1.Clinical research progress on cell therapy induced immune tolerance in liver transplantation

Liver transplantation is the preferred treatment for end-stage liver disease, but recipients require long-term immunosuppressive therapy to control rejection, which may lead to complications and affect their long-term survival. Immune tolerance refers to the ability of organ transplant recipients to maintain their immune system's tolerance to the graft without relying on long-term immunosuppressants. Immune tolerance is an ideal goal pursued in the field of organ transplantation, which can reduce adverse drug reactions and improve long-term survival rates. Cell therapy has emerged as a promising strategy to induce such tolerance after liver transplantation. Therefore, this article reviews the application progress of cell therapies such as regulatory T cells, regulatory dendritic cells, mesenchymal stem cells, hematopoietic stem cells, etc. in inducing immune tolerance after liver transplantation, in order to provide reference for the clinical application of immune tolerance induction after liver transplantation.
2.Neuroprotective effect and mechanism of eleutheroside B on Parkinson’s disease model mice by regulating the IKKβ/NF-κB signaling pathway

OBJECTIVE To investigate the neuroprotective effect and mechanism of eleutheroside B (ELB) on Parkinson’s disease (PD) model mice by regulating the IκB kinase β (IKKβ)/nuclear factor-κB (NF-κB) signaling pathway. METHODS Fifty mice were randomly divided into normal control group, model group, positive control group (selegiline hydrochloride, 10 mg/kg), and ELB low-dose and high-dose groups (80, 160 mg/kg), with 10 mice in each group. Each group was given relevant medicine or normal saline intragastrically for 14 consecutive days. Starting from the 10th day of administration, the model group and all administration groups were intraperitoneally injected with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) 30 mg/kg, for five consecutive days to establish the chronic PD model. After the last administration for 24 h, six mice were randomly selected from each group to test their behavioral abilities; detect the levels of interleukin-1β (IL-1β), IL-10, tumor necrosis factor-α (TNF-α) in brain tissue and their mRNA expressions were measured, and positive expression of tyrosine hydroxylase (TH), protein expressions of TH, α -synuclein ( α -syn), ionized calcium-binding adaptor molecule 1 (Iba-1), as well as phosphorylation levels of IKKβ and NF-κB p65 proteins in the brain tissue were detected. The ultrastructure of neurons in substantia nigra was observed. RESULTS Compared with the model group, rotarod endurance time and climbing score of each administration group (except for the ELB low-dose group) were increased significantly ( P <0.05), while the levels and mRNA expressions of IL-1β, TNF-α, α -syn, and Iba-1, as well as phosphorylation levels of IKKβ and NF-κB p65 proteins in brain tissue were decreased significantly (except for TNF-α in the ELB low-dose group). Conversely, the level and mRNA expression of IL-10 (except for the ELB low-dose group), TH positive expression and protein expressions were significantly increased ( P <0.05). Typical neurodegenerative pathological changes, such as neuronal karyopyknosis, mitochondrial swelling and vacuolization, and endoplasmic reticulum dilation, all showed varying degrees of improvement. CONCLUSIONS ELB may exert neuroprotective effects by inhibiting the activation of the IKKβ/NF-κB signaling pathway, alleviating inflammatory responses, reducing abnormal α -syn aggregation and neuronal loss, and further improving motor dysfunction in PD mice.
3.A scoping review of the relationship between 24 hour movement behaviors and physical and mental health among college students

Objective:
To review the relationship between 24 hour movement behaviors and physical and mental health among college students, in order to provide evidence to support health promotion and further research in universities.
Methods:
Following the Joanna Briggs Institude(JBI) scoping review guidelines, relevant studies published in databases from inception date to December 26, 2025 were searched, including PubMed, Embase, Cochrane Library, Web of Science, China National Knowledge Infrastructure (CNKI) and Wanfang Data. For studies meeting the inclusion and exclusion criteria, a descriptive analysis was conducted to summarize the measurement tools used, adherence rates with guidelines, and the relationship between physical and mental health.
Results:
A total of 30 studies were included. Measurement tools exhibited a high heterogeneity, with questionnaires being the primary method. The rate of full adherence with 24 hour movement behaviors among college students was less than 30%. Moderate to vigorous physical activity and high quality sleep were associated with improvements in physical fitness, cardiopulmonary function, and mental health, while prolonged sitting was negatively associated with obesity and depression. Equivalent time substitution analysis indicated that increasing moderate to vigorous physical activity and reducing prolonged sitting could significantly improve health outcomes.
Conclusions
The adherence rate for 24 hour movement behaviors among college students is low and it is closely associated with physical and mental health. Future studies should standardize measurement tools, and implement targeted interventions based on the optimization of daily activity patterns.
4.Shared genetic architecture between arrhythmia and obsessive-compulsive disorder: a heart-brain axis study based on multi-omics integration

BackgroundArrhythmia and obsessive-compulsive disorder (OCD) frequently co-occur in clinical and epidemiological settings, yet their shared genetic basis and potential heart-brain axis mechanisms remain unclear. ObjectiveTo systematically evaluate the genetic correlation between arrhythmia and OCD, and to elucidate their underlying molecular genetic mechanisms, so as to provide molecular evidence for the "heart-brain axis" to support risk assessment and integrated clinical strategies for these comorbidities. MethodsThe aggregated data from the genome-wide association study (GWAS) of arrhythmia in the UK Biobank (7 207 cases and 477 391 controls) and the GWAS data of OCD released by the Psychiatric Genomics Consortium (2 688 cases and 7 037 controls) were integrated, all of which were limited to individuals of European ancestry. The genome-wide genetic correlations were estimated using the linkage disequilibrium score regression (LDSC) and the high-definition likelihood (HDL). Local genetic correlation analysis was conducted using the local analysis of variance annotation (LAVA). Multi-trait analysis of GWAS (MTAG) was employed to identify pleiotropic loci. Shared risk genes were identified by combining summary-data based Mendelian randomization (SMR) and transcriptome-wide association study (TWAS). Functional enrichment analysis was performed based on the functional mapping and annotation (FUMA) platform. ResultsBoth LDSC (rg=0.248, 95% CI: 0.159–0.336, P=4.82×10-3) and HDL (rg=0.294, 95% CI: 0.237–0.351, P=5.87×10-4) revealed significant positive genetic correlation between arrhythmia and OCD. LAVA identified 23 significantly local correlated regions in the genome (P<2.0×10-5). MTAG discovered 11 genome-wide significant pleiotropic SNPs, among which rs12754189 (intron of KCNN3) had potential functional harmfulness (CADD>12.37). SMR and TWAS jointly identified 20 shared genes, enriched in neural-cardiovascular tissues such as the cerebral cortex, amygdala, and left ventricle, and involved in DNA damage response, RNA metabolism, transcriptional regulation, and FAS signaling pathway (FDR<0.05). ConclusionArrhythmia and OCD share a common genetic basis. The co-morbidity mechanism may involve the common vulnerability of neurons and cardiac muscle cells in terms of gene expression regulation and stress response, supporting the role of the brain-heart axis in the pathophysiology of both conditions.
5.Research on the mechanism of Chaiqi yigan granules against liver cancer via the ferroptosis pathway

OBJECTIVE To explore the mechanism of Chaiqi yigan granules (CQYG) against liver cancer through the ferroptosis pathway. METHODS Network pharmacology combined with ferroptosis-related database was used to screen key targets and main effective components of CQYG against liver cancer via regulating ferroptosis; molecular docking technology was employed to analyze the binding ability of main active components to key targets. Human liver Huh-7 cells were divided into blank serum control (CON) group, CQYG drug-containing serum (CQYGKL) group, ferroptosis inducer (RSL3) group, mammalian target of rapamycin complex 1 (mTORC1) inhibitor (RMC-5552) group, mTORC1 agonist (CCT007093) group, and CCT007093+CQYGKL group. The levels of Fe 2+ , malondialdehyde (MDA), and glutathione (GSH) in the cells were detected in the former three groups; mRNA expressions of mammalian target of rapamycin (mTOR), sterol regulatory element-binding protein 1 (SREBP1), and stearoyl-CoA desaturase 1 (SCD1), protein expressions of SREBP1 and SCD1 as well as phosphorylation levels of mTOR and ribosomal S6 kinase (S6K) proteins were detected in all groups. RESULTS Key targets of CQYG for anti-liver cancer through the ferroptosis pathway were mTOR, SREBP1, SCD1,etc. The main active components included quercetin, tanshinone Ⅱ A , baicalein, etc. The binding energies of main active components to key targets were all less than -5 kJ/mol. Compared with CON group, the levels of Fe 2+ and MDA in the cells in CQYGKL group and RSL3 group were significantly increased, while the levels of GSH were significantly decreased ( P <0.05). mRNA expressions of mTOR, SREBP1 and SCD1, protein expressions of SREBP1 and SCD1, as well as the phosphorylation levels of mTOR and S6K proteins were significantly decreased in the CQYGKL group, RSL3 group, and RMC-5552 group, whereas all the above indicators were significantly increased in the CCT007093 group ( P <0.05). Compared with CCT007093 group, the changes in all the above indicators were significantly suppressed in the CCT007093+CQYGKL group ( P <0.05). CONCLUSIONS CQYG may induce ferroptosis by inhibiting mTORC1/SREBP1/SCD1 axis, thereby exerting anti-liver cancer effects.
6.Efficacy and mechanism of modified Wubei powder combined with Banxia Xie Xin decoction in the treatment of reflux esophagitis

Objective:To investigate the efficacy and underlying mechanism of modified Wubei powder combined with Banxia Xie Xin decoction in the treatment of reflux esophagitis (RE). Methods:A case-control study was conducted involving 82 patients with RE who received treatment at Department of Spleen and Stomach Diseases, Xi'an Hospital of Traditional Chinese Medicine from June 2022 to June 2023. The patients were randomly assigned to either a control group or an observation group, with 41 patients in each group. The control group received conventional treatment, while the observation group was given Wubei powder combined with modified Banxia Xie Xin decoction orally in addition to the conventional treatment. The traditional Chinese medicine symptom scores, lower esophageal sphincter relaxation rate (LESR), percentage of abnormal esophageal contractions, and levels of substance P (SP), cholecystokinin (CCK), motilin (MTL), gastrin (GAS), serum ghrelin, and vasoactive intestinal peptide (VIP) were compared between the two groups before treatment and at 2 and 4 weeks after treatment. Results:There was no statistically significant difference in traditional Chinese medicine symptom scores between the two groups before treatment ( P > 0.05). However, both groups showed lower symptom scores at 2 and 4 weeks after treatment compared with before treatment ( t = 6.87, 10.87, 9.59, 15.39, all P < 0.05). The observation group demonstrated significantly greater improvement in symptom scores at 2 and 4 weeks compared with the control group [(17.68 ± 2.13) vs. (23.76 ± 2.48); (11.44 ± 1.12) vs. (18.82 ± 1.85), t = 4.18, 4.35, both P < 0.05]. There were no statistically significant differences in LESR or percentage of abnormal contractions between the two groups before treatment (both P > 0.05). However, both LESR and the percentage of abnormal esophageal contractions improved at 2 and 4 weeks after treatment compared with before treatment ( t = 4.25, 5.73, 5.86, 6.49, 3.79, 4.84, 4.48, 5.56, all P < 0.05). Additionally, the observation group had significantly lower LESR at 2 and 4 weeks compared with the control group [(71.62 ± 6.37)% vs. (75.46 ± 6.89)%, (64.92 ± 5.82)% vs. (70.78 ± 6.45)%, t = 5.43, 5.86, both P < 0.05]. The percentage of abnormal esophageal contractions was also significantly lower in the observation group [(45.28 ± 5.18)% vs. (59.56 ± 5.84)%; (33.53 ± 4.24)% vs. (47.74 ± 5.31)%, t = 6.08, 7.24, both P < 0.05]. Before treatment, there were no statistically significant differences in levels of SP, MTL, GAS, or CCK between the two groups (all P > 0.05). The levels of SP, MTL, GAS, and CCK improved in both groups at 2 and 4 weeks compared with before treatment ( t = 4.67, 7.15, 7.24, 9.87, 3.62, 5.85, 4.58, 7.17, 3.55, 5.41, 5.42, 6.89, 3.53, 5.43, 5.39, 6.82, all P < 0.05). Furthermore, the observation group had significantly higher levels of SP, MTL, and GAS at 2 and 4 weeks compared with the control group ( t = 3.65, 4.12, 3.86, 4.25, 4.48, 5.36, all P < 0.05), while CCK levels were significantly lower in the observation group ( t = 4.38, 4.51, both P < 0.05). There were no statistically significant differences in ghrelin or VIP levels between the two groups before treatment (both P > 0.05). However, both ghrelin and VIP levels improved in both groups at 2 and 4 weeks after treatment compared with before treatment ( t = 3.35, 3.72, 3.69, 4.28, 3.76, 4.88, 4.11, 5.69, all P < 0.05). Additionally, the observation group had significantly higher levels of ghrelin compared with the control group ( t = 3.43, 4.11, both P < 0.05), while VIP levels were significantly lower in the observation group ( t = 4.59, 5.71, both P < 0.05). Conclusions:The addition of modified Wubei powder combined with Banxia Xie Xin decoction to conventional treatment can considerably alleviate clinical symptoms in patients with RE and improve esophageal pressure. Furthermore, this combined therapy can effectively regulate serum levels of SP, CCK, MTL, GAS, ghrelin, and VIP, thereby modulating the contraction and relaxation of gastrointestinal smooth muscle, enhancing lower esophageal sphincter pressure, and reducing reflux.
7.Analysis of adverse drug reaction monitoring status in 65 medical institutions in guangdong province under the pharmacovigilance system

Objective To investigate the current status of adverse drug reaction(ADR)monitoring in medical institu-tions under China's pharmacovigilance framework in Guangdong Province,and to propose evidence-based strategies for enhancing institutional monitoring capabilities.Methods A cross-sectional survey was conducted using a structured questionnaire devel-oped in alignment with national regulatory requirements and expert consensus.Data were collected from 65 medical institutions,including general hospitals,traditional Chinese medicine hospitals,and maternal/children's hospitals,across 21 prefecture-level cities in Guangdong.Descriptive and comparative analyses were performed to evaluate institutional practices.Results All 65 in-stitutions submitted valid responses.Among them,63(96.9%)had established standardized ADR reporting protocols,with 93.8%(61/65)delegating oversight to pharmacy departments.ADR data were predominantly collected via institutional informa-tion systems(67.7%,44/65),though these systems focused on passive reporting and basic data aggregation,lacking functional-ities for active signal detection or risk alert mechanisms.Significant disparities(P<0.05)were observed across hospitals of dif-ferent tiers in reporting modalities,system sophistication,analytical frequency,and early warning implementation.Notably,mo-nitoring practices for conditionally approved drugs(e.g.,emergency-authorized therapeutics)exhibited systemic deficiencies.Conclusions To address these gaps,the following measures are recommended:Accelerating the adoption of intelligent monito-ring systems to enable real-time ADR detection and predictive analytics;Implementing tiered resource allocation policies to ensure equitable capability development;Establishing specialized protocols for high-risk pharmaceuticals,particularly conditionally ap-proved and fast-tracked drugs;Strengthening interdisciplinary training programs to improve pharmacovigilance literacy among healthcare practitioners.These interventions aim to foster a proactive risk management culture and advance patient safety within China's evolving healthcare landscape.
8.Clinical characteristics of patients with hospital-acquired infections caused by carbapenem-resistant gram-negative bacteria and effectiveness of AMS

OBJECTIVE To understand the clinical distribution of carbapenem-resistant gram-negative bacteria(CR-GNB)hospital-acquired infections and observe the effectiveness of antimicrobial stewardship(AMS)so as to pro-vide theoretical basis for targeted supervision.METHODS The patients who were detected with CR-GNB in the First People's Hospital of Lianyungang from Jan.1,2015 to Dec.31,2023 were monitored by hospital-ac-quired infection informatization monitoring system,the demographic and etiological data were collected from the patients with hospital-acquired infections,the clinical characteristics were analyzed.The incidence of CR-GNB in-fections,isolation rates and drug administration indexes were observed and compared,and the effectiveness of AMS was evaluated.RESULTS A total of 972 633 hospitalized patients were monitored,1319 of whom had hospi-tal-associated CR-GNB infections,and the total incidence of infections was 0.17%,showing a continuously down-ward trend during the years(P<0.001).Carbapenem-resistant Acinetobacter baumannii(43.37%),carbapenem-resistant Pseudomonas aeruginosa(29.72%)and carbapenem-resistant Klebsiella pneumoniae(24.94%)ranked the top 3 species of pathogens.The CR-GNB infections were most prevalent in intensive care unit(ICU),followed by geriatrics department and neurosurgery department.Lower respiratory tract was the predominant infection site.The patients of male,no less than 65 years of age,and ICU stay were at higher risk of CR-GNB infections than other populations(P<0.001).The implementation of AMS reduced the incidence of CR-GNB infections,iso-lation rates,utilization rates of antibiotics,antibiotics prophylaxis rates for type Ⅰ incision surgery and antibiotics use density(P<0.05).CONCLUSIONS The hospital-associated CR-GNB infection show"U"type epidemiological characteristics.It is necessary to focus on the population characterized by male/the elderly/ICU stay.The imple-mentation of AMS may effectively facilitate the control and detection rate of the CR-GNB infections.
9.Progress in mutual regulation of lncRNA and METTL3 in tumors

Epigenetic modifications of RNA play a crucial role in the initiation and progression of tumors.Among these modifications,N6-methyladenosine(m6A)modification catalyzed by the METTL3 methyltransferase is one of the most common RNA modifications.It is essential for regulating RNA transcription,stability,and translation.At the same time,long non-coding RNAs(lncRNAs)also serve as important regulatory molecules and play a significant role in the development and progression of tumors.However,the reciprocal interaction between lncRNAs and METTL3,as well as their functional mechanisms within tumors,are not fully understood.This paper aims to provide a comprehensive over-view of the mutual regulation between lncRNAs and METTL3 in tumors.The goal is to offer new insights and theoretical foundations for a better understanding of tumor pathogenesis.
10.Comparison of three methods for induction of model mice with diminished ovarian reserve

Objective To compare the effects of Tripterygium wilfordii polyglycosides,cyclophosphamide,and cisplatin on the establishment of a mouse model of diminished ovarian reserve(DOR).Methods Mice were randomly divided into the following treatment groups:control(Ctrl),Tripterygium wilfordii polyglycosides(TWP),cyclophosphamide(CTX),and cisplatin(DDP).Mice in the TWP group received a 50 mg/kg suspension of Tripterygium wilfordii polyglycosides by gavage for 14 days,mice in the CTX group received a 20 mg/kg cyclophosphamide suspension by intraperitoneal injection for 14 days,and mice in the DDP group received a 1.5 mg/kg cisplatin solution by intraperitoneal injection for 14 days.The body weight,uterine index,and ovarian index were recorded,the estrous cycle was monitored using the vaginal smear method,and the levels of anti-Mullerian hormone(AMH),estradiol(E2),follicle stimulating hormone(FSH),and luteinizing hormone(LH)were detected using ELISA.Hematoxylin and eosin staining was used to detect ovarian follicle development.The rates of oocyte maturation and fertility were analyzed.Results The three treatment groups of mice all showed the following:significantly decreased body weight and ovarian index(P<0.05);apparent disorder of the estrous cycle;significantly decreased levels of AMH and E2(P<0.05);decreased and increased rates of developing follicles and atretic follicles,respectively(P<0.05);and significantly decreased rates of oocyte maturation,pregnancy,and live birth(P<0.05).Conclusions DOR mouse models were successfully constructed using Tripterygium wilfordii polyglycosides,cyclophosphamide,or cisplatin,as evidenced by decreased body weight and ovarian index,disordered estrous cycle and hormones,and DOR function,resulting in reduced rates of oocyte maturation,pregnancy,and total number of live births.These DOR effects were most appropriate in the cyclophosphamide group.
