Objective To investigate the effect and mechanisms of celecoxib on right heart function in mice with acute high-altitude hypoxia exposure. Methods Male C57BL/6J mice(7 weeks old)were housed in a hypobaric chamber simulating an altitude of 5 800 m for 2 d to establish an animal model of acute hypobaric hypoxia.①Eighteen mice were randomly assigned to plain+saline(P+S),high-altitude hypoxia exposure+saline(H+S),and high-altitude hypoxia exposure+celecoxib(H+Cel).Body weight and routine blood indicators were measured, and cardiac ultrasound examination were performed for heart rate(HR),pulmonary artery acceleration time to ejection time ratio(AT/ET),tricuspid annular plane systolic excursion(TAPSE),tricuspid annular systolic velocity(S'),and left ventricular ejection fraction(LVEF)and fractional shortening(FS).Targeted metabolomic profiling was applied to detect the cardiac arachidonic acid(AA)metabolite levels. The contents of 12,13-dihydroxy-9Z-octadecenoic acid(12,13-diHOME)in the heart, liver, brown adipose tissue, and plasma were quantified by ELISA.② Eighteen mice were randomly assigned into plain+saline(P+S),high-altitude hypoxia exposure+saline(H+S)and high-altitude hypoxia exposure+12,13-diHOME(H+di)groups.Body weight, routine blood tests, and echocardiography were performed as above.③ Thirty-two mice were randomly divided into high-altitude hypoxia exposure+saline(H+S),high-altitude hypoxia exposure+celecoxib(H+Cel),high-altitude hypoxia exposure+soluble epoxide hydrolase inhibitor(sEHI)(H+sEHI),and high-altitude hypoxia exposure+sEHI+celecoxib(H+sEHI+Cel)groups.Body weight, routine blood tests, and echocardiography were performed as above.Cardiac and plasma contents of 12,13-diHOME and epoxyeicosatrienoic acids(EETs)were measured by ELISA.Results ① Compared to the P+S group, the H+S group exhibited significantly reduction of cardiac 12,13-diHOME level(P<0.001),increased counts of white blood cells(WBC)and neutrophils(P<0.01)and decreased TAPSE,S'and AT/ET both at resting state and under stress(P<0.01,P<0.001).Compared to the H+S group, the H+Cel group exhibited significantly increase of cardiac 12,13-diHOME level(P<0.05),reduced WBC and lymphocyte counts(P<0.01,P<0.05)and improved TAPSE and S'levels at resting state and under stress(P<0.01,P<0.001).② Compared to the H+S group, the H+di group demonstrated significantly improvement of TAPSE at basal and under stress(P<0.001)and a trend towards improved TAPSE at resting state(P=0.0532),but no obvious differences was observed in WBC and neutrophil counts between the H+di group and the H+S group.③ Compared to the H+Cel group, both the H+sEHI and H+sEHI+Cel groups exhibited significantly reduction of cardiac 12,13-diHOME level(P<0.01,P<0.05)though no statistical changes in cardiac function indicators.Compared to the H+S group,WBC counts and lymphocyte were decreased, and serum EETs level was incrased in the H+Cel group,H+sEHI group and H+sEHI+Cel group(P<0.01,P<0.001).Conclusion Celecoxib can elevate cardiac level of 12,13-diHOME and improves right heart function in mice after acute high-altitude hypoxia exposure through the CYP450-sEH metabolic pathway.

Objective This study aims to clarify the potential categories of mindfulness of psychiatric nurses through latent profile analysis and to analyse the influencing factors in order to provide a reference for the subsequent design of brief and targeted mindfulness interventions to reduce work stress and improve mental health of psychiatric nurses. Methods 300 psychiatric nurses from a mental health center in Liaoning Province were selected by convenience sampling method to investigate the level of mindfulness through a general demographic questionnaire(gender, age, marital status, educational background, illness, etc.)and the short version of the Five Facet Mindfulness Questionnaire(FFMQ-SF).SPSS Statistics 21.0 was used for descriptive statistics and correlation analysis of variables, and Mplus was used for latent profile analysis(LPA)to explore the potential categories of mindfulness characteristics of psychiatric nurses. The categorized data were subjected to χ2 test and one-way ANOVA to compare the demographic information and differences in mindfulness levels among nurses in different categories, and multifactorial logistic regression was used to analyze the factors affecting the categories of mindfulness characteristics of psychiatric nurses. Results A total of 300 psychiatric nurses were included in the study, with a mean age of 37.98±9.84 years and a mean total mindfulness score of 67.27±7.43.The psychiatric nurses could be categorized into four groups according to their level of mindfulness:the high mindfulness group(38.0%),the nonjudgmental group(36.3%),the judgmental low awareness(5.0%),and the observationally described group(20.7%).Multiple logistic regression analysis showed that, compared with the high mindfulness group, nurses with younger age had a greater likelihood of being in the nonjudgmental group and the judgmental low awareness group;nurses with heavy workload had a greater likelihood of being in the nonjudgmental group;nurses with a monthly income of≤5000 yuan had a greater likelihood of being in the observational descriptive group;and nurses with underlying diseases had a greater likelihood of being in the nonjudgmental group and the observational descriptive group. There were also differences in the total level of mindfulness and the five subdimension scores for nurses in different mindfulness categories. Conclusion There were four potential categories of psychiatric nurses characteristics of mindfulness.Age,gender, monthly income, underlying illness, and workload were all factors that influenced the classification of psychiatric nurses into the high mindfulness group.

OBJECTIVE:To study the effects of different penetration enhancers on in vitro transdermal permeation of Flavaspidic acid BB cream. METHODS :Flavaspidic acid BB cream was prepared ,containing 11 kinds of different penetration enhancers as 1% azone,2% azone,3% azone,4% azone,1% menthol,1% propylene glycol ,1% oleic acid ,1% azone+1% menthol,1% azone+1% propanediol,1% azone+1% oleic acid or 1% menthol+1% propanediol. Modified Franz diffusion cell was adopted using abdominal skin of isolated male rat as transdermal barrier. The content of flavaspidic acid BB was determined by UPLC. The accumulative transdermal amount (Q24 h)and percutaneous permeability (Jss)within 24 h were calculated ;and compared with Flavaspidic acid BB cream without transdermal enhancer ,the enhancement ratio (ER)was calculated. RESULTS : Q24 h of Flavaspidic acid BB cream with above 11 kinds of transdermal enhancers were (82.96±7.15),(80.17±0.66),(78.22± 1.87),(73.53±1.24),(35.65±2.23),(34.02±1.73),(42.68±2.66),(33.94±1.37),(34.16±1.54),(46.78±1.21),(43.66±1.69) μg/cm2,respectively. Jss value were (5.26±0.10),(4.69±0.12),(4.45±0.45),(4.00±0.06),(3.74±0.33),(3.23±0.18), (3.73±0.53),(3.14±0.47),(3.54±0.11),(3.98±0.34),(4.34±0.14)μg(/ cm2·h),respectively. ER were 2.055,1.831,1.738, 1.564,1.462,1.263,1.456,1.227,1.385,1.557,1.698,respectively. CONCLUSIONS :All of the above transdermal absorption enhancers can enhance the percutaneous absorption of Flavaspidic acid BB cream ,among which ,1% azone is the best.
