This study aims to explore protective effects of Lycium barbarum polysaccharides(LBP)on intestinal damage caused by lipopolysaccharide(LPS)-induced inflammatory bowel disease(IBD)in chicks.Network pharmacology was initially employed to determine the target proteins of wolfberry in the prevention and treatment of IBD.Following this,protein-protein interaction analy-sis,GO and KEGG pathway enrichment analysis, and molecular docking studies were conducted.Subsequently,an animal study was conducted:a total of 100 one-day-old male Hy-line brown lay-ing hens were randomly divided into five groups:a blank control group(CON),an LPS treatment group(LPS),a low-dose LBP group(LPS+LBP 0.25 g/L,L-LBP),a medium-dose LBP group(LPS+LBP 0.5 g/L,M-LBP),and a high-dose LBP group(LPS+LBP 1 g/L,H-LBP).Upon reac-hing 21 days old, duodenal, jejunal, ileal, and cecal tissues were collected to determine SOD and GSH-Px levels.Furthermore, the mRNA expression levels of TNF-α,AKT1,IL-6,IL-1β and TP53 in the intestinal tissues were measured using quantitative real-time PCR.The results demonstrated that network pharmacology identified 45 active ingredients in wolfberry that target 116 key protein sites, including TNF,AKT1 and IL6.The primary objectives focus on signaling pathways including AGE-RAGE,IL-17,TNF,HIF-1,and NF-κB.Molecular docking showed excellent ligand-receptor docking scores, with stable binding facilitated by hydrogen bonds and hydrophobic interactions.Compared to the LPS group, the 0.5 g/L LBP exhibited notably higher levels of SOD and T-AOC.In comparison with the LPS group, the medium and high-dose LBP experimental groups showed notably decreased the mRNA expressions of TNF-α,AKT1,IL-6,and IL-1β,while TP53 mRNA expression was significantly upregulated(P<0.01).In summary, wolfberry exerts preventive and therapeutic effects on IBD through a multi-component,multi-target,and multi-pathway mecha-nism.

Objective To explore the effects of physical activity(PA)on carotid intima-media thickness(CIMT)thickening in patients with type 2 diabetes mellitus(T2DM)with poor HbA1c control. Methods A total of 2505 T2DM inpatients with poor HbA1c control who visited the Endocrinology Department of the Integrated Traditional Chinese and Western Medicine Hospital Affiliated with Nanjing University of Traditional Chinese Medicine and Changzhou Second People's Hospital Affiliated with Nanjing Medical University from January 2019 to February 2022 were enrolled in this study. The patients were categorized into the mild-intensity PA(1~599 MET-min/w,n=795),moderate-intensity PA(600~1499 MET-min/w,n=1107)and high-intensity PA(≥1500 MET-min/w,n=603)based on the metabolic equivalence level(MET).Logistic regression was performed to analyze the relationship between PA and CIMT thickening.Statified analysis was used to investigate the effects of blood pressure(BP)and low-density lipoprotein cholesterol(LDL-C)target status on CIMT thickness in different PA groups. Results Restricted cubic spline plots showed a nonlinear relationship between PA levels and the risk of CIMT thickening(P<0.05).Logistic regression showed that with moderate-intensity PA as the control, while high-intensity PA was the influence factor of CIMT thickening(OR 1.581,95%CI 1.244~2.010,P<0.05).Statified analyses showed that under mild-intensity PA,using neither BP nor LDL-C on target as reference, achievement of either index and both were the influence factor of CIMT thickening.Using BP on target as reference, achievements of both indexes, achievements of neither of both indexes, and achievement of LDL-C only were the influence factor of CIMT thickening.Using LDL-C on target as reference, achievement of neither of them and achievement of BP only were the influence factor of CIMT thickening.Under high-intensity PA,achievement of both indexes was negatively associated with CIMT thickening. Conclusions Moderate-intensity PA is the recommended scheme to prevent CIMT thickening in T2DM patients whose HbA1c is not up to standard.Mild-intensity PA is not related to the risk of CIMT thickening, while high-intensity PA is positively related to the risk of CIMT thickening, and focusing on LDL-C compliance is particularly important to avoid CIMT thickening.

Objective To investigate the effect of lidocaine medicated plaster (LMP) combined with pregabalin (PGB) on patients with postherpetic neuralgia (PHN), and the impact on serum pain mediators. Methods 108 PHN patients admitted in our hospital from January 2024 to December 2024 were selected and grouped according to the time point of receiving treatment, 54 PHN patients treated with PGB from January 2024 to June 2024 were included in the PGB group, and 54 PHN patients treated with LMP on top of the PGB group from July 2024 to December 2024 were included in the PGB+LMP group. Comparisons were made between the two groups in terms of pain score, serum pain mediator levels, dosage of PGB, and incidence of adverse reactions. Results After 4 weeks of treatment, both groups showed a decrease in Pain Rating Index scores (sensory score and affective score), Present Pain Intensity score, Visual Analog Scale score, and total score. Meanwhile, above scores of the PGB+LMP group were lower than those of the PGB group (P<0.05). After 4 weeks of treatment, the levels of substance P(SP) and neuropeptide Y (NPY) in both groups were lower than those before treatment, while serum 5-hydroxytryptamine (5-HT) levels were higher than those before treatment. Moreover, the levels of SP and NPY were lower, and 5-HT level was higher in the PGB+LMP group than in the PGB group (P<0.05). The dosages of PGB in the PGB+LMP group at T1, T, T3 and T4 were significantly lower than those in the PGB group (P<0.05). The incidence of adverse reactions was 1.85%(1/54) in the PGB+LMP group. Compared to 5.56%(3/54) in the PGB group, and the difference was not statistically significant (P>0.05). Conclusion LMP combined with PGB was effective in the treatment of patients with PHN, which could effectively alleviate pain and lower the levels of serum pain mediators, with good safety.

Objective:To explore the mediating role of mentoring function between proactive personality and transition shock in new nurses, with the aim of providing reference and basis for managers to develop effective intervention measures for new nurses during their transformational period. Methods:Convenience sampling method was used to select 280 new nurses from the Second Affiliated Hospital of Zhejiang University School of Medicine, and a cross-sectional survey was conducted by applying the General Information Questionnaire, Transition Shock Scale, Proactive Personality Scale, and Mentoring Function Scale. The mediating role of mentoring function between new nurses′ proactive personality and transition shock was evaluated by constructing structural equation modelling.Results:265 valid questionnaires were ultimately collected. Among 265 nurses, there were 25 males and 240 females, with an age of (22.88 ± 2.12) years. The score of transition shock, proactive personality, and mentoring function of new nurses were (83.45 ± 18.95), (58.66 ± 9.96) and (81.72 ± 14.46) respectively; transition shock was negatively correlated with proactive personality and mentoring function ( r=-0.379, -0.340, both P<0.01), and proactive personality was positively correlated with mentoring function ( r=0.452, P<0.01); the mediating effect of mentoring function between proactive personality and transition shock was significant (95% CI -0.085 - -0.015, P<0.01), accounting for 29.14% of the total effect. Conclusions:New nurses' transition shock is at a moderately high level, and proactive personality can affect transition shock directly or indirectly through mentoring function. Managers can mitigate new nurses′ transition shock by improving the quality of mentoring.

To investigate the effect of the polysaccharide derived from Atractylodes macrocephala koidz.(PAMK)on lipopolysaccharide(LPS)-induced acute kidney injury in juvenile laying hens.A total of 100 one-day-old male Hy-Line brown laying hens were randomly divided into five groups:a control group(CON group),an LPS treatment group(LPS group),a PAMK low-dose group(LPS+PAMK 250 mg/L,group A),a PAMK medium-dose group(LPS+PAMK 500 mg/L,group B),and a PAMK high-dose group(LPS+PAMK 1 000 mg/L,group C).The PAMK-treated groups were fed with corresponding doses of PAMK mixed with water daily from 1-20 days of age.At the age of 14,16,18 and 20 days, the chickens in the LPS group and PAMK-treated groups were intraperitoneally injected with LPS,while the chickens in the blank control group were injec-ted with the same volume of normal saline. After feeding for 21 days, the kidneys were collected, and pathological changes in kidney tissue sections were observed using hematoxylin and eosin(HE)staining.Kidney weight and chick weight were measured to calculate the kidney coefficient. The levels of BUN,CRE,MDA,SOD,GSH-Px and T-AOC in kidney tissues were measured. The contents of TNF-α and IL-1β were determined by ELISA.The mRNA expression of inflammatory factor genes(IL-1β,TNF-α and NF-κB)and apoptosis-related factors(p53,Bcl-2,Bax and Caspase-3)in the kidneys was detected using quantitative real-time PCR.The results indicated that PAMK alleviated LPS-induced renal tissue injury.Compared to the LPS group,PAMK significantly re-duced the kidney coefficient and BUN concentration in the kidneys(P<0.05).Additionally,the levels of TNF-α and IL-1β,as well as the mRNA expression of TNF-α,IL-1β,and NF-κB in the PAMK group, were decreased compared to the LPS group.Furthermore,T-AOC,SOD and GSH-Px activities in the PAMK group were significantly increased(P<0.05),while MDA content was significantly decreased(P<0.01).Compared with the LPS group, the mRNA expression of Bax,Caspase-3 and p53 in the PAMK low-dose group and the PAMK medium-dose group decreased, while the mRNA expression of Bcl-2 increased. In conclusion,PAMK can inhibit the inflammatory response and oxidative stress response caused by LPS,and alleviate the apoptosis of kidney cells, and the treatment effect of PAMK in the middle dose group is the best, which provides a theoreti-cal basis for PAMK treatment of acute kidney injury in chickens.
