1.Investigation on mental health status of front-line anti-epidemic medical staff during the COVID-19 outbreak

ObjectiveTo investigate the mental health status of the front-line anti-epidemic medical staff during the COVID-19 outbreak, so as to provide references for the targeted psychological intervention and improvement of mental health status. MethodsA total of 162 front-line medical staff who worked in Lanzhou Heavy Particles Hospital of Gansu Province from October to December 2021 were were enrolled, and assessed using self-designed general information questionnaire, Self-rating Anxiety Scale (SAS) and Self-rating Depression Scale (SDS). ResultsA total of 144 medical staff completed the valid questionnaire survey, and 17 (11.81%) and 19 (13.19%) cases were found to have anxiety and depression, respectively. The detection rate of anxiety yielded statistical difference among medical staff with different anti-epidemic working hours (χ2=10.602, P<0.01), and the detection rate of depression demonstrated statistical differences among medical staff with different occupations, family history of depression and anti-epidemic working hours (χ2=5.486, 4.039, 7.526, P<0.05). ConclusionDuring the COVID-19 outbreak, front-line medical staff develop varying degrees of anxiety and depression. Furthermore, the detection rate of anxiety is relatively high among the staff with long anti-epidemic working hours, and the detection rate of depression is relatively high among the doctors, the staff with family history of depression and long anti-epidemic working hours.
2.Activation of CD8+T cells regulated by γ-aminobutyric acid and its receptors

Objective To evaluate the effect of γ-aminobutyric acid (GABA) and its receptors upon the proliferation of CD8+T cells.Methods The splenic CD8+T cells of Balb/c mice were obtained by CD8+f cell magnetic bead sorting kit.Under the effect of CD3/CD28-activated magnetic bead,CD8+T cells were stimulated by different concentrations of GABA.5-bromo-2-deoxyuridine (BrdU) labeling and flow cytometry were performed to detect the proliferation of CD8+T cells.The expression levels of GABA-A and GABA-B receptor before and after CD8+T cell activation were compared by fluorescent quantitative real-time polymerase chain reaction (PCR).Result Flow cytometry result revealed that GABA could inhibit the proliferation of activated CD8+T cells,manifested as significant decrease in the quantity of CD152+CD8+T cells.Fluorescent quantitative real-time PCR demonstrated that GABA-A receptor subtypes α2,α6 and GABA-B receptor subtype 1a were expressed only when the CD8+T cells were activated.After CD8+T cell activation,the quantity of GABA-A receptor subtypes α3,αs,β2,β3,γ1,γ2 and θ were significantly increased,whereas the quantity of GABA-B2R and GABA-B1b did not significantly differ before and after CD8+T cell activation.Conclusions GABA can suppress the proliferation of activated CD8+T cells.The activation of CD8+T cells is regulated by GABA receptors.However,the underlying mechanism remains to be elucidated.
3.Effect of Psychological Intervention Based on Pharmaceutical Care on the Community Patients with Essential Hypertension

OBJECTIVE:To evaluate the effect of psychological intervention based on pharmaceutical care on the communi-ty patients with essential hypertension. METHODS:172 patients with essential hypertension in a community health service certer in Nanjing Gulou area in 2013 were divided into control group(n=87)and intervention group(n=85)by simple random meth-od. Control group received conventional antihypertensive drugs and management,intervention group additionally received pharma-ceutical care and psychological intervention,it lasted 6 months. Blood pressure controlling,drug compliance,mastery of drug knowledge and health status in 2 groups before and after treatment were compared. RESULTS:Before treatment,there were no significant differences in the systolic blood pressure(SBP)and diastolic blood pressure(DBP)between 2 groups(P>0.05);af-ter treatment, SBP and DBP in 2 groups were lower than before,SBP and DBP in intervention group were lower than control group,the decrease degree of SBP and DBP was bigger than control group,the differences were statistically significant (P<0.01). 1 month after the beginning of intervention,there was no significant difference in the proportion of drug compliance be-tween 2 groups (P>0.05);1 month after the end of the intervention,the proportion of drug compliance in intervention group was higher than 1 month after the beginning of intervention and control group,the differences were statistically significant(P<0.05). Before treatment,there was no significant difference in mastery of drug knowledge between 2 groups (P>0.05);after treatment,the proportion of patients showed good and excellent mastery of drug knowledge was higher than before and control group,the differences were statistically significant (P<0.05). Before treatment,there were no significant differences in scores of physical,mental,social health subscale and SRHMS between 2 groups(P>0.05);after treatment,scores of physical health subscale and SRHMS in control group,each subscale and to-tal scale scores in intervention group were higher than be-fore, and scores of mental, social health subscale and SRHMS in intervention group were higher than control group, the differences were statistically significant (P<0.05). CONCLUSIONS: The psychological intervention based on pharmaceutical care can effectively promote the con-trol of blood pressure,improve drug compliance,mastery of drug knowledge,health status and mental situation,which is worth of popularizing and applying in community health services.
4.Evaluation of axillary lymph node metastasis of breast cancer via an ultrasonic multi parameter regression model

Objective To evaluate conventional ultrasound combined with real-time elastography in differential diagnosis of metastatic axillary lymph nodes of breast cancer with a logistic regression model.Methods Conventional ultrasound and real-time elastography were performed in 112 breast cancer patients with 113 axillary lymph nodes.All the cases were confirmed with pathological examinations after surgery. A binary logistic regression model based on ultrasonographic features was developed.A receiver operator characteristic ( ROC) curve was constructed to assess the performance of the model.Results There were 28 nodes with no metastasis and 85 with metastasis in a total of 113 axillary lymph nodes.Three ultrasonographic features including the elastic strain ratio(SR), elastic score and shape were finally selected into the logistic regression model .The correct rate of the logistic regression model for predicting axillary lymph node metastasis was 93.8%, and the area under ROC curve was 0.962.Conclusion The binary classification logistic regression model has a good diagnostic efficacy in the diagnosis of benign and malignant axillary lymph nodes,while real-time elastography can improve the accuracy of conventional ultrasound in the diagnosis of axillary lymph nodes.
5.Perioperative nursing of percutaneous left atrial appendage closure for preventing thromboembolism of ;the patients with non-valvular atrial fibrillation

Objective To investigate the perioperative nursing of percutaneous left atrial appendage closure for preventing thromboembolism of the patients with non-valvular atrial fibrillation . Methods Preoperative examinations and psychological care were given to 10 non-valvular atrial fibrillation patients who underwent percutaneous left atrial appendage closure . After the operation , vital signs were monitored, more attention was paid on the patients′complaints and complications were watched vigilantly . Discharge guidance was strengthened and regular follow-up was given to the patients .Results Interventional therapies were performed on 9 patients successfully .The effects of 1, 3 and 6 month follow-up after the operation were satisfactory .No complications happened .Left atrial appendage excision was performed on 1 patient due to intraoperative pericardial effusion .Conclusions Strengthening perioperative nursing of percutaneous left atrial appendage closure for preventing thromboembolism of the patients with non-valvular atrial fibrillation can reduce the incidence of complications promote the postoperative rehabilitation of patients and improve the quality of life of patients.
6.CT and MRI findings of littoral cell angioma of spleen

Objective To evaluate the value of CT and MRIin diagnosis of littoral cell angioma(LCA) of spleen.Methods CT and MRI findings the data of 6 patients with LCA were rospectively analyzed,and all patients were proved LCA of spleen by pathology.Then enhanced CT (n =4 cases) and MRI (n =2 cases) were performed.The imaging performance of LCA was observed,and its imaging features was summarized.Results None of patients showed obvious signs and symptoms of hypersplenism,4 cases were with splenomegaly.5 patients had a single lesion,only one had few lesions.There were not any calcification or envelopement of all the lesions.On CT,all lesions demonstrated distinct borders.The enhanced scan for low-density nodules demonstrated slow progressive enhancement.On MRI,all the LAC had well circumscribed borders,and demonstrated T1 · hypointense and T2-equal,hyperintense signals,and progressive enhancement on the post contrast images.Conclusion CT and MRI imaging of littoral cell angioma of spleen have special imaging characteristics,those particular findings is helpful in the diagnosis.
7.Effect of Small Interfering RNA Silencing the Fatty Acid Synthase Gene on Lipid Metabolism in Human Hepatic Cell Line HepG2

Objective: To investigate the effect of the gene interfering technology on fatty acid synthase (FAS) gene silencing for lipid contents in human hepatic cell line HepG2 and to study the lipid metabolism related gene expression in HepG2 cells. Methods: A total of 3 pairs of small interfering RNA (siRNA) targeting different sequences of FAS mRNA were synthesized as FAS-siRNA-1, FAS-siRNA-2 and FAS-siRNA-3, meanwhile, 2 controls were established as Blank control group, in which HepG2 cells were not treated, and Negative control group, in which HepG2 cells were transfected by non-effective siRNA. The mRNA, and protein expression levels of FAS in HepG2 cells were examined by real-time lfuorescence quantitative RCR and Western blot analysis to screen the most effective pair of siRNA for FAS gene silencing; and that speciifc siRNA was transtected to HepG2 cells for 48 hours to detect the intra-/extra-cellular TG, TC levels and the mRNA expression related to lipid metabolism in HepG2 cells. Results: The screening experiment indicated that FAS-siRNA-3 was most effective for FAS gene silencing. Compared with Blank control group, the mRNA and protein expressions in FAS-siRNA-3 transfected HepG2 cells (Transfected group)decreased to (52.33 ± 3.07) % and (51.57 ± 3.14) % respectively. Compared with Blank control group, Transfected group had the reduced intra-/extra-cellular TG levels and reduced extracellular TC level; while increased mRNA expression of hepatic lipase,P<0.0001 and decreased mRNA expression of TG transfer protein in HepG2 microsome,P<0.05. Conclusion: FAS gene silencing could signiifcantly decrease the intra-/extra- cellular TG level and extracellular TC level in HepG2 cells, those ifndings need to be conifrmed by furtherin vivo andin vitro studies.
8.Prevalence and molecular analysis of α-thalassemia in preschool children in Chongqing city.

OBJECTIVETo investigate the incidence and the gene mutation frequencies and patterns of α-thalassemia in preschool children in Chongqing city.
METHODSCluster random sampling was used. A total of 1057 preschool children in three areas of Chongqing were screened by using routine blood test and hemoglobin electrophoresis analysis. Molecular analysis carried out for all the samples.
RESULTSOf the 1057 samples, 55 cases were diagnosed as being carriers of α-thalassemia, which included 80 allele genes. Therefore, the frequency of α-thalassemia carriers in Chongqing was 5.20%. Of the 55 α-thalassemia carriers, five different deletions of α-thalassemia were identified, the three most common deletion types and proportions were 54.55% for the -α(3.7) deletion, 18.18% for --(SEA) deletion, and 9.08% for the -α(4.2) deletion, respectively; eight types of nondeletion defects were determined, containing one case of Hb Quong Sze and seven novel mutations of a-globin gene. Furthermore, 24 cases of α-Triplication were detected with the α-Triplication carrier rate of 2.55%. In addition, in this study we also found two cases of abnormal hemoglobin disorders occurred on α-globin gene, Hb J-Wenchang-Wuming and Hb Arya. Hb Arya was characterized in the Chinese population for the first time confirmed by literature retrieval.
CONCLUSIONIn this study, we have clarified the carrier frequency and molecular spectrum of α-thalassemia in Chongqing, and we first reported the carrier incidence of α-Triplication in Chongqing. The materials obtained from this study would be of valuable reference for genetic counseling and the examination instruction of children in this area.

9.Investigation on the cognition, attitude to disease in outpatient with atrial fibrillation

Objective To investigate and analyze the cognition , attitude to disease in outpatient with atrial fibrillation ( AF) to provide the basis for clinical nursing .Methods One hundred and sixty outpatient with AF from March 2013 to April 2013 were chosen and were surveyed by the self-made questionnaire about the cognition, attitude to disease in AF patients .The related factors which can affect the cognition , attitude to disease in AF patients including age , gender , occupation , education , economic income monthly , payment of medical expenses , type of AF were analyzed .Results The score of the cognition in AF patients was (14.91 ± 4.92), and was in a bad level .The score of the attitude to disease was (18.59 ±4.16), and was in a good level.The differences were found in the cognition in AF patiens with different age , occupation, education, the type of AF (F=1.00, 2.61, 2.20, 4.21, respectively;P<0.05).The differences were found in the attitude to disease in AF patients with different occupation , education, payment of medical expenses (F=0.28, 0.95, 0.92, respectively;P<0.05).Conclusions The nurses should carry out health education according to the different clinical characteristics in AF patients , and improve the patient compliance , and actively cooperate with medical treatment .
10.Parametric imaging of contrast-enhanced ultrasound on recurrent hepatocellular carcinoma after liver transplantation

Objective To explore the diagnostic value of parametric imaging of contrast-enhanced ultrasound(CEUS) on recurrent hepatocellular carcinoma after liver transplantation.Methods CEUS images of 41 recurrent hepatocellular carcinoma after liver transplantation was analyzed by Sonoliver CAP software.The color code image,curve image and quantitative parameter of DVP of each recurrence lesion was recorded,then typed and analyzed statistically.Results The DVP patterns were classified into 3 types,they were washed out types,non-washed out types and negative types.The washed out types,non-washed out types and negative types on color code image and curve image of DVP were 70.73% (29/41),24.39% (10/41),4.88% (2/41) and 63.41%(26/41),34.15% (14/41) and 2.44% (1/41) respectively.The maximum intensity,rise time,time to peak of the recurrence lesion and the surrounding liver parenchyma were (149.98± 65.29) %,(12.32 ± 5.83)s,(13.01 ±6.07)s and (100±0.00)%,(26.10± 10.81)s,(29.69± 11.60)s respectively,and showed statistical differences (P <0.05).Conclusions The difference of blood perfusion between the recurrence lesion and the surrounding liver parenchyma can be displayed by DVP's dynamic,direct and quantitative imaging,which can be used to provide valuable information about the detection of intrahepatic lesion of recurrence after liver transplantation.
