Objective To establish the UPLC fingerprint of 10 batches of Gongliuning capsule and the content determination methods of 10 main components;To conduct network pharmacological research;To provide scientific basis for predicting the quality marker of Gongliuning capsule.Methods UPLC-DAD technique was used to establish the fingerprint of different batches of Gongliuning capsule samples and obtain the complete chemical composition information of Gongliuning capsule.With the help of network pharmacological research techniques,the target of chemical components of Gongliuning capsule and related pathways screened from various databases were predicted and analyzed,and the protein interaction network and composition-target-pathway diagram were constructed to predict the effective components of Gongliuning capsule.Based on the obtained pharmacodynamic substances,the content determination method of Gongliuning capsule was established to evaluate the quality of Gongliuning capsule.Results The fingerprints of Gongliuning capsule identified 17 common peaks.Combined with network pharmacology method,the candidate chemical components were analyzed,10 core targets and 42 metabolic pathways were obtained.Based on the testability of components and the availability of reference substances,10 compounds with high contribution were selected for content determination.Methodological verification results showed that all the 10 compounds had good linear relationship(r2>0.999 4)in the detection range,with good precision,accuracy and stability.Conclusion The established UPLC fingerprint,network pharmacology and multiple component content determination methods are highly specific,accurate and reliable.Combined with chemical pattern recognition,they can be effectively used for quality control and quality marker prediction of Gongliuning capsules.

Objective To explore the effect of multimodal analgesia combined with preemptive analgesia on perioperative pain control and sleep quality in elderly patients with intertrochanteric fracture.Methods A total of 100 elderly patients with femoral intertrochanteric factures were selected as treatment group,and 100 patients were selected as control group.The treatment group was treated with multimodal analgesia and preemptive analgesia,while the control group was treated with the conventional analgesia.The VAS score and the amount of tramadol at preoperative 4 8 h,2 4 h and 2 4 h,3 d,1 week,2 weeks after operation were compared between the two groups.The sleep quality of patients during hospitalization was evaluated by the PSQI score.The length of first time move for exercise and postoperative hospital stay were compared between two groups,and the postoperative Harris hip score at 2 weeks,3 months after treatment and adverse reactions were compared.Results The treatment group had a lower VAS score at preoperative 48 h,24 h and 24 h,3 d,1 week,2 weeks after operation when compared with control group (P ＜ 0.05).Tramadol dose in the control group was significantly higher than the treatment group (P ＜ 0.05),the PSQI score was significantly higher than the treatment group (P ＜ 0.05),the length of postoperative move for exercise was significantly longer than the treatment group (P ＜ 0.05).Harris hip score at 2 weeks after operation in the treatment group was significantly higher than that in control group (P ＜ 0.05),while 3 months after operation,the Harris hip score was similar between the two groups (P ＞ 0.05).The total incidence rate of adverse reactions were similar between the two groups (P ＞ 0.05).Conclusion Application of multimodal analgesia combined with preemptive analgesia is effective and safe in treatment of elderly patients with femoral intertrochanteric fracture,it can improve the sleep quality,promote the hip joint function recovery of patients in early stage.

Objective To set up the quality standard of Renqing Mangjue capsules. Methods TLC was used for qualitative identification of Crocus sativus, Zingiberaceae, Syringa oblata, Radix Aucklandiae and Borneolum. The content of Brucine and Strychnine were determined by HPLC, which conducted with CAPCELL PAK MG C18 column (4.6 mm×250 mm, 5μm). The mobile phase consisted of methanol∶0.01 moL/L potassium dihydrogen phosphate (with 10%phosphoric acid to adjust the pH 2.5)=25∶75 and the flow rate was 1.0 mL/min. The detection wavelength was set at 254 nm and 260 nm. The column temperature maintained at 30 ℃. Results TLC could detect qualitatively Crocus sativus, Zingiberaceae, Syringa oblata, Radix Aucklandiae and Borneolum. The spots were clear and the negative controls had no interference. HPLC determined that Brucine presented a good linear relationship in the range of 0.012 1-0.072 8 μg (r=0.999 1). The average recovery was 97.27%, RSD was 1.20%. Strychnine presented a good linear relationship in the range of 0.045 4-0.272 4 μg (r=0.999 8). The average recovery was 98.69%, RSD was 1.17%. Conclusion The method is simple in operation. The results are accurate, reliable and good in reproducibility. The method can effectively control the quality of Renqing Mangjue capsules.

【Objective】 To assay the naringenin content in different medicinal materials of Exocarpium Citri Grandis (ECG). 【Methods】 High performance liquid chromatography (HPLC) was applied to determine the naringenin content in different medicinal materials of ECG. The chromatographic conditions were: gradient elution being performed with methanol as mobile phase A and water - acetic acid (61: 4) as mobile phase B, flowing rate being 1.0 mL/min; detecting wavelength at 283 nm. 【Results】 In the rage of 0.088～0.880 ?g, naringenin showed a good linearity with the chromatographic peak area, r=0.9999, average recovery rate was 95.49% and RSD=2.00% (n=5). The naringenin content in 23 different samples of ECG was in the range of 0.1～1.0 mg?g~(-1). 【Conclusion】 It is the first time to establish the HPLC method for the determination of naringenin content in ECG and the method is accurate and reproducible. Naringenin content in varieties of ECG is stable and the difference of naringenin content between Citrus grandis Osbeck. Var. tomentosa Hort. and Citrus grandis (L.) Osbeck is not significant.

[Objective] To compare the difference of fingerprints of Pogostemon cablin (Blanco) Benth. from different places of origin. [Methods] The chemical components of Pogostemon cablin (Blanco) Benth. from 6 places of origin were detected by thin-layer chromatography. [ Results ] The thin-layer chromatogram of volatile components of Pogostemon cablin (Blanco) Benth. from different places of origin was similar. The contents of methanol-extract and petroleum-ether-extract from Pogostemon cablin (Blanco) Benth. planted in Gaoyao, Guangdong differed greatly from those in other places of origin, which can be used to identify the herb of Pogostemon cablin ( Blanco) Benth. from Gaoyao, Guangdong. [Conclusion] Thin-layer chromatographic fingerprints can be used as the parameter to differentiate Pogostemon cablin (Blanco) Benth. from different places of origin.

Objective To study the influence of different dosage forms on the release rate of total salvianolic acid of sus- tained-released Danshen extract preparations.Methods With salvianolic acid B as the standard,uhraviolet spec- trophotometry and the release test were applied to determine the accumulated release rate of total salvianolic acid of sus- tained-released Danshen extract preparations(R1～R9)of 9 different dosage forms and to evaluate the release behav- ior of sustained-released Danshen extract preparations in vitro.Results The accumulated release rate of total sal- vianolic acid in R7 was the best,and the in vitro release behavior was consistent with Higuchi equation:M_1/M_∞= 0.363 4t~(1/2)-0.1656,r= 0.996 2.Conclusion R7 is the first choice dosage form for the preparation of the sus- tained-released Danshen extraction preparations.

Objective To study the fingerprint determination method of Pogostemon cablin by HPLC.Methods A RP-HPLC method was established with the mixed solution of methnol and 0.1 % acetic acid aqueous solution as mobile phase(gradient elution),flow rate at 1.0 mL/min,detection wavelength at 254 nm,analytic time being 50 min and column temperature at 25 ℃.Results The results of comparative analysis of 10 batches samples showed that 15 common peaks were shared by Pogostemon cablin.The common peaks were classified to two regions and the second was the main characteristic region which had an amount of characteristic peaks.All samples had the similar fingerprint features.Conclusion The method is accurate,repetitive and can be applied for the identification and quality control of Pogostemon cablin.

Object To establish a new method to determine protocatechualdehyde in Maodongqing Sustained- release Tablet. Methods The RP- HPLC method was applied with CH3OH and 1 % acetic acid as mobile phase, flowrate at 1.0 mL/min, and detection wavelength at 312 nm. Results In the range from 11.7 ng to 140.4 ng, protocatechualdehyde was in a good line with peak area, r=0.99995, the average recovery ratio was 96.41 % , RSD=1.04 % (n=5). Conclusions It was the first time to establish the RP- HPLC method to determine and control the protocatechualdehyde in Maodongqing Sustained- release Tablet with accuracy and a good reproducibility.
