Objective To investigate the effects of open reading frame 1 protein(ORF1p),encoded by long interspersed nuclear element-1(LINE-1),on the proliferation,migration,and invasion of esophageal squamous cell carcinoma(ESCC)cells,and explore the underlying molecular mechanism.Methods① Western blotting was performed to compare the expression of ORF1p between normal esophageal squamous epithelial cells and ESCC cells.② Immunohistochemistry(IHC)assay was used to examine ORF1p expression in ESCC tissues and paired normal tissues adjacent to tumor.③ The effects of ORF1p knockdown and overexpression on malignant behaviors in ESCC cells were determined through functional assays.④ Xenograft tumor model in nude mice was established to evaluate the impact of ORF1p on tumor growth in vivo.⑤ Transcriptome sequencing combined with cell functional rescue experiments were conducted to identify downstream targets regulated by ORF1p.Results ① Western blot analysis demonstrated the expression of ORF1p was significantly higher in the ESCC cell lines than the normal esophageal squamous epithelial cells(P<0.05).② IHC confirmed remarkable up-regulation of ORF1p in ESCC tissues than paired adjacent normal tissues(P<0.000 1).③ Functional assays and experiments on xenograft tumor models revealed that ORF1p substantially enhanced the proliferation,migration,and invasion of ESCC cells,as well as tumorigenic potential in vivo(P<0.05).④ Functional rescue experiments showed that ORF1p facilitated the proliferation,migration,and invasion of ESCC cells by modulating AJUBA expression(P<0.05).Conclusion ORF1p is significantly up-regulated in ESCC and promotes the proliferation,migration,and invasion of ESCC cells by regulating AJUBA expression.

Objective To develop a machine learning model integrating preoperative chest CT radiomic features with clinical data for predicting 5-year postoperative recurrence risk in stage Ⅰ non-small cell lung cancer(NSCLC)patients undergoing surgical resection.Methods A total of 217 patients with pathologically confirmed stage Ⅰ NSCLC(selected from 778 initially screened cases based on our inclusion and exclusion criteria)treated in Army Medical Center of PLA between January 2014 and December 2019 were retrospectively enrolled,including 53 recurrence cases and 164 non-recurrence cases within 5-year follow-up.They were randomly divided into a training set(n=173)and a validation set(n=44)in a ratio of 8:2.Radiomic models were established based on extracted features from tumor-dominant regions of interest(ROI)on CT images,while clinical models were developed using demographic characteristics and preoperative laboratory examinations.A combined model was further constructed by integrating both feature sets,and model performance was compared to identify the optimal predictive model.Results This study screened the features from non-contrast CT images and ultimately selected 7 radiomic features for constructing radiomic model.Among 6 machine learning algorithms,the adaptive boosting(Adaboost)model demonstrated the best overall predictive performance,with an area under the curve(AUC)of 0.866(95%CI:0.808～0.923;accuracy:0.832,specificity:0.884)in the training set and of 0.806(95%CI:0.630～0.983;accuracy:0.795,specificity:0.971)in the validation set.Univariate and multivariate logistic regression analyses identified 4 clinical features for clinical model construction.The clinical model achieved an AUC value of 0.874(95%CI:0.821～0.928;accuracy:0.827,specificity:0.891)in the training set and 0.813(95%CI:0.677～0.948;accuracy:0.636,specificity:0.600)in the validation set.By integrating the 7 radiomic features and 4 clinical features using a feature-level fusion strategy,the combined model exhibited further improved predictive performance,with an AUC value of 0.953(95%CI:0.924～0.983;accuracy:0.884,specificity:0.860)and 0.852(95%CI:0.729～0.976;accuracy:0.682,specificity:0.629),respectively in the training set and the validation set.Conclusion The combined model integrating preoperative CT radiomic features with clinical risk factors may provide an evidence-based framework for evaluating 5-year postoperative recurrence risk in stage Ⅰ NSCLC patients.

Objective To evaluate the current status of compliance with animal experiment reporting guidelines in medical research papers published in Chinese and to enhance the transparency of medical research reporting.Methods Using a predefined literature search strategy,we conducted searches in the China National Knowledge Infrastructure(CNKI)database for literature published in 2019 and 2022 in journals indexed in A Guide to the Core Journals of China published by Peking University Library.We focused on 22 pieces of key information required in the ARRIVE Guidelines 2.0.These pieces of information concerned 6 items of the ARRIVE Essential 10 Items,including study design,sample size,randomization,statistical methods,and experimental animals,and 3 items of the ARRIVE Recommended Set,including abstract,ethical statement,and declaration of interests.We conducted statistical analysis of the reporting rates of the key information in the research publications included in the study.Results A total of 4818 research papers were included in the analysis,and none comprehensively reported all 22 pieces of information investigated in the study.Most of the research papers published in 2019 and 2022 reported information on the control groups,with the reporting rate for the respective years being 99.8％(2461/2467)and 99.7％(2343/2351).Although the sample size reporting rates were 79.2％(1954/2467)for research papers published in 2019 and 77.2％(1815/2351)for those published in 2022,none described the method and rationale of sample size calculation.The reporting rates of randomisation and blinding methods were approximately 20％and 1％,respectively.The reporting of statistical methods increased slightly from 91.8％in 2019 to 96.0％in 2022.The reporting of information on the experimental animals showed mixed trends for 2019 and 2022,including the provenance of animals(93.8％vs.93.7％,P＞0.05),strains and substrains(99.1％vs.99.2％,P=0.514),sex(94.1％vs.92.7％,P=0.044),age(58.1％vs.70.6％,P＜0.001),weight(84.4％vs.81.9％,P=0.020),and health status(66.0％vs.75.5％,P＜0.001).Research papers published in both 2019 and 2022 had relatively low rates of reporting animal ethics review(15.8％vs.38.9％)and animal ethics principles adhered to(9.8％vs.21.3％),declaration of interests(2.3％vs.10.6％),and accurate summaries of animal-related information in the abstracts(9.16％vs.8.13％).In particular,the reporting rate of the animal ethics statement and the declaration of interests increased significantly high in 2022 compared to that in 2019(P＜0.001).Conclusions Since ARRIVE Guidelines 2.0 was released,transparency in the reporting of most of the ARRIVE checklist items of interest in this study has improved significantly.However,the reporting of randomization,blinding,ethical statement,and declaration of interests still need further improvement and should be prioritized for future efforts.

Objective To evaluate the inhibitory effects of prophylactic administration of α-zearalanol(α-ZAL)on bone microarchitecture and bone resorption activity in ovariectomized osteoporotic rats,and to investigate its regulatory effects on the osteogenic and adipogenic differentiation of bone marrow mesenchymal stem cells(BMSCs).Methods A total of 606-month-old unmated female Sprague-Dawley(SD)rats weighing(300±20)g were randomly divided into the sham surgery group(Sham group),ovariectomy group(OVX group),solvent group(Oil group),estradiol benzoate treatment group(Post-E2 group),α-ZAL prevention group(Pre-ZAL group),and α-ZAL treatment group(Post-ZAL group),with 10 rats in each group.An osteoporosis rat model was established using the ovariectomy method.Rats in the Sham group underwent the same surgical procedures except for ovarian removal.Seventy-two hours after ovarian removal,the Oil group received intramuscular injections of 0.5 mL of oil solvent,and the Pre-ZAL group received intramuscular injections of α-ZAL(1.5 mg·kg-1),administered every 3 days for 120 consecutive days.The Post-E2 group and Post-ZAL group began intramuscular injections of estradiol benzoate(1.5 mg·kg-1)and α-ZAL(1.5 mg·kg-1),respectively,90 days after ovariectomy,administered every 3 days for 120 consecutive days.After drug administration,bone density and bone tissue microstructure morphology were analyzed using a micro-CT small animal in vivo imaging system and staining methods.Osteoclasts were isolated and their activity was detected.Femoral BMSCs were obtained to assess their osteoblast and adipocyte differentiation capabilities,and uterine tissue morphological changes were observed via histological sections.Results Compared with the OVX group,BMD in the Sham group,Post-E2 group,Pre-ZAL group,and Post-ZAL group increased by 133.12%,75.97%,69.64%,and 24.69%,respectively(all P<0.01).BMD in the Pre-ZAL group was 36.09%higher than in the Post-ZAL group(P<0.01),and there was no significant difference in BMD between the Post-E2 and Pre-ZAL groups(P>0.05).Tb.N in the Sham group,Post-E2 group,Pre-ZAL group,and Post-ZAL group increased by 160.08%,118.14%,94.76%,and 46.76%,respectively,compared with the OVX group(all P<0.01).Tb.Ar increased by 324.21%,203.83%,177.99%,and 82.71%,respectively(all P<0.01).Tb.N in the Pre-ZAL group increased by 32.71%compared to the Post-ZAL group(P<0.05),while Tb.Ar increased by 52.15%(P<0.01).Tb.Sp in the Sham,Post-E2,and Pre-ZAL groups decreased by 58.53%,42.18%,and 35.61%,respectively,compared with the OVX group(all P<0.01).The MAR of the upper tibial cancellous bone in the Sham,Post-E2,and Pre-ZAL groups increased by 257.81%,156.72%,and 142.63%,respectively,compared with the OVX group(all P<0.01),BFR increased by 192.19%,137.23%,and 88.13%,respectively(all P<0.01).MAR and BFR in the Pre-ZAL group increased by 58.10%and 43.63%,respectively,compared with the Post-ZAL group(both P<0.01).There were no significant differences in MAR and BFR between the Post-E2 group and the Pre-ZAL group(P>0.05).MMP-9,TRAP,and CK mRNA expression was significantly downregulated in both the Post-E2 group and the Pre-ZAL group(P<0.01).The osteoblast differentiation capacity of BMSCs in the Post-E2 group and all Post-ZAL groups was enhanced,with a significant increase in the number of mineralized nodules,and the expression levels of OCN,COL1,and OPN mRNA were significantly increased(P<0.01),while the ability to differentiate into adipocytes was weakened.The number of intracellular lipid droplets in BMSCs was significantly reduced,the lipid droplet volume was smaller,and the expression levels of PPAR-γ2 and aP2 mRNA were decreased(P<0.05).There were no significant differences between the Post-E2 group and the Pre-ZAL group(P>0.05).There was no significant increase in body weight in the Post-E2,Pre-ZAL,and Post-ZAL groups,but uterine weight significantly increased in the Post-E2 group(P<0.05),with marked uterine epithelial hyperplasia.Uterine weight in the Pre-ZAL and Post-ZAL groups showed no significant difference compared to the OVX group(P>0.05),and no significant changes were observed in uterine epithelium.Conclusion α-ZAL can effectively protect bone mass,improve bone microstructure,and reduce estrogen-related uterine adverse reactions by regulating the osteogenic/adipogenic differentiation balance of BMSCs,providing a potential new therapeutic strategy for the prevention and treatment of postmenopausal osteoporosis.

Visual impairment refers to the loss of visual acuity or reduction in visual field in both eyes due to various reasons.Patients with visual impairment experience fall fear,thereby causing ac-tivity limitation.This study reviewed the definition of fall fear,summarized the assessment tools for fall fear,and discussed the current research status and nursing progress regarding fall fear in patients with visual impairment,aiming to provide references for clinical nursing practice and research.

Alzheimer's disease(AD)is a multifactorial degenerative disorder of the central nervous system that mainly manifests as cognitive dysfunction and loss of speech.In recent years,the zebrafish has attracted extensive attention because of its high homology with humans in terms of brain structure and function,nerve conduction,and pathogenic genes of AD.This article reviews the advantages of the zebrafish as an animal model of AD,covering topics in the pathogenesis of AD and the evaluation and screening of drugs for treatment of AD.The overall goal is to provide new insights into the pathogenesis of AD and development of novel drugs.

Depression is a psychosomatic disorder.The rising incidence rate of depression in recent years is placing a heavy economic burden on societies around the world.Morinda officinalis oligosaccharides(MOOs)are active substances extracted from the Chinese herb Morinda officinalis that can soothe depression and calm the mind,tonify the kidneys,and benefit the intellect,as well as improve cognitive disorders in patients to a certain extent.On the basis of the hypothesised pathological mechanism of depression,this study explains the link between MOOs and depression by reviewing existing studies.We propose that MOOs can improve depression through mechanisms that regulate the levels of monoamine neurotransmitters,enhance neuroplasticity,regulate the function of the HPA axis and levels of cytokines,and influence gut microbiota.This paper provides new ideas for research on the antidepressant effects of MOOs.

Objective This study aims to develop a medical patch surface material featuring a microporous polyurethane(PU)membrane and to assess the material's properties and biological performance.The goal is to enhance the clinical applicability of pelvic floor repair patch materials.Methods PU films with a microporous surface were prepared using PU prepolymer foaming technology.The films were produced by optimizing the PU prepolymer isocyanate index(R value)and the relative humidity(RH)of the foaming environment.The surface morphology of the PU microporous films was observed by scanning electron microscopy,and the chemical properties of the PU microporous films,including hydrophilicity,were analyzed using infrared spectroscopy,Raman spectroscopy,and water contact angle measurements.In vitro evaluations included testing the effects of PU microporous film extracts on the proliferation of L929 mouse fibroblasts and observing the adhesion and morphology of these fibroblasts.Additionally,the effect of the PU microporous films on RAW264.7 mouse macrophages was studied.Immune response and tissue regeneration were assessed in vivo using Sprague Dawley(SD)rats.Results The PU films exhibited a well-defined and uniform microporous structure when the R value of PU prepolymer=1.5 and the foaming environment RH=70%.The chemical structure of the PU microporous films was not significantly altered compared to the PU films,with a significantly lower water contact angle([55.7±1.5]°)compared to PU films([69.5±1.7]°)and polypropylene(PP)([104.3±2.5]°),indicating superior hydrophilicity.The extracts from PU microporous films demonstrated good in vitro biocompatibility,promoting the proliferation of L929 mouse fibroblasts.The surface morphology of the PU microporous films facilitated fibroblast adhesion and spreading.The films also inhibited the secretion of tumor necrosis factor-α(TNF-α)and interleukin(IL)-1β by RAW264.7 macrophages while enhancing IL-10 and IL-4 secretion.Compared to 24 hours,after 72 hours of culture,the expression levels of TNF-α and IL-1β were reduced in both the PU film and PU microporous film groups and were significantly lower than those in the PP film group(P＜0.05),with the most notable decreases observed in the PU microporous film group.IL-10 and IL-4 levels increased significantly in the PU microporous film group,surpassing those in the PP film group(P＜0.01),with the most pronounced increase in IL-4.The PU microporous film induced mild inflammation with no significant fibrous capsule formation in vivo.After 60 days of implantation,the film partially degraded,showing extensive collagen fiber growth and muscle formation in its central region.Conclusion The PU microporous film exhibits good hydrophilicity and biocompatibility.Its surface morphology enhances cell adhesion,regulates the function of RAW264.7 macrophages,and promotes tissue repair,offering new insights for the design of pelvic floor repair and reconstruction patch materials.

Objective:To explore the application and practice of "flipped classroom" in the teaching of general surgery interns.Methods:A total of 20 internship groups (3 to 5 people in each group) were randomly selected from the general surgery practice group in the Department of General Surgery of the Second Clinical Medical College of North Sichuan Medical College. They were randomly divided into the flipped group (45 people) and the traditional group (40 people), with 10 subgroups in each group. The flipped group adopted the flipped classroom teaching mode (students' self-study by handing out materials before class, students and teachers' discussion in class, and students and teachers' evaluation after class), while the control group adopted the current conventional teaching mode (students' preview before class, teachers' explanation in class, and teachers' question answering after class). At the end of the teaching, a questionnaire was used to evaluate the participation and completion of each student. The teaching effect was evaluated by medical history collection and case analysis. The participation, completion, and teaching effect between the two groups were compared and analyzed. SPSS 23.0 software was used for t-test and Chi-square test. Results:The participation of the flipped group was better than that of the traditional group [(17.45±1.83) vs. (15.57±1.52)], and the difference was statistically significant ( P < 0.05). There was no statistically significant difference between the flipped group and the traditional group. There was no significant difference in medical history collection scores between the two groups. The case analysis of the flipped group was better than that of the traditional group [(87.30±6.06) vs. (81.50±5.88), P < 0.05]. The questionnaire shows that about 90% of the students think that flipped classroom can improve their interest in learning [96% (43/45)], improve their autonomous learning ability [89% (40/45)], and have better learning effect. At the same time, 78% (35/45) of students think that learning time is too long. Conclusion:The flipped classroom teaching model can improve the teaching participation of general surgery students, improve students' interest in learning, improve their self-learning ability, and improve students' thinking ability of medical record analysis.

Sepsis is characterized by a severe and life-threatening host immune response to polymicrobial infection accompanied by organ dysfunction.Studies on the therapeutic effect and mechanism of immunomod-ulatory drugs on the sepsis-induced hyperinflammatory or immunosuppression states of various im-mune cells remain limited.This study aimed to investigate the protective effects and underlying mechanism of artesunate(ART)on the splenic microenvironment of cecal ligation and puncture-induced sepsis model mice using single-cell RNA sequencing(scRNA-seq)and experimental validations.The scRNA-seq analysis revealed that ART inhibited the activation of pro-inflammatory macrophages recruited during sepsis.ART could restore neutrophils'chemotaxis and immune function in the septic spleen.It inhibited the activation of T regulatory cells but promoted the cytotoxic function of natural killer cells during sepsis.ART also promoted the differentiation and activity of splenic B cells in mice with sepsis.These results indicated that ART could alleviate the inflammatory and/or immunosuppressive states of various immune cells involved in sepsis to balance the immune homeostasis within the host.Overall,this study provided a comprehensive investigation of the regulatory effect of ART on the splenic microenvironment in sepsis,thus contributing to the application of ART as adjunctive therapy for the clinical treatment of sepsis.
