1.Cardiac-targeted liposomes alleviate myocardial ischemia-reperfusion injury by promoting inflammation resolution

Objective To explore the pro-inflammation resolution and protective effects of reactive oxygen species (ROS)-responsive liposomes modified with a cardiac-targeted peptide and loaded with resolvin D1 (RvD1, C-LP-RvD1) on myocardial ischemia-reperfusion (MI/R) injury. Methods The C-LP-RvD1 nanoliposomes were constructed, characterized physically and chemically, and evaluated for in vitro release. Non-targeting peptide-modified drug-loaded liposomes (LP-RvD1) were served as controls. Apoptotic adult mouse cardiomyocytes (AMCMs) were used to verify in vitro targeted binding capacity of C-LP-RvD1. In MI/R mice models, the in vivo distribution and cardiac enrichment of C-LP-RvD1 were assessed. Levels of specialized pro-resolving mediator (SPM) and inflammatory factors in cardiac tissue homogenates and cell culture supernatants were measured using enzyme-linked immunosorbent assay (ELISA). Cardiac function and fibrosis remodeling were evaluated via echocardiography and Masson staining four weeks after treatment. Biosafety was evaluated in healthy mice injected by C-LP-RvD1. Results The C-LP-RvD1 exhibited good nanoscale uniformity and stability, with ROS-triggered accelerated release characteristics. In vitro experiments showed that C-LP-RvD1 had higher binding capacity to apoptotic AMCMs than LP-RvD1, with significantly higher SPM levels (P<0.01) and lower inflammatory factor levels (P<0.05). In vivo experiments indicated enhanced cardiac enrichment of C-LP-RvD1 in MI/R injured hearts, with higher local myocardial SPM levels and lower inflammatory factor levels compared to LP-RvD1 (P<0.05). Four weeks after treatment, compared with LP-RvD1, the C-LP-RvD1 mice group showed improved cardiac function indicators and reduced ventricular fibrosis remodeling ratio (P<0.05). Safety evaluation revealed no significant systemic inflammation, immunogenicity, or coagulation abnormalities in healthy mice, with liver and kidney function and major organ histology showing no notable damage. Conclusions C-LP-RvD1 improves effective delivery of RvD1 to MI/R injured hearts through injury-targeted enrichment and ROS-responsive release, promoting inflammation resolution and suppressing excessive inflammation, thereby improving cardiac function and reducing adverse remodeling, with favorable biosafety.
2.Influence factors of comorbidity between gaming disorder and depression among adolescents

The comorbidity of online game disorder and depression has become a universal problem in the world,which seriously affects the physical and mental health of teenagers.Online game disorder closely relate to depression or depressive mood.There is a certain correlation and interaction between the two in the symptom dimension,leading to the decline of academic performance,and even non-suicidal self-injury,suicidal ideation and other problems.In terms of the neurobiological mechanism of the two comorbidity,there were abnormal functional connection of amygdaloid-dorsolateral prefrontal cortex network,abnormal activity pattern of default mode network and abnormal function of dopamine system in comorbidities.In terms of social psychological mechanism,comorbidities are characterized by loneliness,cognitive bias and poor coping strategies.Exploring the comorbidity mechanism of online game disorder and depression can help to formulate individualized comprehensive intervention measures to reduce the occurrence and development of online game disorder and depression in adolescents.
3.Retrospective clinical analysis of eculizumab treatment for hematopoietic stem cell transplantation-associated thrombotic microangiopathy: a report of 11 cases

Objective:To evaluate the efficacy of eculizumab in treating hematopoietic stem cell transplantation-associated thrombotic microangiopathy (TA-TMA) .Methods:This retrospective study included 11 patients who developed TA-TMA after allogeneic hematopoietic stem cell transplantation and subsequently received eculizumab treatment at Peking University People′s Hospital between June 2018 and May 2024. The incidence of TA-TMA, treatment details, and clinical outcomes were analyzed.Results:Among the 11 included patients [4 males, 7 females; median age: 29 years (range: 9-56) ], underlying diseases were severe aplastic anemia (SAA) in 5 patients, acute lymphoblastic leukemia (ALL) in 3 patients, and acute myeloid leukemia (AML) in 3 patients. The median time to TA-TMA diagnosis was 48 days post-transplantation (range: 4-213 days), and all patients met the diagnostic criteria for high-risk TA-TMA. The median interval from TA-TMA diagnosis to the initiation of eculizumab treatment was 12 days (range: 1-56 days). Patients received a median of 3 doses of eculizumab (range: 1-14). Ten of the 11 patients were assessed as having no response (NR) to eculizumab at the end of treatment or at death. One patient achieved a partial response (PR) but subsequently died after TA-TMA relapsed due to infection. At the last follow-up, all patients were either lost to follow-up or had died. The median follow-up duration was 88 days (range: 33-326 days), and the median time from TA-TMA diagnosis to the last follow-up was 31 days (range: 21-113 days) .Conclusion:Eculizumab demonstrated poor efficacy in this TA-TMA cohort. This might be attributable to the critical and complex condition of the patients, delayed initiation of eculizumab treatment, and insufficient dosage.
4.Diagnostic value of targeted next-generation sequencing for community-acquired respiratory virus infections in patients with hematological diseases

Objective:To evaluate the diagnostic value of targeted next-generation sequencing (tNGS) of throat swab samples for detecting community-acquired respiratory viruses (CARV) in patients with hematological diseases.Methods:Clinical and laboratory data from 64 episodes involving patients with hematological diseases and suspected infections—who underwent both pharyngeal swab tNGS and CARV polymerase chain reaction (PCR) testing concurrently—were retrospectively analyzed. The cases were drawn from the Department of Hematology, Peking University People’s Hospital, between September 2023 and April 2024. Concordance between tNGS and CARV PCR results, as well as the diagnostic performance of tNGS in detecting CARV, were evaluated.Results:Among the 64 episodes, 29 were clinically diagnosed with respiratory tract infections, including one case of cytomegalovirus pneumonia and 28 CARV-positive cases. The remaining 35 episodes involved patients with fever or respiratory symptoms attributed to other causes, including 14 with extrapulmonary infections and 21 with noninfectious etiologies. The median follow-up duration was 215.5 days (range: 7-271 days). PCR detected 24 strains of seven CARV types, whereas tNGS detected 25 strains of eight CARV types. Using PCR results as the reference standard, the sensitivity, specificity, positive predictive value, negative predictive value and accuracy of tNGS were 85.0%, 88.6%, 77.3%, 92.9%, and 87.5%, respectively. The two methods showed good concordance (Kappa=0.717, P<0.001) . Conclusion:Pharyngeal swab tNGS may serve as a viable alternative to PCR for diagnosing CARV infections in patients with hematological diseases.
5.Treatment of Esophageal Cancer Based on ZHANG Jiebin's "Water-Fire of Life Gate Theory"

Guided by ZHANG Jiebin's "water-fire of life gate theory", this paper explored the etiology, pathoge-nesis, and syndrome differentiation-based treatment approach for esophageal cancer. It is proposed that the core pathogenesis of esophageal cancer, derived from the imbalanced state of water and fire in life gate, can be summarized into three manifestations. Firstly, decline of life gate fire leads to failure of qi transformation and accumulation of phlegm and blood stasis; secondly, fire fails to warm the earth resulting in dysfunctional ascending and descending, and qi stagnation and blood stasis; thirdly, deficiency of life gate water contributs to proliferation of cancerous toxins and physical deterioration. In terms of treatment, guided by ZHANG's treatment principles for dysphagia, three treatment methods are proposed, warming and tonifying kidney yang to promote qi transformation through steaming action, warming and nourishing the spleen and stomach to restore central pivot movement, and nourishing kidney water to replenish essence and blood, then support bodily nourishment. Clinical case examples are provided to illustrate the application of these methods.
6.Associations between dietary habits and self-perceived cognitive decline

Objective:To investigate the self-perceived cognitive decline status in the community population,and to explore the association between different dietary habits and self-perceived cognitive decline.Methods:A cross-sectional study was carried out in 11 879 community residents in the three regions of Weifang,Jining,and Zoucheng in Shandong Province.The Ascertain Dementia-8 and dietary habits information questionnaire were used to assess self-perceived cognitive decline and dietary habits,and their association were analyzed using single factor and multivariate logistic regression.Results:The detection rate of self-perceived cognitive decline was 21.4%.Lo-gistic regression showed that smoking in the past was positively associated with self-perceived cognitive decline(OR=1.40,95%CI:1.14-1.73).However,intake of fruits(often,OR=0.70,95%CI:0.52-0.94;everyday,OR=0.60,95%CI:0.44-0.81),nuts(daily,OR=0.62,95%CI:0.44-0.88),mushrooms(often,OR=0.74,95%CI:0.57-0.92)and high tryptophan foods(sometimes,OR=0.79,95%CI:0.68-0.91;everyday,OR=0.54,95%CI:0.34-0.87)were negatively associated with self-perceived cognitive decline.Conclusion:Smoking history might be a risk factor for self-perceived cognitive decline,and high frequency intake of fruits,nuts,mush-rooms,and high tryptophan foods might protective factors for it.
7.Anxiety symptoms and associated factors among relocated elderly residents in new townships

Objective:To investigate anxiety symptoms and associated factors in relocated elderly residents of new townships,and to provide evidence for prevention interventions.Methods:A cross-sectional study was conduc-ted in relocated elderly residents in new townships of three urban areas in Shandong Province from 2021 to 2023.The study instruments included Ascertain Dementia-8,Generalized Anxiety Disorder-7,self-administered de-mographic characteristics information questionnaire.Multivariate analysis of factors associated with anxiety symp-toms in elderly residents was performed using multivariate logistic regression.Results:The prevalence rate of mild anxiety symptoms was 5.8%,and the rate of moderate-to-severe anxiety symptoms was 1.3%in 3 313 resi-dents.Multivariate analysis found that self-assessed general psychological condition(OR=0.52),good family envi-ronment(OR=0.34),no self-perceived cognitive impairment(OR=0.31),no chronic diseases(OR=0.42),and only one chronic disease(OR=0.61)were protective factors for mild anxiety symptoms,and very good dietary structure(OR=2.15)and fair dietary structure(OR=2.39)were risk factors for those.Very good family environ-ment(OR=0.11)and average family environment(OR=0.16),and no self-perceived cognitive impairment(OR=0.14)were protective factors for moderate-to-severe anxiety symptoms,and 0-3 years(OR=3.24)and 4-6 years(OR=3.28)of relocation were risk factors for those.Conclusion:Family environment,dietary structure,and duration since relocation are key factors associated with anxiety symptoms among relocated elderly residents in new townships.Targeted interventions should be implemented to address their mental health needs.
8.Interaction between perceived social support and TMEM161B gene rs768705 polymorphism and its association with risk of developing depression

Objective:To explore the association between perceived social support,TMEM161B gene rs768705 polymorphism,and their interaction with incidence of depressive symptoms in the freshmen.Methods:A total of 9928 freshmen from two medical universities were investigated at baseline and follow-up two years later during 2018-2020 by cluster sampling.A self-report demographic characteristics questionnaire was used to collect information of general demographic characteristics.Perceived Social Support Scale,Beck Depression Inventory,Beck Anxiety Inventory and Adolescent Self-Rating Life Events Check List were used to measure the level of perceived social support,depressive symptoms,anxiety symptoms and the number of negative life events of individuals.Blood sam-ples were collected and typed for DNA by professionals.Results:Family support and other support were all nega-tively associated with depressive symptoms in the freshmen(OR=0.96,95%CI:0.93-0.99;OR=0.94,95%CI:0.91-0.98).There was no correlation between friend support and depressive symptoms in the freshmen(OR=0.99,95%CI:0.95-1.02).TMEM161B gene rs768705 polymorphism(AG)was positively associated with de-pressive symptoms in the freshmen(OR=1.58,95%CI:1.12-2.23).The interactions of friend support with TMEM161B gene rs768705 polymorphism(AG)(OR=1.13,95%CI:1.02-1.26)and other support with TMEM161B gene rs768705 polymorphism(AG)(OR=1.13,95%CI:1.02-1.25)had significant effects on the incidence of depressive symptoms in the females and no significant effect in the males.Conclusion:The perceived social support,TMEM161B gene rs768705 polymorphism and their interaction are associated with influence of de-pressive symptoms,and have sex difference in the freshmen.
9.Impacts of traumatic experiences on panic attack,depression and anxiety symptoms among community residents

Objective:To investigate the impact of traumatic experiences on panic attack,depression and anxi-ety symptoms among community residents.Methods:Totally 11 280 community residents were recruited.The Com-posite International Diagnostic Interview-3.0,Patient Health Questionnaire,7-item Generalized Anxiety Disorder Scale and general information questionnaire were used for face-to-face interview to collect information.Propensity Score Matching was conducted to match 5 184 community residents for analysis.Logistic regression was employed to analyze the impact of traumatic events on panic attack,depressive symptoms,and anxiety symptoms among the study sample.Results:Traumatic events were associated with a higher likelihood of experiencing panic attack(OR=3.26,95%CI:2.26-4.70),depression symptoms(OR=1.92,95%CI:1.56-2.37)and anxiety symptoms(OR=1.57,95%CI:1.24-1.99).Conclusion:Traumatic events extensively impact the mental health of commu-nity residents.Timely psychological support and early intervention are essential to prevent mental health issues in community residents affected by trauma.
10.Effect of social activity participation on self-rated mental health disparity between urban and rural older adults in China

Objective:To explore the effect of different types of social activity participation on self-rated men-tal health disparity between urban and rural older adults in China.Methods:The study data was sourced from the 2018 wave of Chinese General Social Survey(CGSS).Factor analysis was applied to divide social activities into three dimensions:entertainment activities,cultural activities,family and friend gatherings.Logistic regression model was adopted to analyze the effect of social activity on self-rated mental health.Fairlie decomposition model was ap-plied to explore the effect of social activity on urban-rural disparity in self-rated mental health.Results:The self-ra-ted good mental health proportion of urban old persons(n=3 139)was 72.0%,which was higher than that of rural old persons(n=1 549)(53.7%)(P<0.001).Recreational activities and gatherings of friends and relatives sig-nificantly improved self-rated mental health in urban and rural old persons.Fairlie decomposition result revealed that approximately 18.7%of self-rated mental health disparity could be attributed to urban-rural disparity in entertain-ment activities.Moreover,urban-rural disparity in family and friends gatherings could explain about 3.9%of the disparity.Conclusion:Urban older adults have better mental health than their rural counterparts.Furthermore,social activity is beneficial to mental health for both urban and rural older adults.Urban-rural disparity in entertainment ac-tivities make an important contribution to mental health disparity between urban and rural older adults.
