1.Zuogui Wan Improve Ovarian Inflammatory Microenvironment and Stemness of Ovarian Germline Stem Cells in Ovarian Aging via cGAS/STING Signaling Pathway

ObjectiveTo investigate the mechanism of Zuogui Wan (ZGW) in improving ovarian inflammatory microenvironment and stemness of ovarian germline stem cells (OSCs) for treating ovarian aging via the cyclic guanosine monophosphate/adenosine monophosphate synthase (cGAS)/stimulator of interferon genes (STING) signaling pathway. MethodsForty C57BL/6 female mice were randomly divided into a blank group, a model group, a low-dose ZGW group (2.7 g·kg-1), a high-dose ZGW group (5.4 g·kg-1), and an estradiol valerate group (0.15 mg·kg-1), with 8 mice in each group. Except the blank group, all other groups received a single intraperitoneal injection of cyclophosphamide at 120 mg·kg-1 to establish an ovarian aging mouse model. After successful modeling, each group was continuously administered for 4 weeks, once daily. The physiological status of the mice was observed, and the ovarian index was calculated. The estrus cycle of the mice was monitored. Hematoxylin-eosin (HE) staining was used to observe pathological changes in ovarian tissue. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum sex hormone levels. Serum inflammatory factors interleukin-1β (IL-1β), tumor necrosis factor-α (TNF-α), and mouse interleukin-6 (IL-6) levels were detected using kits. Western blot was used to detect the protein expression of ovarian cGAS, STING, p-STING, TANK-binding kinase 1 (TBK1), p-TBK1, interferon-induced transmembrane protein 3 (Fragilis), and Vasa homolog protein (MVH). Quantitative real-time polymerase chain reaction (Real-time PCR) was used to detect the mRNA expression of inflammatory factors in ovarian tissue. Immunofluorescence double labeling was performed to locate OSCs in ovarian tissues, and fluorescence intensities of OSCs markers MVH and octamer binding transcription factor 4 (Oct4) were calculated. ResultsCompared with the blank group, the model group showed reduced body weight, ovarian wet weight, and ovarian index (P<0.01) and a disordered estrus cycle (P<0.01). In addition, the levels of serum follicle-stimulating hormone (FSH), TNF-α, IL-6, and IL-1β were increased (P<0.01), while anti-Müllerian hormone (AMH) and estradiol (E2) levels were decreased (P<0.01). The protein expression of cGAS, p-STING/STING, and p-TBK1/TBK1 in ovarian tissue was increased (P<0.05, P<0.01), while that of OSCs stemness factors MVH and Fragilis was reduced (P<0.01). Immunofluorescence indicated a reduction in MVH and Oct4 expression in OSCs (P<0.01). The mRNA expression of inflammatory factors TNF-α, IL-6, and IL-1β in ovarian tissue was increased (P<0.05, P<0.01). Compared with the model group, the treatment groups exhibited improved body weight, ovarian wet weight, and ovarian index (P<0.05) and a reduced rate of estrus cycle disorder (P<0.05, P<0.01). The levels of serum FSH, TNF-α, IL-6, and IL-1β were decreased (P<0.05, P<0.01), while AMH and E2 levels were increased (P<0.01). The protein expression levels of cGAS, p-STING/STING, and p-TBK1/TBK1 in ovarian tissue were decreased (P<0.05), while the protein expression of MVH and Fragilis was increased (P<0.05), and the fluorescence intensities of MVH and Oct4 were increased (P<0.05, P<0.01). The mRNA expression of inflammatory factors in ovarian tissue was decreased (P<0.05). ConclusionZGW alleviate ovarian inflammatory response, regulate ovarian microenvironment homeostasis, and maintain stemness of OSCs in ovarian aging mice probably by modulating the cGAS-STING signaling pathway, thereby improving ovarian function and delaying ovarian aging.

[Objective]To explore and analyze the thoughts of medical and Yiology of the book Waijing Weiyan written by CHEN Shiduo,a medical doctor from Zhejiang Province in the late Ming and early Qing dynasties.[Methods]By means of literature research method,this paper sorted out and screened the articles related to the thoughts of medicine and Yiology in the book and studied them,discussed and analysed CHEN Shiduo's thoughts of medical and Yiology on"the trigrams of the Zhouyi""The River Vehicle Theory""The Nine Palaces and Eight Winds""Heavenly Stems and Earthly Branches",etc.,discussing to explore the connotation of the thoughts of medicine and Yiology in Waijing Weiyan.[Results]The chapters of Waijing Weiyan,which mainly embody the ideas of medical thinking,include"The Mingmen is the Governor of Meridians""Inability of Water to Restrict Fire""Nomenclature of the Eight King of Wind""Relief of Yang Disease and Relief of Yin Disease",etc.The thoughts of medical and Yiology contained therein is mainly reflected in five aspects,namely,expending the imagery of"Kan"and"Li"to explain the doctrine of Mingmen;using the"Ji-Ji Hexagram"interpreting the water and fire of the human body;innovating the River Vehicle Theory in Taoism to explain the pathway lifting and lowering of Qi;inheriting the thoughts of Nine Palaces and Eight Winds,and rationally treating the eight winds occupancy;proposing the twelve"times of exuberance of Qi"to discuss the prognosis of diseases by day and night.[Conclusion]Waijing Weiyan shows the main contents of CHEN Shiduo's thoughts of medicine and Yiology,reflecting his academic characteristics such as emphasizing the coordination of water and fire and the elevation of Qi,advocating the compatibility of thoughts of medicine and Yiology with the actual medical practice,and being good at explaining the medical theories with the tools of medical and Yiology.The study of the theory of medicine and Yiology in Waijing Weiyan helps to understand and grasp the essence of CHEN Shiduo's medical thoughts,and provides ideas for the study and development of the academic thoughts of the Zhejiang school of traditional Chinese medicine.
4.Research progress on the identification and intervention of non-suicidal self injury behavior among adolescents using artificial intelligence

Abstract
The global prevalence of non-suicidal self-harm (NSSI) among adolescents is becoming increasingly severe. Traditional mental health services are struggling to meet the growing social demand due to limited resource allocation and service accessibility. The rapid development of artificial intelligence (AI) technology provides a new technological path. The article systematically reviews the research progress of AI technology in adolescent NSSI, demonstrating key technologies such as machine learning, natural language processing, and deep learning in predicting NSSI risk prediction, emotion recognition and online intervention for adolescents. However, challenges remain, including algorithm bias, data privacy protection, model interpretability and ethical decision making. Future research should focus on multi disciplinary collaborative cooperation based on artificial intelligence to build a safe, effective and sustainable digital psychological intervention system, so as to provide innovative strategies and technical support for the early warning and intervention of NSSI behavior in adolescents.
5.Effect of adequate vessel preparation on 12-month primary patency rate of drug-eluting stents in patients with lower extremity arteriosclerosis obliterans

Objective To investigate the impact of adequate lumen preparation on the 12-month primary patency rate of drug-eluting stents(DES)in patients with lower extremity arteriosclerosis obliterans(ASO)and to identify risk and protective factors for in-stent restenosis.Methods A retrospective cohort study was conducted on 141 patients who underwent DES implantation for ASO at the Department of Vascular Surgery of the First Affiliated Hospital of Chongqing Medical University between April 2022 and April 2024.According to post-percutaneous transluminal angioplasty(PTA)dissection grade[severe dissection is defined as type C or higher based on National Heart,Lung,and Blood Institute(NHLBI)criteria]and residual stenosis rate(severe stenosis is defined as≥30%),the patients were categorized into an adequate preparation group(no severe dissection or severe residual stenosis;n=59)and an inadequate preparation group(n=82).DES primary patency at 12 months was assessed by color Doppler ultrasonography or angiography,defined as a peak systolic velocity ratio≤2.4 on duplex ultrasound or angiographic stenosis<50%.Kaplan-Meier analysis was used to compare intergroup differences in DES patency rate and freedom from clinically driven target lesion revascularization(CD-TLR).Multivariate logistic regression analysis was employed to identify risk and protective factors for DES restenosis and to evaluate the risk contributions of post-PTA dissection and residual stenosis to DES restenosis.Results Post-PTA,vascular dissection occurred in 94.3%(133/141)of treated limbs,among which 42.1%(56/133)were severe dissections,and severe residual stenosis was present in 44.0%(62/141)of limbs.Kaplan-Meier analysis revealed that the 12-month primary patency rate was significantly higher in the adequate preparation group than the inadequate group(HR=0.322,95%CI:0.132~0.789,P=0.013),while no significant difference was found in the rate of freedom from CD-TLR(HR=0.608,95%CI:0.187~1.937,P=0.407).Multivariate logistic regression analysis identified adequate lumen preparation(OR=0.228,95%CI:0.069~0.747,P=0.015)and Rutherford category 2~3(OR=0.205,95%CI:0.058~0.725,P=0.014)as independent protective factors against DES restenosis.The patency rate in the subgroup with coexisting severe dissection and residual stenosis was significantly lower than that in the uncomplicated group(55.6%vs 89.8%,P<0.001),with a 7.067-fold increased risk of re-stenosis(95%CI:2.424~20.602,P<0.001).Conclusion Adequate lumen preparation significantly improves the 12-month primary patency rate of DES.It,along with Rutherford category 2~3,serves as an independent protective factor against in-stent restenosis.Concurrent severe dissection and significant residual stenosis should be actively avoided during the procedure.
6.A study on the coercive experience of involuntarily hospitalized adolescents with mental disorders

ObjectiveTo explore the coercive experience of involuntarily hospitalized adolescents with mental disorders during the admission process and hospitalization, providing references for formulating targeted nursing interventions. MethodsSemi-structured interviews were conducted with 15 involuntarily hospitalized adolescents with mental disorders selected from October to December 2023, and the themes were summarized and extracted by content analysis. ResultsA total of 3 themes and 10 sub-themes were extracted, which were used to elaborate the essential contents, causes, and improvement methods of coercive experience. These encompassed the multi-dimensional content of coercive experience (complex emotional experience, different physical sensations, and contradictory cognitive evaluation), the multi-faceted causes of coercive experience (insufficient personal preparation, inadequate parental communication, and strict medical management system), as well as the phased improvement of coercive experience (adequate communication before hospitalization, patient notification before coercive intervention, respecting for demands during coercive intervention, and comforting explanation after coercive intervention). ConclusionThe essential content of the coercive experience of involuntarily hospitalized adolescents with mental disorders is complex and has various causes, which require cooperation from multiple parties to improve. Therefore, parents should respect the expression of their children’s self-will, and medical staff should respect patients’ autonomy, establishing a protection-constrained doctor-patient relationship model and collaborating to reduce the use of coercive interventions, to improve the overall medical satisfaction of adolescents with mental disorders.
7.Development of the Chinese Premature Ovarian Insufficiency Quality of Life Scale

Objective To develop a quality of life(QoL)scale for Premature Ovarian Insufficiency(POI)with high reliability and validity in accordance with China's national conditions.Methods The theoretical framework of the scale was constructed under the guidance of traditional Chinese medicine theory with reference to the development method of the QoL scale.Literature analysis method,medical record review method,patient interview method,Delphi expert consultation method and clinical investigation were used to establish,improve and screen the scale items to form the final scale,and the characteristics and differentiation in terms of reliability and validity and feasibility of the final scale were evaluated.Results After screening the entries by the Delphi method and clinical surveys,the Chinese Premature Ovarian Insufficiency Quality of Life Scale was formed,which included three domains of physiology,psychology,and society,and eleven aspects such as menstruation,fertility,and physical symptoms,with 30 entries and two overall evaluations.The Cronbach's coefficient of this scale was 0.910,the fold-half coefficient was 0.956,and the re-test correlation coefficient was 0.868,which showed good internal consistency,fold-half and re-test reliabilities;the scale extracted 9 factors with characteristic root>1,and the cumulative variance contribution rate was 71.063%,the comparative fit index was 0.959,and the goodness-of-fit index was 0.919,and the degree of correlation was tested has showed that the content validity,structural validity and calibration correlation validity were good;the difference in the total scores of the scale between patients with POI and the healthy population was statistically significant,and the scale had a good degree of differentiation.Conclusion As confirmed by this study,the Chinese Premature Ovarian Insufficiency Quality of Life Scale has good reliability,validity,differentiation and feasibility,and can be used as a tool for evaluating the quality of life of POI patients in China as well as enriching the efficacy evaluation system of TCM in treating POI,and it is suitable for clinical popularization and application.

Objective To evaluate the practical application of the Media Fill Test(MFT)in assessing aseptic compounding competency of personnel in Pharmacy Intravenous Admixture Services(PIVAS).Methods A multicenter controlled study was conducted across six tertiary hospitals(center ①-⑥)in China.Participants were divided into an inexperienced group(Group A,n=118)and an experienced group(Group B,n=118),each performing five MFT operations.Positive controls validated medium efficacy.Contamination rates and pass rates were analyzed using chi-square and Fisher's exact tests.Results Valid samples included 584 for Group A and 588 for Group B.The sample pass rate was 66.78%(390/584)in Group A and 91.67%(539/588)in Group B,while personnel pass rates were 46.15%(54/117)and 80.51%(95/118),respectively,with significant intergroup differences(both P<0.01).All centers except Center ⑥ showed significantly higher pass rates in Group B(all P<0.05).Conclusion MFT effectively differentiates technical proficiency levels and is suitable for training evaluation of novice PIVAS staff.
