1.Status quo of general practice postgraduate researches: a visualized analysis of master′s theses

Objective:To understand the status quo of general practice postgraduate researches through visualized analysis of masters′ theses.Methods:The master′s theses in general medicine from January 2007 to December 2022 were retrieved from China Knowledge and Wanfang databases. The number and regional distribution of theses, the degree-granting institutions were analyzed; the performed keywords were visualizde with CiteSpace and VOSviewer softwares.Results:A total of 1 688 master′s theses in general practice were included in the analysis. The highest number of theses ( n=345) were published in 2022. The theses from institutions in the eastern, central and western regions of China accounted for 45.67% (771/1 688), 22.16% (374/1 688) and 32.17% (543/1 688), respectively, and those in Shaanxi Province accounted for the highest proportion at 13.68% (231/1 688). A total of 3 755 keywords were included in the theses, and the top three were diabetes mellitus (115/3 755), risk factors (77/3 755) and clinical efficacy (71/3 755). The results of keyword clustering and timeline mapping showed that the research topics were mainly clustered around five themes: clinical treatment and drug efficacy/safety for diabetes mellitus, training of general practitioners and evaluation of general practice services, risk factors related to common chronic diseases in the community, risk factors and cognitive changes in cerebrovascular diseases, and pathogenic mechanisms of cardiovascular diseases. The results of the emergent words showed that recent research hotspots mainly included obesity, older adults, and cognitive function. Conclusions:The number of general practice master′s theses published in China is increasing rapidly with an uneven regional distribution; and the research topics are diverse, mainly focusing on the diagnosis and treatment of chronic diseases and analysis of related risk factors.

3.Aureane-type sesquiterpene tetraketides as a novel class of immunomodulators with interleukin-17A inhibitory activity.

Interleukin (IL)-17A, a pro-inflammatory cytokine, is a fundamental function in the onset and advancement of multiple immune diseases. To uncover the primary compounds with IL-17A inhibitory activity, a large-scale screening of the library of traditional Chinese medicine constituents and microbial secondary metabolites was conducted using splenic cells from IL-17A-GFP reporter mice cultured under Th17-priming conditions. Our results indicated that some aureane-type sesquiterpene tetraketides isolated from a wetland mud-derived fungus, Myrothecium gramineum, showed remarkable IL-17A inhibitory activity. Nine new aureane-type sesquiterpene tetraketides, myrogramins A-I ( 1, 4- 11), and two known ones ( 2 and 3) were isolated and identified from the strain. Compounds 1, 3, 4, 10, and 11 exhibited significant IL-17A inhibitory activity. Among them, compound 3, with a high fermentation yield dose-dependently inhibited the generation of IL-17A and suppressed glycolysis in splenic cells under Th17-priming conditions. Strikingly, compound 3 suppressed immunopathology in both IL-17A-mediated animal models of experimental autoimmune encephalomyelitis and pulmonary hypertension. Our results revealed that aureane-type sesquiterpene tetraketides are a novel class of immunomodulators with IL-17A inhibitory activity, and hold great promise applications in treating IL-17A-mediated immune diseases.
4.Impact of violent video games and other factors on aggressive behavior of junior high school students in Shanghai

Objective:
To understand the impact of violent video games exposure, self control level and coping style on aggressive behavior of junior high school students in Shanghai, and provide a basis for future interventions to effectively reduce adolescents aggressive behavior.
Methods:
Cluster sampling was used to select 1 886 students of 4 public middle schools in Shanghai from May to June 2019. The data was collected included Violent Video Game Use Habits Questionnaire, Buss & Perry aggression questionnaire, Adolescent Self Control Dual System Scale and Simple Coping Style Questionnaire.
Results:
According to the potential profile analysis, the level of aggressive behavior of junior school students in Shanghai could be divided into four groups, which was low aggressive group,middle aggressive group one, middle aggressive group two and high aggressive group according to level of aggregate. After controlling for gender and age, multivariate Logistic regression showed that exposure to violent video games, impulse system and negative response were risk factors for aggressive behavior( P <0.05); using low aggressive group as control group,the OR of violent video games in other three groups were 1.26, 1.30 and 1.70 respectively. The OR of impulse system were 2.96 , 4.40 and 6.84 respectively. The OR of negative response were 1.17, 1.42 and 1.74 respectively. Positive response was a protective factor( P <0.05). Using low aggressive group as control group, the OR of positive in other three groups were 0.82, 0.52 and 0.49 respectively.
Conclusion
Violent video game exposure, impulsive system and negative coping style can increase the level of aggressive behavior of junior high school students, while positive coping style can reduce the level of aggression of junior middle school students.
5.Study on Protective Mechanism of Dexmedetomidine on Cerebral Injury in Sepsis Model Rats Based on SIRT 1/ Akt/GSK3β/β-catenin Signaling Pathway

OBJECTIVE:To in vestigate the effects of dexmedetomidine (Dex)on SIRT 1/Akt/GSK3β/β-catenin signaling pathway in cerebral injury of sepsis model rats ,and explore the mechanism of its protecitve effect on cerebral injury. METHODS : A total of 80 male SD rats were randomly divided into sham operation group (Sham group ),sepsis group (CLP group ),CLP+Dex group(10 μg/kg Dex),CLP+Dex+Sirtinol group (10 μg/kg Dex+2 μL/100 g SIRT 1 inhibitor sirtinol ),with 20 mice in each group. Two hours before modeling ,CLP+Dex+Sirtinol group was injected with sirtinol via lateral ventricle. Sepsis model was induced by cecal ligation and perforation in each group (in sham group ,only operation was performed but no ligation was performed). At 0,3,6 h after modeling ,CLP+Dex group and CLP+Dex+Sirtinol group were given Dex (10 μg/kg) intraperitoneally,Sham group and CLP group were given constant volume of normal saline intraperitoneally. Cerebral tissue water content,Evans blue (EB)content,apoptosis in cerebral cortex ,the levels of IL- 1β and TNF-α in cerebral tissue as well as the protein expression of SIRT 1,p-Akt,p-GSK3β and β-catenin in hippocampus were detected 24 h after last medication. RESULTS : Compared with Sham group ,cerebral tissue water content ,EB content ,the number of apoptotic cells in cerebral cortex as well as the levels of IL- 1β and TNF-α in cerebral tissue were increased significantly(P<0.05),while the protein expression of SIRT 1, p-Akt,p-GSK3β and β-catenin in hippocampus were decreased significantly (P<0.05). Compared with CLP group ,cerebral tissue water content ,EB content ,the number of apoptotic cells in cerebral cortex as well as the levels of IL- 1β and TNF-α in cerebral tissue were decreased significantly in CLP+Dex group (P<0.05),while the protein expression of SIRT 1,p-Akt,p-GSK3β and β-catenin in hippocampus were increased significantly (P<0.05). Sirtinol could significantly reverse the above-mentioned cerebral protection and factor regulation effects of Dex (P<0.05). CONCLUSIONS :Dex can protect the cerebral tissue of sepsis model rats,which may play an anti-inflammatory and anti-apoptotic role by activating SIRT 1/Akt/GSK3β/β-catenin signaling pathway ,so as to reduce cerebral edema ,protect blood-brain barrier and reduce cerebral injury.
6.Seed-specific expression of heterologous gene DGAT1 increase soybean seed oil content and nutritional quality.

Enhancing soybean (Glycine max) oil production is crucial to meet the market demand of vegetable oil. Diacylglycerol acyltransferase (DGAT) catalyzes the final acylation reaction of triacylglycerol (TAG) synthesis, acting as one of the rate-limiting enzymes for oil biosynthesis in plant seeds. Here, a cDNA clone VgDGAT1A encoding the DGAT1 protein was isolated from the high oil plant Vernonia galamensis. VgDGAT1A was specifically overexpressed in soybean seeds, and several high-generation transgenic lines (T7) were obtained by continuous selection. qPCR analysis showed that VgDGAT1A was highly expressed in the mid-development stage (30-45 DAF) of the transgenic seeds. Accordingly, the DGAT enzyme activity in the transgenic seeds was increased by 7.8 folds in comparison with the wild-type controls. Seed oil and starch contents were, respectively, increased by 5.1% (Dry weight) and reduced by 2%-3% in the transgenic soybeans. Importantly, protein content was not significantly different between transgenic and control seeds. Seed weight and germination rate of the transgenic lines exhibited no negative effect. Fatty acid profiling demonstrated that antioxidant oleic acid (C18:1Δ9) content in the transgenic seed oil was elevated by 8.2% compared to the control, and correspondingly, easily-oxidized linoleic acid (C18:2Δ9,12) and linolenic acid (C18:3Δ9,12,15) were decreased by 6% and 2% respectively. Taken together, seed-specific overexpression of an exogenous VgDGAT1A gene can break the negative linkage of oil and protein contents in soybean seeds, indicating that engineering of this highly-active DGAT enzyme is an effective strategy to improve oil yield and nutritional value in oilseeds.
7.Analysis and expectation of cleaning and disinfection methods of laryngoscope blade

Laryngoscope is an important accessory equipment of trachea cannula. Since laryngoscope is easy to be infected with bacterial microorganism, so it can not be used until it is with highly effective disinfection. However, the bacterial bearing rate is 47.2% in laryngoscope blade after disinfection, which directly related to reuse laryngoscope blade and the situation of cleaning and disinfection. The purpose of this paper was to analyze and expect the cleaning and disinfection methods so as to provide the basis for management of cleaning and disinfection of laryngoscope blade in hospital.
8.Opinion TCM Treatment of Haze Induced Lung Injury

Objective] From the Chinese point of view of haze-induced lung injury pathogenesis, diagnosis and treatment, it provides theoretical support. [Method] To exogenous disease TCM theory, the nature of the haze of evils, a sense of evil ways and pathogenic characteristics, etc. based on the analysis of the haze of the etiology and pathogenesis of lung injury, the basic pathogenesis are analyzed and discussed. [Results] For haze pathogenic proposed basic principles should be taken to the prevention, prevention is greater than cure, the treatment should be Quxie, and to righten. Work using the method of elimination of pathogens through purgation and churesis.[Conclusion]For haze urgent treatment, prevention and treatment of traditional Chinese medicine for haze-induced lung injury has very important theoretical and practical value.
9.Analysis on clinical curative effect of compound polymyxin B ointment combined with basic fibroblast growth factor to treat the wound surface of deepⅡdegree burn of children

Objective To observe the clinical curative effect of compound polymyxin B ointment combined with basic fibroblast growth factor (bFGF) on wound surface of deep second-degree burn in children, and provide reference for the clinical treatment. Methods Eighty cases of children patients with deep second-degree burn were included in this study, who were admitted in Tianjin 4th Center Hospital from March 2015 to March 2016. The cases were divided into control group and combined treatment group, 40 cases for each group. Wherein, the control group was treated with bFGF, the combined treatment group was received compound polymyxin B ointment and bFGF. The occurrence rate of infection of burn wound surface, wound healing time and occurrence rate of scar after healing were compared between two groups. Results There were lower infection rates on day 7, 10, 15 and 20 after burn in combined treatment group than those in control group (P<0.05). The healing time was significantly shorter in combined treatment group than that in control group [(21.53±1.33) d vs. (25.76±1.50) d, t=13.345, P<0.01]. The scar occurrence was significantly lessin combined group than that of control group (U=5.077,P<0.05). Conclusion The compound polymyxin B ointment combined with bFGF show a certain effect of anti-infection, acceleration of wound surface healing and preventing the scar from generating for wound surface on deep second-degree burn in children, which having a significant clinical curative effect.
10.Literature Analysis of ADR Caused by Antigout Agents

OBJECTIVE:To investigate rules and characteristics of ADR induced by antigout agents,and to provide reference for clinical rational drug use. METHODS:A total of 146 literatures,167 putients of ADR induced by antigout agents,which were reported during Jan. 1979-Dec. 2014. RESULTS:Of 167 ADR cases,the ratio of male to female was 5.4:1,and ADR patients aged above 70 years occupied the highest proportion(34.14%);Most ADR were induced by allopurinol(137 cases)ADR induced by antigout agents mainly were dermal system injury(60.00%),systemic reaction(9.19%),urinary system injury(5.95%),etc. and the specific performance manifested as erythra (51 cases),dermatitis exfoliativa (38 cases),drug hypersensitivity syndrome (13 cases),bullous epidermal necrolysis type of drug eruption (11 cases),Stevens-Johnson syndrome (9 cases) and so on. New and severe ADR were 66 cases,13 cases of severe ADR were dead. CONCLUSIONS:ADR induced by antigout agents is common in clinical and should be treated promptly,patient's care also should be strengthen to ensure patient medication safety and improve clinical treatment.
