ObjectiveTo explore the vascular endothelial injury in male mice caused by exposure to polystyrene microplastics (PS-MPs) and the intervention effect of luteolin on vascular remodeling. Additionally, to investigate the mechanism through the oxidative system and metabolomics. MethodsThirty-two C57BL/6 mice (6-8 weeks old) were randomly divided into the saline group (saline group), the 0.1 mg/kg PS-MPs exposure group (0.1PS-MPs group), the 1 mg/kg PS-MPs exposure group (1PS-MPs group), and the 1 mg/kg PS-MPs + luteolin treatment group (1PS-MPs + Lut group), with 8 mice in each group. After 8 weeks of intervention, the body weight, blood pressure, aortic organ coefficient, and aortic histopathological changes of mice in each group were detected; the total cholesterol (TC), triglyceride (TG), and high-density lipoprotein cholesterol (HDL-C) lipid metabolism-related indicators in the aorta of mice were detected; the reactive oxygen species (ROS), glutathione (GSH), and malondialdehyde (MDA) oxidative stress-related indicators were detected; the endothelin (ET-1), nitric oxide (NO), vascular endothelial growth factor A (VEGF-A), vascular cell adhesion molecule-1 (VCAM-1/CD106), and intercellular adhesion molecule-1 (ICAM-1/CD54) endothelial function-related indicators and serum metabolomics were detected. ResultsCompared to the saline group, exposure to PS-MPs resulted in pathological thickening of the mouse aorta, increased aortic organ coefficient, and elevated blood pressure. Lipid metabolism-related indicators, including TC and TG, were elevated, while HDL-C was reduced, indicating lipid metabolism disorder in mice. Oxidative stress markers such as ROS and MDA increased, whereas GSH decreased, demonstrating oxidative damage. Vascular endothelial inflammation and injury markers, including ET-1, VEGF-A, VCAM-1, and ICAM-1, were upregulated, while the vasodilatory substance NO was downregulated, confirming endothelial injury. Furthermore, serum metabolomics results revealed that PS-MPs exposure induced endothelial damage by disrupting metabolic pathways such as the citrate cycle. Compared to the PS-MPs group, luteolin significantly reversed these effects, attenuating oxidative stress and lipid metabolism disorders, and effectively repairing endothelial injury. ConclusionPS-MPs induce vascular toxicity through oxidative stress and lipid metabolism. Luteolin effectively alleviates endothelial damage and vascular remodeling.

Objective Patients with lung cancer often experience interrelated symptoms such as fatigue, pain, and sleep disturbances during chemotherapy, forming a core symptom cluster that significantly impacts daily functioning, treatment experience, and quality of life. Currently, evidence on non-pharmacological management of this core symptom cluster is scattered, and there is a lack of standardized, patient- and caregiver-friendly guidance tools that are easy to understand and implement. This patient guideline aims to provide decision support for self-management of the core symptom cluster in patients undergoing chemotherapy for lung cancer. Methods Guided by evidence-based methodology, qualitative interviews and on-site questionnaire surveys were conducted to identify the health-related needs of stakeholders. Domestic and international guideline websites, relevant professional association websites, and comprehensive databases were systematically searched according to the inclusion and exclusion criteria. The included literature was evaluated for quality, and recommendations were extracted, integrated, and graded to form a summary of the best evidence. Based on the identified health issues and evidence summary, preliminary recommendations for the patient guideline were developed. The recommendations were then revised through two rounds of Delphi expert consultation, and consensus was reached on the strength of recommendations, forming the final version of the patient guideline. Results The patient guideline covered four major sections: screening and assessment of core symptom clusters, daily management, intervention measures, and timing of referral. The core content included nine aspects: screening of core symptom clusters, assessment, lifestyle habits, dietary interventions, sleep environment, exercise interventions, psychological interventions, Traditional Chinese Medicine interventions, and referral indications. In total, the guidelines addressed 16 health issues and provided 25 recommendations. Conclusion The development process of this patient guideline is rigorous and fully considered the needs of patients. Expert review concludes that the recommendations are highly feasible and appropriate.

To analyze the relationship between serum vitamin A and E levels and asthma risk in children aged 1-14 years. Based on a multi-center case-control study design, general information and vitamin A and E test data were collected from asthmatic children in the respiratory outpatient departments of four medical institutions in Hunan Province and control children undergoing health check-ups in the child healthcare departments during the same period from July 2017 to October 2019.After 1∶1 matching age and gender using propensity scores, a total of 2 454 research subjects were included. 1 730 boys and 724 girls, with an age range of 1-14 years, a median age of 3 years and a mean age of (3.58±2.83) years. The levels of vitamin A and E in the asthma group and the control group were compared. Binary Logistic regression was used to analyze the association between vitamin levels and asthma, and stratified analysis by age and gender was conducted. The results showed that a total of 3 447 research subjects were initially included in this study, and 2 454 research subjects were included after propensity scores matching (PSM). After matching, there was no statistically significant difference in age and gender between the two groups ( χ2=0.00 ,P>0.05), and the standardized mean difference (SMD) values were all less than 10%.The levels of vitamin A and E in the asthma group were significantly lower than those in the control group [(0.32±0.09)mg/L vs.(0.34±0.08)mg/L, t=4.94 ,P<0.001;(8.90±2.47)mg/L vs.(9.19±2.58) mg/L, t=2.79 ,P=0.005]. Compared to the normal vitamin A group, the high-risk subclinical deficiency group ( OR=1.53, 95% CI: 1.28-1.83) and the subclinical/clinical deficiency group ( OR=2.68, 95% CI: 1.81-3.97) were significantly positively associated with asthma risk. Compared to the normal vitamin E group, the vitamin E insufficient group was significantly associated with increased asthma risk ( OR=1.28, 95% CI: 1.03-1.57). Subgroup analysis revealed that subclinical/clinical vitamin A deficiency was significantly associated with increased asthma risk across all age groups and both genders (all P<0.05), whereas vitamin E insufficiency was only associated with higher asthma risk in the 1-3 age group and among girls ( P<0.05).In conclusion, subclinical or clinical deficiency of vitamin A may significantly increase the risk of asthma. Vitamin E deficiency is mainly associated with an increased risk of asthma in younger age groups (1-<3 age group) and girls.

The Golgi apparatus(GA),an essential membrane organelle in eukaryotes positioned between the endoplasmic reticulum and the plasma membrane,is responsible for transporting and modifying proteins and lipids generated by the endoplasmic reticulum. The homeostasis of the endoplasmic reticulum and GA plays a crucial role in regulating cellular life. When GA is unable to bear the load of protein or lipid processing and transportation,it enters a state of stress.Cells sense this and activate Golgi-related quality control mechanisms.By regulating the structure and function of GA,it provides important protection for the cell or induces programmed cell death, known as the GA stress response.Understanding the mechanisms and outcomes of cellular GA stress is of great significance for exploring Golgi dynamics and its impact on human diseases. This review summarizes the structure and function of GA under physiological conditions, the phenomenon of GA stress under pathological conditions, and the role of GA in infectious diseases, aiming to provide a basis for the clinical study of new strategies for the prevention and treatment of infectious diseases.

This study was aimed at establishing a method of ultra-fast quantitative PCR for Brucella detection. We used an exogenous recombinant plasmid as the internal reference and targeted the T4SS secretion system,an important Brucella viru-lence factor,to design specific primers and probes. The sensitivity, specificity, and repeatability of this method were evaluated, and a standard curve was constructed. The coincidence rate of detection findings with this method versus quantitative PCR was determined. This method markedly decreased the detection time to only 10 minutes. The standard curve demonstrated a good linear relationship(Y=-3.410 7x+38.357,R2=0.998 5)with a low minimum detection limit of 10 copies/μL.The method exhibited good specificity and did not specifically amplify several common clinical bacteria other than Brucella. The de-tection of three concentrations of positive plasmids yielded coefficients of variation(CVs)of 0.20%to 0.91%,thus demonstra-ting the method's excellent repeatability.Furthermore,140 clinical samples were analyzed concurrently with the fluorescence PCR method, which yielded a 100%compliance rate and consistent results. Our findings indicated that the Brucella ultra-fast quantitative PCR was ultrafast;had high sensitivity, high specificity, and good specificity;and can be used for the clinical de-tection of Brucella and emergency investigation of epidemics. Therefore, this method is valuable for the early diagnosis of Bru-cella.

To investigate the antimicrobial resistance of Enterococci isolated from 596 anal swabs, feces and environmental samples were collected from pig, cattle, chicken and quail farms in Chang-sha,Hunan Province,Enterococci were isolated and identified by mass spectrometry. The minimum inhibitory concentrations(MICs)of 10 antimicrobial agents were determined by AGAR diffusion method.Whole genome sequencing(WGS)was used to detect the distribution of multilocus se-quence typing(ST),drug resistance genes and virulence genes.A total of 272 strains of Enterococ-cus were isolated(45.6%).The isolates were resistant to cefoxitin(68.9%)and cefotiofur(58.5%),followed by trimethoprim/sulfamethoxazole(52.2%),vancomycin(4.4%),and linezolid(13.6%).In this study, six linezolid highly resistant Enterococci isolates were analyzed by whole genome sequencing to explore the transmission mechanism of linezolid resistance because linezolid is forbidden to be used in aquaculture.ST403(4/6),ST16(1/6)and ST476(1/6)were the most common ST types, which all originated from the same farm.Three oxazolidinone resistance genes(cfr, poxtA and optrA)were found in all 21 strains. One strain(Ecc60)carried all three oxazo-lidinone resistance genes, but none of them were located on the plasmid.tet(M),aph(3')-Ⅲ,and lsa(A)were found in all six Enterococcus isolates.Interestingly, the present study is the first to i-dentify enterococci carrying the optrA gene in quail fecal samples. The analysis of the genetic envi-ronment of oxazolidinone resistance genes showed that the genetic environment of cfr(D),poxtA and optrA of isolates from the same farm was similar.A total of 19 virulence genes were detected in 6 isolates,of which 12 genes(ElrA,SrtA,ace,agg,cCF10,cOB1,cad,camE,ebpA,ebpC,efaAfs,tpx)were carried by all 6 isolates, and the types of virulence genes in strains from the same farm were extremely similar. The results showed that the drug resistance of Enterococci iso-lated from the fish farms in Changsha was serious, and the resistance rate to linezolid, which was prohibited in the fish farms, was high. The oxazolidinone resistance gene loci were accompanied by other resistance genes, especially the florfenicol resistance gene(FexA),which may be related to the abuse of florfenicol in the fish farms.

Objective:To investigate whether escin(ESC)and dextromethorphan(DEX)have the protective effects on the progression and symptoms of Alzheimer disease(AD).Methods:The AD model of Caenorhabditis elegans(C.elegans)was established by transgenic amyloid β-protein(Aβ protein).Different concentrations of ESC or DEX or 50 μmol/L memantine(MEM)were used to treat the AD model worms, and their lifespan was detected. The movement ability of AD model C.elegans was evalua-ted by body bending frequency and head swinging frequency. The changes in cognitive functions of AD model C.elegans before and after treatment were detected by chemotaxis experiments. The changes in A βprotein and reactive oxygen species(ROS)content in C.elegans were detected. The changes in gene pathways related to oxidative stress were detected by Real-time quantitative polymerase chain reaction(RT-qPCR).Results:At high dose 1 000 μmol/L,ESC or DEX treatment showed no significant effects on the activity of C.elegans.Compared with untreated worms, the survival time of AD model C.elegans in the 20 μmol/L ESC and 60 μmol/L DEX intervention groups was significantly extended. In the middle stage of AD progression, the body bending frequency and head swinging frequency of AD model worms after ESC or DEX treatment was significantly increased compared with the untreated control group with DEX being more effective in the recovery of head swinging frequency. For the early cognitive function tests, the chemotaxis index of ESC or DEX treated worms was significantly higher than that of the untreated worms, which correlated with marked reductions in the Aβ protein levels. The reactive oxygen species content in the drug intervention group was also lower than that in the control group.RT-qPCR results showed that ESC could inhibit oxidative stress in the AD model C.elegans by a 2-fold upregulation of skn1 expression. Conclusion:ESC and DEX could improve the reductions of movement ability and cognitive function in the AD model worms and delay the aggravation of AD-related symptoms.ESC delays the progression of AD pos-sibly by activating the SKN-1/Nrf2 pathway to protect against oxidative injury in the AD model.
