Objective To construct a lung cancer surgery-oriented disease-specific database covering the entire perioperative care pathway, thereby improving the quality and usability of key surgical data elements. Methods Real-world clinical data were extracted from a single-center thoracic surgery department. A standardized data model was established based on the open electronic health record (openEHR) standard. Large language model (LLM), optical character recognition (OCR), and artificial intelligence (AI)-driven techniques were employed to extract, structure, and perform quality control on unstructured clinical narratives, imaging reports, and radiological data, with a focus on capturing surgically relevant perioperative indicator. Results A multimodal database comprising 19 917 patients was established, including 7 930 males and 11 987 females, with ages ranging from 15 to 97 (61.7±9.7) years. The database includes 582 structured data variables, textual report data corresponding to 69 clinical indicators, 13 000 pulmonary function test PDF reports, and chest CT imaging data from 16 884 patients. This database comprehensively covers major information relevant to surgical diagnosis and treatment of lung cancer, significantly improving the completeness and granularity of surgical detail data. Large language models (LLMs) and optical character recognition (OCR) technologies enhanced the efficiency of converting unstructured data into structured formats, while a multi-level manual verification process ensured data accuracy and traceability. The database supports real-world research including comparisons of surgical procedures, prediction of postoperative complications, prognosis assessment, and multimodal data association analyses.

Recent studies have shown that the physiological homeostasis of oral mucosal cells is regulated by the circadian clock.Dis-ruption or dysfunction of the circadian clock is closely associated with the development of oral squamous cell carcinoma(OSCC).Research based on the circadian clock offers a novel perspective on the pathogenesis and therapeutic strategies for OSCC.However,there is current-ly limited research on this topic,and people generally have insufficient understanding and recognition of the circadian clock.Given the complexity and challenges of circadian clock which is the fourth dimension of medical research,we organize relevant experts based on summarizing the current research results of circadian clock in the pathogenesis and precision diagnosis and treatment of OSCC,combining the scientific principles of the circadian clock's role and their long-term research experience,then summarizes and recommends the con-sensus opinions for the research of circadian clock in the pathogenesis mechanism and precision diagnosis and treatment of human OSCC,with the hope of providing guidance for the basic research and clinical application of circadian clock or circadian rhythm in the pathogene-sis mechanism and precision diagnosis and treatment of oral and maxillofacial squamous cell carcinoma.

Objective To investigate the role and mechanisms of acyl-CoA synthetase long-chain family member 3(ACSL3)m6A modification in suppressing ferroptosis during cisplatin resistance in non-small cell lung cancer.Methods In vitro,cisplatin-resistant human lung adenocarcinoma A549/DDP cells were subjected to the following interventions:transduction with lentiviral vectors expressing ACSL3 short hairpin RNA,transfection with Wilms'tumor 1-associating protein(WTAP)small interfering RNA(siRNA)plasmids,or co-transfection with WTAP siRNA plasmids and ACSL3 overexpression plasmids.After 48 hours,all the groups were replaced with complete medium containing 1 μg/mL cisplatin and continued to culture for 24 h.A549(or A549/DDP)cells infected with ACSL3 short hairpin RNA lentivirus were subcutaneously inoculated into the backs of nude mice to establish a subcutaneous xenograft model.Immunofluorescence assay was performed to detect ACSL3 protein expression in cisplatin-treated or normally cultured A549 and A549/DDP cells.Cisplatin sensitivity was evaluated using colony formation assays.FerroOrange staining and total iron assay kits were used to measure intracellular and tissue iron levels.Lipid peroxidation was detected using the BODIPY 581/591 C11 probe.ACSL3 m6A methylation was analyzed by MeRIP-qPCR,while RNA decay assays evaluated ACSL3 mRNA stability.qRT-PCR and Western blotting were used to measure mRNA and protein expressions of ACSL3 and WTAP.Ferritin and COX2 levels were measured by immunohistochemistry.Results After cisplatin treatment,the decrease in ACSL3 expression was significantly smaller in A549/DDP cells[(48.70±3.81)％]than in A549 cells[(65.62±2.53)％].ACSL3 knockdown significantly reduced the colony formation capacity of A549/DDP cells,but increased FerroOrange intensity,total iron levels and lipid peroxidation levels(P＜0.05).Cisplatin reduced ACSL3 m6A methylation in A549/DDP cells,with a less decline than in A549 cells(P＜0.05).WTAP knockdown accelerated ACSL3 mRNA decay and reduced m6A enrichment of ACSL3.Compared to A549/DDP cells with only WTAP knockdown,those with combined si-WTAP knockdown and ACSL3 overexpression showed increased colony formation capacity,but decreased FerroOrange intensity,total iron levels,and lipid peroxidation levels.In vivo,ACSL3 knockdown upregulated COX2 and ferritin levels,increased Fe2+and ROS levels in tumor tissues,and suppressed tumor growth.Conclusion WTAP suppresses ferroptosis and reduces cisplatin sensitivity in A549 cells by mediating m6A modification of ACSL3 mRNA,thereby regulating the progression of lung cancer.

Methods: The visibility of the C6, C7 upper, and C7 lower endplates was graded and compared. Baseline data such as height, weight, body mass index (BMI), age, and sex were analyzed for their impact on the visibility of the C7 lower endplate. Values of C2-6 Cobb angle, SYS-G angle, and C2-7 Cobb angle were measured. The intra- and interobserver reliability, differences, and efficacy of evaluation on cervical lordosis of the parameters were compared, and the correlations among the parameters were analyzed. Furthermore, reference ranges for the SYS-G angle were established based on lateral cervical spine x-rays of 825 asymptomatic Chinese adults across different age groups and sexes. Results: The visibility of the C7 lower endplates was significantly reduced compared to the C6 lower and C7 upper endplates.Age, weight, BMI, and male sex were identified as factors negatively influencing the visibility of the C7 lower endplate. Both intraobserver and interobserver reliability demonstrated excellence for all tested parameters. The linear regression model unveiled a stronger association of the SYS-G angle with the C2-7 Cobb angle compared to the C2-6 Cobb angle. Furthermore, the SYS-G angle exhibited excellent efficacy in evaluating cervical lordosis. Age displayed a positive correlation with the SYS-G angle, and across every age bracket from 20 to 69 years, men exhibited a higher mean SYS-G angle compared to women. Conclusions The visibility of the C7 lower endplate diminishes with increasing age, weight, BMI, and male sex. Objective : To analyze the distribution characteristics of free water ( FW) and FW-corrected fractional anisotropy (FAt) in the white matter of the brain in patients with anti-N-methyl-D-aspartate receptor ( NMDAR) encephalitis and to explore their correlation with cognitive function． Methods : A total of 38 patients with anti- NMDAR encephalitis and 30 controls were recruited from three hospitals in Hefei．Diffusion tensor imaging data and neuropsychological assessment results were collected．Tract-Based Spatial Statistics was applied to compare group differences in FW and FAt across the whole brain white matter．Correlation analyses were further performed to ex- amine the relationships between FW/ FAt metrics and cognitive function． Results : Compared to the control group， patients with anti-NMDAR encephalitis showed significantly lower scores on the montreal cognitive assessment，im- mediate recall，delayed recall，and the verbal fluency test (all P＜0. 05) ，as well as significantly longer comple- tion times for color naming and word reading tasks in the stroop color word test (all P＜0. 05) ．Diffusion tensor im- aging analysis revealed significantly elevated FW and reduced FAt in widespread white matter regions in the patient group (all P＜0. 000 1) ．Further correlation analysis showed that increased FW was positively associated with the completion time of the color-switching condition in the color digital trail making test (P = 0. 044 ) and with the difference between color-switching and number sequencing conditions ( P = 0. 016 ) ，while negatively correlated with semantic fluency scores (P = 0. 002) ．Additionally，FAt was positively associated with delayed recall perform- ance (P = 0. 012) ，and negatively correlated with the completion times for color naming (P = 0. 018 ) and word reading (P = 0. 046) tasks in the SCWT． Conclusion Patients with anti-NMDAR encephalitis exhibit significantly elevated FW and significantly reduced FAt in white matter tracts，both of which are closely linked to cognitive im- pairment．

Resistance to poly adenosine diphosphate(ADP)-ribose polymerase inhibitor(PARPi)presents a considerable obstacle in the treatment of ovarian cancer.F-box and tryptophan-aspartic(WD)repeat domain containing 11(FBXW11)modulates the ubiquitination of growth-and invasion-related factors in lung cancer,colorectal cancer,and osteosarcoma.The function of FBXW11 in PARPi therapy is still ambiguous.In this study,RNA sequencing(RNA-seq)showed that FBXW11 expression was raised in ovarian cancer cells that had been treated with PARPi.FBXW11 was abnormally expressed at low levels in high-grade serous ovarian cancer(HGSOC)tissues,and low levels of FBXW11 were associated with shorter overall survival(OS)and progression-free survival(PFS)in HGSOC patients.Overexpressing FBXW11 made ovarian cancer more sensitive to PARPi,while knocking down FBXW11 made it less sensitive.The four-dimensional(4D)label-free quantitative proteomic analysis revealed that FBXW11 targeted S100 calcium binding protein A11(S100A11)and promoted its degradation through ubiquiti-nation.The increased degradation of S100A11 led to less efficient DNA damage repair,which in turn contributed to increased PARPi-induced DNA damage.The role of FBXW11 in promoting PARPi sensitivity was also confirmed in xenograft mouse models.In summary,our study confirms that FBXW11 promotes the susceptibility of ovarian cancer cells to PARPi via affecting S10OA11-mediated DNA damage repair.

BACKGROUND: Previous evidence showed that ambient air pollution and cardiovascular mortality are related. However, there is a lack of evidence towards the modification effect of long-term lifestyle on the association between short-term ambient air pollution and death from cardiovascular events. METHOD: A total of 14,609 death from major adverse cardiovascular events (MACE) were identified among the China Kadoorie Biobank participants from 2013 to 2018. Ambient air pollution exposure including particulate matter 2.5 (PM_2.5), SO₂, NO₂, CO, and O₃ from the same period were obtained from space-time model reconstructions based on remote sensing data. Case-crossover design and conditional logistic regression was applied to estimate the effect of short-term exposure to air pollutants on MACE mortality. RESULTS: We found MACE mortality was significantly associated with PM_2.5 (relative percent increase 2.91% per 10 µg/m³ increase, 95% CI 1.32-4.53), NO₂ (5.37% per 10 µg/m³ increase, 95% CI 1.56-9.33), SO₂ (6.82% per 10 µg/m³ increase, 95% CI 2.99-10.80), and CO (2.24% per 0.1 mg/m³ increase, 95% CI 1.02-3.48). Stratified analyses indicated that drinking was associated with elevated risk of MACE mortality with NO₂ and SO₂ exposure; physical inactivity was associated with higher risk of death from MACE when exposed to PM_2.5; and people who had balanced diet had lower risk of MACE mortality when exposed to CO and NO₂. CONCLUSIONS The study results showed that short-term exposure to ambient PM_2.5, NO₂, SO₂, and CO would aggravate the risk of cardiovascular mortality, yet healthy lifestyle conduct might mitigate such negative impact to some extent.
