1.Discovery and investigation of six polio vaccine derived viruses in Guangzhou City

Objective To understand the surveillance situation of poliovirus in Guangzhou from 2011 to 2024, and to further strengthen polio surveillance and ensure the continued maintenance of a polio-free status. Methods An analysis was conducted on the discovery and investigation results of six cases of vaccine-derived poliovirus (VDPV) detected in Guangzhou. Results A total of 6 VDPV incidents were reported in Guangzhou from 2011 to June 2024, among which 5 incidents were from sewage sample testing in the Liede Sewage Treatment Plant in Guangzhou, all of which were confirmed as VDPV, with 1 for type I, 1 for type II, and 3 for type III. In addition, one confirmed HFMD case was identified as a type VDPV II carrier. No presence of any wild poliovirus (WPV), VDPV cases, or circulating VDPV (cVDPV) was reported. Conclusion Guangzhou City has maintained a high level of vigilance and effectiveness in the monitoring and prevention of polio. Continuously strengthening the construction of the polio monitoring network, optimizing vaccination strategies, and comprehensively improving public health awareness are still the focus of the prevention and control work in the future.
2.Association Between the Protein Expressions of MutS Homologs and Villin and the Clinicopathological Characteristics in 310 Colon Cancer Patients

Objective To examine the relationship between the expressions of mismatch repair proteins,MutS homolog 2(MSH2)and MutS homolog 6(MSH6),and villin and the pathological features in patients with colon cancer.Methods A total of 310 cases of colon cancer patients who were treated at our hospital between January 2017 and September 2021 were selected.The diagnosis of colon cancer of all patients was verified by pathological evaluation.Immunohistochemistry was used to determine the protein expressions of MSH2,MSH6,and villin.The correlation between the expressions of MSH2,MSH6,and villin and the clinicopathological parameters in patients with colon cancer was analyzed accordingly.Multivariate logistic regression was used to analyze the correlation between the expressions of MSH2,MSH6,and villin and the clinicopathological parameters of colon cancer.Kaplan-Meier survival curve was used to compare the 2-year survival rates of colon cancer patients with different expression levels of the proteins.Results Among the 310 patients with colon cancer,the negative expression rates of MSH2,MSH6,and villin proteins in cancer tissues were 8.71%(27/310),9.35%(29/310),and 46.13%(143/310),respectively.The negative expression rates of the three proteins in tissues adjacent to cancer were 3.23%(10/310),4.19%(13/310),and 9.68%(30/310),respectively.The negative expression rates of the three proteins in cancer tissues were all higher than those in adjacent tissues(P<0.05).Regression analysis showed that the expression of MSH2 and MSH6 in cancer tissues was correlated with the age,the location of tumor lesions,tumor differentiation degree,and lymph node metastasis in colon cancer patients(P<0.05).The expression of villin in the cancer tissue is correlated with the depth of tumor infiltration,lymph node metastasis,distant metastasis,and clinical staging status in colon cancer patients(P<0.05).The 2-year survival rates of patients with negative expressions of MSH2 and MSH6 were 51.85%and 44.83%,respectively,which were lower than those of patients with positive expression of MSH2 and MSH6(79.51%and 80.43%,P<0.05).Thirteen patients(4.1%)had negative expression of MSH2,MSH6,and villin(referred to as"triple negative expressions")in the cancer tissues,and their 2-year survival rate was 30.77%,which was lower than that of colon cancer patients who did not meet the criteria for triple negative expressions(79.12%[235/297],P<0.05).Conclusion The expressions of MSH2,MSH6,and villin are closely correlated with the pathological features of colon cancer patients.Evaluating the expression of the three proteins may assist in the clinical diagnosis,treatment,and prognosis evaluation of colon cancer.
3.Influence of occupational stress on mental health of medical staff: mediating effect of affective commitment and moderating effect of overcommitment

Background Occupational stress has been shown to be an important factor affecting the mental health of workers. The role of affective commitment to the organization and overcommitment to work cannot be ignored. However, there is a lack of research on this topic in China. Objective To explore a potential mediating effect of affective commitment on how occupational stress affects the mental health of medical staff and a potential moderating effect of overcommitment on the mediating effect of affective commitment. Methods A total of 1372 health care workers in a tertiary Grade A hospital in Lanzhou City were selected as study subjects for a cross-sectional survey. The occupational stress, emotional commitment, and psychological distress of the subjects were evaluated by the Effort-Reward Imbalance Scale, Affective Commitment Scale, and Kessler 10 Scale. SPSS 26.0 was used for correlation analysis, mediation analysis, and moderated mediation analysis. Common method bias wasevaluated by Harman one-factor test. Results A total of 1372 questionnaires were distributed, of which 1277 valid questionnaires were returned, with a valid recovery rate of 93.08%. The mean occupational stress score was 1.14±0.23, the mean overcommitment score was 20.26±3.21, the mean affective commitment score was 20.25±3.34, and the mean psychological distress score was 26.26±7.90. The Spearman correlation analysis results showed that occupational stress among medical staff was positively correlated with overcommitment and psychological distress (r=0.153, 0.410, P<0.01) and negatively correlated with affective commitment (r=−0.341, P<0.01); overcommitment was negatively related to affective commitment and positively related to psychological distress (r=−0.107, 0.312, P<0.01); affective commitment was negatively related to psychological distress (r=−0.464, P<0.01). The positive effect of occupational stress on psychological distress of medical staff was significant (b=0.41, t=15.42, P<0.001); affective commitment presented a partial mediating effect on the relationship between occupational stress and psychological distress (effect value=0.13), accounting for 31.71% of the total effect; overcommitment moderated the process of occupational stress-affective commitment-psychological distress (P<0.01). Conclusion Affective commitment of medical staff has a partial mediating effect on the relationship between occupational stress and psychological distress, and overcommitment plays a significant role in moderating the process of occupational stress-affective commitment-psychological distress.

5.Relationship and prognostic study between E-cadherin gene expression and lymphatic hyperplastic reaction in gastric cancer

Objective To study the relationship and prognosis between E-cadherin gene expression and lymphatic hyperplastic reaction in gastric cancer. Methods The degree of lymphocytosis and draining lymph node from 86 cases of gastric cancer were observed and the expression of E-cadherin gene in gastric cancer were detected by SP method of immunohistochemistry (IHC). Results Lymphocyte infiltration degree around gastric cancers was positively related to the reactive hyperplasia of the lymphnodes and was inversely related to lymphatic metastasis. The expression of E-cadherin has relationship with the infiltration degree of stomach carcinoma. To compare with T1/T2 and T3/T4 phases, the result is significantly different (P <0.01).Meanwhile, the expression of E-cadherin was positively correlated with tumor-infiltrated lymphocytes and reactive hyperplasia of the lymph nodes, was negatively correlated with lymphatic metastasis in drainage area.Conclusion The over-expression of E-cadherin gene is significantly related to lymphoproliferation and lymph node metastasis. The abnormal expression of E-cadherin can be used as an index to determine prognosis of gastric carcinoma.
6.Clinical significance of the level of CD4+CD25+ regulatory T cells and C-reactive protein in patients with acute exacerbation of chronic obstructive pulmonary disease

Objective To study the changes and significance of CD4+CD25+ regulatory T cells and C-reactive protein(CRP) in patients with acute exacerbation of chronic obstructive pulmonary disease(COPD). Methods Flow cytometry was used to detect the frequency of CD4+CD25+ regulatory T cells in peripheral blood from 36 patients with acute exacerbation of COPD( COPD group) and 36 patients with clinical stability of COPD(control group one)and 36 normal individuals(control group two). The level of CRP was detected routinely. Results The ratio of CD4+CD25+ regulatory T cells number in peripheral blood of COPD group to the total number of CD4+T cell was (2.56±1.83 )%, and it was significantly decreased compared to the other two groups (P all<0.01 ). The level of CRP in COPD group was markedly higher than that in the other two groups (P all<0.01 ). The level of CD4+CD25+ regulatory T cells in patients with acute exacerbation of COPD had negative relation with CRP. Conclusions CD4+CD25+ regulatory T cells participate in inflammatory response. The proportion of CD4+CD25+ regulatory T cells decreases in patients with acute exacerbation of COPD, and it may result in maladjustment of cytoimmunity.
