1.Moderating effect of interacting partner and personality traits on social interaction and emotional experience in the elderly

Objective:To investigate the relationship between social interaction and emotional experience in older adults,and the moderating effects of social interaction objects and personality traits on their relationship.M-ethod:Totally 208 elderly individuals were selected and assessed with a self-designed demographic information questionnaire and the NEO Personality Inventory(NEOPI).The Rochester Interaction Record(RIR)was used to record 14 days of social activity,and the Positive-Negative Affect Scale(PANAS)was used to evaluate emotions after each interaction.Using Mplus Multilevel Model(MLM)for model construction.Results:Through model con-struction,it was found that the interaction between openness and communication quality was statistically significant(β=0.45,P<0.05).When the dependent variable was positive emotion,the higher the openness of elderly people,the stronger the connection between communication quality and positive emotion.When interacting with non-family members,the interaction between openness and social quality was statistically significant(β=0.52,P<0.05).The higher the openness of older adults,the stronger the connection between social quality and positive emotions.Conclusion:It suggests that older adults with moderate or higher levels of openness have a stronger con-nection between social interaction quality and positive emotions when interacting with non-family members.That is,the higher the quality of interaction,the more positive emotions could be promoted,but negative emotions could not be reduced.
2.Research progress on intelligent diagnosis of eye diseases based on facial photos

The number of patients with eye diseases in China is enormous,and the negative effects of these conditions,such as impaired visual function,psychological burdens,and restricted social participation,are becoming increasingly severe.Due to the limited and unevenly distributed ophthalmic resources,and the significant limitations of traditional diagnostic and therapeutic approaches in terms of accuracy and efficiency,there is an urgent need for more sensitive and efficient modalities.With the rapid advancement of artificial intelligence technology,ophthalmic diagnosis has entered a new stage of intelligent transformation.Facial photos,as a noninvasive and convenient medium,show unique advantages in eye disease diagnosis.Artificial intelligence systems based on facial photo analysis have been applied to the screening and diagnosis of conditions such as myopia,strabismus,ptosis,and thyroid eye disease,showing promising results.This review introduces the workflow of intelligent diagnosis for ocular diseases based on facial photographs,with a focus on recapitulating relevant research findings both domestically and internationally in recent years.It summarizes the innovative features and application advantages of intelligent diagnosis systems for eye diseases based on facial photos,analyzes the current technical bottlenecks and challenges in application,proposes corresponding countermeasures,and discusses future development directions,aiming to provide references and new insights for the intelligent screening and diagnosis of eye diseases.

4.Therapeutic Effect of Wenweishu Granules on Functional Dyspepsia Rats with Spleen-stomach Deficiency Cold Syndrome Based on Bioinformatics Analysis and Experimental Validation

ObjectiveThis study aims to investigate the therapeutic effects of Wenweishu granule (WWSG) on functional dyspepsia (FD) with spleen-stomach deficiency cold syndrome in rats by integrating network pharmacology, molecular docking, and animal experiments. MethodsActive components and corresponding targets of WWSG were collected from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP) and the Bioinformatics Analysis Tool for Molecular Mechanism of Traditional Chinese Medicine (BATMAN-TCM). Disease-related targets for FD with spleen-stomach deficiency cold syndrome were screened using GeneCards and the Integrative Pharmacology-based Research Platform of Traditional Chinese Medicine (TCMIP). Core therapeutic targets were identified via Cytoscape and validated by molecular docking. A rat model of FD with spleen-stomach deficiency cold syndrome was established using vinegar gavage combined with tail-clamping. The rats were randomly divided into a model group, low-, medium-, and high-dose WWSG groups (2.0, 4.0, 8.0 g·kg-1), a domperidone group (3.0 mg·kg-1), a Fuzi Lizhong pillwan (0.8 g·kg-1), and a normal control group (n=10 per group). Drugs were administered once daily by gavage for 14 consecutive days. After treatment, body weight, symptom scores, and gastrointestinal motility indices were recorded. Gastric and duodenal pathologies changes were observed via hematoxylin-eosin (HE) staining. Brain-gut peptides were measured in serum and tissue using enzyme-linked immunosorbent assay (ELISA). Immunohistochemistry and Western blot were performed to assess stem cell factor (SCF) and receptor tyrosine kinase (c-Kit) protein expression in gastric tissues. ResultsA total of 305 drug targets, 1 140 disease targets, and 116 overlapping targets were identified. Cytoscape analysis revealed 104 core targets. Enrichment analysis indicated that the SCF/c-Kit signaling pathway was the key mechanism. Molecular docking confirmed a strong binding affinity between active components of WWSG and SCF/c-Kit proteins (binding energy<-5.1 kcal·mol-1). Compared with the normal group, model rats exhibited slower weight gain (P<0.05), reduced gastric emptying and intestinal propulsion (P<0.01), mild gastric mucosal shedding, duodenal inflammatory cell infiltration, decreased levels of gastrin (GAS), 5-hydroxytryptamine (5-HT), and vasoactive intestinal peptide (VIP) (P<0.05, P<0.01), and elevated somatostatin (SS) expression (P<0.05, P<0.01). WWSG treatment ameliorated weight gain, symptom scores, and low-grade inflammation in gastric/duodenal tissues. High-dose WWSG significantly improved gastric emptying and intestinal propulsion, upregulated GAS, 5-HT, and VIP, and downregulated SS expression in serum and tissues (P<0.05, P<0.01). Immunohistochemistry and Western blot demonstrated that SCF and c-Kit protein expression was decreased in the model group (P<0.05, P<0.01), which was reversed by WWSG intervention (P<0.05). ConclusionWWSG exerts therapeutic effects on FD with spleen-stomach deficiency cold syndrome in rats, potentially by regulating the SCF/c-Kit signaling pathway to enhance gastrointestinal motility.

6.Production of 2-18F-fluorobutyric acid as a PET imaging agent for prostate cancer

[Objective] In view of the crucial role of indole propionic acid in the treatment of tumor immune checkpoint blockade and further revealing its mechanism, our study intended to design and synthesize 1-[18F]-fluoroethyl-indole propionic acid (1-[18F]-IPA), and evaluate it as a tumor PET imaging agent. [Methods] The precursor 1-(2-p-toluenesulfonic acid oxygen ethyl)-methyl indole propionate underwent nucleophilic substitution reaction with 18F-. The crude product was separated and purified by high-performance liquid chromatography and the intermediates were collected. Finally, 1-[18F]-IPA was obtained by hydrolysis. The clarity of the product was measured by visual inspection, the pH value was determined by precision test paper, and the radiochemical purity and stability were determined by high-performance liquid chromatography. In order to determine the biodistribution of 1-[18F]-IPA in normal mice, ICR mice were intravenously injected with 1-[18F]-IPA (0.2 mL, 7 MBq), and sacrificed at 5, 15, 25, 45, 75 and 120 min and dissected. Micro-PET imaging was performed and analyzed in BxPC-3 tumor-bearing nude mice. Student t test was used to compare the biodistribution of tissues and organs at different time points. [Results] The total preparation time of 1-[18F]-IPA was 35-40 min, the radiochemical yield was (45±5)%, and the radiochemical purity was more than 95%. The product solution was clear without particles, and the pH value was 6.5, which had good stability in vitro and in vivo. The results of biodistribution in healthy ICR mice showed that except for the brain, 1-[18F]-IPA had a certain uptake in all major organs, with the most obvious uptake in the liver, gallbladder and kidneys. The radioactivity in the gallbladder gradually increased with time and reached (39.86±6.56)%ID/g at 120 min, but bone uptake did not change significantly with time. Micro-PET/CT showed that there was radioactive uptake at the tumor 30 min after injection of 1-[18F]-IPA in Dutch BxPC-3 nude mice, but it was not obvious. At this time, SUVmax was about 55.18±14.62. Consistent with the results of biodistribution, the brain uptake was low at each time point. [Conclusion] In summary, 1-[18F]-IPA with short preparation time and high yield is expected to be a tool to probe tryptophan indole metabolism pathway and further reveal tumor immune resistance.
7.Establishment of prediction model for long-term prognosis of acute pulmonary embolism in internal medicine inpatients with type 2 diabetes mellitus

Objective To investigate the long-term prognostic factors of acute pulmonary embolism (APE) among internal medicine inpatients with type 2 diabetes mellitus (T2DM),and establish a prediction model of long-term prognosis. Methods This study included 156 internal medicine inpatients with T2DM and APE (T2DM+APE group) admitted to internal medicine of Xinjiang 474 Hospital and The Fifth Affiliated Hospital of Xinjiang Medical University from July 2019 to June 2022. At the same time,156 inpatients with APE from the department of hematology were selected as APE group. All patients were fo llowed up for 1 year to evaluate the prognosis. Patients in T2DM+APE group were divided into good prognosis subgroup (n=108) and poor prognosis subgroup (n=48). The risk factors for poor prognosis were analyzed and a prediction model was established. Results The incidence of adverse events in T2DM+APE group was higher than that in APE group (P<0.05). Logistic regression analysis found that N-terminal precursor of B-type natriuretic peptide (NT-proBNP),cardiac troponin T (c-TnT),heart-type fatty acid binding protein (H-FABP) and simplified Pulmonary Embolism Severity Index (sPESI) score were risk factors for poor long-term prognosis of T2DM complicated with APE. The area under the curve(AUC),sensitivity and specificity of this model for predicting poor prognosis of T2DM complicated with APE were 0.938,83.3% and 96.3%,which were higher than those of prediction with single indicator. Conclusions Patients with T2DM and APE have worse long-term prognosis than those with simple APE.The establishment of a prediction model is helpful for clinical evaluation of long-term prognosis.

9.Effects of temperature management based on comfort theory in the prevention of hypothermia in patients undergoing transurethral resection of prostate

Objective:To explore the effect of temperature management based on comfort theory on hypothermia prevention in patients undergoing transurethral resection of prostate (TURP) .Methods:From March to November 2021, 242 patients with benign prostatic hyperplasia who were treated with TURP in Affiliated Jinhua Hospital of Zhejiang University School of Medicine were selected as the study subject by convenience sampling. The patients were divided into control group and observation group according to the method of random number table, 121 patients in each group. The control group carried out routine temperature management, while observation group was given temperature management based on comfort theory on the basis of the control group. We compared the temperature of the two groups when they entered the Operating Room (T 1) , 0.5 h after anesthesia (T 2) , 1 h after anesthesia (T 3) , and at the end of the operation (T 4) . The recovery time of anesthesia was compared between the two groups, and the postoperative complications were counted. Results:The temperature of the observation group at T 2, T 3, T 4 was higher than those of the control group, with statistically significant differences ( P<0.01) . The temperature changes of patients in both groups showed a downward trend from T 1 to T 4. The incidence of shivering and bladder spasm in the observation group were lower than those in the control group, also with statistically significant differences ( P<0.05) . The duration of stay in the Recovery Room, the recovery time of spontaneous respiration and the recovery time of consciousness in the observation group were shorter than those in the control group, with statistical differences ( P<0.01) . Conclusions:Temperature management based on comfort theory can effectively prevent hypothermia in TURP patients, promote postoperative anesthesia recovery, and reduce postoperative complications.
10.Construction of a stable target interfering long non-coding RNA LINC01224 colorectal cancer cell line and its effect on apoptosis

Objective:
To establish colorectal cancer LoVo and SW620 cell lines that stably interfere with the expression of LINC01224, and to explore the effect of down-regulating the expression of LINC01224 on cell apoptosis.
Methods:
The GEPIA2 database was used to analyze the expression of LINC01224 in colorectal cancer tissues; qPCR method was used to detect the expression of LINC01224 in 10 human colorectal cancer cells. Three different LINC01224 siRNAs were respectively transfected into human colorectal cancer LoVo cells, and the LINC01224 shRNA lentiviral vector was constructed with the siRNA sequence with the most obvious inhibitory effect of LINC01224 expression. Recombinant lentiviral particles were packaged in HEK293 T cells and then infected with LoVo and SW620 cells. After selection by puromycin, the monoclonal cells that stably interfere with LINC01224 were obtained by limiting dilution method. MTS method detects cell proliferation ability, and flow cytometry detects cell apoptosis rate.
Results:
The expression of LINC01224 in colorectal cancer tissues was higher than that in normal colorectal tissues, and its expression in 10 types of colorectal cancer cells was also higher than that in normal colorectal epithelial cells HCOEPic. The inhibition rate of siRNA-3 on the expression of LINC01224 in LoVo cells was higher than that of siRNA-1 and siRNA-2. Therefore, siRNA-3 was chosen to design LINC01224 shRNA.Compared with the control group(sh-NC group), the expression level of LINC01224 in the LoVo and SW620 cells of the stable interference LINC01224 group(sh-LINC01224 group) was reduced(P<0.01), and the cell growth rate was slowed down(P<0.01), the rate of apoptosis also increased(P<0.01).
Conclusion
The shRNA lentiviral interference vector of LINC01224 is successfully constructed, which can stably infect LoVo and SW620 cells, down-regulate the expression of LINC01224 and induce cell apoptosis.
