1.Intervention effects of Naoxintong capsules on psychological and cardiac status in depressed rats after heart failure

Background: Depression is a common clinical phenomenon in the patients with heart failure (HF). In traditional Chinese medicine (TCM), diseases in the brain and heart are thought to be correlated and interact. Naoxintong capsules (NXT) has been used for treating cardio-cerebrovascular diseases, while its therapeutic effect on depression after HF remains unclear. Objective: The aim of the study is to evaluate the intervention effect of NXT on depression after HF. Methods: Sprague-Dawley rats were assigned into the following 5 groups: sham, model, NXT (250, 1000 mg/kg), and valsartan (8 mg/kg). Coronary artery occlusion was performed to induce HF and subsequent depression in rats. The cardiac function was evaluated by echocardiography, hematoxylin-eosin staining, and Masson trichrome staining. The sucrose preference test and Morris water maze test were carried out to assess the depressive behaviors in rats. The ultrastructure of hippocampal CA1 neurons was observed and the levels of corticotropin-releasing hormone in the hypothalamus, brain-derived neurotrophic factor in the cortex, and adrenocorticotropic hormone (ACTH) in the plasma were determined by enzyme-linked immunosorbent assay. The levels of dopamine, 5-hydroxytryptamine, norepinephrine, and γ-aminobutyric acid in the hippocampus were measured by UPLC-QQQ-MS. Results: NXT reduced myocardial injury and pathological changes in the cardiac tissue and increased the left ventricular ejection fraction, left ventricular fractional shortening, and cardiac output. NXT increased the sugar preference rate and number of crossings and shortened the escape latency. Furthermore, the NXT treatment restored the levels of corticotropin-releasing hormone, adrenocorticotropic hormone, brain-derived neurotrophic factor, dopamine, and γ-aminobutyric acid to the baseline values. Conclusions: NXT not only demonstrates cardioprotective effect but also attenuates depression in the rats after HF. It may exert the antidepressant effect by inhibiting the hyperactivity of the hypothalamic-pituitary-adrenal axis and recovering the levels of neurotrophic factors and neurotransmitters.
2.Role of the APEX family in hepatocellular carcinoma and its potential as a biomarker

Objective To analyze the role of APEX family in hepatocellular carcinoma(HCC).Methods After expression data of HCC and normal liver tissues from public databases(TCGA and TNM plot)were collected,the expression patterns,functional networks,and potential biological pathways of the APEX family in HCC were investigated through differential expression analysis,construction of protein-protein interaction networks(PPI),and Gene Set Enrichment Analysis(GSEA).Receiver operating characteristic(ROC)curve analysis was used to assess the potential of APEX family members as diagnostic markers for HCC,and their correlation with HCC prognosis was anlyzed through survival analysis.RT-qPCR and Western blotting we performed to detect the expression of APEX1 and APEX2 at mRNA and protein levels in liver cells and HCC cells.The effect of silencing APEX1 and APEX2 in HCC cells were determined through CCK-8 assay,EdU assay,scratch healing test,and Transwell assay.Results The expression levels of the 2 members of the APEX family,APEX1 and APEX2,were significantly higher in the HCC samples than the normal liver tissues(P<0.01).The results of ROC curve analysis on public databases showed that the AUC value of APEX1 and APEX2 in predicting HCC was 0.922 and 0.917,respectively.PPI network analysis revealed that APEX family members regulated cell cycle through various signaling pathways.Survival analysis indicated that HCC patients with high expression levels of APEX1 and APEX2 had significantly shorter overall survival than those with low levels(P<0.05),and APEX1 and APEX2 were highly correlated with traditional tumor survival prognostic factors Ki-67 and TP53(P<0.001).The mRNA and protein levels of APEX1 and APEX2 were significantly higher in the liver cancer cells than the normal liver cells(P<0.05).In addition,silencing APEX1 and APEX2 resulted in notably reduced proliferation,migration,and invasion abilities of HCC cells(P<0.05).Conclusion APEX1 and APEX2 can serve as potential biomarkers for HCC diagnosis and prognosis assessment,and their involvement in the regulation of HCC proliferation and metastasis provides new strategies and targets for the early diagnosis and individualized treatment of HCC.
3.Mutation characteristic of STK11 gene in Chinese with Peutz-Jeghers syndrome

Objective To identify the mutation characteristic of STK11 gene in Chinese with Peutz-Jeghers syndrome(PJS) and establish the base of the gene diagnosis of PJS.Methods STK11 germline mutation was analysed by DNA sequencing in 18 unrelation patients with PJS.Results Six novel mutations of STK11 gene were detected in six unrelation patients. These mutations will lead to production of truncated protein.Conclusion STK11 gene mutation accounts for one third of the Chinese with PJS. The content of mutation includes single base substitution or deletion and one or two bases insertion. The mutations were widely found in different regions of the whole coding sequence, and 2/3 of those concentrate in exon 1. Mutation frequency is 66.7% in the family suffering PJS in two or more generations, and 16.7% in the disseminated cases.
