1.Protection of exosomes derived from bone marrow mesenchymal stem cells of different mouse ages on radiation-induced lung injury

BACKGROUND:Mesenchymal stem cells show extremely therapeutic potential for radiation-induced lung injury through delivering exosomes.Age is a primary factor affecting the function and biological efficacy of mesenchymal stem cells.OBJECTIVE:To investigate the protective effects of exosomes derived from bone marrow mesenchymal stem cells of different mouse ages on radiation-induced lung injury in mice.METHODS:Bone marrow mesenchymal stem cells of young mice and old mice were obtained by whole bone marrow adherent culture.The exosomes were isolated from the supernatant of passage 3 bone marrow mesenchymal stem cells.Ten 2-month-old C57BL/6J mice were randomly selected as the control group after anesthesia and not irradiated.The remaining 30 2-month-old C57BL/6J mice were used to establish a mouse radiation-induced lung injury model and were randomly divided into three groups.Exosomes derived from bone marrow mesenchymal stem cells of young mice,exosomes derived from bone marrow mesenchymal stem cells of old mice,and PBS were injected through the tail vein,respectively.The survival rate of mice was monitored.The lung function,lung inflammation and fibrosis were assessed at 1 and 12 weeks after irradiation.RESULTS AND CONCLUSION:(1)The concentrations of particles and proteins in exosomes derived from bone marrow mesenchymal stem cells of young mice were higher than those in exosomes derived from bone marrow mesenchymal stem cells of old mice.(2)Compared with the control group,the survival rate of mice in the PBS group was low,and lung inflammation was obvious at week 1 after irradiation,and the levels and mRNA expressions of interleukin-1β,interleukin-6,and tumor necrosis factor-α were increased.Collagen deposition in lung tissues was observed at week 12 after irradiation,and the mRNA level of E-cadherin was decreased,while the mRNA levels of α-smooth muscle actin,transforming growth factor-β1,and β-catenin were increased.(3)Compared with the PBS group,the survival rate of mice in the exosome group was significantly improved,and the level of proinflammatory factors and their mRNA expression were reduced at week 1 after irradiation,the mRNA level of E-cadherin was increased,and the mRNA levels of α-smooth muscle actin,transforming growth factor β1 and β-catenin were reduced at week 12 after irradiation.(4)Among all the above indicators,the therapeutic effect of exosomes derived from bone marrow mesenchymal stem cells of young mice was better than that of exosomes derived from bone marrow mesenchymal stem cells of old mice.(5)The results showed that exosomes derived from bone marrow mesenchymal stem cells of young mice contained more particles and proteins,and the effect of alleviating early inflammation and late fibrosis of radiation-induced lung injury in mice was better than that of exosomes derived from bone marrow mesenchymal stem cells of old mice.

3.Factors associated with anxiety symptoms among Filipino farmers in Central Luzon: An analytical cross-sectional study.

BACKGROUND AND OBJECTIVE
Filipino farmers face unique occupational challenges that increase the risk of mental health issues, particularly anxiety. This study aims to determine the different personal, environmental, socioeconomic, occupational, and psychosocial factors associated with anxiety symptoms among Filipino farmers in Central Luzon.
METHODSChain referral sampling method was used to recruit participants for the study, who underwent screening based on the eligibility criteria. Eligible participants were then asked about anxiety symptoms using the Generalized Anxiety Disorder-7 (GAD-7), while the validated, researcher-constructed Data Collection Tool for Factors Associated with Anxiety Symptoms (DCFAAS) was used to determine the farmers’ exposure to a variety of factors. Microsoft Excel was utilized in computing for frequency and percent distribution of participants, in each factor. Binary logistic regression was used to compute crude and adjusted odds ratio of each factor thru IBM SPSS Statistics®.
RESULTSAmong the 113 eligible farmers enrolled in the study, only 19 (16.8%) experienced anxiety symptoms, with excessive worrying, which was seen among 45 participants (39.8%). The mental health of Filipino farmers was significantly affected by the presence of physical illness (OR = 10.70 [95% CI 1.367, 83.773]) and having relatives affecting work completion (OR = 6.45 [95% CI 1.346, 30.896]).
CONCLUSIONDespite the low prevalence of anxiety symptoms in this study, the findings suggest government policies to improve mental health service access to farmers, to integrate psychosocial support into agricultural programs, and to address family-related work pressures. By addressing these factors, it can improve farmer productivity and promote overall well-being, putting emphasis on the mental health of the Filipino farmers.

4.Data Mining Analysis of TCM Compound Patents for Treatment of Myocardial Infarction

Objective To analyze the medication law and compatibility characteristics of TCM compounds for the treatment of myocardial infarction in the national patent database.Methods TCM compounds for treating myocardial infarction were retrieved from CNIPA patent publication website.A prescription database was built using Excel 2019 software to statistically analyze the frequency of medicinal use and their properties,taste and meridian tropism;SPSS Modeler 18.0 software was used to analyze the association rules of drugs;a network of Chinese materia medica co-occurrence was constructed using Cytoscape 3.10.0,and systematic clustering analysis was performed on the Chinese materia medica in the core network.Results A total of 146 patents of TCM compounds were included,involved 440 kinds of Chinese materia medica.High frequency drugs included Salviea Miltiorrhizae Radix et Rhizoma,Chuanxiong Rhizoma,Angelicae Sinensis Radix,Glycyrrhizae Radix et Rhizoma,etc.The main property was warm,the main tastes were bitter,sweet and pungent,and the medicines mostly belongs to the liver meridian,heart meridian and spleen meridians.Commonly used medicinal pairs included Angelicae Sinensis Radix-Chuanxiong Rhizoma,Astragali Radix-Salviea Miltiorrhizae Radix et Rhizoma,Paeoniae Radix Rubra-Angelicae Sinensis Radix,etc.Commonly used tripartite combinations included Paeoniae Radix Rubra-Chuanxiong Rhizoma-Angelicae Sinensis Radix,Carthami Flos-Angelicae Sinensis Radix-Chuanxiong Rhizoma,Carthami Flos-Chuanxiong Rhizoma-Angelicae Sinensis Radix,etc.Clustering analysis showed four types of combinations.Conclusion TCM compound patents for the treatment of myocardial infarction is based on promoting blood circulation,removing blood stasis,and relieving pain,while also using methods such as eliminating phlegm,tonifying qi,warming yang,and nourishing yin.It can provide references for clinical medication.
5.Clinical predictive model for poor visual function recovery in patients after refractive cataract surgery

Objective:To investigate the factors influencing poor visual function recovery in patients after refractive cataract surgery, and to provide an effective clinical predictive model for improving and preventing poor postoperative visual function in such patients.Methods:A retrospective study was conducted on 314 patients who underwent refractive cataract surgery in Xi'an Ai'ergucheng Eye Hospital Affiliated to Northwest University from January 2021 to June 2024. The best corrected visual acuity (BCVA) was used to evaluate the postoperative visual function of patients. Patients were divided into a good visual function recovery group of 252 cases and a poor visual function recovery group of 62 cases based on their visual function recovery. The differences in general clinical data, surgical-related information and aqueous humor cytokines between the two groups were compared. Correlation tests and Logistic regression analysis were used to identify factors closely associated with poor visual function recovery in patients after refractive cataract surgery. The predictive efficacy of these factors for poor visual function recovery was evaluated using the receiver operating characteristic (ROC) curve, area under the curve (AUC) and decision curve.Results:The age, proportion of diabetes, proportion of severe cataracts, preoperative fasting blood glucose (FBG) and preoperative glycosylated hemoglobin (HbA 1c) levels in the poor visual function recovery group were significantly higher than those in the good visual function recovery group: 66 (64, 68) years old vs. 64 (61, 66) years old, 32.3% (20/62) vs. 17.9% (45/252), 48.4% (30/62) vs. 32.1% (81/252), (5.73 ± 0.94) mmol/L vs. (5.29 ± 0.84) mmol/L, (6.39 ± 0.76)% vs. (5.86 ± 0.64)%, and the differences were statistically significant ( P<0.05). The operation time, proportion of intraoperative vitreous leakage and phacoemulsification time in the poor visual function recovery group were significantly higher than those in the good visual function recovery group: (34.23 ± 4.13) min vs. (32.55 ± 2.20) min, 45.2% (28/62) vs. 26.2% (66/252), (19.81 ± 2.96) min vs. (18.62 ± 1.49) min, and the differences were statistically significant ( P<0.05). The levels of aqueous humor interleukin (IL)-1β and vascular endothelial growth factor (VEGF)-A in the poor visual function recovery group were significantly higher than those in the good visual function recovery group: (20.17 ± 3.71) ng/L vs. (18.54 ± 2.16) ng/L, (130.11 ± 15.54) ng/L vs. (122.35 ± 6.74) ng/L, and the differences were statistically significant ( P<0.05). Spearman correlation analysis, univariate and multivariate Logistic regression analysis confirmed that age ( OR = 1.762, 95% CI 1.430 to 2.172), preoperative FBG ( OR = 2.272, 95% CI 1.387 to 3.721), preoperative HbA 1c ( OR = 2.823, 95% CI 1.517 to 5.254), diabetes ( OR = 5.413, 95% CI 1.162 to 25.222), intraoperative vitreous leakage ( OR = 4.751, 95% CI 1.877 to 8.309) and aqueous humor IL-1β ( OR = 1.195, 95% CI 1.031 to 1.386) were important risk factors for poor visual function recovery after refractive cataract surgery ( P<0.05). ROC and decision curve analysis found that the combined application of these risk factors had a high predictive efficacy for poor visual function recovery in patients after refractive cataract surgery, with an AUC (95% CI) of 0.885 (0.839 to 0.931) ( P<0.05). Conclusions:Older age, higher preoperative FBG and HbA 1c levels, diabetes, intraoperative vitreous overflow and higher levels of aqueous humor IL-1β are important factors contributing to poor visual function recovery in patients undergoing refractive cataract surgery. The model constructed based on these indicators has a high predictive efficacy for postoperative visual function recovery in patients undergoing refractive cataract surgery.
6.An exploration of the historical development of modern Chinese dental societies

Modern Chinese dental societies have been a crucial part of the progress of stomatology in China, serving as both witnesses and key drivers of its development. By reviewing historical materials related to these societies, this paper focuses on analyzing their establishment background, developmental stages, overall scale, and major activities. The findings reveal that dental societies exhibited regional, phased, professional, and academic characteristics. Furthermore, the study highlights their contributions to strengthening dental education, promoting standardization within the industry, facilitating academic exchanges, and advocating for the integration of oral and systemic health. This research enriches the study of modern Chinese stomatology history.
7.Clinical features analysis of patients with inflammatory bowel disease accompanied by axial spondyloarthritis

Objective:To summarize and analyze the clinical features of patients with inflammatory bowel disease (IBD) accompanied by axial spondyloarthritis (ax-SpA) .Methods:A descriptive case series study was conducted. Consecutive IBD patients with ax-SpA admitted to Peking Union Medical College Hospital from January 2012 to December 2023 were enrolled, and the clinical data were collected and analyzed descriptively.Results:A total of 21 IBD patients with ax-SpA were enrolled, including 14 males (66.7%) and 7 females (33.3%). The median age at IBD diagnosis was 29 (24, 42) years. Among them, 10 had ulcerative colitis (UC) and 11 had Crohn's disease (CD). In 11 patients (52.4%), ax-SpA was diagnosed before IBD, while in 7 patients (33.3%), IBD was diagnosed before ax-SpA; the remaining 3 patients (14.3%) were diagnosed simultaneously. Thirteen patients (61.9%) had other extraintestinal manifestations, with 12 involving mucocutaneous manifestations. Nineteen patients including 9 UC and 10 CD were followed up for a median duration of 34 (12, 57) months. Among UC patients, 5 (55.6%) were escalated from conventional therapy to biologics or small-molecule drugs, and 6 (66.7%) developed opportunistic infections. Among CD patients, 3 (30.0%) were escalated to biologic therapy, and 2 (20.0%) developed opportunistic infections. Three patients (15.8%) underwent intestinal resection surgery.Conclusions:IBD patients with ax-SpA are predominantly male, with a similar proportion of UC and CD. There is no clear sequential pattern in the diagnosis timing of IBD and ax-SpA. These patients often present with other extraintestinal manifestations of IBD, primarily mucocutaneous manifestations.
8.Effect of high glucose on polarization of Raw264.7 macrophages in vitro

Objective:To investigate the induction effect of different concentrations of glucose on the polarization of Raw264.7 macrophages in vitro.Methods:The Raw264.7 cells cultured in DMEM medium were divided into control group(5.5 mmol·L-1 glucose),different high-glucose groups(15.0,25.0,35.0,and 45.0 mmol·L-1 glucose),and positive control group[lipopolysaccharide(LPS)].The cells were cultured for 3,6,and 9 h,respectively.Cell morphology was observed in various groups.Cell counting kit-8(CCK-8)method was used to assess the survival rates of cells in various groups.Real-time fluorescence quantitative PCR(RT-qPCR)method was used to detect the expression levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),and interleukin-10(IL-10)mRNA in cells in various groups.Enzyme-linked immunosorbent assay(ELISA)was used to measure the levels of IL-6,TNF-α,and IL-10 in the cell supernatants in various groups.Flow cytometry was used to determine the percentages of M1 and M2 macrophage markers,CD86+and CD163+cells,in various groups.Results:In control group,theRaw264.7 cells adhered to the culture dish,with a predominantly round morphology.The cells in 35.0 mmol·L-1 high-glucose group and positive control group showed elongated shapes and pseudopodium formation,indicating inflammatory changes.Compared with control group,the survival rates in different high-glucose groups at 6,12,24,and 48 h after treatment were increased(P<0.05).Compared with control group,after 3 h of treatment,the expression levels of IL-6 and IL-10 mRNA in the cells in 35.0 mmol·L-1 high-glucose group were increased(P<0.05 or P<0.01),while no significant changes were observed in IL-6,TNF-α,and IL-10 levels in the cell supernatants(P>0.05);after 6 h of treatment,the expression level of TNF-α mRNA in the cells in 35.0 mmol·L-1 high-glucose group was increased(P<0.001),and the levels of IL-6,TNF-α,and IL-10 in the cell supernatants were significantly increased(P<0.05 or P<0.001);after 3 h of treatment,the percentages of macrophage markers CD86+cells and CD163+cells in 35.0 mmolL-1 high-glucose group were significantly increased(P<0.01 or P<0.001).Conclusion:A certain high concentration of glucose may induce the polarization of Raw264.7 macrophages towards the M1 subtype in vitro.
