1.Investigating the impact of silencing an RNA-binding protein gene SlRBP1 on tomato photosynthesis through RNA-sequencing analysis.

Photosynthesis in plants directly affects the synthesis and accumulation of organic matter, which directly influences crop yield. RNA-binding proteins (RBPs) are involved in the regulation of a variety of physiological functions in plants, while the functions of RBPs in photosynthesis have not been clearly elucidated. To investigate the effect of a glycine-rich RNA-binding protein (SlRBP1) in tomato on plant photosynthesis, a stably inherited SlRBP1 silenced plant in Alisa Craig was obtained by plant tissue culture using artificial small RNA interference. It turns out that the size of the tomato fruit was reduced and leaves significantly turned yellow. Chlorophyll(Chl) content measurement, Chl fluorescence imaging and chloroplast transmission electron microscopy revealed that the chloroplast morphology and structure of the leaves of tomato amiR-SlRBP1 silenced plants were disrupted, and the chlorophyll content was significantly reduced. Measurement of photosynthesis rate of wild-type and amiR-SlRBP1 silenced plants in the same period demonstrated that the photosynthetic rate of these plants was significantly reduced, and analysis of RNA-seq data indicated that silencing of SlRBP1 significantly reduced the expression of photosynthesis-related genes, such as PsaE, PsaL, and PsbY, and affected the yield of tomato fruits through photosynthesis.

2.Dynamic phenotype of lung type Ⅱ alveolar epithelial cells in radiation-induced lung injury and its role in the formation of fibrosis

Objective:To explore the dynamic phenotype of type Ⅱ alveolar epithelial cells(AEC Ⅱ)in radiation-induced lung fibrosisand its role in the formation of fibrosis.Methods:Totally 90 C57BL/6J female mice were divided into 2 groups: irradiation group (50, thoracic irradiation with a single dose of 20 Gy X-rays), control group (40, sham irradiation). At 24 h, 4 and 12 weeks after irradiation, 5 mice were euthanized and the lungs were collected for pathological observation. The other lungtissues were collected for the isolation of primary AEC Ⅱ cells with microbeadssorting.The mRNA expressions of proSP-C, HOPX, vimentin, β-catenin and TGF-β1 in AEC II cells were detected by RT-PCR.Results:Acute pneumonitis was observed in the lungs at 24 h after irradiation and alleviated in accompany with partial alveolar septal thickening and a small amount of collagen deposition at 4 weeks after irradiation. The collagen deposition became more pronounced at 12 weeks after irradiation, together with collapsed and fused alveolar cavities, alveolar septal hyperplasia, and pulmonary fibrosis formation.The mRNAexpression levels of proSP-C and HOPX in primary AEC Ⅱ cells increased at 24 hours after irradiation and then approached to a peak value at 4 weeks after irradiation ( F=8.441, 3.586, P=0.036). The mRNA expression levels of vimentin, a biomarker of EMT, was increased significantly at 4 weeks and continued up to 12 weeks after irradiation( F=8.358, P=0.001). The mRNA expression levels of profibrotic factors β-catenin and TGF-β1 were both significant increased at 12 weeks after irradiation( F=4.62, 3.279, P=0.044). Conclusions:The phenotypeof AECⅡ cells could not only be transformed from proSP-C+ to HOPX+ /proSP-C+ , HOPX+ /proSP-C+ /vimentin+ , and vimentin+ /proSP-C, but also differentiated into mesenchymal cells with highly expressed profibrotic factors, thereby inducing EMT process, which either played a role in the repair of radiation-induced lung injury or triggered radiation-induced fibrosis.
3.Genetic variation analysis of Nsp2, ORF5 and ORF3 of PRRSV SC-GY strain from Sichuan province

To monitor genetic variation of porcine reproductive and respiratory syndrome virus (PRRSV),RT-PCR was used to identify a sample suspected of PRRSV infection.A PRRSV named SC-GY strain was obtained,and its Nsp2,ORF5 and ORF3 genes were used for sequence alignment and phylogenetic tree construction.The results showed that SC-GY strain is highly pathogenic PRRSV American variant strains with Nsp2 gene discontinuous deletion of 30 amino acids,ORF3 gene aa17 a serine (S) insert.Comparing to VR2332,CH-1a,JXA1,HUN4,NADC30,HENAN-XINX and SC2012,the Nsp2,ORF5 and ORF3 of SC-GY shared 70.3%-97.9%,82.4%-97.6% and 83.1%-98.2% of nucleotide similarity,and 62.3%-96.3%,78.0%-95.7% and 81.6%-96.5% of deduced amino acid similarity;and compared to LV they shared only 18.9%,60.8% and 63.7% of nucleotide similarity,and 14.0%,54.9% and 57.2% of deduced amino acid similarity.The phylogenetic tree revealed that the SC-GY formed independent small branches although it belonged to the same subgroup as highly pathogenic PRRSV strains.The results showed that in high frequency live vaccine immunization of currently PRRSV,the gene of PRRSV epidemic strain is still in constant variation.Vaccination of live PRRSV vaccines should be reduced and surveillance of PRRSV strains should be enhanced.
4.Establishment and effect of nursing information technology platform

Objective To explore nurses' bonus assessment and distribution methods based on the information technology platform and to improve the efficiency of nursing management. Methods We build a hospital informa-tion platform in order to collect nursing work efficiency data by using existing computer information network. We distributed the bonus through the information platform according to evaluate nursing quality and performance effi-ciency. Results After the information flatform used for 2 years,the satisfaction of nurses with different professional titles and different experience showed no significant difference (P>0.05). The time of department management person-nel collecting and distributing data change from [103.75 (180.00,215.00)] min per month to [42.50 (60.00,90.00)] min per month.The difference showed statistical significance(P<0.05). Conclusion The technology platform can im-prove the satisfaction and the nursing management efficiency.
5.Investigation and influencing factors of the core competence of registered nurses in class Ⅲ grade A hospitals in Xi′an city

Objective To investigate the core competence of nurses in class Ⅲ grade A level hospitals in Xi′an City and its influencing factors so as to provide evidences for managers to enhance the core competence of clinical nurses and promote the quality and safety of nursing.Methods A total of 890 registered nurses from class Ⅲ grade A hospitals in Xi′an city were investigated using "the competency inventory for registered nurse scale" (CIRN) from July to September 2015. Statistical software SPSS 19.0 was used to analyze the data. Influencing factors of the core competence of nurses were analyzed using multiple linear regression analysis. Results The total score of core competences of clinical registered nurses was (159.12±33.21). The dimension with highest score was ethical/legal practice ability with an average score of (3.01±0.64). The dimension with lowest score was critical thinking/scientific research ability with an average score of (2.70±0.67). The scores of nurses from emergency and critical care were higher than those of nurses from general departments in the dimensions including clinical nursing,interpersonal relationship,ethical/legal practice, professional development and critical thinking/research ability. The differences were statistically significant (Z=-3.496, -2.688,-3.727,-2.969,-3.911;P<0.05). Multivariate linear regression analysis showed that technical title, age, department training, and professional qualification were influencing factors of the nurses′ core competence (P<0.05).Conclusions The core competence of registered nurses in class Ⅲ grade A hospitals in Xi′an city is at above medium level. The scores of nurses from emergency and critical care are higher than nurses from general departments in several dimensions. Age, technical title, department training and professional qualification are main influencing factors of core competence of nurses. Interventions to improve the core competency of nurses with low grade, low technical title and non-specialist qualification are necessary.
6.Establishment of a transgenic heterozygous mouse model of ApcMin/+pre-cancerosis of colorectal cancer with p110δmutation

[ABSTRACT]AIM:Toestablishatransgenicheterozygousmousemodelofprecancerouslesionsofcolorectal cancer with p110δmutation in the C57BL/6J background for serving the studies on colorectal cancer research mediated by p110δ.METHODS:The transgenic heterozygous mice were generated by crossing in p110δD910A/D910A mouse and ApcMin/+mouse, and the genotype was detected by PCR .Compared with ApcMin/+mice, transgenic heterozygous mice ( ApcMin/+;p110δD910A/D910A)were counted, and the number and size of intestine polyps were analyzed after methylene blue staining . The intestinal tissue structure was assessed by HE staining .RESULTS:The transgenic heterozygous mouse model of pre-cancerous lesions of colorectal cancer with p 110δmutation was established .The number and size of polyps in the transgenic heterozygous mice were declined .CONCLUSION: A transgenic heterozygous mouse model of precancerous lesions of colorectal cancer with p 110δmutation was successfully established .The initial phenotype of intestinal tumors in transgenic mice was observed .This model will greatly contribute to the related research of colorectal cancer in mice .
7.Contrast study of middle school students ’ oral health education and pro-motion in Xixiang street of Baoan district

Objective To analyze the effect of different educational models on oral health education and health promo-tion among middle school students. Methods Junior students from January to June 2011 as the object of study average from 3 middle school in Xixiang street Baoan District were randomly selected, that's each school selected 101 students, and male to female ratio was about 1:1. The students were randomly divided into in three groups, form-teacher model group (group A), mother-model group (group B) and control group (group C). After education, measured the 3 groups students' oral hygiene indexes and gingival indexes before and after 3 months, in order to compare and judge the effect of different educational models. Results According to analyze oral hygiene index, gingival index and mastery of knowl-edge on oral health education and promotion of A,B,C group, group A and B were better after education, but they were worse than group C (P all<0.05). After education in knowledge, passing percent of group A was 89.11%, while group B and C were 72.43% and 52.48%. It was indicated that group A and B were both better than group C (P all < 0.01). Conclusion Form-teacher model and mother-model education on oral health education and promotion are significant results, both of whom are competitive advantage and practical significance in clinical trials.
8.Comparative studies on photosynthetic characteristics of Notopterygium incisum and N. forbesii.

OBJECTIVETo compare the photosynthetic characteristics of Notopterygium incisum and N. forbesii in order to provide basic data for introduction and cultivation of the two wild medicinal species.
METHODThe light-response, CO2-response and Chlorophy II fluorescence parameters of leaves at the booting stages between N. incisum and N. forbesii, were analyzed in situ by Li-6400 Portable Photosynthesis system under natural conditions.
RESULT1) The light saturation point (LSP) was 1539 micromol x m(-2) x s(-1) for N. incisum and 1464 micromol x m(-2) x s(-1) for N. forbesii, the maximum net photosynthetic rate (Pmax) was 22.95 micromol x m(-2) x s(-1) for N. incisum and 19.65 micromol x m(-2) x s(-1) for N. forbesii, the apparent quantum yield (AQY) was 0.0509 for N. incisum and 0.0470 for N. forbesii, LSP, AQY and Pmax of N. incisum were significantly higher than those of N. forbesii; the light compensation point (LCP) was 17.92 micromol x m(-2) x s(-1) for N. incisum and 26.69 micromol x m(-2) x s(-1) for N. forbesii, LCP of N. incisum was significantly lower than that of N. forbesii. 2) The carbondioxide compensation point (CCP) were 33.41 micromol x mol(-1) for N. incisum and 37.82 micromol x mol(-1) for N. forbesii, the carbon dioxide saturation point (CSP) were 988 micromol x mol(-1) for N. incisum and 1150 micromol x mol(-1) for N. forbesii, CCP and CSP of N. incisum were significantly lower than N. forbesii; the carboxylation efficiency (CE) were 0.0591 for N. incisum and 0.0459 for N. forbesii; the maximum rate of RuBP regeneration (Jmax) were 28.18 micromol x m(-2) x s(-1) for N. incisum and 25.32 micromol x m(-2) x s(-1) for N. forbesii; the light respiration rate (Rd) were 1.971 micromol x m(-2) x s(-1) for N. incisum and 1.736 micromol x m(-2) x s(-1) for N. forbesii, CE, Jmax and Rd of N. incisum were higher than those of N. forbesii. 3) The primary light energy conversion of PS II (Fv/Fm) was 0.8213 for N. incisum and 0.8257 for N. forbesii, wihich didn't showed significant difference, between N. incisum and N. forbesii there was no photoinhibition.
CONCLUSIONBoth N. incisum and N. forbesii were C3 type plant, could perfectly acclimate to light condition. However, the weak light of N. incisum was absorbed significantly higher than that of N. forbesii, strong photosynthesis ability causes assimilation products accumulation of N. incisum obviously to be higher than that of N. forbesii.

9.Characteristics of Molecular Recognition of Norfloxacin Template Polymer

A molecular template synthetic polymer highly selective for norfloxacinum was prepared by a molecular imprinting technique. The selective binding characteristics of the template polymer was evaluated by Scatchard analysis. The multiple-sites binding model was used to calculate the maximum numher of binding sites and the dissociation constant. The results showed that the template polymer using methacrylic acid (MAA) as functional monomer could form two kinds of binding sites. The dissociation constants were estimated to be Kd1 = 2.9 × 10-5mol/L and Kd2 = 3.2 × 10-3 mol/L. The selective binding experiment for substrates indicated that the polymer gave much higher affinity and selectivity for norfloxacin than for acid pipemidic and cefalexin. It is possible to be a good adsorption and binding material in the selective enrichment and determination of trace norfloxacin in complex biosamples.
10.Role of heme oxygenase-1/carbon monoxide system in pulmonary ischemiareperfusion injury

AIM: To investigate the effect of heme oxygenase-1 (HO-1)/carbon monoxide (CO) system on pulmonary ischemia-reperfusion injury (PIRI) in rabbits. METHODS: Single lung ischemia and reperfusion animal model was used in vivo. The rabbits were randomly divided into three groups (n=10 in each), control group (C), PIR group (I-R), PIR+ hemin group (H) and PIR+zinc protoporphyrin IX (ZnPP) group (Z). Changes of several parameters which included plasma carboxyhemoglobin (COHb) at different time points, wet to dry ratio of lung tissue weight (W/D), the injured alveoli rate (IAR) and the HO-1 enzymatic activity were measured at 180 min after reperfusion in lung tissue. The tissue slides were also stained by immunohistochemistry (IHC) and in situ hybridization (ISH) for HO-1 to detect the expression of HO-1 in lung and to analyze the optical density. The lung tissue was prepared for electron microscopic observation at 180 min after reperfusion. RESULTS: The plasma content of COHb in I-R, H, and Z group increased in a time-dependent manner after I-R. But the increment of H group was higher than that of I-R group, while that of Z group was lower.
