1.Transcriptome Sequencing and Bioinformatics Analysis of Herba Chlorophyti Laxi

Objective To study the transcriptional expression of Chlorophyti Laxi R.Br.using high-throughput sequencing technology.Methods Total RNA of Chlorophtum laxum R.Br.was extracted,followed by library construction,sequencing,and de novo assembly to obtain unigene sequences.These sequences were then annotated and compared to acquire genetic information of the Chlorophtum laxum R.Br.transcriptome.Results A total of 29 325 unigene were identified,with an average length of 1 026 bp and an N50 of 1 697 bp.Among them,20 338 unigene were functionally annotated in at least one database,with Chlorophtum laxum R.Br.showing the highest sequence similarity to Asparagus officinalis.In the Kyoto Encyclopedia of Genes and Genomes(KEGG)database,13 392 unigene were annotated,including genes involved in flavonoid biosynthesis and ubiquinone and other terpenoid-quinone biosynthesis pathways.Thirty-five upstream genes related to Chlorophtum laxum R.Br.saponin biosynthesis were identified.In the Gene Ontology(GO)database,16 728 unigene were annotated,covering cellular anatomical entities,binding,and biological processes.Using the Microsatellite Identification Tool(MISA),11 486 assembled unigene longer than 1 000 bp were analyzed for simple sequence repeats(SSRs),resulting in the identification of 5 178 SSR loci,for which primers were designed using Primer 3.0.By comparing Chlorophtum laxum R.Br.unigene with the transcription factor database,657 transcription factors,including bHLH,MYB,and WRKY,were identified.Conclusion The transcriptome data provide a foundation for studying the biosynthetic pathways and functional genes of medicinal active components in Chlorophtum laxum R.Br.,and also contribute to the conservation and development of its resources.
2.Clinical observation on the treatment of 50 cases of phlegm-dampness type insomnia with salt-partitioned moxibustion at Shenque acupoint combined with warm needling method at Sanyinjiao acupoint

Objective To explore the clinical application of Shenque acupoint salt moxibustion combined with Sanyinjiao acu -puncture and moxibustion in the treatment of phlegm-dampness type insomnia .Methods Ninety-five patients with phlegm-dampness type insomnia who sought medical care in the Department of Traditional Chinese Medicine from January 2001 to February 2017 were chosen as study subjects .The patients were divided into the control group ( n=45 ) and the treatment group ( n=50 ) in accordance with different treatment methods .The control group received Shenque acupoint salt moxibustion therapy , once a day, 30 minutes a ses-sion, while the treatment group was treated with Sanyinjiao acupuncture in addition to the same treatment as received by the control group, also once a day, 50 minutes a session.The 2 groups were treated 5 times a week, 1 week of treatment as a treatment course , for a total of 5 courses.After treatment, the Pittsburgh sleep quality index (PSQI) was used to evaluate treatment efficacy .Results Fol-lowing treatment, significant differences in sleep quality , sleep time, sleep disorder, daytime function and total PSQI scores could be seen, as compared with those before treatment for the same treatment group (P<0.05).After therapy, significant differences in sleep quality, sleep time, sleep disorder, daytime function and total PSQI scores in the PSQI evaluation scores could also be note in the con-trol group, as compared with those before treatment (P<0.05).Statistical significance in sleep quality , sleep time, daytime function and total PSQI scores in the PSQI evaluation scores could be noticed , when comparisons were made between the treatment group and the control group following treatment (P<0.05).Conclusion Shenque acupoint salt moxibustion combined with Sanyinjiao acupuncture and moxibustion in the treatment of phlegm-dampness type insomnia could effectively improve sleep quality of the patients , which was worth further clinical extension .
