1.Modified Xiaoyaosan Alleviates Neuronal Dysfunction in Rat Model of Post-myocardial Infarction Depression by Regulating Mitochondrial Quality Control Through Drp1/PINK1/Parkin Signaling Pathway

ObjectiveTo investigate the effects of modified Xiaoyaosan (JJXYS) on behavioral abnormalities and hippocampal mitochondrial quality control (MQC) in the rat model of post-myocardial infarction depression (PMD) and preliminarily explore its potential mechanism. MethodsA rat model of PMD was established by left anterior descending coronary artery ligation combined with chronic unpredictable mild stress (CUMS). Rats were randomized into a control group, a model group, a fluoxetine (FLX, 10 mg·kg-1) group, and low-, medium-, and high-dose JJXYS (JJXYS-L/M/H, 1.12, 2.24, 4.48 g·kg-1, respectively) groups. Depressive-like behaviors were evaluated by body weight monitoring, sucrose preference test, open field test, and forced swimming test. Hematoxylin-eosin staining and Nissl staining were used to observe hippocampal histomorphology and neuronal changes. Enzyme-linked immunosorbent assay was conducted to determine the serum levels of 5-hydroxytryptamine (5-HT), dopamine (DA), interleukin-1 beta (IL-1β), interleukin-6 (IL-6), and tumor necrosis factor-alpha (TNF-α). The mRNA levels of MQC-related genes including peroxisome proliferator-activated receptor-gamma coactivator-1 alpha (PGC-1α), nuclear respiratory factor 1 (Nrf1), and transcription factor A, mitochondrial (TFAM) in the hippocampal tissue were measured by real-time PCR. The expression of proteins related to the dynamin-related protein 1 (Drp1)/PTEN-induced putative kinase 1 (PINK1)/Parkin signaling pathway was determined by Western blot. ResultsCompared with the control group, the model group showed restricted body weight gain, aggravated depressive-like behaviors, declined serum 5-HT and DA levels, evident hippocampal neuronal damage and reduced Nissl bodies, as well as downregulated expression of MQC-related genes and proteins (P<0.05). Compared with the model group, both FLX and JJXYS alleviated the above changes to varying degrees. Moreover, the JJXYS-M and JJXYS-H groups showed more pronounced effects, improving behavioral performance, restoring 5-HT and DA levels, alleviating hippocampal pathological injury, and upregulating the expression of PGC-1α/Nrf1/TFAM mRNA and Drp1/PINK1/Parkin signaling pathway-related proteins (P<0.05). ConclusionJJXYS can significantly alleviate depressive-like behaviors and neurotransmitter imbalance in the rat model of PMD by regulating hippocampal MQC and upregulating the Drp1/PINK1/Parkin-related pathway. This study provides experimental evidence for the intervention of PMD with JJXYS.
2.Serum levels and significance of Kisspeptin and LH in girls with central precocious puberty

Objective To explore the expression significance of serum Kisspeptin and luteinizing hormone(LH)in girls with central precocious puberty(CPP).Methods A total of 98 CPP girls were included as the study group,and 91 healthy girls during the same period were used as the control group.Under the condition of no intervention,CPP girls were followed up for 6 months and assigned into the fast progressive CPP(RP-CPP)group(n=46)and the slowly progressive CPP(SP-CPP)group(n=52)based on their developmental status during the follow-up period.ELISA was applied to detect serum Kisspeptin level.Chemiluminescence method was applied to detect serum LH and follicle stimulating hormone(FSH)levels.The correlation of serum Kisspeptin,LH baseline(B-LH)and growth development and imaging indicators was analyzed.Receiver operating characteristic curve(ROC)was applied to analyze the value of serum Kisspeptin and LH in predicting RP-CPP.Results Compared with the control group,body mass index(BMI),serum levels of Kisspeptin,B-LH,and FSH baseline(B-FSH)were significantly higher in the study group(P<0.05).Compared with the SP-CPP group,BMI,bone age index(BAI),serum Kisspepti and B-LH levels were higher in the RP-CPP group,and ovarian volume was larger(P<0.05).Correlation analysis showed that serum Kisspeptin level was positively correlated with BMI,BAI,ovarian and uterine volume of CPP girls(P<0.05),and serum B-LH level was positively correlated with ovarian and uterine volume of CPP girls(r=0.218,0.381,0.300,P<0.05).ROC curve showed that the sensitivity and specificity of Kisspeptin combined with B-LH in predicting RP-CPP were 95.65%and 82.69%,respectively.Conclusion Serum Kisspeptin and B-LH levels are higher in CPP girls,and the combined of the two has a high predictive value for RP-CPP,which can provide a certain reference for clinical diagnosis and treatment.

4.Metabolic profiles of serum lysophosphatidylcholine and amino acids in rats with nonalcoholic fatty liver disease

Objective To use metabolomics method to study the metabolic profiles of amino acids and lysophosphatidylcholine(LPC)in the serum of rats with nonalcoholic fatty liver disease(NAFLD),to identify biomarkers for NAFLD,and to speculate on the possible mechanism responsible for its occurrence.Methods NAFLD rats were prepared by feeding a high-fat diet and intraperitoneal injection of carbon tetrachloride.Levels of 15 LPCs and 18 amino acids in the serum were determined in control and NAFLD rats by liquid chromatography-mass spectrometry.Changes in serum LPC and amino acid metabolic profiles in NAFLD rats were analyzed by principal component analysis and orthogonal partial least squares discriminant analysis.Correlations between biomarkers and NAFLD were analyzed by Pearson's correlation analysis.Results The metabolic profiles of serum LPC and amino acids differed significantly between the NAFLD group and the control group and were completely distinct.LPC(20∶1),arginine,and glutamic acid had significant contributions to NAFLD and were identified as biomarkers.Furthermore,LPC(20∶1)and arginine were significantly correlated with serum biochemical indicators such as aspartate transaminase,alanine transaminase,low-density lipoprotein,and total bilirubin.Conclusions The metabolic profiles of serum LPC and amino acids may be closely related to NALFD.
5.Establishment and application of a simulation system for auricular reconstruction with autogenous costal cartilage

Objective:To design and develop a digital auricle reconstruction simulation training system and to evaluate the application effect of the system.Methods:Based on 3D reconstruction and 3D printing technology, a new auricle reconstruction simulation training system was designed and developed from September 2019 to December 2021. Thirty-six basic volunteers (including 15 males and 21 females, aged from 22 to 27 years, with mean 23.1 years) without auricle reconstruction (surgical clinical standardized training students) were selected and divided into two groups (experimental group and control group) for clinical teaching experiment, and the teaching effect of simulation system was preliminarily evaluated.Results:Theoretical test scores before and after the intervention in the experimental group were 8.00 (4.75, 11.25) compared to 16.50 (13.00, 19.25) , P<0.001; design test scores were 2.00 (0, 4.25) compared to 7.00 (6.00, 10.00), P<0.001. In the control group, theoretical test scores before and after the intervention were 5.50 (3.75, 12.25) compared to 11.00 (9.50, 16.25), P<0.001; design test scores were 1.00 (1.00, 2.25) compared to 5.00 (3.75, 6.00), P<0.001. The improvement in the experimental group compared to the control group was significantly more pronounced with a statistically significant difference ( P<0.001). Conclusions:This system can assist teachers in teaching more systematically in auricle reconstruction training with autologous costal cartilage, and the teaching effect is better.
6.Comparative Experiment on Physical Properties of Red Blood Cells in Newborn Infants and Elderly People

Objective By comparing the physical properties (cell area, volume and elastic modulus) of red blood cells (RBCs) between newborn infants and the elderly over 80 years old, and correlation with the physiological and biochemical parameters such as total cholesterol and glycosylated hemoglobin, the effects of different ages and biochemical parameters on RBC physical properties were analyzed. Methods The mcropipette aspiration was used to measure the surface area, volume and elastic modulus of erythrocytes in newborn infants and the elderly over 80 years old, and the data were analyzed by statistical distribution analysis, correlation analysis and regression analysis. Results The mean values of RBC volume, surface area and elastic modulus in the elderly over 80 years old were smaller than those in newborn infants, and the mean values of RBC mechanical parameters in the same age group were not significantly different. The erythrocytes geometric parameter distribution of newborn infants was more concentrated than that of the elderly, while the elastic modulus distribution of newborn infants was more dispersed than that of the elderly. The mechanical properties of RBCs in newborn infants were highly correlated with the total cholesterol and gestational week; the mechanical properties of RBCs in the elderly were highly correlated with diastolic blood pressure and glycated hemoglobin. Conclusions There are significant differences in physical properties of RBCs between newborn infants and the elderly over 80 years old, and the biochemical parameters that affect physical properties of RBCs at different ages are also different.
7.The association between myopic refraction and near work among children and adolescents based on genetic risk score

Objective:To explore the relationship between myopic refraction and near work in children and adolescents with different genetic risks.Methods:From September to December 2016, Nankai District and Hongqiao District of Tianjin were taken as the study sites. Using the method of stratified cluster random sampling, 533 children and adolescents aged 6-14 years from one primary school and one junior middle school in each of the two districts were included as the study subjects. Refraction measurements by an auto-kerato-refractor and questionnaire survey about near work were conducted. 11 single nucleotide polymorphisms in the selected myopia susceptibility genes were detected, and the genetic risk of each individual was scored. After grouping by genetic risk score, the relationship between myopia and near work was analyzed by the multivariate logistic regression, and the relationship between near work and refraction was analyzed by the multivariate linear regression.Results:The age of 553 subjects was (9.8±2.5) years, including 295 boys (53.3%). The overall detection rate of myopia was 62.0%. The spherical equivalent refraction (SER) was (-1.30±1.85) D. The results of the multivariate logistic regression showed that in the low risk group of GRS, compared with those with continuous near work time less than half an hour, those with continuous near work time no less than half an hour had a higher risk of myopia [ OR ( 95%CI) = 2.64 (1.07, 6.52)]. In the moderate risk group of GRS, the risk of myopia increased with the increase of daily computer use [ OR ( 95%CI) = 2.14 (1.03, 4.77)]. In the high risk group of GRS, the risk of myopia increased with the increase of the total daily reading and writing time [ OR ( 95%CI) = 1.27 (1.01, 1.59)]. The results of the multivariate linear regression showed that in the low risk group of GRS, with increase of 1 hour in the total daily reading and writing time and mobile phone time, the SER decreased by 0.18 D (95% CI:-0.30, -0.07) and 0.95 D (95% CI:-1.51, -0.39), respectively. In the moderate risk group of GRS, with increase of 1 hour in the total daily reading and writing time and computer use time, the SER decreased by 0.25 D (95% CI:-0.31, -0.18) and 0.57 D (95% CI:-0.97, -0.18), respectively. In the high risk group of GRS, with increase of 1 hour in the daily total reading and writing time, the SER decreased by 0.33 D (95% CI:-0.43, -0.22). Conclusion:Continuous near work time no less than half an hour, daily computer use time, the total daily reading and writing time, and daily mobile phone use time were associated with myopic refraction in children and adolescents.

9. Interference research of umbilical cord mesenchymal stem cells on the pulmonary fibrosis in silicosis rats

Objective:
To explore the effect of the umbilical cord mesenchymal stem cells(UC-MSCs) on the pulmonary fibrosis in silicosis rats.
Methods:
SPF male Sprague Dawley rats were randomly divided into control group, silica model group and UC-MSCs treatment group with 12 rats each group. SiO2 intra-tracheal injection(0.5 ml of 50 mg/ml/rat) were applied to silica model group and UC-MSCs treatment groups. After that UC-MSCs treatment group received 1 ml UC-MSCs suspension (3×106 cells/ml) by tail vein injection on the 29th, 36th, 43th and 50th day after exposure to the first silica suspension. On the 60th and 75th day after exposure to silica suspension, all animals were examed for pulmonary CT. Then the rats were euthanized on 75th day after the first exposure to silica.Lung's histopathological examination of the rats from all the groups were carried out. The content of hydroxyproline in lungs, TGF-β1 and IL-6 in serum were examined.
Results:
The lung's histopathological examination showed no obvious inflammatory cell and no fibrosis in the lung tissue of the control group, there were a lot of inflammatory cell aggregation and collagen fiber deposition in silica model group, while in the UC-MSCs intervention group and treatment group, there were less inflammatory cells and collagen fiber. The rats from silica model groups had higher HYP, TGF-β1 and IL-6 than the rats from UC-MSCs treatment group and control group. Lung fields of rats in the control group were clear and no obvious high-density shadow. Different-sized granular high-density shadows or reticular fibrous shadows were found diffusely distributed in the lungs of the rats in silica model group. Lung field of rats in UC-MSCs intervention group and treatment group were less high density shadows, and more clear.
Conclusion
UC-MSCs can alleviate the pulmonary fibrosis in silica model rats through regulating the secretion of some fibrosis related cytokines.
