1.Analysis of T cell and natural killer cell characteristics in cerebrospinal fluid of multiple sclerosis patients based on single-cell transcriptome sequencing

Objective:To explore the characteristics of T cells and natural killer (NK) cells in cerebrospinal fluid of patients with multiple sclerosis (MS).Methods:Cerebrospinal fluids from patients with relapsing-remitting multiple sclerosis (RRMS) and healthy controls attending the Second Hospital of Lanzhou University from January 2023 to October 2024 were collected and analyzed by single-cell RNA sequencing (scRNA-seq) and flow cytometry, and T and NK cell characteristics were summarized and compared in the two groups. The proposed time-series analysis was used to explore the differentiation trajectories of T cells and NK cells.Results:A total of 3 patients with RRMS and 3 healthy controls underwent scRNA-seq, and 6 patients with RRMS and 4 healthy controls underwent flow cytometry. The composition of cerebrospinal fluid T and NK cell subtypes was similar in the RRMS group and the healthy control group, but the proportion of the cell subtypes differed. The RRMS group exhibited significantly higher frequencies of CD4 +Naive T- IL7R [1 529/9 055(16.89%) vs 1 423/9 910(14.36%), χ2=22.980, P<0.001], CD4 +Naive T- CCR7 [1 573/9 055(17.37%) vs 948/9 910(9.57%), χ2=250.114, P<0.001], and CD4 +Naive T- LTB [1 369/9 055(15.12%) vs 1 079/9 910(10.89%), χ2=75.336, P<0.001] subsets compared to controls. Conversely, the control group demonstrated greater proportions of CD4 +Th1- GZMA [1 255/9 910(12.66%) vs 719/9 055(7.94%), χ2=113.213, P<0.001] and CD8 +Tem- GZMK cells [1 607/9 910(16.22%) vs 1 232/9 055(13.61%), χ2=25.326, P<0.001] than the RRMS group. The transcription factors and gene expression of each T cell subtype were also different between the 2 groups, and CD4 initial T cells and CD8 effector T cells were located at the beginning and the end of the differentiation trajectory, respectively. Conclusions:The cerebrospinal fluid of MS patients is characterized by increased expression of genes involved in T cell differentiation and over-activation of immune cells.
2.Effect of Yishen Paidu Formula on inflammatory response in chronic renal failure rats by regulating Calcineurin/NFAT signal pathway

Objective:To explore effect of Yishen Paidu Formula on inflammatory response in chronic renal failure(CRF)rats and role of Calcineurin/nuclear factor of activated T cell(NFAT).Methods:CRF rat model was constructed,and randomly grouped into model group,low,medium and high doses Yishen Paidu Formula groups,with 10 rats in each group,another 10 rats were fed normally as a blank group;general situation of rats was recorded;urine protein quantification kit was applied to detect 24-hour urine protein level of CRF rats in each group;serum creatinine and urea nitrogen levels of CRF rats were detected;ELISA was applied to detect serum IL-1β,IL-6 and TNF-α levels in CRF rats;pathological changes of renal tissue were observed by HE staining;Western blot was applied to detect expressions of fibroblast growth factor 23(FGF23),Klotho protein,Calcineurin,NFAT protein in renal tissue of CRF rats in each group.Results:Compared with blank group,levels of creatinine,urea nitrogen in serum,24 h urine protein in urine,IL-1β,IL-6,TNF-α in serum,and protein levels of FGF23,Calcineurin,NFAT in kidney tissue were obviously increased in model group,level of Klotho protein was obviously decreased(P<0.05).Compared with model group,levels of creatinine,urea nitrogen in serum,24 h urine protein in urine,IL-1β,IL-6,TNF-α in serum,and protein levels of FGF23,Calcineurin,NFAT in kidney tissue were obviously decreased in low,medium and high doses Yishen Paidu Formula groups,level of Klotho protein was obviously increased,which were more significant with dosage increase(P<0.05).Conclusion:Yishen Paidu Formula may alleviate inflammatory response in CRF rats by inhibiting Calcineurin/NFAT signaling pathway.

5.Accuracy assessment of refractive status in patients implanted with extended depth of focus intraocular lens

AIM: To evaluate the refractive status through computer refractometer and OPD-Scan III auto refractometer in cataract patients after extended depth of focus(EDOF)intraocular lens implantation.METHODS: Retrospective observational study. A total of 61 cases(76 eyes)that received phacomulsification and implanted with TECNIS® Symfony ZXR00 intraocular lens in Shanghai Eye Diseases Prevention & Treatment Center from May 2022 to May 2023 were collected. Measurements from the computer refractometer, OPD-Scan III auto refractometer, and subjective refraction, were taken from all patients on the same day postoperatively.RESULTS: There were statistical significant difference in sphere(S)and spherical equivalent(SE)readings from the computer refractometer and subjective refraction(all P<0.01), with mean differences of -0.67±0.37 D and -0.75±0.35 D, respectively, and the S and SE obtained from computer refractometer more incline to myopia than those from subjective refraction; there were statistical significant difference in computer refractometer and subjective refraction(P<0.01), with a relative small absolute difference(0.21±0.24 D). The S, cylinder(C)and SE of computer refractometer(S, C, SE)were positively correlated with subjective refraction(r=0.7994, 0.7929, and 0.8118, respectively, all P<0.01). Additionally, there were statistical significant differences in S, C and SE of OPD-Scan Ⅲ and subjective refraction(P<0.01), and the absolute differences of S(0.63±0.36 D), C(0.35±0.26 D)and SE(0.53±0.36 D)were small. Furthermore, the S, C and SE of OPD-Scan Ⅲ were positively correlated with subjective refraction(r=0.4410, 0.4982, 0.5224, all P<0.01).CONCLUSION: In patients who received implantation of EDOF lenses, the consistency of computer refractometer, OPD-Scan III auto refractometer and subjective refraction was good. The average difference of the S and SE obtained via computer refractometer was large, but both exhibited a myopic shift relative to those derived from subjective refraction, and the C values demonstrated minimal discrepancy. Furthermore, the differences between OPD-Scan III auto refractometer and subjective refraction were small, but the direction of the difference is unstable, sometimes it is myopic deviation, while sometimes it is hyperopic deviation.
6.Sampling inspection results of disinfection products in Shaanxi Province in 2020 - 2023

Objectives To understand the quality status of disinfection products in the market in Shaanxi Province, and to provide a basis for taking targeted management measures. Methods From 2020 to 2023, in accordance with the national disinfection product management regulations and standards, the physicochemical and microbial tests of disinfection products in Shaanxi Province were carried out through supervised sampling, and the test results were analyzed according to the active ingredients, illegal addition, stability, and bactericidal or bacteriostatic performance. Results The overall qualification rate of the active ingredient content of disinfectants sampled in Shaanxi Province was 90.48%, with the lowest qualification rate in 2023 (84.38%). The overall qualification rate of quantitative sterilization test of disinfectants was 90.12%, showing a decreasing trend year by year (P=0.272). The overall qualification rate of anti-bacterial products was 88.59%, and the bactericidal test results of 20 antibacterial products were all qualified. The overall qualification rate of bacteriostatic performance testing for bacteriostatic products was 86.17%, with the pass rate of bacteriostatic product germicidal efficacy testing of different components in descending order being quaternary ammonium salt/silver ion > guanidine > others > lysozyme, and there was statistically significant difference (P=0.004). The detection rate of illegal additives in antimicrobial products was 7.07%, with the main detection indicators being miconazole nitrate, clobetasol propionate, and dexamethasone acetate. Conclusion The qualified rate of disinfection products in Shaanxi Province is relatively high, but there is a downward trend. It is necessary to continue to strengthen the daily supervision and product testing of disinfection product manufacturers, and promote the continuous improvement of disinfection product quality.
7.Analysis on the spatial-temporal distribution characteristics of hepatitis B incidence in a county of western Liaoning province from Jan 2011 to Dec 2021

Objective:To understand the incidence and spatial-temporal distribution characteristics of hepatitis B among residents in a county of western Liaoning province from Jan 2011 to Dec 2021.Methods:Data on hepatitis B incidence were obtained from the Disease Prevention and Control Center of the county,and its crude incidence and standardized incidence were calculated.ArcGIS software was used to analyze the global spatial autocorrelation analysis and local spatial autocorrelation analysis of hepatitis B incidence in the county.A space-time scan analysis was performed using SaTScan software to identify the case distribution and spatiotemporal clustering patterns of hepatitis B incidence.Results:From Jan 2011 to Dec 2021,there were 2 978 cumulative cases of hepatitis B in the county,with a crude incidence of 69.25 per 100 000 and a standardized incidence of 70.13 per 100 000.Among different regions,Toutai Manchu Town had the highest crude incidence of hepatitis B,followed by Gaotaizi Town and Chengguan Street.The global autocorrelation showed an aggregated distribution in 2014,2016,2019,and 2021.The local autocorrelation hotspots were clustered in the central and western parts of the county,the H-H clustering areas included Toutai Manchu Town and Jiudaoling Town from 2014 to 2016,Toudaohe Manchu Town,Yizhou Street and Chengguan Street from 2017 to 2019,and Qianyang Town and Yizhou Street from 2020 to 2021.The L-L clustering areas were mostly concentrated in the southwest of the county,including Dizangsi Manchu Township,Dadingbao Manchu Township and Liulonggou Manchu Township.A total of 4 clustering areas were obtained by temporal-spatial scanning analysis,the first-level area was Zhangjiapu Township in 2017,the second-level area included Qianyang Town and Chengguan Street from 2011 to 2015,the third-level area was Gaotaizi Town from 2013 to 2016,and the fourth-level area was Toutai Manchu Town from 2014 to 2017.Conclusions:From Jan 2011 to Dec 2021,the incidence of hepatitis B among residents in a county in western Liaoning Province is mainly concentrated in the central and northwest regions.The hepatitis B cases are clustered in the central and western areas of the county,and has a trend of gradually spreading to the northern areas over time.It is necessary to pay attention to the townships with high incidence of hepatitis B and carry out effective prevention and surveillance.
8.Clinical characteristics of 315 Han and Uyghur ulcerative colitis patients in a single center in Xinjiang Uygur Autonomous Region

Objective:To analyze the clinical characteristics of ulcerative colitis (UC) in Han and Uyghur patients in Xinjiang Uygur Autonomous Region.Method:Data of UC patients hospitalized at the People′s Hospital of Xinjiang Uygur Autonomous Region from January 2015 to June 2023 were retrospectively collected, with a focus on the differences in basic information, extraintestinal manifestations and biochemical laboratory indicators between Han and Uyghur patients.Results:A total of 315 UC patients were included, with 172 Han cases (54.6%) and 143 Uyghur cases (45.4%) . Compared to the Han group, the Uyghur group had a higher proportion of females, a lower average age, and lower rates of smoking and alcohol consumption (all P<0.05) . The Uyghur group showed a higher prevalence of hepatobiliary diseases and joint injuries as extraintestinal manifestations, although the differences were not statistically significant ( P all>0.05) . The Han group had a statistically significant higher level of intraepithelial neoplasia (χ 2 = 5.075, P<0.05) . The Uyghur group exhibited significantly higher levels of white blood cell count, platelet count, neutrophils, lymphocyte count, globulin, C-reactive protein, erythrocyte sedimentation rate, highly sensitive C-reactive protein, anti-neutrophil cytoplasmic antibodies, and H.pylori antibodies compared to the Han group (all P<0.05) . Both Han and Uyghur UC patients achieved satisfactory clinical efficacy. Conclusion:Uyghur UC patients have distinct characteristics, including a younger age of onset and more severe disease course, yet appropriate treatment can lead to satisfactory clinical outcomes.
9.Long non-coding RNA AW112010 improves insulin resistance in adipocytes of aging mice through the miR-204/POU2F2 axis

Objective:To investigate whether long non-coding RNA(lncRNA) AW112010 can improve insulin resistance in aging adipocytes through the miR-204/POU2F2 signaling pathway.Methods:In vivo experiment: C57BL/6 mice were divided into young control group(4 months old) and aging model group(18 months old) based on body weight. The expression levels of AW112010, miR-204-5p, POU2F2, aging related indicators(p16, p21), and insulin signaling pathway genes [insulin receptor(INSR), insulin receptor substrate 1(IRS1), phosphatidylinositol kinase(PI3K), protein kinase B(AKT)] in epididymal adipose tissue were detected using real-time fluorescence quantitative PCR(RT-qPCR) and Western blotting. In vitro experiment: Using adriamycin(ADR) to induce 3T3-L1 aging adipocyte model, β-gal staining was used to observe cellular senescence, and miR-204 inhibitor and miR-204 mimic small interfering RNA were successfully constructed and transfected into 3T3-L1 adipocytes. Results:RT-qPCR and Western blot results showed that compared with the young group, the expression of AW112010 in the adipose tissue of aging mice was increased, while the expression of miR-204-5p was decreased. The expressions of POU2F2, p16, and p21 in the adipose tissue of aging mice were increased, while the expressions of INSR, IRS1, PI3K, GLUT4 mRNA and protein were decreased. The β-gal stainging results showed that the number of 3T3-L1 senescent adipocytes induced by ADR was significantly increased, and the expression levels of AW112010, POU2F2, p16, and p21 in ADR-induced senescent adipocytes were increased compared with the control group, while the expression levels of miR-204-5p, INSR, IRS1, PI3K, GLUT4 were decreased, and remaining glucose in the culture medium was increased. Compared with control, overexpression of miR-204 resulted in decreased expressions of aging indicators p16, p21, and target gene POU2F2 while the expressions of INSR and GLUT4 were increased.Conclusion:Upregulation of lncRNA AW112010 in adipocytes of aging mice may induce insulin resistance by targeting miR-204-5p/POU2F2/IRS1.
10.Multiple Single Cannulation Technique Improves the Outcomes of Arteriovenous Graft in Hemodialysis Patients:A Retrospective Study

Objective To evaluate the effects of the multiple single cannulation technique(MUST)on the outcomes of arteriovenous graft(AVG).Methods A retrospective study of AVG created between January 2018 and December 2021 at the First Affiliated Hospital of Zhengzhou University was conducted.The clinical data of patients and their follow-up data for venous access were analyzed.Subjects were divided into the MUST group or the non-MUST group according to whether MUST was used.The cumulative patency rate and complication incidence were compared between the two groups.Logistic regression was applied to analyze the influencing factors of applying MUST in AVG.Results The MUST group included 115 AVG and the non-MUST group,122 AVG.The 1-year,2-year,3-year,and 4-year cumulative patency rates of the MUST group were 100%,99.1%,95.2%,85.4%,and 73.2%,respectively,while those for the non-MUST group were 97.5%,92.7%,77.7%,69.7%,and 50.0%,respectively,with the 2-year and 3-year patency rates showing significant difference(P=0.022,P=0.004).The standard intervention rate expressed in(median[interquartile range])in the MUST group was significantly lower than that in the non-MUST group(0.46[0.00,0.94]vs.0.97[0.60,1.59],Z=-5.808,P<0.001).A total of 24(20.9%)AVG in the MUST group and 60(49.2%)AVG in the non-MUST group had a standard intervention rate>1.0 per patient-year,with significant difference between the two groups.Three(2.6%)AVG in the MUST group and 7(5.7%)AVG in the non-MUST group were complicated by aneurysm(χ2=20.737,P<0.001).One(0.9%)AVG in the MUST group and 6(4.9%)AVG in the non-MUST group had graft infection,with the difference between the groups showing no significance(P=0.121).Multivariate logistic regression showed that dialysis in the alliance facilities(odds ratio[OR]=2.713,95%confidence interval[CI]:1.698-4.336,P<0.001],and excellent follow-up[OR=2.189,95%CI:1.221-3.927,P=0.009]were the influencing factors of applying MUST in AVG.Conclusion MUST improves the cumulative patency of AVG and decreases the intervention frequency and the incidence of aneurysm without increasing the risk of graft infection.
