1.Mechanisms of bone marrow mesenchymal stem cells in counteracting D-galactose-induced brain aging

Objective To investigate the effect and potential mechanism of rat mesenchymal stem cells(MSC)on D-galactose-induced brain-tissue aging.Methods A rat brain-aging model was established by injecting D-galactose,and rats in the treatment group received MSC injections via the tail vein.Superoxide dismutase(SOD)activity and malondialdehyde(MDA)levels were assessed in rat brain tissue at the end of the experiment,and pathological changes in brain tissue were observed by hematoxylin-eosin(HE)staining.Expression levels of the inflammatory factors interleukin(IL)-1 and IL-6,the pathway proteins brain-derived neurotrophic factor(BDNF)-tropomyosin receptor kinase B(TrkB),the negative growth regulators p53 and p16,as well as vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF)were observed by polymerase chain reaction(PCR)and Western Blot.Results Brain levels of SOD activity were significantly increased and MDA levels were significantly decreased in rats in the modle group compared with the treatment group(P<0.05).The pathological state of the cerebral cortex and hippocampus were improved and the number of neurons and nucleus pulposus ratio in the brain were increased in the treatment group,as shown by HE staining.Expression levels of IL-1,IL-6,p53,and p16 were significantly decreased,while BDNF,TrkB,VEGF,and bFGF were significantly increased in the treatment group compared with the model group,as shown by PCR and Western Blot(P<0.05).Conclusions These result suggest that MSCs potentially mitigate D-galactose-induced cerebral senescence by concurrently modulating the BDNF-TrkB axis to attenuate oxidative/inflammatory damage,while enhancing the secretion of vasculotrophic(VEGF)and neurotrophic(bFGF)factors for neuronal maintenance.
2.A qualitative study on family fusion experience of breast cancer patients and inspiration for nursing care

Objective To explore the ways of breast cancer patients and family fusion and self-adjustment strategies,and to provide reference information for the adjustment of breast cancer patients and their families.Methods By purposive and theoretical sampling,we selected patients who sought medical treatment at a tertiary hospital from September 2020 to July 2023.The semi-structured interviews were conducted in the suspicion,diagnosis,treatment,and recovery periods,respectively.Charmaz's constructivist grounded theory was used for data analysis.Results The behavioural characteristics of breast cancer patients'fusion with their families throughout the disease trajectory were anxiety transmission,information sharing,emotional connection and experience review.The patient's fusion behaviour can bring dual psychological experiences to the patients,including a sense of relaxation and anxiety,a sense of trust and loss of control,a sense of intimacy and constraint,and a sense of meaning and burden.Self-adjustment strategies,such as staying calm,opening communication,setting boundaries and self-identification,can promote patients'and families'adaptation.Conclusion The adjustment model of breast cancer patients and family fusion suggests medical staff to pay attention to the identification of emotional reactions of patients and their families in family fusion according to the behavioural characteristics of patients and family integration,and timely apply self-adjustment strategies to improve patients'behavioural management ability,reduce negative emotions in the family system,and promote the adjustment of patients and families.

5.Clinical characteristics, diagnosis and treatment of nasal cartilage mesenchymal hamartoma in infants

Objective:To explore the clinical characteristics, pathological features, and diagnosis and treatment strategies of nasal chondromesenchymal hamartoma (NCMH) in infants and young children.Methods:A retrospective analysis was conducted on seven cases of NCMH infants and young children admitted to Beijing Children′s Hospital, Capital Medical University from April 2015 to January 2022. The cohort included 5 males and 2 females, aged from 6 days to 2 years and 3 months. General information, clinical symptoms, imaging findings, treatment plans, postoperative complications, recurrence and follow-up time were collected, summarized and analyzed. Additionally, immunohistochemical characteristics of the lesion were examined.Results:The clinical symptoms of 7 children included nasal congestion, runny nose, open mouth breathing, snoring during sleep, difficulty feeding, and strabismus. All patients underwent electronic nasopharyngoscopy examination, with 5 cases of tumors located in the right nasal cavity and 2 cases in the left nasal cavity. No case of bilateral nasal cavity disease was found. All 7 patients underwent complete imaging examinations, with 5 patients underwent MRI and CT examinations, 1 patient underwent CT examination only, and 1 patient underwent MRI examination only. The CT results showed that all tumors were broad-based, with uneven density, multiple calcifications and bone remodeling, and some exhibited multiple cystic components. The MRI results showed that the tumor showed low signal on T1 weighted imaging and high or slightly high signal on T2 weighted imaging. All patients were diagnosed through histopathological examination and immunohistochemistry, including 7 cases of Ki-67 and SMA (+), 5 cases of S-100 and Vimentin (+), and all EMA and GFAP were negative. All patients underwent endoscopic resection surgery through the nasal approach, with 3 cases using navigation technology. Five cases of tumors were completely removed, and two cases of tumors were mostly removed. No nasal packing was performed after surgery, and no postoperative nasal, ocular, or intracranial complication occurred in all patients. Follow up assessments conducted 6 to 84 months post-surgery revealed no instances of tumor recurrence in any of the patients.Conclusions:The clinical symptoms of children with NCHM mainly depend on the size and location of the tumor. Nasal endoscopic surgery is the main treatment method. In cases where critical structures like the skull base or orbit are implicated, staged surgical interventions may be warranted. Long-term follow-up is strongly advised to monitor for any potential recurrence or complications.
6.Antidepressant effects of Peiyuan Jieyu formula in a mouse model of chronic stress in conjunction with lipopolysaccharide-induced depression

Objective:
To explore the mechanism of the Peiyuan Jieyu formula in treating depression by assessing its impact on a lipopolysaccharide-induced (LPS-induced) depression mouse model.
Methods:
We created a mouse model of depression by exposing mice that had previously received chronic stress to intraperitoneal LPS injections. The mice were divided into the following groups: control, model, fluoxetine, Tiansi Yin, Sini powder, and low-, medium-, and high-dose Peiyuan Jieyu formula groups. Forced swim and tail suspension tests were used to assess the efficacy of the depression (despair) model, and weight gain rates were also measured. Furthermore, serum levels of various depression and inflammation-associated molecules, including tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), tryptophan, 5-hydroxytryptamine, kynurenine (KYN), and kynurenic acid (KA) were assessed. Furthermore, the expression levels of ionic calcium-binding adaptor molecule-1 (IBA-1) and indoleamine 2,3-dioxygenase (IDO) mRNA in hippocampal microglia were measured.
Results:
The model group displayed greater despair-associated immobility, which was shortened in response to various doses of Peiyuan Jieyu formula. Furthermore, formula administration significantly reduced serum TNF-α levels and hippocampal IDO mRNA expression. The high formula dose also reduced IFN-γ and IBA-1 levels, the latter was also decreased in response to the medium formula dose. However, the low formula dose reduced serum KYN level and KYN/tryptophan (TRP) and KYN/KA ratios.
Conclusion
The Peiyuan Jieyu formula holds immense potential in treating depression in a mouse model, potentially inhibiting inflammation and improving TRP-KYN metabolic disorders.
7.Quality Difference Analysis of Raw and Steamed Products of Citri Reticulatae Pericarpium Based on Electronic Eye Technique and Chemical Fingerprint

ObjectiveTo explore the quality differences between steamed products and raw products of Citri Reticulatae Pericarpium(CRP). MethodThe color of steamed products and raw products of CRP was determined from the perspective of appearance by electronic eye technique, and the quality differences between them was objectively characterized by the luminous value(L*), yellow-blue value(b*), red-green value(a*) and total chromatic value(E*ab). Based on this, ultra-high performance liquid chromatography(UPLC) was used to establish a fingerprint evaluation method with the mobile phase of acetonitrile(A)-0.1% formic acid aqueous solution(B) for gradient elution(0-5 min, 5%A; 5-30 min, 5%-20%A; 30-60 min, 20%-52%A), detection wavelength at 270 nm, flow rate of 0.3 mL·min-1 and column temperature of 30 ℃. The quality differences between steamed products and raw products of CRP were compared from the perspective of chemical composition, and correlation analysis was used to reveal the correlation between the difference in appearance color and the difference in internal chemical composition. ResultAfter being steamed, L*, b* and E*ab of CRP showed an overall decreasing trend, indicating that the color of the steamed products darkened and deepened from yellow to blue but still tended to be yellow, while a* showed an overall increasing trend, indicating that the color of the steamed products tended to red. A total of 24 peaks were identified in the fingerprint profiles of raw products and steamed products of CRP, and 13 of the main peaks were identified. The precision, stability and repeatability studies showed that compared with the reference peak (peak 14, hesperidin), the relative standard deviations(RSDs) of the relative peak area and relative retention time of the remaining peaks were<3.0%.The results of chemometric statistical analysis showed that there were some differences between raw products and steamed products of CRP, and 7 main differential components were identified, among which 5-hydroxymaltol(peak 1) and 5-hydroxymethylfurfural(peak 2) were the characteristic components of steamed products. The correlation analysis results showed that, in addition to the above two characteristic components, four components of peak 4, peak 10 (vicenin-2), peak 23 (tangeretin) and peak 24 (5-demethylnobiletin) also correlated significantly with the color change (E*ab) of the samples (P<0.05, P<0.01). ConclusionBefore and after steaming, not only the chemical composition changes, but also the color. Comparing the characteristic peaks of chemical composition difference and color difference before and after steaming of CRP, it is found that 5-hydroxymaltol, 5-hydroxymethylfurfural and peak 4 are common characteristic difference components, which can provide a reference for establishing the characteristic quality control method of steamed products, and quickly evaluating the quality difference between raw products and steamed products of CRP.
8.Experience of young cancer patients using mobile health applications: a qualitative research

Objective:To explore the experience of young cancer patients using mobile health applications, so as to provide a basis for improving the utilization rate and sustainability of health applications.Methods:From May to June 2020, purpose sampling was used to select 12 young cancer patients in the Internal Medicine Ward of a ClassⅢ Grade A cancer hospital in Nanjing, Jiangsu Province. The semi-structured interview with patients was conducted with the descriptive phenomenological method. The Colaizzi 7-step analysis method was used to analyze the data and refine the theme.Results:A total of four themes were summarized, namely, there was a difference in usage attitudes (being used for rehabilitation management, as a means of verification, generating resistance, and reducing the frequency of use) , insufficient function development and reduced user experience (lack of autonomy, insufficient intelligent evaluation) , desiring a professional operating agency, recognizing the development prospects and looking forward to perfection.Conclusions:In follow-up studies, researchers need to fully consider the patient's preference and usage habits for health applications on the basis of meeting the needs of disease management.
9.The experience of self-identity in young adults with cancer: a qualitative study

Objective:To explore the real experience of developing self-consciousness, self-discovery, self-identity crisis and self-cognition improvement of young cancer patients from the initial stage of diagnosis, and to provide reference for medical staff to construct the intervention mode of psychosocial support.Methods:Using phenomenological method in qualitative research, 14 young cancer patients aged 18-25 years from April 2019 to August 2020 in Jiangsu Cancer Hospital were interviewed in semi-structured way separately at three time points: initial diagnosis, 6 months after diagnosis and 12 months after diagnosis. 7-step Colaizzi analysis was used to analyze the data.Results:Three themes were extracted as following. The dilemma of self-cognition, self-identity biases and the exploration of self-identity.Conclusions:Attention should be paid to the psychological problems of young cancer patients at all stages, and meet the needs of patients to talk, cooperate with professional psychotherapists, encourage patients to recognize themselves, tap the inner positive power, so as to improve their sense of self-identity.
10.The value of combined detection of AFP ,GGT and GP73 in the diagnosis of early primary liver cancer

Objective To investigate the value of combined detection of alpha fetoprotein (AFP) ,gamma-glutamyl transferase (GGT)and Golgi glycoprotein (GP73)in the diagnosis of primary hepatocellular carcino-ma.Methods 109 cases of primary liver cancer treated in the hospital from June 2015 to June 2017 were en-rolled in the study as primary liver cancer group ,76 cases of liver cirrhosis were enrolled in the study as liver cirrhosis group and 70 cases of healthy people who underwent healthy assessment were enrolled in the study as control group.The peripheral venous blood in each group was extracted fasting in the morning ,and the ser-um was separated.The levels of GP73 were measured by enzyme linked immunosorbent assay.The level of AFP was measured by chemiluminescent immunoassay ,and the level of GGT was measured by rate method. Results The serum levels of AFP ,GGT and GP73 in the primary liver cancer group were higher than those in the liver cirrhosis group and the control group ,and the serum levels of AFP ,GGT and GP73 in the liver cir-rhosis group were higher than those in the control group ,and the differences were statistically significant (P<0.05).The specificity and the sensitivity of the combined detection of AFP ,GGT and GP73 were higher than those of single detection of AFP ,GGT ,and GP73.Conclusion The combined detection of AFP ,GGT and GP73 has important value in the diagnosis of early primary liver cancer ,and has relatively high sensitivity and specificity ,which is worthy of further clinical study.
