1.Nonspecific peripheral T cell lymphoma combined with pure red cell aplasia: a case report and literature review

Nonspecific peripheral T-cell lymphoma (PTCL-NOS) combined with pure red cell aplasia (PRCA) is reported by only few in China. This article described a patient with PTCL-NOS who presented with severe anemia as the initial symptom, accompanied by multiple lymphadenopathy. Laboratory tests confirmed decreased red blood cell count and reticulocyte proportion, markedly reduced bone marrow red blood cell proliferation, and markedly increased proportion of bone marrow CD3 +CD8 + T lymphocytes. On lymph node pathology examination, immunohistochemistry was consistent with PTCL-NOS, with positive TCRβ gene rearrangement. First-line chemotherapy with four courses of ECHOP regimen led to significant reductions in lymph node size, partial remission on whole-body computed tomography evaluation, normalization of hemoglobin and bone marrow hematopoietic erythroid proliferation, and absence of CD3 +CD8 + T lymphocytes in bone marrow. Currently, the patient remains to have good prognosis with maintenance oral lenalidomide.
2.Expert Consensus on the Diagnosis and Treatment of Chronic Coronary Syndrome with Traditional Chinese and Western Medicine in Yunnan Province

Chronic coronary syndrome(CCS)is one of the most prevalent cardiovascular disease(CVD)in clinical practice.With the accelerating aging of the population and the increasing prevalence of cardiovascular risk factors,CVD has become the leading cause of death among Chinese residents.To optimize diagnosis and treatment strategies through integrated traditional Chinese and Western medicine approaches,and to further improve the clinical management of CCS,the expert group reviewed recent domestic and international guidelines on CCS diagnosis and treatment.Incorporating advances in traditional Chinese medicine(TCM)and leveraging the unique characteristics of Yunnan's local herbal medicine(Yunyao),this consensus document was developed.It aims to guide clinical practice and enhance the overall management of CCS patients in the province.
3.Identification of Mycobacterium abscessus subsp.abscessus and subsp.massiliense based on MALDI-TOF MS and analysis for their characteristics

Objective To perform the identification at the subspecies-level of Mycobacterium abscessus(M.abscessus)and analyze its characteristics based on matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS).Methods Bi-otyper software was used to construct the predicted peak spectrum of M.abscessus subsp.abscessus and M.abscessus subsp.massiliense.The predicted peak spectrum was constructed with expected maximum peak value number of 70 and peak frequency of 100%in the ex-perimental group and control group,respectively.A blind test was performed on 31 strains of M.abscessus that were not used to con-struct predictive peak spectra to evaluate the identification efficiency of predictive peak spectra.FlexAnalysis software was used to sum-marize and analyze the list of mass spectral peak value of M.abscessus,and screen the specific peaks in mass spectra of different sub-species of M.abscessus.The principal component analysis(PCA)algorithm was used to perform the cluster analysis for the data from mass spectrometry of M.abscessus,and explore the feasibility of PCA clustering in distinguishing the subspecies of M.abscessus.Results In the experimental group,96.8%(30/31)of the strains were correctly identified,and one strain of M.abscessus subsp.massiliense with rough colony form was mistakenly identified as M.abscessus subsp.abscessus.In control group,77.4%(24/31)of the strains were correctly identified,but 7 strains of M.abscessus subsp.massiliense were incorrectly identified or unable to be identified.The identification efficiency in the experimental group was significantly better than that in the control group with statistical difference(X2=5.167,P=0.026).M.abscessus subsp.abscessus exhibited three specific peaks(m/z 4 001.67,4 386.81 and 4 963.17),and M.abscessus subsp.massiliense also exhibited three specific peaks(m/z 4 950.48,4 381.78 and 5 214.90).In the PCA 3D scatter plot,the data points of M.abscessus subsp.abscessus and M.abscessus subsp.massiliense were relatively dispersed without obvious clus-tering.The PC A dendrograph could be divided into six branches in which only four branches were composed of a single subspecies.The minimum level value of distance between M.abscessus subsp.abscessus and M.abscessus subsp.massiliense was about 0.1.Conclusion The predicted peak spectrum based on MALDI-TOF MS with the expected maximum peak number of 70 could accurately identify M.abscessus at the subspecies level.The specific peak of mass spectrometry method in this study should be feasible to distinguish the subspecies of M.abscessus subsp.abscessus and the subspecies of M.abscessus subsp.Massiliense,but PCA cluster analysis cannot be used as a means to distinguish M.abscessus subsp.abscessus from M.abscessus subsp.massiliense.
4.Changes in serum HSP27 levels in patients with multiple myeloma and correlation analysis with diagnosis, treatment, and prognosis

Objective:To investigate the levels of serum heat shock protein 27 (HSP27) in patients with multiple myeloma (MM) and its relationship with disease diagnosis, treatment, and prognosis.Method:Retrospective cohort study. We select 85 newly diagnosed MM patients from the Second Affiliated Hospital of Fujian Medical University from December 1, 2021 to June 30, 2023, including 43 males and 42 females, aged 60 (48, 81) years. Their serum HSP27 levels were measured at the time of initial diagnosis, and 100 healthy individuals who underwent physical examinations during the same period were recruited as controls. The correlation between serum HSP27 levels in MM patients and common clinical indicators such as gender, age, hemoglobin (Hb), serum creatinine (Cr), calcium (Ca), lactate dehydrogenase (LDH), β2-microglobulin levels (β2-MG), and bone marrow plasma cell ratio were analyzed; grouping 85 MM patients according to disease types and stages, and the differences in HSP27 levels before and after treatment in different MM classifications, stages, and treatments were compared; using receiver operating characteristic (ROC) curves to obtain the predictive value of initial HSP27 levels for remission in MM patients after standardized treatment, and calculating the optimal critical value. MM patients were divided into a high HSP27 group of 37 cases and a low HSP27 group of 48 cases based on the cutoff value, and Kaplan-Meier was used to compare the survival differences between the two groups.Result:The serum HSP27 levels in newly diagnosed MM patients were higher than those in healthy individuals [(281.33±27.69) pg/ml, (52.11±8.12) pg/ml, t=73.7, P0.001]. There was no correlation between HSP27 levels and patients' MM classification, gender, age, Hb serum Cr, Ca, LDH, and the proportion of bone marrow plasma cells, but positively correlated with β2-MG levels ( r=0.703, P0.05). There are significant differences in HSP27 levels at different stages of MM and before and after treatment; ROC curve results showed that serum HSP27 levels have certain predictive value for whether MM patients can achieve remission after treatment: sensitivity of 75.0%, specificity of 63.8%, and area under the curve of 0.720; the optimal cut-off value is 281.19 pg/ml. The survival time of the low HSP27 level group is longer than that of the high level group. Conclusion:Serum HSP27 is highly expressed in MM, and is correlated with the disease condition, treatment, and prognosis of MM.

8.Incidence of abdominal obesity among primary and middle school students and its association with parental weight related knowledge, attitude and practice

Objective:
To investigate the association of parental weight related knowledge, attitude and practice (KAP) with abdominal obesity among primary and middle school students and to provide evidence for related intervention measures.
Methods:
In September 2013, the multistage random sampling method was used to select 17 307 primary and secondary school students with normal waist circumference in seven provinces/municipalities in China. After nine month follow up, 14 118 students aged 6-18 years were included in the final analysis. Questionnaires were used to assess parental weight related KAP, and students’ waist circumference was measured objectively at baseline and follow up. Multiple linear regression and multivariate Logistic regression were conducted to analyze the relationship between parents’ KAP with the change of students waist circumference and abdominal obesity, respectively.
Results:
The nine month cumulative incidence of pre and abdominal obesity among students was 6.67% and 0.85%, respectively. The scoring rates of parents knowledge, attitude and practice were 80.17%, 78.17%, 50.50%, respectively. The score of parents weight related attitude was inversely associated with the changes of waist circumference ( β=-0.18, 95%CI = -0.26- -0.10) and cumulative incidence of pre and abdominal obesity ( OR=0.90, 95%CI =0.82-0.98)( P <0.05).
Conclusion
Parental positive and correct weight related attitude is inversely associated with the incidence of abdominal obesity among children and adolescents. Interventions targeting establishing parents correct beliefs may be an effective way to prevent abdominal obesity in children and adolescents.
9.Microneedles in diagnosis and treatment: a review.

Microneedles have been developed rapidly in the field of transdermal administration in the past few decades. In recent years, the development of microelectronics technology has expanded the applications of microneedles by combining with microelectronic systems, especially in biological diagnosis and treatment. Different types of microneedles have been designed to extract blood and tissue fluids for detection, or as electrodes to directly detect blood sugar, melanoma and pH in real-time in vivo, both show good prospects for real-time detection applications. In this paper, we review the design of materials and structure of microelectronic-based microneedles, and discuss their advances in biological diagnosis.

10. Research progress of monoclonal antibodies in pharmacokinetic characteristics, analysis methods and method validation

In recent years, monoclonal antibodies (mAbs) have been developing rapidly and widely used in fields of tumor, immunity, blood and other systemic diseases. Global share of mAbs in prescription drug market had reached up to $ 140 billion, 15.3% by 2019. As macromolecule proteins, with special structures and physiological properties, mAbs have great differences in absorption, distribution, metabolism and excretion in vivo compared with small molecule drugs, including characteristics of relatively large molecular mass, high specificity and selectivity in target combination, non-linear pharmacokinetics, time dependence, long half-life and so on. Fully understanding of these special pharmacokinetic characteristics shall effectively guide analysis of mAbs. Meanwhile, the particularity and complexity of disposal mechanism in organisms greatly increase the difficulty of biological detection. As a result, it is necessary to establish exclusive, sensitive, accurate and repeatable quantification methods in biological samples. This article focuses on pharmacokinetic characteristics, main analytical methods, and methodology validation of quantification for mAbs in biological samples, expounding with comparisons of small molecular drugs, so as to prompt development of pharmacokinetic study of this kind of drugs.
