Objective:To study the expression of T follicular helper (Tfh) cells, B cells and their subsets in the peripheral blood of patients with systemic lupus erythematosus (SLE), and to preliminarily explore the relationship between Tfh cells and their subsets and the occurrence and development of SLE.Methods:A total of 38 SLE patients who were hospitalized in the Nanjing Drum Tower Hospital from January 2020 to January 2021 and 23 healthy volunteers matched by age and gender were included. The expression of Tfh cells, B cells, their subsets and cytokines in the peripheral blood of healthy volunteers and SLE patients was analyzed by flow cytometry. The clinical data of SLE patients were collected to evaluate the correlation between Tfh cells and their subsets and disease activity. Results:The proportions of Tfh cells, primary B cells, memory B cells and plasma blasts in the peripheral blood of SLE patients and the level of interleukin-21 (IL-21) were significantly higher than those in healthy volunteers (all P<0.05). Among them, Tfh cells were mainly characterized by an increase in the proportion of Tfh17 cell subsets, and the proportion of Tfh17 cells was positively correlated with the systemic lupus erythematosus disease activity index 2000 (SLEDAI-2K), primary B cells, and plasmatocytes ( r=0.592, 0.300, 0.379, all P<0.05). The proportion of Tfh cells was also positively correlated with SLEDAI-2K ( r=0.469, P<0.05). The proportions of Tfh cells and Tfh17 cells in the peripheral blood of SLE patients with medium to high disease activity were significantly higher than those in the low disease activity group (all P<0.05). After 3 months of treatment for SLE patients with medium to high disease activity, the proportions of Tfh ( P=0.04) and Tfh17 cells ( P=0.003) in the peripheral blood decreased compared with before. Conclusions:The disorder of Tfh cells and B cell subsets is involved in the occurrence and development of SLE. Peripheral blood Tfh17 cells can be used as biomarkers for evaluating the disease activity of SLE patients.

Bovine rotavirus(BRV)is an important pathogen causing diarrhea in calves. To understand the genomic charac-teristics and genetic variations in bovine rotavirus, and to further enrich data on the biological characteristics of rotavirus,we aimed to amplify 11 gene segments of the isolated and cultured G6P[1]bovine rotavirus BLL strain, perform whole genome se-quencing,and analyze the molecular characteristics.MEGA7.0 and DNAMAN software were used for homology and typing a-nalysis,and the whole genome phylogenetic tree was constructed to analyze genetic evolution relationships. The complete geno-type of the BLL strain was G6-P[1]-I2-R2-C2-M2-A3-N2-T6-E2-H3.Phylogenetic analysis of the VP7 and VP4 genes of the BLL strain showed that the VP7 gene had the highest homology with RVA/Cow-wt/HB01/China/2021,and the VP4 gene of the BLL strain was in the same branch as RVA/Human-tc/ISR/Ro8059/1995.From the sequence alignment of VP8*amino acids, the sialic acid domain of the BLL strain was found to be similar to that in other P[1]strains,but different from those in other types of strains, except for residue 189,which was the same as that in Ro8059 but different from that in other strains. The results suggested that the BLL strain might potentially infect humans. Therefore, continued monitoring and study of the biological characteristics of this strain are necessary to provide more information and evidence supporting further research on the cross-species transmission of group A rotavirus in China.

Aim To investigate the expression of al-pha-enolase 1(ENO1)in FLT3-mutated acute myeloid leukemia(AML)and its impact on prognosis,as well as to assess the influence of ENO1 on the antitumor effects of FLT3 inhibitors. Methods Data from the Cancer Genome Atlas(TCGA)and Gene Expression Omnibus(GEO)databases were analyzed to compare the expression differences of ENO1 in various tumors and normal tissues.ENO1 knockdown cells were con-structed using the AML cell line MOLM13.The impact of ENO1 on AML cell proliferation, apoptosis, and drug sensitivity was evaluated through CCK-8 assays, flow cytometry, drug synergy experiments, transcriptome se-quencing,and lipid peroxide and reactive oxygen spe-cies(ROS)levels were detected. Results The ex-pression level of ENO1 was significantly elevated in FLT3 internal tandem duplication mutation(FLT3-ITD)positive AML patients and was associated with poor prognosis.Knockdown of ENO1 significantly in-hibited AML cell proliferation and induced apoptosis.Inhibition of ENO1 significantly increased the chemo-sensitivity of AML cells to FLT3 inhibitors.Transcrip-tome sequencing analysis revealed differential expres-sion of genes in the NADPH oxidase-related pathway following ENO1 knockdown, and a positive correlation between ENO1 and glutathione peroxidase 4(GPX4)expression.Inhibition of ENO1 significantly increased the levels of lipid peroxides and ROS induced by FLT3 inhibitors. Conclusions High expression of ENO1 is significantly associated with poor prognosis in FLT3(+)AML.Inhibition of ENO1 can enhance the anti-tumor effects of FLT3 inhibitors by regulating the an-tioxidant defense capacity of AML cells, providing a new target and therapeutic strategy for the treatment of FLT3/ITD(+)AML.

Aim To investigate the effects of rutae-carpine(RUT)on lung inflammation in septic mice and the underlying mechanisms. Methods The sepsis mouse model was generated by intraperitoneal injection of lipopolysaccharide(LPS)at 5 mg·kg-1.The mice were randomly divided into the Control group,Model group,Low-dose,Medium-dose,High-dose RUT(5,10,20 mg·kg-1)treatment group and dexamethasone(DEX,2 mg·kg-1),with 10 mice in each group. The mice were intraperitoneally injected with RUT 30 min before LPS injection.HE staining was used to observe the morphology of lung tissues, and activity of my-eloperoxidase was determined to assess the neutrophil infiltration.Wet/dry weight ratio and Evan's blue ex-travasation of lung tissues were examined to assess lung edema.Survival analysis was performed to determine the in vivo protective effects of RUT.ELISA and quan-titative RT-PCR analysis were employed to determine the contents and gene expression of pro-inflammatory mediators, including tumor necrosis factor-α(TNF-α),interleukin-1 β(IL-1 β),IL-6,and IL-18 in lung tis-sues.Western blot was used to detect the protein levels of p38 MAPK,NF-κB,Caspase-1,NOD-like receptor family pyrin domain containing 3(NLRP3)and IL-18.Results RUT at 10-20 mg·kg-1 could dose-de-pendently inhibit leukocyte infiltration, reduce pro-in-flammatory mediator production, vascular permeability and wet/dry weight ratio in lungs, similar to the effects induced by DEX.The mice treated with RUT exhibited increased survival,down-regulated expressions of p-p38 MAPK,p-NF-κB,Caspase-1,NLRP3,and IL-18 pro-teins in lungs, with decreased IL-18 mRNA level. Conclusions RUT exhibits protective effects on sep-sis-induced lung injury, manifested by reduced inflam-mation and edema, potentially via inhibition of p38 MAPK signaling pathway and inflammasome formation.

Soy is a vital source of plant carbohydrates. However,it poses significant allergenic risks, particularly to young children and animals.Among the various proteins in soy,β-conglycinin,which con-stitutes approximately 30%of total soy carbohydrates,is a primary allergen.Undigested β-conglycinin can lead to intestinal damage by inhibiting cell growth, disrupting the cytoskeleton, and inducing apopto-sis.It can also enter the lymphatic and circulatory systems, triggering allergic reactions.Conventional ELISA methods for detecting β-conglycinin rely on polyclonal or monoclonal antibodies, which are limited by their large molecular weight, difficulty in accessing the protein core, and sensitivity to acidic and bas-ic conditions. To address these limitations, this study aimed to develop nanobodies(Nbs)against β-con-glycinin.Nbs,derived from the variable regions of heavy-chain antibodies found in camelids, have a mo-lecular weight approximately one-tenth that of conventional antibodies. They offer advantages such as small size, stable structure, high specificity, and strong affinity.A female alpacas was immunized five times using β-conglycinin,which showed a heavy chain antibody potency of 1∶16 000 by ELISA.Pe-ripheral blood lymphocytes were subsequently isolated and total RNA was extracted. The variable region of the heavy-chain antibody was amplified via PCR,and recombinant plasmids were constructed and transformed into the E.coli competency strain ER2738.The resulting library contained about 3.5×108 CFU/mL,which increased to 1.15×1012 PFU/mL after phage rescue, with a 100%Nbs gene insertion rate, indicating high diversity. Its Nbs phage output was significantly enriched by four rounds of solid-phase elution with an enrichment rate of 155.9.Four rounds of solid-phase panning yielded 35 positive clones, all of which shared the same amino acid sequence upon sequencing. The selected Nb was ex-pressed in a prokaryotic system, and its binding ability to β-conglycinin was confirmed using Western blotting and ELISA.The results demonstrated excellent specificity and affinity. This research lays the groundwork for developing a rapid and efficient detection method for β-conglycinin using Nbs,potentially enhancing food safety and allergen management.
