1.Effect of GSTP1 and MTHFR Gene Polymorphism on Side Effects of HD-MTX in ALL Children.

OBJECTIVETo Study the effect of C677T and MTHFR gene polymorphism on side effects of HD-MTX in ALL children.
METHODSThe gene polymorphism of C677T A303G and MTHFR C677T were detected by PCR in 98 ALL children from January 2014 to January 2016. The side effects during HD-MTX therapy were observed, and the relationship among GSTP1, MTHFR gene polymorphism and incidence of side effect of HD-MTX were analyzed.
RESULTSAmong 98 ALL children, the gene variation was observed in 61 ALL children (62.24%). Polymorphism study on C677T A303G showed that the gene frequency of A was 84.69%, while that of G was 15.31%; for polymorphism of MTHFR C677T, gene frequency of C was 66.33%, and that of T was 33.67%. Seven patients(7.14%) experienced with bone marrow supression, 23 patients(23.47%) with liver function damage, 15 patients(15.31%) with renal function damage, 48 patients(48.98%) with gastrointestinal reactions and 46 patients(46.94%) with mucosal lesions. After adjustment of sex, age, risk stratification and dosage of MTX, the gene polymorphism had no significant relationship with bone marrow suppression, gastrointestinal reactions and mucosal lesions(P>0.05). However, the number of the mutant genes had statistically significant relationship with liver and renal function damage(P<0.05).
CONCLUSIONThe risk of side effects during HD-MTX therapy increases in ALL children with combined mutation of MTHFR and C677T.
2.Analyze moisture transformation and transport rules during processing of Paeoniae Radix Alba by using low-field NMR.

Moisture status and content during the processing of Paeoniae Radix Alba were studied by nuclear magnetic resonance (NMR) and nuclear magnetic resonance imaging (MRI) to investigate the changes of transverse relaxation time (T ₂) and MRI images during boiling and drying processes of Paeoniae Radix Alba. The results showed that water in Paeoniae Radix Alba fresh products was major of free water, and in the boiling process, the content of free water increased whereas the content of bound water declined. At the end of boiling, content of free water reached over 90%. During the drying process, T ₂ moved to the left, and moisture mobility was reduced. The MRI image directly showed that moisture transfer was outside-in process for both increase and decrease. At the end of drying, remaining moisture was mainly present in inner layer of Paeoniae Radix Alba. Quality and appearance were affected by the change of moisture during processing process of medicinal herbs. NMR and MRI could provide direct reference evidence for its moisture changes, and the results of this study could provide direct references and technical support for optimization of processing process of root medicinal materials and evaluation of Chinese herbal pieces.
3.Changes of Peripheral Blood Marrow-Derived Suppressor Cell Level after Chemotherapy Induction Remission by VDLP Regimen and Their Relationship with Immune System in B-ALL Children.

OBJECTIVETo investigate the changes of peripheral blood marrow-derived suppressor cell level after chemotherapy induction remission by regimen consisting of vincristine, daunorubicin, L-asparaginase and prednisone (VDLP) and to analyze their relationship with immume system in B-ALL children.
METHODSThirty B-ALL children after induction remission by VDLP regimen from August 2015 to August 2016 were selected as B-ALL group and 30 normal healthy children were selected as control group. The peripheral blood in 2 groups was collected and detected by flow cytometry, then the ratios of CD30cells and CD33HLA-DRmarrow-derived suppressor cells, CD14CD33HLA-DRmarrow-derived suppressor cells and CD15CD33HLA-DRmarrow-derived suppressor cells were calculated, and their changes after induction remission by VDLP regimen and the relationship with immune system were analyzed.
RESULTSAfter treatment the ratio of CD33cells in peripheral blood of B-ALL group and control group was not significantly different (P> 0.05), moreover, the ratio of CD33cells in B-ALL group was significantly higher than that before treatment (P<0.05), while the ratios of CD33HLA-DRmarrow-derived suppressor cells, CD14CD33HLA-DRmarrow-derived suppressor cells and CD15CD33HLA-DRmarrow-derived suppressor cells in B-ALL group were significantly lower than those in control group (all P<0.05), but the ratios of these cells in B-ALL group were higher than those before treatment, and yet there was no statistical significance (P>0.05).
CONCLUSIONThe ratios of marrow-derived suppressor cells in peripheral blood of B-ALL children in complete remission after treatment with VDLP regimen are higher than those before treatment, but are significantly lower than normal value, which may be related with non-complese recovery of immune system in B-ALL children after treatment.
4.Ovarian Damages Produced by Aerosolized Fine Particulate Matter (PM) Pollution in Mice: Possible Protective Medications and Mechanisms.

BACKGROUNDAmbient aerosol fine particulate matter (PM2.5) is associated with male reproductive toxicity in experiments and may have adverse effects in the female. However, studies evaluating the protective effects and precise mechanisms of aspirin, Vitamin C, Vitamin E, or ozone against toxic effects of PM2.5are sparse. This study was conducted to investigate the possible protective effects and mechanisms of aspirin, Vitamin C, Vitamin E, or ozone on fertility in female mice treated with PM2.5.
METHODSEighty-four ICR mice were divided into six groups: control group, PM2.5group, PM2.5 + aspirin group, PM2.5 + Vitamin C group, PM2.5 + Vitamin E group, and PM2.5 + ozone group. PM2.5was given by intratracheal instillation every 2 days for 3 weeks. Aspirin, Vitamin C, and Vitamin E were given once a day by oral gavage for 3 weeks, and ozone was administered by intraperitoneal injection once a day for 3 weeks. The levels of anti-Müllerian hormone (AMH), interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and 8-hydroxy-2'-deoxyguanosine (8-OHdG) were measured using enzyme-linked immunosorbent assay. Western blotting analysis was used to analyze the expressions of Bcl-2, Bax, and caspase-3 in ovaries. Changes in histological structure were examined by light microscope and electron microscopy was used to detect ultramicrostructure.
RESULTSThe results demonstrated that PM2.5 decreased AMH levels (P < 0.001); however, aspirin (P < 0.001), Vitamin C (P < 0.001), Vitamin E (P = 0.001), and ozone (P = 0.002) alleviated the decrease. Changes of IL-6, TNF-α, 8-OHdG, Bax/Bcl-2, and caspase-3 in PM2.5group were increased compared to control group (P < 0.001), while in PM2.5 + aspirin, PM2.5 + Vitamin C, PM2.5 + Vitamin E, and PM2.5 + ozone groups, they were statistically decreased compared to PM2.5group (P < 0.001 or P< 0.05).
CONCLUSIONSPM2.5cause the damage of ovaries, and aspirin, Vitamin C, Vitamin E, and ozone antagonizes the damage. The protective mechanism is probably due to its ability to blunt the inflammatory and oxidative stress caused by PM2.5, which subsequently suppressing the expression of apoptotic regulatory protein and reducing the incidence of ovary apoptosis.
5.Early Kinetics of Procalcitonin in Predicting Surgical Outcomes in Type A Aortic Dissection Patients.

BACKGROUNDIn cardiac surgery, elevation of procalcitonin (PCT) could be observed postoperatively in the absence of any evidence of infection and also seems to be a prognostic marker. PCT levels measured in patients undergoing Type A aortic dissection (TAAD) were used to determine prognostic values for complications and surgical outcomes.
METHODSMeasurements of PCT, C-reactive protein (CRP), and leukocyte count were observed in TAAD surgery patients (n = 251; average age: 49.02 ± 12.83 years; 78.5% male) at presurgery (T0) and 24 h (T1), 48 h (T2), and 7 days (T3) postsurgery. PCT clearance (PCTc) on days 2 and 7 was calculated: (PCTday1- PCTday2/day7)/PCTday1 × 100%. Endotracheal intubation duration, length of stay (LOS) in the Intensive Care Unit (ICU)/hospital, and complications were recorded.
RESULTSPCT peaked 24 h postsurgery (median 2.73 ng/ml) before decreasing. Correlation existed between PCT levels at T1 and duration of cardiopulmonary bypass (P = 0.001, r = 0.278). Serum PCT concentrations were significantly higher in nonsurvivor and multiple organ dysfunction syndrome groups on all postoperative days. PCT levels at T1 correlated with length of time of ventilation support and ICU/hospital LOS. Comparing PCT values of survivors versus nonsurvivors, a PCT cutoff level of 5.86 ng/ml at T2 had high sensitivity (70.6%) and specificity (74.3%) in predicting in-hospital death. PCTc-day 2 and 7 were significantly higher in survivor compared with nonsurvivor patients (38% vs. 8%, P= 0.012, 83% vs. -39%, P< 0.001). A PCTc-day 7 cutoff point of 48.7% predicted survival with high sensitivity (77.8%) and specificity (81.8%).
CONCLUSIONSPCT level and PCTc after TAAD surgery might serve as early prognostic markers to predict postoperative outcome. PCT measurement may help identify high-risk patients.

6.Design and implementation of information management system for Chinese materia medica resources survey.

By means of the established Information Management System for Chinese materia medica resources survey, the national, provincial and county level organization, personnel and the implementation of the program of Chinese materia medica resources survey, and the survey team of medicinal plant investigation, photos, Chinese herbal medicine market survey, the traditional Chinese materia medica resources knowledge survey, germplasm resources investigation and the data collation and summary specimen have been realised. Throughout the whole working process of the fourth national Chinese materia medica resources survey, it is ensured that all data were no missing, no repeat, and well stored and managed. The Information Management System can improve the standardization degree of Chinese materia medica resources survey, and maintain the continuity.
7.Detection and analysis of effect of geographical environmental factors on spatial distribution of artemisinic acid in Artemisia annua.

Dao-di herbs are preferred by long-term clinical application of Chinese medicine, they are produced in a specific area and affected by the specific production and processing methods. Dao-di herbs have a high reputation, compared with other regions produced by the same kind of herbs with good quality, good efficacy and stable quality characteristics. Geo-herbal is a unique concept that has been formed by the history of Chinese civilization for thousands of years and the history of Chinese medicine. According to the definition of geo-herbal, it can be seen that there are some differences in the quality and efficacy of the same kind of medicinal herbs in different regions, and there is some similarity in the specific area. In this study, based on the content of artemisinic acid in sampling points and its potential environmental factors, the effects of geographical environment factors on the spatial distribution of artemisinic acid content in Artemisia annua were studied by using the geophysical model. The results show that the spatial distribution of artemisinic acid content is the result of a combination of multiple factors. The effects of environmental factors on the spatial distribution of artemisinic acid were in the order of soil type (0.233)> radiation (0.208)> vegetation type (0.192)> elevation (0.171)> sunshine (0.170)> annual mean temperature (0.153) >annual precipitation (0.111)> slope (0.110)> relative humidity. Among them, the soil type and the amount of radiation are the main influencing factors, and the main influencing area is in the soil type as the initial soil and the average annual radiation of 1 200-1 400 kWh•m⁻². The main influencing factors selected in this study can be used to monitor the spatial distribution of artemisinic acid by remote sensing technology, so as to provide the theoretical basis for the cultivation of A. annua.
8.Analysis of spatial distribution of artemisinin in Artemisia annua in China.

In this study, the spatial distribution characteristics of artemisinin in Artemisia annua were analyzed by spatial statistical analysis. ①The spatial autocorrelation of artemisinin content was analyzed by "I coefficient". The results showed that the content of artemisinin in each province was significant and positive spatial autocorrelation. The content of artemisinin in each province was not randomly distributed, but with a clear spatial aggregation characteristics. ②The spatial variability of artemisinin content was analyzed by "G statistic". The results showed that the area with high content of artemisinin was distributed in the southwest region, and the content of artemisinin was concentrated in the space, which tends to accumulate at high value. The content of artemisinin in Tianjin and Liaoning province is low, and it tends to accumulate at low value, while the content of artemisinin in other provinces is the transition zone of high and low value. ③Based on the index of artemisinin in each province, the spatial correlation of artemisinin content in each region was analyzed by "moran scatter plot". The results showed that the content of artemisinin in nine provinces such as Guangxi and Chongqing belonged to high value gathering area. ④Based on the artemisinin content and ecological environment factor of sampling point, the "geophysical detector" was used to analyze the continuous quantitative data and discontinuous qualitative data. The results showed that the sunshine, temperature and precipitation were the main factors affecting the content of artemisinin.
9.Location information acquisition and sharing application design in national census of Chinese medicine resources.

In literature, there are many information on the distribution of Chinese herbal medicine. Limited by the technical methods, the origin of Chinese herbal medicine or distribution of information in ancient literature were described roughly. It is one of the main objectives of the national census of Chinese medicine resources, which is the background information of the types and distribution of Chinese medicine resources in the region. According to the national Chinese medicine resource census technical specifications and pilot work experience, census team with "3S" technology, computer network technology, digital camera technology and other modern technology methods, can effectively collect the location information of traditional Chinese medicine resources. Detailed and specific location information, such as regional differences in resource endowment and similarity, biological characteristics and spatial distribution, the Chinese medicine resource census data access to the accuracy and objectivity evaluation work, provide technical support and data support. With the support of spatial information technology, based on location information, statistical summary and sharing of multi-source census data can be realized. The integration of traditional Chinese medicine resources and related basic data can be a spatial integration, aggregation and management of massive data, which can help for the scientific rules data mining of traditional Chinese medicine resources from the overall level and fully reveal its scientific connotation.
10.Application of image recognition technology in census of national traditional Chinese medicine resources.

With the development of computer and image processing technology, image recognition technology has been applied to the national medicine resources census work at all stages.Among them: ①In the preparatory work, in order to establish a unified library of traditional Chinese medicine resources, using text recognition technology based on paper materials, be the assistant in the digitalization of various categories related to Chinese medicine resources; to determine the representative area and plots of the survey from each census team, based on the satellite remote sensing image and vegetation map and other basic data, using remote sensing image classification and other technical methods to assist in determining the key investigation area. ②In the process of field investigation, to obtain the planting area of Chinese herbal medicine was accurately, we use the decision tree model, spectral feature and object-oriented method were used to assist the regional identification and area estimation of Chinese medicinal materials.③In the process of finishing in the industry, in order to be able to relatively accurately determine the type of Chinese medicine resources in the region, based on the individual photos of the plant, the specimens and the name of the use of image recognition techniques, to assist the statistical summary of the types of traditional Chinese medicine resources. ④In the application of the results of transformation, based on the pharmaceutical resources and individual samples of medicinal herbs, the development of Chinese medicine resources to identify APP and authentic herbs 3D display system, assisted the identification of Chinese medicine resources and herbs identification characteristics. The introduction of image recognition technology in the census of Chinese medicine resources, assisting census personnel to carry out related work, not only can reduce the workload of the artificial, improve work efficiency, but also improve the census results of information technology and sharing application ability. With the deepening of the work of Chinese medicine resources census, image recognition technology in the relevant work will also play its unique role.
