1.Dimeric natural product panepocyclinol A inhibits STAT3 via di-covalent modification.

Homo- or heterodimeric compounds that affect dimeric protein function through interaction between monomeric moieties and protein subunits can serve as valuable sources of potent and selective drug candidates. Here, we screened an in-house dimeric natural product collection, and panepocyclinol A (PecA) emerged as a selective and potent STAT3 inhibitor with profound anti-tumor efficacy. Through cross-linking C712/C718 residues in separate STAT3 monomers with two distinct Michael receptors, PecA inhibits STAT3 DNA binding affinity and transcription activity. Molecular dynamics simulation reveals the key conformation changes of STAT3 dimers upon the di-covalent binding with PecA that abolishes its DNA interactions. Furthermore, PecA exhibits high efficacy against anaplastic large T cell lymphoma in vitro and in vivo, especially those with constitutively activated STAT3 or STAT3Y640F. In summary, our study describes a distinct and effective di-covalent modification for the dimeric compound PecA to disrupt STAT3 function.
2.Difficulties and achievements of wound repair discipline in China in the first five years after its establishment

The aging population and changes in the disease spectrum have contributed to an increasing number of chronic refractory wounds, which not only cause physical and mental pain to patients and diminish their life quality, but also place a heavy burden on both their families and the society. Clinical practice has proven that establishing wound repair department for specialized treatment of chronic refractory wounds can effectively enhance treatment outcomes. As a newly established tertiary surgical discipline, wound repair discipline in China has been in development for 5 years. The authors outlined the main difficulties encountered during the establishment of wound repair discipline and summarized the key issues over the past 5 years. To solve these difficulties, the authors reviewed measures and achievements in areas such as raising people′s awareness, building standardized wards, exploring the multiple models of discipline construction, advancing research-oriented discipline construction, defining the scope of treatment, striving for service projects and promoting establishment of specialized level IV surgical systems, formulating a series of consensus guidelines for standardized treatment, developing and translating new wound repair technologies, creating a talent education and training system, expanding academic exchanges and cooperation, as well as fostering the growth of high-level disciplines, so as to offer references to support ongoing advancement in the establishment of wound repair discipline.
3.Further promotion of the construction of the discipline system of trauma medicine with Chinese characteristics through multiple initiatives

The construction of the discipline system of trauma medicine with Chinese characteristics is a continuous process of development and improvement. In the paper, the author discussed the eight aspects concerning further promotion of the construction of the discipline system of trauma medicine with Chinese characteristics including the construction of the discipline system, development of trauma care procedure, institutional construction of the specialty of clinical trauma care, cultivation of innovative talents, breakthrough of traditional concepts and innovation of first-aid theories, improvement of rescue technology and equipment development capabilities, efficient transformation of trauma care achievements, nationwide trauma care training and regular first-aid exercises, aiming to provide reference for construction of the discipline system of trauma medicine with Chinese characteristics.
4.Biomaterial is becoming a promising research field for trauma, burns, and wound repair

I learned from some specialists in fields of trauma, burns, and tissue repair in different conferences that the new research fields in trauma, burns, and tissue repair combined with biomaterials have been established in recent years. I feel that the direction of scientific research in the subject field characterized by the treatment of "injury" is undergoing new changes, expanding into new fields related to "injury" treatment such as biomaterials and biomedical engineering, especially the close integration with biomaterials. These reflect the characteristics of cross-industry, cross-field, and multi-disciplinary intersection and integration. These new developments could lead to revolutionary breakthroughs in future research and clinical treatment of trauma, burns, and tissue repair. In this paper, I would like to summarize the fruitful achievements in these fields and put forward some suggestions for the next development of the discipline.
5.Talking again about the academic concept and its practice of treating burns in wartime and wounds in peacetime

The transformation and development of burn specialty is an important topic. In 2018, I officially published my academic concept which was treating burns in wartime and wounds in peacetime in the Chinese Journalof Burns. All works related to the view are progressing smoothly and have achieved important phased results. In the paper, I would like to further elaborate on the background for the proposal of this academic view, the academic concerns, the important phased results achieved, and the future development of this discipline. It is my hope that it can provide some reference for the transformation and development of burn specialty and its related disciplines.

10.Reliability and Validity Analysis on Symptom Identification Scale for Vascular Cognitive Impairment with Kidney Yang Deficiency

Objective To evaluate the feasibility,reliability and validity of the symptom identification scale for kidney yang deficiency syndrome.Methods The symptom identification scale for kidney yang deficiency syndrome developed in previous research was administered to 200 vascular cognitive impairment patients from September 2020 to September 2022 to assess feasibility through recall rate,completion rate,and completion time;reliability was measured using retest reliability,split-half reliability,homogeneity reliability,and inter-rater reliability;and validity was evaluated based on discriminant and structural validity.Results A total of 200 scales were sent out,and all of them cooperated and were completed and retrieved within 20 min.The results of reliability analysis showed that the retest reliability of the scale was 0.828 for the dimension of yang deficiency and 0.718 for the dimension of kidney qi deficiency;the Spearman-Brown coefficient of split-half reliability was 0.784;the Cronbach coefficient of the dimension of yang deficiency was 0.799,and the Cronbach coefficient of the dimension of kidney qi deficiency in the homogeneity reliability was 0.670.The results of the analysis showed that the differences between the kidney yang deficiency syndrome group and the non-kidney yang deficiency syndrome group in the yang deficiency dimension,kidney qi deficiency dimension scores and total scale scores in the discriminant validity were statistically significant(P<0.001);the KMO value in the structural validity was 0.842,and a total of two factors with eigenvalues greater than 1 were extracted,with a cumulative variance contribution rate of 58.227%.Conclusion The symptom identification scale for kidney yang deficiency syndrome demonstrates adequate reliability and validity,potentially enhancing the prediction of kidney yang deficiency in vascular cognitive impairment.However,the validity of the scale is somewhat limited and requires further refinement for clinical application.
