Objective: To analyze the impact of premature death due to four major chronic diseases on life expectancy in Yangpu District, Shanghai Municipality from 2010 to 2021, so as to provide the evidence for formulating chronic disease prevention and control strategies. Methods : Mortality data of registered residents in Yangpu District from 2010 to 2021 were collected through the Death Information Registration and Management System of the Shanghai Municipal Disease Control and Prevention Information Management Platform. The premature death probability of malignant tumors, diabetes, cardiovascular and cerebrovascular diseases, and chronic respiratory diseases, and life expectancy of residents were calculated using the abridged life table method. Trends in premature death probability for four types of chronic diseases were analyzed using the average annual percent change (AAPC). The impact of premature death probability due to four chronic diseases on life expectancy was assessed by Arriaga's decomposition method. Results : The premature death probability due to four major chronic diseases in Yangpu District decreased from 9.88% in 2010 to 9.22% in 2021, showing an overall declining trend (AAPC=-0.540%, P<0.05). Among females, the premature death probability declined from 6.71% to 4.90% (AAPC=-2.715%, P<0.05), whereas no statistically significant trend was observed in males (P>0.05). Life expectancy increased from 82.52 years in 2010 to 84.50 years in 2021, with an overall upward trend (AAPC=0.244%, P<0.05). Life expectancy rose by 1.71 years for males and 2.34 years for females (AAPC=0.197% and 0.303%,both P<0.05). Declines in premature death probability from malignant tumors (AAPC=-0.967%, P< 0.05) and chronic respiratory diseases (AAPC=-3.071%, P<0.05) contributed to gains in life expectancy of 0.30 years and 0.03 years, with contribution rates of 12.18% and 1.29%, respectively. Changes in premature death probability due to diabetes as well as cardiovascular and cerebrovascular diseases were not statistically significant (both P>0.05), resulting in reductions in life expectancy of 0.05 years and 0.10 years, with contribution rates of -2.40% and -5.05%, respectively. Notably, an increase in premature death probability due to cardiovascular and cerebrovascular diseases among males (AAPC=1.673%) contributed to a decrease of 0.22 years in male life expectancy, whereas a decrease among females (AAPC=-3.824%) contributed to an increase of 0.03 years in female life expectancy, with contribution rates of -13.03% and 1.14%, respectively. Conclusions From 2010 to 2021, Yangpu District experienced an overall decline in premature death probability due to four major chronic diseases and an increase in life expectancy. Greater attention should be paid to the negative impacts of premature death probability from diabetes as well as cardiovascular and cerebrovascular diseases among males on life expectancy.

Objective:To observe the therapeutic efficacy of point catgut-embedding therapy combined with cupping therapy in treating simple obesity due to stomach heat and dampness obstruction,as well as its effects on Chinese medicine symptoms,obesity indicators,and blood lipid parameters.Methods:A total of 135 cases with simple obesity due to stomach heat and dampness obstruction were allocated into an embedding group,a cupping group,and a combined group using the random number table method,with 45 cases in each group.The embedding group received point catgut-embedding therapy alone,the cupping group was treated with cupping therapy alone,while the combined group underwent both embedding therapy and cupping therapy.The same points were used in the three groups.The treatment was performed once daily,and consecutive 28-day treatment was taken as 1 treatment course.The therapeutic efficacy was evaluated,and the changes in the traditional Chinese medicine(TCM)symptom score,obesity indicators[body mass(BM),body mass index(BMI),body fat percentage(BFP),waist circumference,and waist-to-hip ratio],and blood lipid parameters[triglyceride(TG)and total cholesterol(TC)]were observed after 3 consecutive courses.Results:After 3-course treatment,the total effective rate in the combined group was 93.3%,higher than 82.2%in the embedding group and 73.3%in the cupping group(P<0.05),and the total effective rate of the embedding group was higher than that of the cupping group(P<0.05).Compared to those before treatment,the TCM symptom score,obesity indicators,and blood lipid parameters reduced significantly in the three groups(P<0.05 or P<0.01),and indicators in the combined group were lower than those in the other two groups(P<0.05).Conclusion:The synergistic application of point catgut-embedding therapy and cupping therapy can improve TCM symptoms,obesity-related indicators,and blood lipid parameters of patients with simple obesity due to stomach heat and dampness obstruction;compared to the use of either point catgut-embedding therapy or cupping therapy alone,the combined application of the two therapies shows a more significant therapeutic effect.

AIM To investigate the effects of Xuesaitong Capsules(Panax notoginseng saponins)on ischemia/reperfusion injury in a mouse model of skin frostbite.METHODS The mice were randomly divided into the control group,the model group,the dexamethasone group(1 mg/kg),and the low-dose,medium-dose,and high-dose Xuesaitong Capsules groups(0.036,0.072,and 0.144 g/kg),with eight mice in each group.A frostbite model was established using a dry ice-cooled ceramic(ferrite)magnet.On the 2nd day after modeling,each group started its corresponding dosing by gavage for 14 consecutive days.The wound healing,histopathological changes,and serum levels of high-sensitivity C-reactive protein(hs-CRP),thromboxane B2(TXB2),6-keto-prostaglandin F1α(6-K-PGF1α),nitric oxide(NO)and endothelin(ET)were assessed using ELISA.The superoxide dismutase(SOD)activity and malondialdehyde(MDA)levels in skin tissues were measured biochemically.The protein expressions of tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,Toll-like receptor(TLR)4 and phosphorylated nuclear factor-KB p65(p-NF-κB p65)in skin tissues were determined by Western blot.Additionally,LncRNA H19 mRNA expression in skin tissues was evaluated using RT-qPCR.RESULTS After the final administration,compared with the control group,the model group exhibited partial scab detachment,wound healing,and larger wound areas;hyperkeratosis with incomplete keratinization,detachment of the dermis and subcutaneous tissue,partial loss of appendages,subcutaneous edema,and dilated,congested,and hemorrhagic stromal vessels with extensive lymphocyte infiltration revealed by the histopathological examination;elevated serum levels of hs-CRP,TXB2,and ET(P＜0.05,P＜0.01);decreased 6-K-PGF1α and NO levels(P＜0.05,P＜0.01);reduced SOD activity in skin tissues(P＜0.01);increased MDA levels(P＜0.01);and upregulated protein expressions of TNF-α,IL-1β,IL-6,TLR4 and p-NF-κB p65,as well as LncRNA H19 mRNA expression(P＜0.05,P＜0.01).Compared with the model group,the group intervened with high-dose Xuesaitong Capsules displayed reduced wound areas(P＜0.01);decreased serum levels of hs-CRP,TXB2 and ET(P＜0.05,P＜0.01);increased 6-K-PGF1α and NO levels(P＜0.05,P＜0.01);enhanced SOD activity(P＜0.05,P＜0.01);reduced MDA level in skin tissues(P＜0.05,P＜0.01);and down-regulated TNF-α,IL-1β,IL-6,TLR4 and p-NF-κB p65 protein expressions and suppressed LncRNA H19 mRNA expression in skin tissues as well(P＜0.05,P＜0.01).CONCLUSION Xuesaitong Capsules alleviate ischemia/reperfusion injury in frostbite-injured mice by exerting anti-inflammatory and anti oxidative stress effects and restoring vascular endothelial function mediated by the downregulation of LncRNA H19 expression and inhibition of the TLR4/NF-κB signaling pathway.

Aim To investigate the protective effect of Gualou Guizhi granules(GLGZG)on neuronal injury induced by LPS-activated microglia based on Notch signaling pathway.Methods LPS-activated microglia were co-cultured with neurons to construct neuron inju-ry models,and the cells were divided into the control group,model group,Notch inhibitor(DAPT)group,GLGZG(50,100,200 mg·L-1)group,DAPT+100 mg·L-1GLGZG group.After intervention,the activity of HT22 cells was detected by CCK-8 method,and rel-ative mRNA expression was detected by real-time PCR.The relative protein expression was detected by Western blot.Results Compared with the model group,after GLGZG intervention,the cell activity was significantly improved,GLGZG decreased IL-6,IL-12,Bax,Notch 1,caspase-3,Delta-1,NICD,RBPSUH,HES1 expression,and increased Bcl-2 expression(P＜0.05).Compared with the model group,the NICD,RBPSUH and HES1 mRNA and protein expressions significantly decreased after DAPT treatment(P＜0.05),and there was no superposition effect with GLG-ZG.Conclusion GLGZG may play a neuroprotective role by inhibiting inflammatory factors and apoptosis,and inhibiting Notch signaling pathway.

The pathogenesis of central precocious puberty(CPP)is due to the early initiation of the function of the hypothalamic-pituitary-gonadal axis,which is driven by the gonadotrophin-releasing hor-mone(GnRH)from the hypothalamus.The increase of GnRH secretion will promote the secretion of gon-adotropin by the pituitary gland,which leads to the development of reproductive organs and the secretion of sex hormones.The timing of puberty onset is influenced by the interaction between nutritional,envi-ronmental,and socioeconomic factors.In the study of pathogenic genes,loss-of-function mutations of MKRN3,LIN28 and DLK1 genes can lead to early onset of puberty,among which loss-of-function muta-tions of MKRN3 are one of the most common single-gene causes of familial CPP.Since 2013,when the MKRN3 mutation was first identified in five CPP families,the role of MKRN3 in adolescence has gradu-ally been discovered.MKRN3 is a member of the E3 ubiquitin ligase family and is expressed in the cen-tral nervous system of many eukaryotes.MKRN3 may act as an E3 ubiquitin ligase to inhibit GnRH activ-ity.Recently,more and more studies have explored the molecular mechanism of MKRN3 in CPP,revea-ling a variety of genes and proteins that interact with MKRN3.For example,MKRN3 can inhibit KISS1 and TAC3 activities,thereby affecting the expression of kisspeptin and neurokinin B to regulate GnRH se-cretion.MKRN3 also inhibited GNRH1 mRNA translation through PABPC1 ubiquitination.In addition,there are other targets such as MBD3,IGF2BP1 and NPTX1,all of which are involved in GnRH regula-tion downstream of MKRN3.Upstream of MKRN3,miR-30 can bind to three sites in the 3'-untranslated region of the MKRN3 gene to block MKRN3 transcription.Here we review the role of MKRN3 in the path-ogenesis of CPP and the research progress of its molecular mechanism,which will help to better under-stand the pathogenesis of CPP,provide new ideas and treatment methods for CPP-related fields in the fu-ture,and lay a theoretical foundation for further exploration of the molecular mechanism of MKRN3.

Objective:To evaluate the role of tumor necrosis factor-α-induced protein 8-like molecule-2 (TIPE2) in the endogenous protective mechanism against acute lung injury in septic mice and the relationship with ferroptosis.Methods:Twenty SPF healthy wild-type male C57BL/6N mice and 20 TIPE2 gene knockout C57BL/6N mice, aged 6-8 weeks, weighing 20-25 g, were assigned to wild-type sham operation group (WT-Sham group), wild-type sepsis group (WT-SEP group), TIPE2 knockout sham operation group (KO-Sham group), and TIPE2 knockout sepsis group (KO-SEP group) using a random number table method, with 10 mice in each group. Acute lung injury was induced by cecal ligation and perforation in anesthetized mice. The animals were sacrificed after anesthesia at 24 h after operation and lung tissues were obtained for examination of the morphological results of lung tissues (with a light microscope) and for determination of wet to dry lung weight (W/D) ratio, contents of ferrous ions (Fe 2+ ), glutathione (GSH) and malondialdehyde (MDA) and expression of solute carrier family 7 member 11 (SLC7A11), glutathione peroxidase 4 (GPX4) and Acyl-CoA synthetase long-chain family member 4 (ACSL4) (by Western blot). Results:Compared with WT-Sham group, the lung injury score, W/D ratio and contents of Fe 2+ and MDA were significantly increased, the content of GSH was decreased, the expression of GPX4 and SLC7A11 was down-regulated, and the expression of ACSL4 was up-regulated in WT-SEP group ( P<0.05). Compared with WT-SEP group, the lung injury score, W/D ratio and contents of Fe 2+ and MDA were significantly increased, the content of GSH was decreased, the expression of GPX4 and SLC7A11 was down-regulated, and the expression of ACSL4 was up-regulated in KO-SEP group ( P<0.05). Conclusions:TIPE2 is involved in the endogenous protective mechanism against acute lung injury, which may be related to the inhibition of ferroptosis in lung tissues of septic mice.
