1.A cross-sectional study of anxiety disorders in adults in Inner Mongolia Autonomous Region

Objective:To describe the prevalence of anxiety disorders and its distribution in Inner Mongolia Autonomous Region,and to explore the relevant factors of anxiety disorders.Methods:From June 2019 to Decem-ber 2019,representative multi-stage disproportionate stratified sampling procedure was used to sample in residents aged 18 and over in the Inner Mongolia Autonomous Region.All respondents were face-to-face interviewed by trained interviewers.Composite International Diagnostic Interview-3.0(CIDI-3.0)was used to diagnose anxiety disorders according to the criteria and definition of the Diagnostic and Statistical Manual of Mental Disorders,Fourth Edition(DSM-Ⅳ).Chi-square test and multivariate logistic regression analysis were used for statistical anal-ysis.Results:Totally 12 315 people were interviewed in the survey.The weighted 12-mouth prevalence rate of any anxiety disorder was 4.64%,and the lifetime prevalence rate was 6.25%.The weighted 12-month prevalence rate of anxiety disorders was higher in female than that in male(5.38%vs.3.92%).The rate was higher in rural resi-dents than that in urban residents(5.67%vs.3.95%).The rate was higher in people with chronic diseases than that in people without chronic diseases(6.81%vs.2.29%).Logistic regression analysis showed that unmarried(OR=2.32,95%CI:1.31-4.10),separated/divorced(OR=2.49,95%CI:1.33-4.67),in debt(OR=1.55,95%CI:1.04-2.32),chronic disease(OR=2.22,95%CI:1.39-3.53),family history of anxiety disorders(OR=12.05,95%CI:8.78-16.53),poor sleep(OR=2.64,95%CI:1.97-3.54)were risk factors of occurrence of anxiety disorders,while junior high school(OR=0.65,95%CI:0.44-0.96)was protective factor of anxiety disor-ders.Conclusion:Adults with chronic diseases,poor sleep,unmarried or separated/divorced,family history of anxi-ety disorders,and financial debt are at higher risk groups of anxiety disorder in Inner Mongolia Autonomous Re-gion.

Clinical pharmacist teacher training is an important mean to improve the quality of clinical pharmacy talent cultivation and ensure the service ability and level of the clinical pharmacist team.The Pharmacy Administration and Pharmacy Practice in Healthcare Institutions-Part 4-8-2:Pharmacy Administration-Pharmacy Training Management-Clinical Pharmacist Teacher Training was based on the newly revised management document for clinical pharmacist teacher training of the Chinese Hospital Association.After sorting out relevant materials,such as standards,policies and regulations,technical specifications,liter-ature,documents of the Chinese Hospital Association,expert opinions,and the current situation of clinical pharmacist teacher training in China,the standard was formulated.In the standard,12 key elements,which can be divided into 3 parts of base manage-ment,training process and assessment,quality management and evaluation improvement,were standardized.This article aimed to introduce the construction method and content of the standard,to facilitate the understanding of the standard content for medical institutions which joined or willing to join the clinical pharmacist teacher training base,and to provide a reference for other medi-cal institutions to carry out related work.

4.The Synergistic Effect of Astaxanthin and Curcumin on the Outcome of IVF-ET Treatment in Patients with Poor Ovarian Response

Objective:To investigate the synergistic effect of astaxanthin and curcumin on assisted reproduc-tive technology(ART)outcomes in patients with poor ovarian response(POR).Methods:A prospective cohort study was conducted,involving 123 POR patients enrolled at the Reproductive & Women-Children Hospital,Chengdu University of Traditional Chinese Medicine between March 6,2023 to May 1,2024.According to patient preference,participants were divided into the treatment group(41 cases)and the control group(82 cases)at a ratio of 1∶2.The treatment group took astaxanthin and turmeric compound tablets orally for 60 to 90 days before ovulation induction treatment.The ovulation induction parameters and pregnancy outcomes were compared between the two groups,and a self-controlled analysis was conducted within the treatment group.Results:During the observation period,5 and 10 participants withdrew from the treatment and control groups,respectively,resul-ting in 108 patients included in the final analysis.There were no statistically significant differences between the two group in terms of endometrial thickness on the day of human chorionic gonadotropin(HCG),number of oocytes retrieved,oocyte maturation rate,normal fertilization rate,high-quality embryo rate,or cycle cancellation rate(P>0.05).However,the treatment group demonstrated significantly higher clinical pregnancy rate(61.54%vs.28.57%),fresh embryo implantation rate(40.91%vs.18.97%),and cumulative clinical pregnancy rate(65.38%vs.33.33%)(P<0.05).The early miscarriage rate was observed with no significant difference between groups(12.5%vs.10.0%)(P>0.05).Furthermore,after treatment,basal FSH(bFSH),basal E2(bE2),antral follicles count(AFC),anti-Müllerian hormone(AMH),and the number of oocytes retrieved within the treatment group were significantly improved compared with those before treatment(P<0.05).Conclusions:Pretreatment with astaxanthin and turmeric compound tablets in POR patients did not significantly improve the ovulation induction re-sponse,but was associated with increased fresh embryo clinical pregnancy rate,implantation rate,and cumulative pregnancy rate.The underlying mechanism may involve modulation of basal endocrine profiles and improvement of ovarian reserve function,though further investigation is required to elucidate precise pathways.

High mobility group box protein 1(HMGB1)is one of the most widely expressed protein member in the HMGs family,which is well known for its involvement in the body inflammatory response.Previous researches have found that it plays a significant role in cell migration,immune identification and neuroprotection.Spinal cord injury is a disease that causes severe damage to the nervous system,and neural circuits are disrupted after a spinal cord injury,which leads to many conditions including ischemia and hypoxia,inflammatory responses,demyelinating lesions,and glial scar formation that are detrimental to nerve regeneration and repair,making it one of the most difficult diseases to treat in the modern spinal surgery field.HMGB1 is upregulated after spinal cord injury,thereby regulating neuroinflam-matory responses,and participating in the neuronal apoptosis,promoting neuronal regeneration,and inducing neural stem cell differentiation and migration,which plays an important role in the process of neural function recovery.This paper summarizes the structure and function of HMGB1,as well as its role in spinal cord injury,in order to provide direction for founding therapeutic target for neurological function recovery after spinal cord injury.

8.A Study of a New Variation of α-1,3-Galactosyltransferase Gene in Pedigrees

Objective:To investigate the association between genetic variation sites and blood group phenotypes in a family with the ABw subtype.Methods:A 22-year-old male proband and six family members who underwent health examinations at the Department of Transfusion Medicine,960th Hospital of the PLA Joint Logistics Support Force on April 8,2023 were enrolled.ABO blood group phenotyping of the proband and family members was performed using the tube method.Direct sequencing of PCR products covering the ABO gene promoter region,intron 1,and exons 1-7 was conducted for the proband and family members.Clonal sequencing of exons 6 and 7 was performed for the proband.Results:The serological phenotype of the proband was identified as ABw.Direct sequencing of PCR products revealed that the proband's ABO gene promoter region contained two variants(c.-105G>C and c.-106G>C),intron 1 contained the variant c.28+5708G>A,and exon 6 contained the variant c.255C>T.Blood group genotype of the proband was ABO*A1.02/ABO*B with the c.255C>T variant.Family analysis showed that the proband's mother,brother,elder niece,and younger niece also carried the c.255C>T variant in exon 6.The proband's mother had variants c.-105G>C and c.-106G>C in the promoter region,c.28+6127T>C and c.28+6154A>G in the intron region,with a genotype of ABO*O.01.01/ABO*B and the c.255C>T variant.The proband's brother,elder niece,and younger niece exhibited no promoter region abnormalities but carried variants c.28+6272C>T,c.28+6173A>G,c.28+6284T>C,and c.28+6297A>T in intron 1.The brother's genotype was ABO*A1.02/ABO*B with c.255C>T,the elder niece's genotype was ABO*B.01/ABO*B with c.255C>T,and the younger niece's genotype was ABO*O.01.01/ABO*B with c.255C>T.The proband's father had a genotype of ABO*A1.02/ABO*A1.02,and the sister-in-law's genotype was ABO*O.01.01/ABO*B.01.Conclusion:The c.255C>T variant in exon 6 of the ABO blood group B allele(α-1,3-galactosyltransferase gene)exhibits hereditary characteristics and exerts a negative regulatory effect on glycosyltransferase activity to a certain extent.
9.Effect of Yuxuebi Tablets on mice with inflammatory pain based on GPR37-mediated inflammation resolution.

In order to investigate whether the effect of Yuxuebi Tablets on the peripheral and central inflammation resolution of mice with inflammatory pain is related to their regulation of G protein-coupled receptor 37(GPR37), an inflammatory pain model was established by injecting complete Freund's adjuvant(CFA) into the paws of mice, with a sham-operated group receiving a similar volume of normal saline. The mice were assigned randomly to the sham-operated group, model group, ibuprofen group(91 mg·kg~(-1)), and low-, medium-, and high-dose groups of Yuxuebi Tablets(60, 120, and 240 mg·kg~(-1)). The drug was administered orally from days 1 to 19 after modeling. Von Frey method and the hot plate test were used to detect mechanical pain thresholds and heat hyperalgesia. The levels of interleukin-10(IL-10) and transforming growth factor-beta(TGF-β) in the spinal cord were quantified using enzyme-linked immunosorbent assay(ELISA), and the mRNA and protein expression of GPR37 in the spinal cord was measured by real-time quantitative reverse transcription PCR(qRT-PCR) and Western blot. Additionally, immunofluorescence was used to detect the expression of macrosialin antigen(CD68), mannose receptor(MRC1 or CD206), and GPR37 in dorsal root ganglia, as well as the expression of calcium-binding adapter molecule 1(IBA1), CD206, and GPR37 in the dorsal horn of the spinal cord. The results showed that compared with those of the sham-operated group, the mechanical pain thresholds and hot withdrawal latency of the model group significantly declined, and the expression of CD68 in the dorsal root ganglia and the expression of IBA1 in the dorsal horn of the spinal cord significantly increased. The expression of CD206 and GPR37 significantly decreased in the dorsal root ganglion and dorsal horn of the spinal cord, and IL-10 and TGF-β levels in the spinal cord were significantly decreased. Compared with those of the model group, the mechanical pain thresholds and hot withdrawal latency of the high-dose group of Yuxuebi Tablets significantly increased, and the expression of CD68 in the dorsal root ganglion and IBA1 in the dorsal horn of the spinal cord significantly decreased. The expression of CD206 and GPR37 in the dorsal root ganglion and dorsal horn of the spinal cord significantly increased, as well as IL-10 and TGF-β levels in the spinal cord. These findings indicated that Yuxuebi Tablets may reduce macrophage(microglial) infiltration and foster M2 macrophage polarization by enhancing GPR37 expression in the dorsal root ganglia and dorsal horn of the spinal cord of CFA-induced mice, so as to improve IL-10 and TGF-β levels, promote resolution of both peripheral and central inflammation, and play analgesic effects.

10.Construction of Saccharomyces cerevisiae cell factory for efficient biosynthesis of ferruginol.

Diterpenoid ferruginol is a key intermediate in biosynthesis of active ingredients such as tanshinone and carnosic acid.However, the traditional process of obtaining ferruginol from plants is often cumbersome and inefficient. In recent years, the increasingly developing gene editing technology has been gradually applied to the heterologous production of natural products, but the production of ferruginol in microbe is still very low, which has become an obstacle to the efficient biosynthesis of downstream chemicals, such as tanshinone. In this study, miltiradiene was produced by integrating the shortened diterpene synthase fusion protein,and the key genes in the MVA pathway were overexpressed to improve the yield of miltiradiene. Under the shake flask fermentation condition, the yield of miltiradiene reached about(113. 12±17. 4)mg·L~(-1). Subsequently, this study integrated the ferruginol synthase Sm CYP76AH1 and Sm CPR1 to reconstruct the ferruginol pathway and thereby realized the heterologous synthesis of ferruginol in Saccharomyces cerevisiae. The study selected the best ferruginol synthase(Il CYP76AH46) from different plants and optimized the expression of pathway genes through redox partner engineering to increase the yield of ferruginol. By increasing the copy number of diterpene synthase, CYP450, and CPR, the yield of ferruginol reached(370. 39± 21. 65) mg·L~(-1) in the shake flask, which was increased by 21. 57-fold compared with that when the initial ferruginol strain JMLT05 was used. Finally, 1 083. 51 mg·L~(-1) ferruginol was obtained by fed-batch fermentation, which is the highest yield of ferruginol from biosynthesis so far. This study provides not only research ideas for other metabolic engineering but also a platform for the construction of cell factories for downstream products.
