1.Current research status of ferroptosis in endometriosis

Endometriosis presents a common challenge in global women's health.Ferroptosis,as an emerging form of regulated cell death,has recently attracted attention in relation to endometriosis.In terms of disease mechanisms,ferroptosis drives ovarian endometrial fibrosis via the induction of iron overload,and promotes disease progression by regulating multiple signaling pathways,such as p38 mitogen-activated protein kinase/signal transducer and activator of transcription 6(p38MAPK/STAT6),promote angiogenesis,autophagy,etc,fueling disease progression.In terms of infertility,ferroptosis affects embryo development and ovarian function,thereby reducing fertility.From a diagnostic perspective,high expression of ferroptosis-related genes provides potential biomarkers for the early diagnosis of endometriosis,effectively distinguishing patients from healthy individuals,and showing important clinical value.In terms of treatment strategies,non-natural compounds such as Erastin,as well as natural compounds such as resveratrol,ursolic acid,and baicalein,have shown potential therapeutic effects in relieving the pathological process and related symptoms of endometriosis.This review comprehensively elucidates the key role of ferroptosis in endometriosis in terms of the disease mechanisms,infertility,diagnosis,and treatment,and explores its potential as a diagnostic biomarker and therapeutic target,thus providing new theoretical support and treatment strategies for the management of endometriosis.

3.Long non-coding RNA CTC-338M12.4 inhibition on activation of JAK/STAT signaling pathway via miRNA-27a-5p makes cell cycle arrest, cell proliferation and migration inhibition in tongue squamous cell carcinoma

Objective:To investigate the expression level of long non-coding RNA (lncRNA) CTC-338M12.4 in tongue squamous cell carcinoma tissues, and its effects on the cell cycle, cell proliferation, and migration of tongue squamous cell carcinoma cells in vitro as well as its molecular mechanisms.Methods:The Gene Expression Omnibus (GEO) database was used to obtain the lncRNA series data set GSE139869, and the differential expression of CTC-338M12.4 in tongue squamous cell carcinoma tissues and adjacent tissues was analyzed. The transcriptional expression levels of CTC-338M12.4 in human immortalized oral keratinocytes (HOK) and tongue squamous cell carcinoma cell lines HN13, TSCCa, CAL-27, Tca8113, SCC15 were detected by using reverse transcription quantitative real-time polymerase chain reaction (qRT-PCR). CAL-27 cells with the lowest expression level of CTC-338M12.4 were selected and were divided into the control group (co-transfected with vectors containing negative sequence) and CTC-338M12.4 group (co-transfected with CTC-338M12.4 overexpression vectors). The proliferation ability of CAL-27 cells in each group was detected by using cell colony formation assay, and the cell cycle distribution of CAL-27 cells was detected by using flow cytometry. The migration ability of CAL-27 cells was detected by using scratch test. The lncACTdb database was used to predict the complementary binding sites between CTC-338M12.4 and miRNA-27a-5p (miR-27a-5p), and dual-luciferase reporter gene assay was used to verify. The expression level of miR-27a-5p in CAL-27 cells of all groups was detected by using qRT-PCR. The protein expression level of related factors on JAK/STAT signaling pathway in CAL-27 cells of all groups was detected by using Western blot.Results:Analysis of GEO database data showed that transcriptional level CTC-338M12.4 in tongue squamous cell carcinoma tissues was lower than that in adjacent tissues, and the difference was statistically significant ( P < 0.01). Transcriptional level CTC-338M12.4 in tongue squamous cell carcinoma HNl3, TSCCa, CAL-27, Tca8113, and SCC15 cells was lower than that in HOK cells, and the differences were statistically significant (all P < 0.05). The transcriptional level relative expression level of CTC-338M12.4 in CAL-27 cells in the CTC-338M12.4 group was higher than that in the control group, and the difference was statistically significant ( P < 0.01). Cell colony formation assay showed that the colong number of CAL-27 cell in the CTC-338M12.4 group was less than that in the control group [(51±10) vs. (114±21)], and the difference was statistically significant ( t = 2.71, P = 0.035). Flow cytometry showed that the proportion of G 0/G 1 phase cells in CAL-27 cells in the CTC-338M12.4 group was higher than that in the control group [(64±3)% vs. (43±4)%], and the difference was statistically significant ( t = 4.87, P = 0.003). The scratch test showed that when scratching, the scratch width of both groups was similar ( P > 0.05); after scratch for 25 h, the scratch width of CAL-27 cells in the CTC-338M12.4 group was wider than that in the control group [(133±15) μm vs. (64±10) μm], and the difference was statistically significant ( t = 3.78, P = 0.009). The dual luciferase reporter gene assay showed that the relative luciferase activity of CAL-27 cells co-transfected with wild-type CTC-338M12.4 sequence and miR-27a-5p sequence was lower than that of CAL-27 cells co-transfected with wild-type CTC-338M12.4 sequence and miR-27a-5p irrelevant sequence, and the difference was statistically significant ( P < 0.001). The relative expression level of transcriptional level miR-27a-5p of CAL-27 cells in the CTC-338M12.4 group was lower than that in the control group, and the difference was statistically significant ( P = 0.003). Western blot showed that the protein expression levels of JAK/STAT signaling pathway p-JAK, p-STAT, p-Raf, p-ERK, and p-mTOR were lower than those in the control group. Conclusions:The level of lncRNA CTC-338M12.4 is low in tongue squamous cell carcinoma. CTC-338M12.4 mediates the inactivation of JAK/STAT signaling pathway via inhibiting miR-27a-5p expression in tongue squamous cell carcinoma CAL-27 cells, thereby leading to cell cycle arrest and inhibiting the cell proliferation and migration of CAL-27 cells.
4.Present situation and hotspots of KM mouse research based on the CiteSpace map of scientific knowledge

Objective The present situation and hotspots of KM mouse research were visualized by CiteSpace software,which provided a reference for breaking through the bottleneck of KM mouse research and finding new ideas in China.Methods A total of 3981 articles were retrieved from CNKI and Web of Science databases from January 1,2004 to June 10,2023,of which the number of publications,countries(regions),institutions,author cooperation network,keyword co-occurrence,clustering,burst words,and cited English literature for analysis.Results The number of documents published in Chinese was decreased and the number of documents published in English was increased gradually in KM mouse research.The three countries with the most published articles were China,the United States,and Japan,where the leading research institutions were the China Agricultural University,Chinese Academy of Sciences,and Nanjing University of Chinese Medicine.The main authors at home and abroad were Liu Weihui,Li Guangwu,and Zhao Xin.The domestic research focused on the toxicology,immunology,and reproduction of KM mouse,and the foreign research focused on the cell biology and physiology of KM mouse.Conclusions KM mouse have been used and popularized in our country for more than 70 years,mainly focusing on toxicology,immunology,and reproduction.Although the use of KM mouse has led tove made allowed some achievements in experiments,certain it is necessary to solve their problems in the future,such as the standardization of population breeding,the discovery of the dominant research fields,and the further promotion of animal ethics,should be resolved.
5.Research status and trend of depression models in animals:a bibliometric analysis based on CiteSpace

Objective Using literature metrology,scientific literature in the field of depression models in animals was analyzed to better understand the development trends and hot topics in this field.Methods We obtained publications on depression and animal models from 2013 to 2022 from the Web of Science core set database.CiteSpace 6.1 R1(64-bit)Basic was used to analyze annual publications,countries,institutions,authors,and keywords related to this field.Results A total of 1000 articles were included in this study.From 2013 to 2022,the number of articles published increased gradually and then stabilized.In terms of the number of articles,the United States had the most published articles(256).Wegener Gregers was the most influential author in the field with 23 published articles.Conclusions The field focuses on signaling pathways and therapeutic approaches to determine the pathogenesis of depression and better treatments.This study provides a visual analysis of trends in depression research to help researchers keep up with the latest developments.
6.Effects of task-oriented balance training on balance dysfunction in patients with ankle sprain

Objective:To explore the effect of task-oriented training on balance dysfunction in patients with ankle sprain.Method:Forty subjects with ankle sprain were divided into experimental group (group A) and control group (group B) according to the random number table method,with 20 cases in each group. On the basis of rou-tine training in both groups,group A was given task-oriented balance training,and group B was given bal-ance board training. Two groups of subjects performed static balance function[LOS:maximum excursion (MXE),movement velocity(MVL),reaction time (RT)],dynamic balance (Y-Balance) and Cumberland An-kle Instability Tool (CAIT) assessment before and after training for 4 weeks.Result:Before treatment,there was no significant difference in the scores of limbs between the two groups(P>0.05).After 4 weeks of training,the balance function MXE,MVL and RT scores of group A were significantly im-proved compared with those before treatment(P<0.05);but there was no significant difference in the scores of group B(P>0.05). The Y-balance scores of group A and group B after treatment were improved compared with those before treatment,and the difference was statistically significant(P<0.05). The CAIT scores of group A were improved compared with those before treatment(P<0.05),and those of group B were improved but had no signifi-cant difference(P>0.05). The changes in Y-balance and CAIT scores of group A were better than those in group B (P<0.05);there was no significant difference in the changes of LOS score between group A and group B(P>0.05).Conclusion:Task-oriented training can improve the balance dysfunction and motor function of patients with an-kle sprain.

8.Effect of Gegen Qinliantang-medicated Serum on Nonalcoholic Steatohepatitis Based on Nrf2/TXNIP Signaling Pathway

ObjectiveTo investigate the effect of Gegen Qinliantang (GGQLT)-medicated serum on free fatty acid (FFA)-induced nonalcoholic steatohepatitis (NASH) in vitro model of human hepatoma cells HepG2. MethodNASH model of HepG2 cells was established in vitro, and the cells were intervened with different volume fractions of GGQLT-medicated serum and resveratrol. Intracellular lipid deposition in each group was detected by oil red O staining, the level of reactive oxygen species (ROS) in each group were detected by flow cytometry, the levels of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), triglyceride (TG) and malondialdehyde (MDA) in each group were detected by kits. Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR) was used to measure the mRNA expression levels of nuclear transcription factor (NF)E2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), quinone oxidoreductase 1 (NQO1), Kelch-like epichlorohydrin-associated protein-1 (Keap1), NF-κB, thioredoxin interacting protein (TXNIP), interleukin-1β (IL-1β) in HepG2 cells of each group. The protein expression of Nrf2, TXNIP in cells of each group was detected by Western blot. ResultFFA induced large accumulation of intracellular lipids. Compared with the normal group, the activities of GSH-Px and SOD were significantly decreased (P<0.01) and the contents of TG, ROS and MDA were significantly increased (P<0.05, P<0.01) in the model group. Compared with the model group, all GGQLT groups and resveratrol group could elevate intracellular SOD activity to different degrees (P<0.05, P<0.01) and significantly reduce the levels of intracellular ROS and MDA (P<0.05, P<0.01), GGQLD high- and medium-dose groups and resveratrol group significantly elevated GSH-Px activity (P<0.01), GGQLD medium- and low-dose groups and resveratrol group significantly decreased TG content (P<0.05, P<0.01). Compared with the model group, GGQLT high- and medium-dose groups and resveratrol group could significantly upregulate the mRNA expression levels of Nrf2, HO-1 and NQO1 (P<0.01), all GGQLT groups and resveratrol group could significantly downregulate the TXNIP protein expression level, as well as significantly downregulate the mRNA expression levels of Keap1, NF-κB (P<0.05, P<0.01). Nrf2-siRNA transfection of cells revealed that Nrf2 expression was significantly downregulated (P<0.01) in the Nrf2-siRNA group of cells by comparing with NC-siRNA group at the corresponding dose of drugs, and the inhibitory effects of GGQLT and resveratrol on TXNIP, IL-1β were attenuated. ConclusionFFA induces the production of ROS and inflammatory factors in HepG2 cells, and GGQLT can improve the anti-inflammatory and antioxidant capacities of cells, and its mechanism may be related to the regulation of Nrf2/TXNIP signaling pathway, so as to improve NASH.
9.Clinicopathological features and endoscopic treatment in patients with portal hypertension and gastroesophageal varices with unknown etiology

Objective:To analyze the clinicopathological characteristics of patients with unknown etiology of portal hypertension and investigate the efficacy of endoscopic management of gastroesophageal varices in these patients.Methods:Patients with unknown etiology of portal hypertension and gastroesophageal varices who received liver biopsy between January, 2017 and January, 2020 in Zhongshan Hospital were included. The characteristics of pathology, portal computed tomography (CT) angiography, and endoscopy were recorded and follow-up for the occurrence of bleeding after treatment.Results:A total of 31 patients were included and divided into cirrhosis with unknown etiology group ( n=10) and non-cirrhotic portal hypertension group ( n=21). Patients in the non-cirrhotic group were younger [28.0(29.5-49.5) vs 58.5(43.5-65.8), P=0.004] and mostly male (71.4%), and fewer comorbidities including diabetes (4.8% vs 40.0%, P=0.027). The features of pathology finding including vasculopathy, cholestasis, and hepatic sinusoidal dilatation as well as the Sarin classification and bleeding rate of gastroesophageal varices, proportion of patients receiving endoscopic treatment were shown similar between the two groups ( P>0.05). The hepatic venous pressure gradient (HVPG) was significantly lower in the non-cirrhotic group [4.5(2.8-12.8)mmHg vs 12(8-18)mmHg, P=0.018]. Among them, 21 patients received endoscopic treatment, and the bleeding rate had no difference between these two groups after endoscopic treatment ( P=0.751). Conclusions:Non-cirrhotic portal hypertension in a predominantly young male population has similar clinicalpathological characteristics when compared to cirrhotic portal hypertension with unknown etiology. HVPG can not reflect the actual portal pressure in these patients. Endoscopic treatment is the effective treatment option for the prevention of variceal bleeding.
10.Clinical experience in transoral endoscopic thyroidectomy via sublingual and vestibular approach: a report of 98 cases

Objective:To summarize the clinical experience in transoral endoscopic thyroidectomy via sublingual and vestibular approach (TOETSLVA).Methods:We retrospectively reviewed the medical record of patients who underwent TOETSLVA in our department from November 2011 to May 2020, including 3 males and 95 females, aged from 18 to 57 years old. Initial 81 cases were categorized in "Period A (November 2011-November 2015)" and subsequent 17 cases in "Period B (August 2019-May 2020)" . Data about demographics, operation time and complications were collected. SPSS 22.0 software was used for statistical analysis.Results:In Period A, the average age of patients was (34.2±9.4) years old; the mean tumor diameter was (2.33±0.80) cm; postoperative pathology showed benign nodules in 76 cases and malignant carcinoma in 5 cases; there were 65 cases of unilateral subtotal thyroid lobectomy, 6 cases of isthmus lobectomy, and 5 cases of bilateral subtotal thyroid lobectomy, with the mean operation time of (132.70±47.22) min; in 5 cases of unilateral thyroid lobectomy with central lymph node dissection, the mean operation time was (185.4±31.40) min; postoperative neck infections occurred in 6 cases; temporary vocal cord paralysis occurred in 1 case, which it recovered within two months; and CO 2 gas embolism occurred in 2 cases. In Period B, the average age of patients was (35.1±8.5) years old; mean tumor diameter was (1.32±0.67) cm; postoperative pathology indicated malignant nodules in 15 cases and benign nodules in 2 cases; in 2 cases of unilateral thyroid lobectomy, the mean operation time was (153.5±34.64) min; in 15 cases of unilateral thyroid lobectomy with central lymph node dissection, the mean operation time was (123.73±14.26) min; and none of patients developed postoperative neck infections or CO 2 gas embolism. All patients had different degree of cutaneous numbness in the submandibular region after surgery, which recovered within 1-2 weeks. There were no complications such as postoperative secondary hemorrhage, permanent vocal cord paralysis, hypoparathyroidism in both the periods. The median follow-up time was 86 months (57-105 months) in Peroid A and 5 months (3-12 months) in Peroid B. During the follow-up periods, there were no obvious abnormalities in swallowing, chewing, oral sensory function and neck activity, and also no tumor recurrence or metastasis. Conclusions:TOETSLVA is a safe and feasible surgery method, with a good cosmetic result. This approach will not lead to a postoperative cutaneous numbness of the submandibular region for a long time.
