1.UPLC-Q-TOF-MS Reveals Mechanisms of Modified Qing'e Formula in Delaying Skin Photoaging and Regulating Circadian Rhythm

ObjectiveTo reveal the active substances and mechanisms of modified Qing'e formula (MQEF) in delaying skin photoaging by ultra-performance liquid chromatography-quadrupole-time of flight mass spectrometry (UPLC-Q-TOF-MS),network pharmacology, and cell experiments. MethodsUPLC-Q-TOF-MS and a literature review were employed to analyze the transdermally absorbed components in mice after the topical application of MQEF. The potential targets of MQEF in treating skin photoaging were retrieved from databases.The compound-potential target network and protein-protein interaction network were constructed to screen the key components and core targets. A photoaging cell model was established by irradiating HaCaT cells with medium-wave ultraviolet B (UVB). The safe doses of bakuchiol (BAK) and salvianolic acid B (SAB) for treating HaCaT cells and the effects of BAK and SAB on the viability of cells exposed to UVB irradiation were determined by the cell counting kit-8 (CCK-8) method.The reactive oxygen species (ROS) fluorescent probe was used to measure the ROS production in the cells treated with BAK and SAB.The expression levels of genes related to oxidative stress,inflammation,collagen metabolism,and circadian rhythm clock were measured by Real-time PCR. ResultsA total of 24 transdermally absorbed components of MQEF were identified,which acted on 367 potential targets,and 417 targets related to skin photoaging were screened out,among which 47 common targets were predicted as the targets of MQEF in treating skin photoaging. MQEF exerted the anti-photoaging effect via key components such as BAK and SAB,which acted on core proteins such as serine/threonine kinase 1 (Akt1) and mitogen-activated protein kinase 3 (MAPK3) and intervened in core pathways such as the tumor necrosis factor (TNF) and phosphatidylinositol-3-kinase (PI3K)/protein kinase B (Akt) signaling pathways.Compared with the model group,the administration of BAK and SAB increased the survival rate of HaCaT cells (P<0.01),down-regulated the mRNA levels of cyclooxygenase-2 (COX-2),interleukin-6 (IL-6),tumor necrosis factor-α (TNF-α),matrix metalloproteinase-1 (MMP-1),and matrix metalloproteinase-9 (MMP-9) (P<0.01),and up-regulated the mRNA levels of heme oxygenase-1 (HO-1) and NAD(P)H quinone dehydrogenase 1 (NQO-1) (P<0.05,P<0.01) in photoaged HaCaT cells.In addition,it eliminated excess ROS production induced by UVB and up-regulated the mRNA levels of brain and muscle ARNT-like 1 (BMAL1) and circadian locomotor output cycles kaput (CLOCK) associated with circadian clock (P<0.05,P<0.01). ConclusionMQEF delays skin photoaging through the coordinated effects of various components,multiple targets,and diverse pathways.The key components BAK and SAB in MQEF exhibit anti-photoaging properties,which involve inhibiting oxidative stress,preventing collagen degradation,mitigating inflammation,and maintaining normal skin circadian rhythms by regulating clock gene expression.
2.Effect of temperature changes between neighboring days on mortality risk of respiratory diseases

Objective:
To investigate the impact of temperature changes between neighboring days (TCN) on the mortality risk of respiratory diseases, so as to provide the evidence for the study of deaths from respiratory diseases caused by climate change.
Methods:
The monitoring data of deaths from respiratory diseases in Zibo City from 2015 to 2019 were collected from Shandong Provincial Management Information System for Chronic Diseases and Cause of Death Surveillance. The meteorological and air pollutant data of the same period were collected from China Meteorological Data Website and ChinaHighAirPollutants dataset. The effect of TCN on the risk of deaths from respiratory diseases was examined using a generalized additive model combined with a distributed lag non-linear model, and subgroup analyses for gender and age were conducted. The disease burden attributed to TCN at different intervals was assessed by calculating attributable fraction.
Results:
Totally 11 767 deaths from respiratory diseases were reported in Zibo City from 2015 to 2019, including 6 648 males (56.50%) and 5 119 females (43.50%). There were 1 307 deaths aged <65 years (11.11%), and 10 460 deaths aged 65 years and older (88.89%). A monotonically increasing exposure-response relationship was observed between TCN and deaths from respiratory diseases in the general population, females, and the population aged 65 years and older. The 95th percentile of TCN (P95, 3.84 ℃) reached the peak at a cumulative lagged of day 11 (RR=2.063, 95%CI: 1.261-3.376). The results of subgroup analyses showed greater impacts on females and the population aged 65 years and older, with cumulative lagged effects peaking at day 12 (RR=3.119, 95%CI: 1.476-6.589) and day 11 (RR=2.107, 95%CI: 1.260-3.523). The results of attributional risk analysis showed that next-day warming might increase the attributable risk of deaths from respiratory diseases, and next-day cooling might decrease the attributable risk.
Conclusion
Next-day warming may increase the mortality risk of respiratory diseases, and has greater impacts on females and the population aged 65 years and older.
3.Research on the RIS-PACS environment operation and big data testing of artificial intelligence diagnostic model of bone age imaging

Objective To explore the feasibility and effectiveness of the artificial intelligence(AI)diagnostic model for bone age imaging in the radiology information system-picture archiving and communication system(RIS-PACS)environment operation.Methods The optimized bone age AI model was integrated into the RIS-PACS platform.The bone age imaging data of 88 038 patients aged 0-18 years old were automatically evaluated.The reference bone age was determined by the consensus of two experienced radi-ologists based on GP map,with an error of±1.0 year old.The success rate,accuracy,system compatibility,stability,and influen-cing factors of results were further analyzed.Results The time for bone age AI evaluation of each case did not exceed 3 seconds,and the success rate of automatic evaluation reached 100%.AI model of bone age and RIS-PACS in hospital could be well integrated.Accord-ing to the readings evaluated by pediatric radiologists based on GP maps,the accuracy rate was 93.05%for girls and 89.53%for boys,with a mean absolute error(MAE)of(0.42±0.54)years old for girls and(0.45±0.60)years old for boys,respectively.The AI model could run efficiency in the RIS-PACS,which significantly reduced the burden of radiologists.The factors that affect the accuracy of the model were image position,exposures,multiple images in a single sequence and hand deformity,etc.Conclusion The bone age imaging AI diagnostic model can be seamlessly embedded into RIS-PACS in hospital,achieving one click bone age imaging diagnosis.
4.Prognostic Value of ABAT mRNA Expression and ABAT Methylation Level in Bone Marrow of Patients with Myelodysplastic Syndrome

Objective To detect the expression level of 4-aminobutyrate aminotransferase(ABAT)in bone marrow of patients with myelodysplastic syndrome(MDS),and analyze its influence on clinicopathological features and prognosis of patients.Methods From January 2016 to March 2020,92 patients with MDS and 30 patients with acute myeloid leukemia(AML)from the First Affiliated Hospital of Xingtai Medical College were retrospectively collected.Meanwhile,30 patients with immunothrombocytopenia who did not develop MDS or other clonal diseases of the blood system during a 3-year follow-up were collected as control group.Real-time quantitative fluorescent PCR(qRT-PCR)was used to detect the relative expression level and methylation level of ABAT mRNA of all patients,and the relative expression level and methylation level of ABAT mRNA among different clinical characteristics of MDS patients were compared.Multivariate logistic regression analysis was used to analyze the risk factors affecting the adverse prognosis of MDS.The clinical value of detecting ABAT methylation level in predicting poor prognosis of MDS patients was analyzed by receiver operating characteristic(ROC)curve.Kaplan-Meier method was used to calculate the 3-year survival rate between groups with different ABAT mRNA relative expression levels and methylation levels,and log-rank test was used for their comparison.Results The expression level of ABAT mRNA in MDS group(0.42±0.08)was lower than that in control group(0.56±0.15)and AML group(0.52±0.10),while the methylation level of ABAT(32.51±5.32)was higher than that of AML group(26.21±4.58)and control group(10.25±4.31),and the differences were significant(t=4.251,4.562;10.415,8.326,all P<0.001).The methylation level of ABAT in high-risk patients(42.65±5.32)was higher than that in low-risk patients(25.63±4.16),intermediate-risk-1 patients(30.59±2.51)and intermediate-risk-2 patients(33.25±3.69)by IPSS risk grade,and the differences were significant(t=8.329,7.077,15.874,all P<0.001).Poor Karyotype analysis result[OR(95%CI):4.973(1.524~8.581),P=0.004],high IPSS risk grade[OR(95%CI):8.542(2.365~14.521),P<0.001]and ABAT hypermethylation level[OR(95%CI):6.178(1.589~13.021),P<0.001]were the risk factors affecting the poor prognosis of MDS.The cut-offvalue of ABAT methylation level to predict the poor prognosis of MDS were 30.54,and the area under the curve(AUC),the sensitivity and specificity were 0.92,0.874 and 0.851,respectively.The 3-year survival rate of the high ABAT methylation group(>30.54)was 66.67%,which was lower than that of the low ABAT methylation group(≤30.54)was 93.18%,with significant difference(Log-rank x2=9.814,P=0.002).Conclusion The ABAT methylation levels in MDS bone marrow increase,which is a risk factor affecting the poor prognosis of patients.ABAT basal level>30.54 is expected to become a factors predicting the poor prognosis of patients.
5.CT and MRI manifestations of soft tissue Kaposiform hemangioendothelioma in children

Objective To observe CT and MRI manifestations of soft tissue Kaposiform hemangioendothelioma(KHE)in children.Methods Data of 11 children with KHE confirmed by postoperative pathology were retrospectively analyzed,including 7 case underwent CT scanning,3 case underwent MR scanning and 1 case underwent both CT and MR scanning.The tumors originated from the skin with adjacent soft tissue invasion in 10 cases,while from pharyngeal soft tissue without skin involvement in 1 case.Nine cases were found with single and 2 cases were found with multiple lesions,with a total of 13 lesions,including 6 in the limbs,4 in the trunk and 3 in the head and neck,5 were focal type and 8 were diffuse type.CT and MRI manifestations of KHE were observed.Results Involvements of subcutaneous fat and deep fascia were noticed in all 8 cases(8/8,100%)who underwent CT scanning,while bone involvements were found in 3 cases(3/8,37.50%).The tumors present as equal density in 7 cases(7/8,87.50%)and equal-low density in 1 case(1/8,12.50%),with CT value of(30.25±14.32)HU,and no obvious venous stones or calcification was observed.Significant enhancement was observed in all 8 cases(8/8,100%)after administration of contrast agents,with CT value of(124.30±82.96)HU,and blood vessels around the lesions were noticed in 4 cases(4/8,50.00%).Among 4 cases who underwent MR examination,tumors invaded adjacent muscles in 3 cases(3/4,75/00%)and affected bones in 1 case(1/4,25.00%),presenting as uneven high signal on T2WI,high signal on diffusion weighted imaging(DWI)without significant decreased of apparent diffusion coefficient(ADC),with sustained significant enhancement in all 4 cases and empty blood vessel shadows in 3 cases.Conclusion CT and MRI manifestations of soft tissue KHE in children were characteristic,especially on contrast-enhanced scanning and DWI.
6.Effects of heat waves and cold spells on the incidence of hemorrhagic stroke

Objective:
To examine the impact of heat waves and cold spells on the incidence of hemorrhagic stroke, so as to provide insights into prevention of hemorrhagic stroke.
Methods:
Data pertaining to the incidence of hemorrhagic stroke in Zibo City from 2015 to 2019 were collected from Shandong Provincial Management Information System for Chronic Diseases and Cause of Death Surveillance, and the meteorological data during the period from 2015 to 2019 were captured from National Meteorological Information Center of China. The air quality index (AQI) was collected from the National Daily Report of Urban Air Quality in China. Heat wave was defined as the highest daily temperature that was no less than the 90th percentile (P90), P92.5, P95 and P97.5 of the highest daily temperature in the warm season for at least 2, 3 or 4 days, and cold spell was defined as the lowest daily temperature that was no more than the P10, P7.5, P5 and P2.5 of the lowest daily temperature in the cold season for at least 2, 3 or 4 days. The effect of heat waves and cold spells on the incidence of hemorrhagic stroke was evaluated using a generalized additive model and described with relative risk (RR) and its 95%CI.
Results:
A total of 8 844 case with first-onset hemorrhagic stroke were recorded in Zibo City from 2015 to 2019. The lowest daily temperature that was no more than P10, P7.5 or P5 of the lowest daily temperature in the cold season for at least two days, or that was no more than P10 or P7.5 of the lowest daily temperature for at least 3 days resulted a remarkably increased risk of hemorrhagic stroke (lowest RR=1.187, 95%CI: 1.031-1.366; highest RR=1.242, 95%CI: 1.042-1.480), and after adjusting the effect of daily mean temperature, the lowest daily temperature that was no more than P10 or P7.5 of the lowest daily temperature in the cold season for at least two days, or that was no more than P10 of the lowest daily temperature for at least 3 days resulted a remarkably increased risk of hemorrhagic stroke (lowest RR=1.236, 95%CI: 1.009-1.513; highest RR=1.274, 95%CI: 1.023-1.585). However, there was no significant association between heat waves and the risk of hemorrhagic stroke.
Conclusion
Cold spells may increase the risk of hemorrhagic stroke, while no significant association is examined between heat waves and the risk of hemorrhagic stroke.
7.Clinical study on effect of Shumu Peitu herb-partitioned moxibustion on diarrhea-predominant irritable bowel syndrome and negative emotions

Objective:To explore the intervention effect of Shumu Peitu herb-partitioned moxibustion on clinical signs and symptoms and negative emotions of diarrhea-predominant irritable bowel syndrome (IBS-D) patients with liver-stagnation and spleen-deficiency pattern.Methods:A total of 72 patients with IBS-D of liver-stagnation and spleen-deficiency pattern treated in the Department of Gastroenterology of Nanjing Vniversity of Chinese Medicine from September 2021 to June 2022 were selected for randomized controlled trial. The patients were randomly divided into the observation group (2 cases dropped off, 34 cases in total) and control group (1 case dropped off, 35 cases in total) by random number table method. The patients in control group were treated with Tongxieyaofang (TXYF). The patients in observation group were treated with oral administration of TXYF and Shumu Peitu herb-partitioned moxibustion, and both groups were treated for 4 weeks. The clinical efficacy, Traditional Chinese Medicine (TCM) syndrome integral, IBS Quality of Life Questionnaire (IBS-QOL), IBS Symptom Severity Scale (IBS-SSS), Bristol Stool Form Scale and Hospital Anxiety and Depression Scale (HADS) were compared before and after treatment.Results:After treatment, the total effective rate of the observation group was 94.12%(32/34), which was higher than the 71.43%(25/35) in the control group, the difference was significant ( χ2 = 6.18, P<0.05). After treatment, the TCM syndrome integral in the observation group was (7.62 ± 4.08), which was lower than the (9.89 ± 4.71) in the control group, the difference was significant ( t = 2.14, P<0.05). After treatment of 3 days, the scores of quality of life in the five dimensions of dysthymia, behavior disorder, health worry, avoidance of eating and social function in the observation group were (82.44 ± 11.46), (80.25 ± 11.67), (76.23 ± 12.67), (59.80 ± 15.14) and (79.23 ± 11.59) points, which were different with the (73.57 ± 12.39), (72.35 ± 15.48), (69.76 ± 13.11), (50.00 ± 16.17) and (73.04 ± 13.11) points in the control group, the difference were significant ( t values were -3.09 - -2.08, all P<0.05). Three days after treatment, the score of IBS-SSS and Bristol fecal character in the observation group were (118.24 ± 40.64) and (5.09 ± 0.62) points, which were lower than the (146.86 ± 60.09) and (5.51 ± 0.66) points in the control group, the difference were significant ( t = 2.31 and 2.76, both P<0.05). After treatment, the score of HADS-A and HADS-D in the observation group were (6.26 ± 1.75) and (5.29 ± 1.47), which were different with the (7.26 ± 2.19) and (6.17 ± 2.11) in the control group, the difference were significant ( t = 2.08 and 2.00, both P<0.05). Conclusions:Shumu Peitu herb-partitioned moxibustion can effectively improve IBS-D patients with liver-stagnation and spleen-deficiency pattern, relieve clinical symptoms, reduce negative emotions, and improve quality of life.
8.Group risk perception and selfefficacy with individual prevention/control behaviors among college students during the COVID 19 pandemic

Objective:
To explore the relationship between group risk perception, self efficacy and attitudes among college students, and its associations with individual prevention and control behaviors regarding COVID 19 pandemic.
Methods:
From October to November 2021, data were collected among 4 092 undergraduate students from eight colleges and universities in Anhui Province,using a multi stage stratified cluster sampling method. A t test and analysis of variance were performed for comparison between groups, while multiple linear regression was employed to investigate group risk perception, self efficacy and attitudes among college students in terms of individual COVID 19 prevention and control measures.
Results:
The total scores of group emotional risk perception (ARP), cognitive risk perception (CRP), distance perception (DIP) and self efficacy were (19.72±4.01)(14.56±3.64)(18.04±5.33) and (27.27±6.74), respectively. The total score of attitudes and beliefs was (26.15±4.54), while individual prevention and control behavior was (25.67±4.45). Emotional risk perception ( β=0.16 ), cognitive risk perception ( β=0.08 ), attitudes ( β=0.23 ) and self efficacy ( β=0.13 ) were positively correlated with individual prevention and control behaviors of college students( P <0.05). However, distance risk perception had no significant effect ( P >0.05).
Conclusion
Risk perception, self efficacy and attitude of college students are correlated with individual COVID 19 prevention and control behaviors. Therefore, colleges and universities should strengthen general self efficacy and attitude,and improve risk awareness towards COVID 19 among college students, and promote the effective prevention and control of campus epidemic.
9.Effect of forgetting curve based self-management on cognitive function, daily living ability and treatment efficacy of patients with mild cognitive impairment

ObjectiveTo discuss the effect of forgetting curve based self-management on cognitive function, daily living ability and treatment efficacy of patients with mild cognitive impairment (MCI). MethodsSimple random sampling method was adopted to enroll 162 MCI patients who met the diagnostic criteria of "Expert Consensus on the Prevention and Treatment of Cognitive Impairment in China" in Nanchong Physical and Mental Hospital and Gaoping Ledele Seniors-Oriented Apartment from April 2020 to June 2021. The selected individuals were classified into study group and control group according to random number table methods, each with 81 cases. Both groups received routine intervention, based on this, study group received the forgetting curve based self-management. The interventions lasted for 3 months in two groups, and patients were assessed using Montreal Cognitive Assessment Scale (MoCA) and Activity of Daily Living Scale (ADL) at the baseline and end of interventions. Then the clinical efficacy was compared between groups. ResultsAfter intervention, an increase was found in MoCA and ADL scores in both groups (tcontrol group=25.004, 12.503, tstudy group=48.211, 24.949, P<0.01), and post-intervention MoCA and ADL scores in study group were higher than those in control group (t=28.527, 9.433, P<0.01). The overall efficacy rate was 86.42% in control group, which was lower than 96.30% in study group, with statistical difference (χ²=5.004, P<0.05). ConclusionForgetting curve based self-management may ameliorate the cognitive function and daily living ability in MCI patients, thus improving the treatment efficacy.
10.Relationships among psychological stress, achievement motivation and psychological capital in college students

Objective:
This study explored the relationships among psychological stress, achievement motivation and psychological capital in college students to provide a reference for improving the level of psychological capital in college students.
Methods:
A multi stage stratified cluster sampling method was used to select 9 940 college students from ten universities in Anhui Province. The achievement motivation scale (AMS), psychological stress scale (SRQ-20) and psychological capital scale (PPQ) were applied. The moderating effect of the questionnaire was analyzed with χ 2 tests, Spearman rank correlation and stratified regression.
Results:
Statistical differences were found in psychological stress according to major, whether students leader, family economic status and whether students had left behind experience ( χ 2=15.50, 10.25, 28.61, 25.55, P <0.05). The rank correlation results indicated that psychological stress was negatively correlated with the pursuit of success ( r =-0.27) and four dimensions of self efficacy,optimism,hope and resilence in psychological capital ( r =-0.43, -0.41,-0.36,-0.45)( P <0.05), and was positively correlated with the avoidance of failure ( r =0.25, P <0.05). The stratified regression model indicated that psychological stress in the dimensions of college students achievement motivation (pursuit of success: β =0.02, Δ R 2=0.01, P <0.01; failure avoidance: β = 0.03 , Δ R 2=0.01, P <0.01) played a moderating role in the relationship between psychological capital and psychological capital.
Conclusion
Being female, senior students, low household economic status, and left behind experience are associated with more psychological stress among college students. Psychological stress is correlated with achievement motivation and psychological capital, and has a moderating effect on the relationship between achievement motivation and psychological capital.
