Aim To study whether intravenous injec-tion of miR-129-3p mimetic(agomir)can resist bone loss caused by hind limb disuse, and to provide new i-deas for preventing bone loss in microgravity environ-ment.Methods Forty-eight C57BL/6J male mice were randomly divided into the control group(CON),tail suspension model group(TS),tail suspension+miR-129-3p agomir administration group(miRNA)and tail suspension+miR-129-3p negative sequence agomir control group(NC).The miRNA group was given 4 mg·kg-1 miR-129-3p agomir by intravenous injection into the medial canthus twice a week. The NC agomir group were consistent with those in the miR-129-3p agomir group, and the CON and TS groups were given only equal volumes of normal saline. After four weeks, all mice were sacrificed and samples were collected.Micro-CT scan of femur,three-point femur bending test, serum bone metabolism index detection, oxidative stress index detection and osteogenesis-related protein expression analysis in bone tissue were per-formed.Results After four weeks, the number of tra-becular bone in the TS group was significantly re-duced,and Tb.BMD,Tb.Th,Tb.N,Tb.BS/TV and Tb.BV/TV were significantly lower than those in the CON group(P<0.01).While Tb.Sp TS group was significantly higher than the CON group(P<0.05),the maximum load and flexural strength of the femur significantly decreased(P<0.01),the content of ser-um bone formation index PINP was significantly lower than that of the CON group(P<0.01),and the con-tent of bone resorption index CTX-I was significantly higher than that of the CON group(P<0.01),the content of serum oxidative damage indexes 8-iso-PGF2α and 8-OHdG significantly increased(P<0.01),and the expression of osteogenesis-related pro-teins in bone tissue markedly decreased(P<0.01).However,the increase or decrease of all indexes in miRNA group was significantly lower than that in TS group. Conclusions miR-129-3p mimetic can signifi-cantly reduce bone loss caused by hind limb disuse. This experiment provides a new idea and method for preventing bone loss in microgravity environment.

The development of artificial intelligence(AI)technology has provided new avenues for the public to access health information. While both domestic and international research and practical applications of health conversational AI have emerged, there remains a lack of consensus on the key issues surrounding their development.Based on a comprehensive review of domestic and international literature and multiple rounds of expert consultations, the study focuses on six issues:the definition, characteristics, application scenarios, evaluation elements, evaluation methods,as well as prospects and challenges of AI-driven health consultation systems, and develops the practice guideline for health conversational artificial intelligence. The findings aim to provide scientific reference for promoting the innovative application of AI technology in the healthcare field.

Objective To investigate the expression of arginase Ⅱ(ArgⅡ)in kidney tissue of rats with diabetic nephropathy(DN)and its significance in the development of DN.Methods A total of 10 male SD rats were randomly divided into the control group and the model group, with 5 rats in each group. An rat model of DN was developed by feeding with high-sugar and high-fat diet combined with intra-peritoneal injection of low-dose streptozotocin(45 mg/kg),and they were sacrificed after 11 weeks of continued feeding. The body weight, and biochemical indexes of blood and urine of rats were determined. The right kidney was weighed and histopathological examination was performed. The pathological changes of kidney tissues and protein expression of ArgⅡ and CD68+were observed, and the immunofluores-cence double staining was used to observe the distribution and expression of ArgⅡand a marker of renal macrophage activation CD68+;the protein expression of ArgⅡ,NF-κB,TNF-α and IL-6 in kidney tissues was determined by Western blot. Results Compared with the control group, the ratio of kidney weight to body weight,24-hour urine volume,24-hour urine protein, fasting blood glucose, urea nitrogen and insulin level in the model group were significantly increased(P<0.05).The renal histopathology showed that the mesangial cells of the renal glomerular were necrotic with vascular dilatation, and the renal tubular epithelial cells were steatosis and congestion.Compared with the control group, the protein expression of ArgⅡ,CD68+,NF-κB,TNF-α and IL-6 in the kidney tissues of the model group were significantly increased(P<0.05).Immunofluorescence double staining demonstrated the co-expression of ArgⅡ and CD68+in renal tissue, and the fluorescence intensities of both ArgⅡ and CD68+in the model group were significantly stronger than those in the control group(P<0.01).Conclusion The expression of ArgⅡ is increased in DN,which may be participated in the occurrence of inflammatory lesions in DN.

The incidence rate of kidney diseases in China has always remained high.At present, the clinical treat-ment mainly focuses on symptomatic treatment to delay the progression of the disease, and there is a lack of eco-nomical and effective treatment methods.MicroRNA plays an important regulatory role in the occurrence and devel-opment of diseases. This study aims to explore the role and regulatory mechanism of miR-142a-3p in adriamycin(ADR)-induced renal tubular epithelial cell(TCMK-1)injury,with a focus on its potential as a therapeutic target for ADR nephropathy.First, cell viability was assessed using the CCK-8 kit,and a mouse renal tubular epithelial cell model induced by ADR was established.Subsequently, alterations in miR-142a-3p and its target gene ATG16L1 mRNA levels were quantified using RT-qPCR.Western blotting was used to detect the protein levels of autophagy marker proteins and pyroptosis marker proteins.Monodansylcadaverin(MDC)staining was performed and the autophagy of cells was detected by flow cytometry. The results showed that the relative expression of miR-142a-3p in TCMK-1 cells induced by ADR was increased and the relative expression of its target gene ATG16L1 was decreased(P<0.0001).Western blotting results showed that the levels of p62(P<0.001)and pyroptosis-related proteins(P<0.001)were increased, while the protein levels of autophagy-related proteins were decreased(P<0.05).The flow cytometry results showed that there was no difference in the mean fluorescence intensity of autoph-agosomes between the ADR group and the autophagosome inhibitor group(3-MA group)(P>0.05),indicating that after ADR induction, cell autophagy was inhibited and pyroptosis was enhanced. When the expression of miR-142a-3p was inhibited by transfecting miR-142a-3p inhibitor, the relative expression level of the target gene ATG16L1 was restored(P<0.001).Western blotting showed that the protein level of p62(P<0.01)and pyropto-sis-related proteins(P<0.01)were decreased, and the protein level of autophagy-related proteins was restored(P<0.001).Flow cytometry results further indicated that cell autophagy was restored(P<0.0001).In conclusion,ADR targets A TG1 6L1 through miR-142a-3p to reduce the autophagy level of TCMK-1,and simultaneously activates GSDMD-mediated pyroptosis.

The cribriform fascia is formed by the fusion of deep and superficial fascias in the axilla, and it forms the base of the axilla together with the subcutaneous tissue and skin, which is a key part of the system of the membranous anatomy of breast surgery. The loose anatomical structure of the cribriform fascia makes it an important anatomical landmark in the axillary region, which plays an important role in accurate localization of lymph nodes. The sentinel lymph node biopsy through cribriform fascia has less intraoperative blood loss and shorter catheterization time compared with the traditional axillary fold approach. However, the anatomical structure of the cribriform fascia and its significance in the membranous anatomy of breast surgery have not been fully elucidated. This article analyzes the anatomical structure of the cribriform fascia and its adjacent tissues, and emphasizes the anatomical structure that needs to be paid attention to in sentinel lymph node biopsy through the axillary cribriform fascia,so as to provide a reference for clinicians to fully understand the cribriform fascia structure.
