2.High-efficient discovering the potent anti-Notum agents from herbal medicines for combating glucocorticoid-induced osteoporosis.

Notum, a negative feedback regulator of the Wnt signaling, has emerged as a promising target for treating glucocorticoid-induced osteoporosis (GIOP). This study showcases an efficient strategy for discovering the anti-Notum constituents from herbal medicines (HMs) as novel anti-GIOP agents. Firstly, a rapid-responding near-infrared fluorogenic substrate for Notum was rationally engineered for high-throughput identifying the anti-Notum HMs. The results showed that Bu-Gu-Zhi (BGZ), a known anti-osteoporosis herb, potently inhibited Notum in a competitive-inhibition manner. To uncover the key anti-Notum constituents in BGZ, an efficient strategy was adapted via integrating biochemical, phytochemical, computational, and pharmacological assays. Among all identified BGZ constituents, three furanocoumarins were validated as strong Notum inhibitors, while 5-methoxypsoralen (5-MP) showed the most potent anti-Notum activity and favorable safety profiles. Mechanistically, 5-MP acted as a competitive inhibitor of Notum via creating strong hydrophobic interactions with Trp128 and Phe268 in the catalytic cavity of Notum. Cellular assays showed that 5-MP remarkably promoted osteoblast differentiation and activated Wnt signaling in dexamethasone (DXMS)-challenged MC3T3-E1 osteoblasts. In dexamethasone-induced osteoporotic mice, 5-MP strongly elevated bone mineral density (BMD) and improved cancellous and cortical bone thickness. Collectively, this study constructs a high-efficient platform for discovering key anti-Notum constituents from HMs, while 5-MP emerges as a promising anti-GIOP agent.

Osteosarcoma (OS), chondrosarcoma (CS), and Ewing sarcoma (ES) represent primary malignant bone tumors and pose significant challenges in oncology research and clinical management. Conventional research methods, such as two-dimensional (2D) cultured tumor cells and animal models, have limitations in recapitulating the complex tumor microenvironment (TME) and often fail to translate into effective clinical treatments. The advancement of three-dimensional (3D) culture technology has revolutionized the field by enabling the development of in vitro constructed bone tumor models that closely mimic the in vivo TME. These models provide powerful tools for investigating tumor biology, assessing therapeutic responses, and advancing personalized medicine. This comprehensive review summarizes the recent advancements in research on 3D tumor models constructed in vitro for OS, CS, and ES. We discuss the various techniques employed in model construction, their applications, and the challenges and future directions in this field. The integration of advanced technologies and the incorporation of additional cell types hold promise for the development of more sophisticated and physiologically relevant models. As research in this field continues to evolve, we anticipate that these models will play an increasingly crucial role in unraveling the complexities of malignant bone tumors and accelerating the development of novel therapeutic strategies.

4.Establishment of an in vitro model for Erastin and RSL3-induced ferroptosis in hepatocellular carcinoma

Objective:
To establish anin vitromodel of ferroptosis induced by Erastin and RAS-selective lethal 3(RSL3) in hepatoma cells, and to provide theoretical basis for the development of novel therapeutic strategies for HCC.
Methods:
Hepatoma cells(HCCLM3, HepG2, Hep3B, Huh7 and PLC/PRF/5) in logarithmic growth phase were treated with Erastin(0-40 μmol/L) and RSL3(0-10 μmol/L) at double concentrations respectively. After 24 h, CCK-8 method was used to detect cell viability, draw growth curve, calculate IC50, and HCC cells sensitive to inducers were selected for follow-up experiments. The effect of inducer on the state of hepatoma cells was observed under light microscope, and immunoblotting and flow cytometry were used to verify whether the ferroptotic modelin vitrowas successfully constructed.
Results:
Huh7, Hep3B and HepG2 cells were sensitive to Erastin and RSL3, but HCCLM3 and PLC/PRF/5 were insensitive to Erastin and RSL3. When the concentration of Erastin and RSL3 reached the maximum, the survival rate was still above 65%. Huh7, Hep3B and HepG2 cells were selected for subsequent experiments. Compared with the control group, the expression of Glutathione peroxidase 4(GPX4), a ferroptotic marker, was down-regulated in a concentration-dependent manner. In Huh7, Hep3B and HepG2 cells, lipid reactive oxygen species(ROS) levels significantly increased after 24 h treatment with 10 μmol/L and 20 μmol/L Erastin, respectively; in Huh7 cells, lipid ROS levels significantly increased after 24 h treatment with 0.5 μmol/L and 1 μmol/L RSL3, respectively; in Hep3B and HepG2 cells, lipid ROS levels significantly increased after 24 h treatment with 1 μmol/L and 2 μmol/L RSL3, respectively, compared with control group.
Conclusion
Huh7, Hep3B and HepG2 cells are highly sensitive to Erastin and RSL3. Huh7, Hep3B and HepG2 cells treated with 10 μmol/L Erastin for 24 h are good models for simulating ferroptosis induced by Erastinin vitro, Huh7 cells treated with 0.5 μmol/L RSL3 for 24 h and Hep3B and HepG2 cells treated with 1 μmol/L RSL3 for 24 h are good models for simulating ferroptosis induced by RSL3in vitro.
5.Research on the mechanism by which the Y chromosome gene KDM5D inhibits the malignant progression of male bladder cancer

Objective:
To investigate the expression of the Y chromosome gene lysine-specific demethylase 5D(KDM5D) in male bladder cancer and its effects on cancer progression, thereby explaining gender-related prognostic differences in bladder cancer.
Methods:
Bioinformatics analysis was conducted to identifyKDM5Das a differentially expressed Y chromosome gene with prognostic significance in bladder cancer based on data from the TCGA database. The expression ofKDM5Din bladder cancer cell lines, collected clinical bladder cancer tissues, and adjacent normal epithelial tissues were examined using quantitative Real-time PCR(qPCR), Western blot, immunofluorescence, tumor microenvironment analysis, and bioinformatics pathway analysis. The effects ofKDM5Don bladder cancer cell proliferation and migration were validated through cell experiments followingKDM5Dknockdown and overexpression. Additionally, clinical data onKDM5Dexpression in bladder cancer tissue arrays were statistically analyzed in relation to metastasis and recurrence.
Results:
KDM5Dwas generally downregulated in male bladder cancer but exhibited variable expression in different bladder cancer cell lines and between recurrent and non-recurrent bladder cancers. Overexpression ofKDM5Dinhibited tumor cell proliferation and migration, and lowerKDM5Dexpression was associated with higher recurrence and metastasis rates. Bioinformatics analysis suggested thatKDM5Dmight influence tumor metastasis or recurrence by affecting immune cell aggregation and epithelial-mesenchymal transition.
Conclusion
KDM5Dis a protective gene specific to male bladder cancer and may be associated with the generally better prognosis observed in male compared to female bladder cancer patients.
6.Rapid Analysis of the Chemical Composition of Zhangbang Nine Steaming Nine Sun-dying Rehmanniae Radix Based on UPLC-Q-TOF-MS Technique and Study on the Function of Solid Excipiemts in Alleviating the "Nourishing and Spleen-impairing" in Rehmanniae Radix

OBJECTIVE
To study the chemical composition and the function of solid excipients in relieving the "nourishing and spleen-impairing" effect of Zhangbang nine steaming nine sun-dying Rehmanniae Radix(RR) by using UPLC-Q-TOF-MS technique.
METHODS
UPLC-Q-TOF-MS was used to collect the data of Zhangbang nine steaming nine sun-dying RR, and UNIFI platform and self-built database were used for the rapid qualitative analysis and identification of the chemical components of Zhangbang nine steaming nine sun-dying RR. The effect of solid excipients on the function of RR "nourishing and spleen-impairing" was investigated by establishing a mouse model of low gastric motility and measuring its gastric residual rate and small intestine propulsion rate.
RESULTS
The UPLC-Q-TOF-MS and UNFI platforms identified 76 chemical components in nine steaming nine sun-dying RR, including 9 iridoid glycosides, 11 phenylethyl glycosides, 31 flavonoids, 6 saccharides and 19 other compounds; the results of gastric emptying and small intestinal propulsion experiments on mice showed that Zhangbang nine steaming nine sun-dying RR had gastric emptying and intestinal propulsion effects.
CONCLUSION
This method can be used for the rapid identification and analysis of nine steaming nine sun-dying RR, which has solid excipients Citri Reticulatae Pericarpium and Amomi Fructus to alleviate the "nourishing and spleen-impairing" function of RR. This study can provide a reference for quality control and elucidation of the material basis of the medicinal effects of specialty beverage of Zhangbang nine steaming nine sun-dying RR.
7.Risk assessment and prediction model construction of pregnant women with IVF-ET and GDM

Objective To explore the adverse pregnancy outcomes of gestational diabetes mellitus (GDM) patients with different conception methods,and to construct a risk prediction model.Methods A ret-rospective cohort study was carried out to analyze the clinical data of 1453 cases of GDM patients who re-ceived regular prenatal care and delivery in the obstetrics department of the First People's Hospital of Yunnan Province from January 2018 to August 2022.A total of 436 patients with GDM who underwent in vitro fertili-zation-embryo transfer (IVF-ET) were served as the observation group,1017 GDM patients with natural ges-tation during the same period were selected as the control group.Chi-square test and t-test were used to com-pare the basic data,weight change,blood glucose reaching standard rate,maternal and infant pregnancy out-comes and related complications between the two group.The risk factors causing adverse pregnant outcomes were analyzed after controlling the confounding variables by adopting multivariate logistic regression.The pre-diction model was constructed according to the regression coefficient weight of each risk factor.The receiver operating characteristic (ROC) curve and the area under the curve (AUC) were used to evaluate the predictive efficiency of each independent risk factor and combined predictive factors.Results IVF-ET was an independ-ent risk factor for the first cesarean section (OR=1.809,95%CI:1.339-2.442),preterm birth (OR=1.622,95%CI:1.090-2.413),postpartum hemorrhage (OR=2.377,95%CI:1.406-4.018),adherent placenta (OR=1.971,95%CI:1.211-3.209),low birth weight infant (OR=2.232,95%CI:1.354-3.680) in the pa-tients with GDM (P<0.05).A combined prediction model was established by parity,age,fetus number,ges-tational weight gain,IVF-ET and scar uterus.The combined prediction probability was P=eLogit(P)/(1+eLogit(P)),Logit(P)=β1X1+β2X2+…+βnXn.The combined prediction efficacy of this study model for pre-term birth,cesarean section and low birth weight infants was good,AUC was approximately 0.8,which was greater than AUC of each independent risk factor.Conclusion The combined model of parity,age,fetus num-ber,gestational weight gain,IVF-ET and scar uterus has a certain predictive value for the risk of cesarean sec-tion,preterm birth and low birth weight infant in the patients with GDM.
8.Effect of Orem self-care mode on disease management ability and quality of life in patients with diabetes

Objective To explore the effect of Orem self-care mode on disease management ability and quality of life of diabetic patients.Methods A total of 130 patients with diabetes were divided into the control group and the observation group by random number table method,65 cases in each group.The control group underwent routine nursing intervention,and the observation group were treated with Orem self-care mode.The intervention effects of the two groups were compared.Results After the intervention,the knowledge of diabetes management,the self-care ability and the improvement of quality of life in the observation group were better than those in the control group(P<0.05).Conclusion The application of Orem self-care mode can significantly improve patients'cognition of the disease and self-management ability,which is of great significance to improve patients'condition and improve their quality of life.
9.Current status and influencing factors of rumination in inpatients with metastatic prostate cancer based on the disease coping mechanism

Objective:To explore the current status of rumination in inpatients with metastatic prostate cancer (mPCa) under the disease coping mechanism, and analyzing its influencing factors.Methods:From March 2021 to December 2022, 267 mPCa inpatients at the Tenth People's Hospital of Tongji University were selected as the study subject using convenience sampling. The survey was conducted using the General Information Questionnaire, Chinese Version of Event Relate Rumination Inventory (C-ERI), Medical Coping Modes Questionnaire (MCMQ), International Prostate Symptom Score (IPSS), and Social Support Revalued Scale (SSRS), and the factors influencing the rumination in patients were analyzed.Results:This study collected 260 valid questionnaires, with an effective response rate of 97.4% (260/267). The total score of rumination in 260 mPCa patients was (31.08±6.46), with invasive rumination and purposeful rumination scores of (14.13±4.44) and (16.95±4.55), respectively. The initial prostate-specific antigen index, confrontation medical coping modes were the influencing factors of invasive rumination in mPCa patients ( P<0.05). The initial prostate-specific antigen index, course of disease, social support, and confrontation medical coping style were the influencing factors of purposeful rumination in mPCa patients ( P<0.05) . Conclusions:The rumination of mPCa patients is above the medium level, and is influenced by multiple factors under the disease coping mechanism. Clinical medical and nursing staff should combine the clinical situation and characteristics of mPCa patients, provide personalized interventions as early as possible, and assist patients in improving their positive cognitive processing abilities.
10.Intervention effect of kushenol F on ulcerative colitis mice

OBJECTIVE To investigate the intervention effect of kushenol F (KSC-F) on ulcerative colitis (UC) mice. METHODS Totally 30 male C57BL/6J mice were randomly divided into the normal group, model group, positive drug group (sulfasalazine, 703 mg/kg), KSC-F 50 mg/kg group (KSC-F50 group), and KSC-F 100 mg/kg group (KSC-F100 group), with 6 mice in each group. Except for the normal group, the mice in the remaining groups were given 3% dextran sulfate sodium solution continuously for 7 days to induce UC model. Concurrently, administration groups received corresponding drug solution intragastrically, once a day, for 10 consecutive days. During the experiment, the changes in body weight and bowel movements of the mice were observed. Disease activity index scoring was performed after the last administration. The histopathological morphology of colonic tissue was examined. The levels of inflammatory factors in the serum and colon tissue were measured. Additionally, the mRNA expression of inflammatory factors, and the protein expressions of inflammation-related proteins [interleukin-1β (IL-1β), forkhead box O1(FOXO1), phosphoinositide 3-kinase(PI3K), phosphorylated PI3K(p-PI3K), p38 mitogen-activated protein kinase(p38 MAPK), phosphorylated p38 MAPK(p-p38 MPAK) and phosphorylated protein kinase B(p- Akt)] were determined in colonic tissue. RESULTS KSC-F could alleviate weight loss and colonic tissue damage in UC mice. KSC- F reduced the levels of IL-1β, IL-6, IL-8 and tumor necrosis factor-α (TNF-α) in serum, as well as IL-1β, IL-6, IL-17 and TNF- α in colonic tissue to varying degrees and increased the levels of IL-10 in both serum and colonic tissue (P<0.05 or P<0.01). Moreover, KSC-F decreased the expression levels of IL-1β, IL-17 and TNF-α mRNA, as well as p-PI3K, p-p38 MAPK, and p- Akt proteins in colonic tissue to varying degrees, and increased the expression levels of IL-10 mRNA and FOXO1 protein in colonic tissue (P<0.05 or P<0.01). CONCLUSIONS KSC-F effectively alleviates UC symptoms in mice by inhibiting PI3K, Akt and p38 MAPK activation, mitigating the release of pro-inflammatory factors such as IL-1β, IL-6, TNF- α,promoting the anti-inflammatory factor IL-10 secretion, and reducing inflammation-induced colonic tissue damage.
