Objective This study aimed to investigate the seasonal variation of mosquitoes in Yantai, Shandong Province, and provide a scientific basis for mosquito and mosquito-borne disease control. Methods From March to November for each of the years between 2020 and 2023, adult mosquitoes were collected from different habitats, including parks, hospitals, urban residential areas, rural households and livestock sheds, in 13 counties of Yantai City using a mosquito trap lamp method, among which, female mosquitoes were separated and identified, and the numbers were counted. We used the circular distribution method to analyze the temporal distribution characteristics of mosquito density. Results The γ values obtained for mosquitoes in Yantai from 2020 to 2023, the distribution of mosquitoes in different habitats, and dominant mosquito species in different months showed strong seasonality, with values ranging from 0.7461 to 0.8531(P < 0.05). The peak period of seasonal variation in mosquitoes occurred from June 28thto September 13th, with August 5thbeing identified as the peak day. The peak periods of seasonal variation in mosquitoes in residential areas, parks, hospitals, peasant households, and animals sheds were observed from June 24thto September 14th, June 25thto September 10th, June 22ndto September 12th, July 1stto September 9th, and July 2ndto September 17th, respectively, with corresponding peak days on August 4th,3rd, 2nd, 5th, and 9th, respectively. The peak periods of Culex pipiens pallens, Aedes albopictus, and Anopheles sinensis were from June 26thto September 11th, June 28thto September 17th, and July 12thto September 17th, respectively, with corresponding peak days on August 3rd,7th, and 14th, respectively. Conclusions Our findings in this study revealed that the peak of mosquito activity in Yantai was concentrated in early August, with a peak period extending from June 28thto September 13th, and there were differences between different habitats(with peak periods for rural households and livestock sheds occurring later than those in residential areas, parks, and hospitals). Moreover, with respect to the mosquito species, populations of Cx. pipiens pallens and Ae. albopictus peaked earlier than those of An. sinensis. Based on these findings, we recommend that city-wide mosquito control work should commence in late June, and include appropriate early prevention and control measures for key habitats, such as livestock sheds, and dominant mosquito species, such as Cx. pipiens pallens, to minimize the density of mosquito vectors and the risk of disease transmission.

Objective This study conducted molecular biological identification of the viruses carried by Culex pipiens pallens specimens collected in Shunyi District, Beijing in 2023, and observed the changes in the infection rate of the viruses carried by Cx. pipiens pallens at different collection times. Methods Cx. pipiens pallens were collected using carbon dioxide mosquito traps. The mosquito samples were ground in batches and analyzed by molecular biology technologies. The virus infection rate at different collection times was analyzed statistically. Results 17 strains of Culex pipiens pallens densovirus(CppDNV)were identified from Cx. pipiens pallens samples collected in Shunyi District, Beijing, in 2023. The nucleotide sequence analysis of the virus genome coding region showed that CppDNV was a single-stranded DNA virus with a total length of 3 335 nt, encoding 2 non-structural proteins(NS1, NS2)and 1 capsid protein(VP). The nucleotide(amino acid)sequence lengths of the three proteins were 2 376 nt(791 aa),1 092 nt(363 aa)and 1 071 nt(356 aa), respectively. Phylogenetic analysis showed that CppDNV was located in genus Brevihamaparvovirus. Statistical analysis showed no significant difference in infection rates across collection times(χ2=4.429, P=0.194). Conclusions CppDNV was identified in Cx. pipiens pallens in Beijing, and it was stably maintained in this natural population.

OBJECTIVE To investigate the effects of metformin (Met) on liver injury in non-alcoholic steatohepatitis (NASH) rats by regulating the PI3K/AKT/PDGF signaling pathway. METHODS NASH model was constructed by feeding rats with a high- glucose and high-fat diet, and assigned into Model group, Met low-dose group (Met-L group, 100 mg/kg), Met medium-dose group (Met-M group, 200 mg/kg), Met high-dose group (Met-H group, 400 mg/kg), and high dose of Met+PI3K activator group (Met-H+740 Y-P group, 400 mg/kg Met+50 mg/kg 740 Y-P), with 12 rats in each group. Another 12 rats were regarded as the Control group. Each group of rats was orally administered/injected with the corresponding medication once a day for 6 consecutive weeks. The changes in body weight and liver index of rats were recorded and analyzed. The pathological damage [evaluation of non-alcoholic fatty liver disease activity score (NAS)], lipid deposition (calculation of the proportion of oil red O-positive staining area), and fibrosis (calculation of collagen deposition score) were observed in liver tissue of rats. The levels of inflammatory factors [interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α)] in serum and liver tissue, the levels of serum lipid metabolism indicators [total cholesterol (TC), triglyceride (TG), and low-density lipoprotein cholesterol (LDL-C)] and liver function indicators [aspartate aminotransferase (AST) and alanine Δ 基金项目 武汉市知识创新专项项目(No.2022020801010588); aminotransferase (ALT)] were measured. The expression levels of PI3K/AKT/PDGF signaling pathway-related proteins and Caspase-3 in liver tissue of rats were determined. RESULTS Compared with the Control group, body weight, liver index, the levels of serum lipid metabolism indicators and liver function indicators, the levels of IL-6 and TNF-α in serum and liver tissue, the NAS, the proportion of oil red O-positive staining area, the collagen deposition fraction, and the levels of phosphorylated PI3K and AKT proteins, as well as the expression levels of PDGF and Caspase-3 proteins in liver tissue, were all significantly increased (P<0.05). The liver tissue showed severe pathological damage, characterized by an abundance of lipid droplets and pronounced collagen deposition. After the intervention with Met, the aforementioned quantitative indicators and pathological changes in rats were significantly improved in a dose- dependent manner (P<0.05). 740 Y-P could reverse the improvement effects of high dose of Met on the above indexes of rats (P< 0.05). CONCLUSIONS Met can improve liver damage, and alleviate inflammatory reactions and liver fibrosis of NASH rats, the mechanism of which may be associated with inhibiting PI3K/AKT/PDGF signaling pathway.

Objective:To investigate the predictive value of telomerase reverse transcriptase (TERT) promoter C228T/C250T mutations in tumor tissues of patients with hepatocellular carcinoma (HCC) for postoperative recurrence after hepatectomy. Methods:Clinical data of 66 patients with HCC who underwent curative surgical resection at the Cancer Hospital, Chinese Academy of Medical Sciences, between January 2013 and May 2016 were retrospectively analyzed, including 54 males and 12 females, aged (53.5±11.1) years. Tumor tissues were collected from all patients. Droplet digital ploymerase chain reation (ddPCR) was employed to detect the TERT promoter C228T/C250T mutations. Survival outcomes were estimated using the Kaplan-Meier method and compared by the log-rank test. Univariate and multivariate Cox regression were used to analyze the impact of TERT promoter C228T/C250T mutations on postoperative recurrence. The predictive performance of TERT mutations for postoperative recurrence was further assessed using receiver operating characteristic (ROC) curve analysis. Results:The prevalence of TERT C228T and C250T mutations in tumor tissues was 43.9% (29/66) and 3.0% (2/66), respectively. Patients were stratified into a TERT promoter mutation group ( n=31) and a non-mutation group ( n=35). Those harboring C228T/C250T mutations exhibited significantly lower recurrence-free survival compared with non-mutated cases ( χ2=10.10, P=0.002). Multivariate Cox regression analysis showed that TERT promoter C228T mutation ( HR=2.24, 95% CI: 1.18-4.25, P=0.013) and TERT promoter C228T/C250T mutations in tumor tissue ( HR=2.49, 95% CI: 1.31-4.75, P=0.006) were associated with an increased risk of postoperative recurrence in patients with HCC. ROC analysis demonstrated the predictive accuracy for recurrence, with an area under the curve of 0.68 (95% CI: 0.55-0.81) for TERT C228T mutation and 0.71 (95% CI: 0.58-0.84) for combined C228T/C250T mutations. Conclusion:TERT promoter C228T/C250T mutations in tumor tissues of HCC patients detected by ddPCR are risk factors for postoperative recurrence and may serve as indicators for predicting recurrence.

OBJECTIVE:
This study investigated the antidepression mechanisms of Xiaoyaosan (XYS), a classic Chinese prescription, from the perspective of energy metabolism in the brain and intestinal tissues.
METHODS:
Chronic unpredictable mild stress model-a classic depression rat model-was established. Effects of XYS on behaviors and gastrointestinal motility of depressed rats were investigated. Effects of XYS on energetic charge (EC), adenosine triphosphate-related enzymes, and key enzymes of energy metabolism in both hippocampus and jejunum tissues of depressed rats were investigated using high-performance liquid chromatography, biochemical analysis, and real-time quantitative polymerase chain reaction, respectively. Spearman correlation analysis was conducted to construct a correlation network of "behavior-brain energy metabolism-intestinal energy metabolism" of depression.
RESULTS:
XYS significantly reduced the abnormal behaviors that observed in depressed rats and increased the EC and the activity of Na+-K+-adenosine triphosphatase (ATPase) and Ca2+-Mg2+-ATPase in hippocampus and jejunum tissues of depressed rats. XYS restored the key energetic pathways that had been interrupted by depression, including glycolysis, tricarboxylic acid cycle, and oxidative phosphorylation. Furthermore, XYS exhibited antidepressive effects in terms of regulating energy metabolism in tissues of both brain and intestine.
CONCLUSION
XYS significantly corrected the disturbances in EC and energy metabolism-related enzymes of both brain and intestinal tissues, alleviating both core and concomitant symptoms of depression. The current findings underscore the role of energy metabolism in the antidepressive activity of XYS, providing a fresh perspective on depression, and novel research strategies for revealing the mechanism of actions of traditional Chinese medicines on multi-site and multi-symptom diseases. Radiopharmaceuticals operate by combining radionuclides with carriers. The radiation energy emitted by radionuclides is utilized to selectively irradiate diseased tissues while minimizing damage to healthy tissues. In comparison to external beam radiation therapy, radionuclide drugs demonstrate research potential due to their biological targeting capabilities and reduced normal tissue toxicity. This article reviews the applications and research progress of radiopharmaceuticals in cancer treatment. Several key radionuclides are examined, including 223Ra, 90Y, Lutetium-177 (177Lu), 212Pb, and Actinium-225 (225Ac). It also explores the current development trends of radiopharmaceuticals, encompassing the introduction of novel radionuclides, advancements in imaging technologies, integrated diagnosis and treatment approaches, and equipment-medication combinations. We review the progress in the development of new treatments, such as neutron capture therapy, proton therapy, and heavy ion therapy. Furthermore, we examine the challenges and breakthroughs associated with the clinical translation of radiopharmaceuticals and provide recommendations for the research and development of novel radionuclide drugs.

Acta Physiologica Sinica 2025;77(3):419-431
Parkinson's disease (PD) is a common neurodegenerative disorder mainly related to mitochondrial dysfunction of dopaminergic neurons in the midbrain substantia nigra. This study aimed to investigate the effects of exercise preconditioning on motor deficits and mitochondrial function in a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mouse model. Eight-week-old male C57BL/6J mice were randomly divided into four groups: sedentary + saline (SS), sedentary + MPTP (SM), exercise + saline (ES), and exercise + MPTP (EM) groups. Mice in the ES and EM groups received 4 weeks of treadmill training, and then SM and EM groups were treated with MPTP for 5 days. Motor function was assessed by behavioral tests, and morphological and functional changes in dopaminergic neurons and mitochondria in the substantia nigra of the midbrain were evaluated using immunohistochemistry, Western blot, and transmission electron microscopy technology. The results showed that, compared with the SM group, the EM group exhibited significantly improved motor ability, up-regulated protein expression levels of tyrosine hydroxylase (TH) and dopamine transporter (DAT) in the midbrain, and down-regulated protein expression of α-synuclein (α-Syn) in the mitochondria of substantia nigra. Compared with the SM group, the EM group showed up-regulated protein expression levels of mitochondrial fusion proteins, including optical atrophy protein 1 (OPA1) and mitofusin 2 (MFN2), and biogenesis-related proteins, including peroxisome proliferator activated receptor gamma coactivator 1α (PGC-1α) and mitochondrial transcription factor A (TFAM), while the protein expression levels of dynamin-related protein 1 (DRP1) and mitochondrial fission protein 1 (FIS1) were significantly down-regulated. Compared with the SM group, the EM group showed significantly reduced damage to substantia nigra mitochondria, restored mitochondrial membrane potential and ATP production, and decreased levels of reactive oxygen species (ROS). These results suggest that 4-week treadmill pre-training can alleviate MPTP-induced motor impairments in PD mice by improving mitochondrial function, providing a theoretical basis for early exercise-based prevention of PD.
