Nigella sativa L. seeds have been traditionally utilized in Chinese folk medicine for centuries to treat vitiligo. This study revealed that the ethanolic extract of Nigella sativa L. (HZC) enhances melanogenesis and mitigates oxidative stress-induced cellular senescence and dysfunction in melanocytes. In accordance with established protocols, the ethanol fraction from Nigella sativa L. seeds was extracted, concentrated, and lyophilized to evaluate its herbal effects via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assays, tyrosinase activity evaluation, measurement of cellular melanin contents, scratch assays, senescence-associated β-galactosidase (SA-β-gal) staining, enzyme-linked immunosorbent assay (ELISA), and Western blot analysis for expression profiling of experimentally relevant proteins. The results indicated that HZC significantly enhanced tyrosinase activity and melanin content while notably increasing the protein expression levels of Tyr, Mitf, and gp100 in B16F10 cells. Furthermore, HZC effectively mitigated oxidative stress-induced cellular senescence, improved melanocyte condition, and rectified various functional impairments associated with melanocyte dysfunction. These findings suggest that HZC increases melanin synthesis in melanocytes through the activation of the MAPK, PKA, and Wnt signaling pathways. In addition, HZC attenuates oxidative damage induced by H2O2 therapy by activating the nuclear factor E2-related factor 2-antioxidant response element (Nrf2-ARE) pathway and enhancing the activity of downstream antioxidant enzymes, thus preventing premature senescence and dysfunction in melanocytes.

Chinese Journal of Forensic Medicine 2024;39(6):751-755
This paper aimed to identify etomidate and its structural analogues in hair and urine by HPLC-MS/MS.Hair samples were extracted by ultrasonic grinding and urine samples were extracted by methanol. The samples were on ACQUITY UPLC? HSS T3 column with 10 mmol/L ammonium acetate and 0.5 ‰ formic acid-methanol as mobile phases for gradient elution.Multiple reaction monitoring mode was used for analysis. The separation and detection of structural analogues were realized, especially the isomers propoxate and isopropoxate. There was good linear relationship of the etomidate and its structural analogues in hair and urine, with R greater than 0.998.And the recovery, accuracy and precision all met the requirements. When applied to practical cases, etomidate, metomidate, isopropoxate and etomidate acid were detected simultaneously in the hair samples of 3 cases. And correspondingly,in the urine samples, high content of etomidate acid and low content of the precursor substance metomidate were detected in 2 cases, only etomidate acid was detected in 1case.Etomidate structural analogues have higher content of precursor substances and lower content of metabolite in hair samples, while the opposite was true in urine samples. We cannot reverse confirm the structure of the precursor substances through the detection of the common metabolite etomidate acid in the urine samples. Therefore, ingestion of etomidate structural analogues cannot be accurately assessed from urine samples alone.

Objective To compare the clinical effect and safety of peroral endoscopic myotomy (POEM) with surgical therapy in treatment of esophageal achalasia. Methods 78 patients diagnosed as esophageal achalasia from January 2012 to October 2014 were enrolled in the study and divided into POME group and Heller group. There were 42 patients in POEM group and 36 patients in Heller group. The clinical symptom remission rate, LES resting pressure, Eckardt scores, complication rate, length of hospital stay and the hospitalization expenses were analyzed between the two groups. Results The patients in POEM group and Heller group both got clinical remissions after the treatment. There was no statistical difference in the rate of complication occurrence, Eckardt scores and LES resting pressure between the two groups. Patients in POEM group had shorter operation time, hospital stay and less expenses compared with the Heller group. Conclusions Compared with Heller group, the POEM group has the similar curative effect in treatment of esophageal achalasia. The POEM as a minimally invasive surgery has the advantages of less pain and trauma, shorter hospital stay, well tolerated and low cost. Therefore, the POEM is worth to be popularized and applied in treating esophageal achalasia.
