significantly increased (P<0.05). CONCLUSIONS Arbutin can improve cardiac fibrosis and cardiac function of MF model rats, the mechanism of which may be associated with up-regulating protein and mRNA expressions of ADA and ADK,influencing the nucleotide metabolism and collagen generation. zhangminghao@hactcm.edu.cn
2.Improved extraction efficiency of bakuchiol with antioxidant and whitening bioactivities from the seeds of Psoralea corylifolia L.

Background: Mining new whitening ingredients from plants is an effective way to develop new cosmetic products. Bakuchiol, as one of the main active ingredients from the seeds of Psoralea corylifolia L, displays whitening, antioxidant, antiaging, and free radical-scavenging activities and attracts wide attention especially in the cosmetics industry. However, environmentally friendly extraction technologies with high efficiency and high product purity remain to be developed for the scale production of bakuchiol. Objective: In this study, a new approach (acetic acid-steaming-assisted technique) was developed to improve the extraction efficiency of bakuchiol. Methods: The factors influencing the extraction efficiency of bakuchiol, including seed powder granularity, acetic acid volume, temperature, and time, were optimized based on single-factor experiments. Results: The extraction conditions were optimized as steaming the seed powder (100 g) of 40-100 meshes with acetic acid (50 mL) for 10 minutes, from which a crude extract (25.1 g) containing bakuchiol was obtained. High purity (99.1) of bakuchiol (6.02 g) was then prepared by silica gel column chromatography in a one-step process. The method established in this study had higher extraction rate than other methods. The antioxidant potential of bakuchiol was evaluated based on the ABTS free radical scavenging activity and ferric reducing antioxidant power. Furthermore, bakuchiol demonstrated stronger tyrosinase inhibitory activity than the positive control (arbutin). Conclusions: Compared with the available methods, the method established in this study has increased extraction rate of bakuchiol and is environmentally friendly and economical, demonstrating the potential for industrial production.
3.Diagnosis and Treatment of Phyllode Tumor of Breast

Objective To study the histopathological features, diagnosis criteria, the relationship of surgery pattern and prognosis of phyllode tumor of breast. Methods To analyze the histopathological features and clinical outcome of different surgery patterns in 203 patients with phyllode tumor of breast by Chi square test, Cluster, Focater, Logistic and Cox multivariate regression according to the request of SPSS 10. 0. Results 203 patients with phyllode tumor of breast were divided into three groups, i.e. benign 133 cases, borderline 42 cases and malignant 28 cases. Local recurrences in three groups were 28, 19 and 18, respectively. The patients died from tumor were 0, 2 and 16, and circulatory metastases were 0, 1 and 10, respectively. Five-year survival were 100 %,92. 0% and 33. 3% in the three groups of 131 patients by a 5 years' follow-up survey. Conclusions Tumor necrosis has important value in the diagnosis of phyllode tumor. Nature of tumor margin, cellular pleomorphism, frequency of mitoses and tumor necrosis were statistically appropriate composition in histological diagnosis of phyllode tumor. Wide local excision is preferred for the benign and borderline phyllode tumor, while simple mastectomy is indicated for recurred borderline and malignant, but tylectomy should be abolished in the treatment of phyllode tumor. Correlation of histotypes of phyllode tumor with local recurrence and tumor death was statistically significant at a level of P<0. 001; correlation of infiltrative growth of the tumor with local recurrence was statistically significant at a level of P<0. 001. Tumor necrosis and mitotic activity were independent prognostic factors.
4.Expression of p73 Protein in Giant Cell Tumor of Bone

Objective: Our aims were to study the expression of p73 protein in giant cell tumor of bone, and to investigate its function in the carcinogenesis and development of giant cell tumor of bone. Methods: The expression of p73 was detected by using S-P immunohistochemical method in 40 cases of paraffin-embedded sections of giant cell tumor of bone. Results: The positive rate of p73 in 40 patients with giant cell tumor of bone was 30.0%,and 20.0% in osteochondroma.The difference between them was not significant(P<0.05). There was significant difference between the expression of p73 in different stages of giant cell tumor of bone(P>0.05). Conclusion: The expression of p73 in giant cell tumor of bone increased prominently in this experiment. It suggests that p73 may play an important role in the carcinogenesis and development of giant cell tumor of bone, and p73 may also have a certain value for the diagnosis of giant cell tumor of bone.
5.Histopathological study of colorectal injury after endoscopic polypectomy

Objective The study was attempted to observe the colorectal injury due to endoscopic snare polypectomy. Methods Histopathological study was conducted on 47 snared colorectal polyps as well as on the corresponding sites in 34 patients who underwent surgery thereafter for malignancy. Results The injury to all snared specimens was confined within submucosa,while that to surgically resected specimens at the snared sites extended to muscular and serosal layers in 28 out of 34 cases, which manifested as focal necrosis in inner muscularis and hemorrhage between inner and outer muscle layers, bearing no continuity with submucosal change.The mean size of polyps snared with injury to muscularis and serosa at the site was larger than those without ( P10mm.
