1.Mechanistic study on ITGA6 regulation of abdominal wall endometriosis via the PI3K/AKT signaling pathway

ObjectiveTo investigate the differential expression of integrin alpha-6(ITGA6) in abdominal wall endometriosis (AWE) tissues and its molecular mechanisms in regulating AWE. Methods36 AWE lesions were designated as the experimental group, while 36 cases of normal endometrial tissues served as the controls. Differential expression of ITGA6 between the two groups was assessed through immunohistochemical (IHC) staining. Human ITGA6 gene-specific interference sequences were designed, synthesized, and packaged into lentiviral vectors to establish the Ishikawa cell line with ITGA6-knockdown. Similarly, the ITGA6-overexpression cell line was constructed using the coding sequence (CDS) of the gene. Real-time PCR and Western blot were performed to detect changes in epithelial-mesenchymal transition(EMT)-related markers and angiogenesis-related indicators. Cell invasion and migration capabilities were assessed by Cell Scratch and Transwell assays. Furthermore, Western blot was conducted to profile PI3K/AKT pathway dynamics. ResultsEctopic endometrial tissues exhibited a marked increase in the number of ITGA6-positive cells and their expression intensity compared to eutopic endometrium (each P < 0.001). Compared with the NC group, the ITGA6-knockdown group showed significantly reduced expression of N-cadherin, VEGF, and TGF-β1 (all P < 0.01), while E-cadherin expression was markedly increased (P < 0.01). Concomitantly, the invasion and migration capacities of ITGA6-low expression were significantly impaired (P < 0.001 for both), accompanied by a marked reduction in AKT and phosphorylated AKT(p-AKT) levels (P < 0.001). Conversely, overexpressing ITGA6 resulted in opposite effects. ConclusionITGA6 modulates EMT and angiogenesis in Ishikawa cells via the PI3K/AKT signaling pathway, thereby enhancing cell invasion and migration capabilities, which contributes to the pathogenesis of AWE.

Drug resistance is one of the key factors affecting the effectiveness of cancer treatment methods, including chemotherapy, radiotherapy, and immunotherapy. Its occurrence is related to factors such as mRNA expression and methylation within cancer cells. If drug resistance in patients can be accurately identified early, doctors can devise more effective treatment plans, which is of great significance for improving patients' survival rates and quality of life. Cancer drug resistance prediction based on artificial intelligence (AI) technology has emerged as a current research hotspot, demonstrating promising application prospects in guiding clinical individualized and precise medication for cancer patients. This review aims to comprehensively summarize the research progress in utilizing AI algorithms to analyze multi-omics data including genomics, transcriptomics, epigenomics, proteomics, metabolomics, radiomics, and histopathology, for predicting cancer drug resistance. It provides a detailed exposition of the processes involved in data processing and model construction, examines the current challenges faced in this field and future development directions, with the aim of better advancing the progress of precision medicine.
3.Discovery of orally active and serine-targeting covalent inhibitors against hCES2A for ameliorating irinotecan-triggered gut toxicity.

Human carboxylesterase 2A (hCES2A) plays pivotal roles in prodrug activation and hydrolytic metabolism of ester-bearing chemicals. Targeted inhibition of intestinal hCES2A represents a feasible strategy to mitigate irinotecan-triggered gut toxicity (ITGT), but the orally active, selective, and efficacious hCES2A inhibitors are rarely reported. Here, a novel drug-like hCES2A inhibitor was developed via three rounds of structure-based drug design (SBDD) and structural optimization. Initially, donepezil was identified as a moderate hCES2A inhibitor from 2000 US Food and Drug Administration (FDA)-approved drugs. Following two rounds of SBDD and structural optimization, a donepezil derivative (B7) was identified as a strong reversible hCES2A inhibitor. Subsequently, nine B7 carbamates were rationally designed, synthesized and biologically assayed. Among all synthesized carbamates, C3 showed the most potent time-dependent inhibition on hCES2A (IC50 = 0.56 nmol/L), excellent specificity and favorable drug-like properties. C3 could covalently modify the catalytic serine of hCES2A with high selectivity, while this agent also showed favorable safety profiles, high intestinal exposure, and impressive effects for ameliorating ITGT in both human intestinal organoids and tumor-bearing mice. Collectively, this study showcases a rational strategy for developing drug-like and serine-targeting covalent inhibitors against target serine hydrolase(s), while C3 emerges as a promising orally active drug candidate for ameliorating ITGT.
4.Explore the Rules of Acupoint Selection in the Treatment of Chemotherapy-Induced Nausea and Vomiting Based on Data Mining

[Objective]To explore the common auricular points and the rules of auricular point selection in the treatment of chemotherapy-induced nausea and vomiting by data mining technology,so as to provide reference for clinical treatment date of chemotherapy-induced nausea and vomiting in patients with malignant tumors.[Methods]China National Knowledge Infrastructure(CNKI),Wanfang Database,China Science and Technology Journal Database(VIP),China Biomedical Literature Database(CBM),PubMed,Web of Science,the Cochrane Library database were searched by computer.The search time limit was from the establishment date of the database to August 25,2024.The frequency statistics,association rule analysis and cluster analysis of auricular points in the literature were carried out to describe the compatibility association rules between auricular points.[Results]A total of 92 articles were included,involving 29 auricular points,with a total frequency of 497 times.The most commonly used auricular points were stomach point,Shenmen point,sympathetic point,spleen point,etc.In addition,the commonly used auricular points for chemotherapy-induced nausea and vomiting in breast cancer patients include liver point,subcortical point and endocrine point;the commonly used auricular points for chemotherapy-induced nausea and vomiting in lung cancer patients include subcortical point,lung point and large intestine point;the commonly used auricular points for chemotherapy-induced nausea and vomiting in colorectal cancer and gastric cancer patients included small intestine point,large intestine point and duodenal point.Association rule analysis showed that the combination with the highest support for auricular point sticking in the treatment of chemotherapy-induced nausea and vomiting was Shenmen point-sympathetic point.Cluster analysis classified high-frequency acupoints into four categories.[Conclusion]Auricular points application is effective in treating chemotherapy induced nausea and vomiting and has been widely used in clinical practice.The acupoint compatibility rules obtained based on data mining technology have certain guiding significance for the precise intervention of chemotherapy-induced nausea and vomiting,which can provide reference for acupoint selection.
5.Targeted Delivery of Triptolide Alleviates Diabetic Nephropathy via Inactivation of JAK2-STAT1 Signaling

Objective Inflammation and fibrosis are key features of diabetic nephropathy(DN).Triptolide(TP)exhibits anti-inflammatory and anti-fibrotic properties,though its mechanisms of action in DN remain unclear.CREKA(Cys-Arg-Glu-Lys-Ala)is a pentapeptide that specifically binds to fibronectin(FN),and the CREKA-modified liposome(CREKA-Lip)represents a novel FN-targeted drug delivery system.This study aimed to investigate the role of TP in diabetic db/db mice and determine whether encapsulation within CREKA-Lip enhances therapeutic efficacy while reducing the multi-organ toxicity of TP.Methods Eight-week-old diabetic db/db mice received tail vein injections twice weekly with vehicle,free TP,or CREKA-Lip/TP for 10 weeks.Urine and serum parameters were measured,and kidney,heart,liver,and testis tissues were collected for pathological evaluation.Protein-protein interaction networks were constructed using Cytoscape and its plug-ins to identify core targets and elucidate the therapeutic mechanism of TP against DN.Inflammatory,fibrotic,apoptotic,and lipid metabolism markers were evaluated in the kidneys of diabetic mice with DN and in high glucose-treated mouse mesangial cells and podocytes using qPCR,Western blot,immunohistochemistry,and immunofluorescence assays.Results TP administration reduced fasting blood glucose levels and glomerular mesangial expansion in diabetic mice.TP significantly suppressed renal inflammation,fibrosis,and apoptosis while enhancing lipid metabolism.Integration of network pharmacology,molecular docking,and transcriptomics revealed that TP ameliorated DN by inhibiting the JAK2-STAT1 signaling pathway.In vitro,TP inhibited high glucose-induced phosphorylation of JAK2 and STAT1,reduced collagen production in mesangial cells,decreased apoptosis,and improved lipid metabolism in podocytes.Moreover,CREKA-Lip/TP exhibited superior efficacy compared with free TP,with a more sustained reduction in urine albumin-to-creatinine ratio and greater inhibition of mesangial expansion.Notably,CREKA-Lip/TP treatment did not induce systemic toxicity.Conclusion TP improves renal inflammation,fibrosis,apoptosis,and lipid homeostasis,thereby ameliorating DN by inhibiting JAK2-STAT1 activation.Targeted delivery of TP via FN-binding CREKA-Lip enhances therapeutic efficacy while minimizing multi-organ toxicity.

[Objective]Using data mining technology,to investigate the common syndrome types and acupoint compatibility rules of auricular point pressing beans in the treatment of pediatric enuresis,and provide reference for auricular point pressing beans in the treatment of pediatric enuresis.[Methods]A computer-based search of the literature on auricular point pressing beans for the treatment of pediatric enuresis from the establishment date of the database to January 8,2025.The search databases including Web of Science,PubMed,Cochrane library,CNKI,China Biology Medicine disc,VIP Database and Wanfang Medical Data.Descriptive analysis and association rule analysis were performed on the auricular data in the literature.[Results]A total of 90 articles were included,among which 88 were in Chinese and 2 in English.There were four main syndrome types in pediatric enuresis:syndrome of non-consolidation of kidney qi,syndrome of qi deficiencey of spleen and lung,syndrome of dampness-heat of liver channel,spleen-kidney deficiency pattern.There were 31 auricular points involved in the treatment of pediatric enuresis by auricular point pressing with beans,among which the commonly used auricular points were Bladder,Kidney,Subcortical Yuanzhong and Shenmen.The commonly used auricular acupoints for the treatment of pediatric enuresis were Bladder-Urethra-Yuanzhong-Subcortical-Kidney,Kidney-Shenmen-Yuanzhong-Bladder,and so on.[Conclusion]Data mining effectively identified common syndrome types of pediatric enuresis,frequently used auricular acupoints,and their combination patterns in auricular acupressure therapy.The findings revealed the characteristics and regularities of syndrome-based auricular nursing interventions,providing a scientific basis for acupoint selection in clinical practice and supporting the precise application of auricular acupressure in the nursing care of pediatric enuresis.

9.Association between dietary diversity and the risk of MACE after PCI in patients with coronary heart disease

Objective:To investigate the association between dietary diversity and the risk of major adverse cardiovascular events (MACE) after percutaneous coronary intervention (PCI) in patients with coronary heart disease (CHD) .Methods:A total of 553 patients diagnosed with CHD and undergoing PCI in the Department of Cardiology at the Second Affiliated Hospital of Harbin Medical University between May and November 2023 were enrolled using a convenience sampling method. A Semi-Quantitative Food Frequency Questionnaire was used to assess patients' dietary intake after PCI, and the Dietary Diversity Score (DDS) was calculated. Patients were followed up for one year to determine the incidence of MACE.Results:History of hypertension, history of hyperlipidemia, body mass index, use of antiplatelet agents, use of diuretics, triglycerides, smoking index and DDS were identified as factors influencing the occurrence of MACE after PCI ( P<0.05) . Among these, higher dietary diversity had a protective effect against MACE. Conclusions:After PCI, patients with lower DDS experienced MACE more frequently than those with higher scores. Increased dietary diversity can effectively help prevent MACE in patients after PCI.
