1.Honokiol inhibits the malignant progression of gastric cancer cells by regulating the ATF4/CHOP/TRIB3 pathway

ObjectiveTo investigate the effect of honokiol on proliferation, apoptosis, migration, and invasion of gastric cancer cells and its underlying mechanistic. MethodsHuman gastric cancer cell lines HGC-27 and AGS were treated with Honokiol at concentrations of 0, 15, and 25 μmol/L. CCK-8 assays were conducted to determine the half maximal inhibitory concentration (IC50) for both cell lines. Cell viability, proliferation, migration, and invasion capabilities were assessed using CCK-8, colony formation, wound healing, Transwell migration and Transwell invasion assays. Apoptosis rates were measured via flow cytometry. Western blot analysis examined proteins related to proliferation, apoptosis, migration, invasion, and the endoplasmic reticulum stress pathway ATF4-CHOP-TRIB3. ResultsCompared with the control group, treatment with 15 and 25 μmol/L Honokiol significantly reduced the proliferation, colony formation, migration, and invasion capabilities of the two gastric cancer cell lines, while significantly increasing the apoptosis rate (P<0.05). Additionally, compared to the control group, the protein expression levels of neural cadherin(N-cadherin), Vimentin, proliferating cell nuclear antigen(PCNA), and B-cell lymphoma/leukemia-2 protein(Bcl-2)decreased in the two gastric cancer cell lines after treatment with 15 and 25 μmol/L Honokiol, while the protein expression levels of epithelial cadherin(E-cadherin), Bcl-2-associated X protein(Bax), activating transcription factor 4(ATF4), endoplasmic reticulum stress-related protein(CHOP), and tribbles homolog 3(TRIB3)increased(P<0.05). ConclusionHonokiol promotes apoptosis and inhibits proliferation, migration, and invasion of gastric cancer HGC-27 and AGS cells by regulating the ERS signaling pathway ATF4/CHOP/TRIB3.
2.Role of autophagy in regulating phagocytosis in Vibrio vulnificus-infected murine macrophages

Methods:The expression of cellular autophagy-related proteins in PBS-treated and V. vulnificus-infected RAW264.7 and BMMφ cells was detected by Western blot. The co-localization of V. vulnificus-GFP and LC3Ⅱ protein in V. vulnificus-GFP-infected RAW264.7 and BMMφ cells were detected using confocal microscopy. The phagocytosis of V. vulnificus in V. vulnificus-GFP-infected RAW264.7 and BMMφ cells with or without autophagy inhibition using Bafilomycin A1 was detected by flow cytometry. Results:The up-regulated levels of Atg7, Atg12 and Atg16L1 proteins, increased LC3Ⅱ/actin ratio, as well as down-regulated p62 protein levels were significantly detected in V. vulnificus-infected RAW264.7 and BMMφ cells. The co-localization of V. vulnificus-GFP and LC3Ⅱ protein was clearly observed in V. vulnificus-GFP-infected RAW264.7 and BMMφ cells. Enhanced phagocytosis of V. vulnificus and increased autophagy were exhibited in V. vulnificus-GFP-infected RAW264.7 and BMMφ cells, while weakened phagocytosis, accumulation of Atg7, Atg12, Atg16L1, LC3Ⅱ and p62 protein levels, as well as blocking autophagy flux were detected in those cells within autophagy inhibition using Bafilomycin A1. Conclusion:Autophagy induced by V. vulnificus infection could promote phagocytosis of V. vulnificus in macrophages.

4.Incidence and mortality of lung cancer in cancer registration areas of Yunnan province in 2020 and the trends from 2012 to 2020

Objective The aim of this study was to analyze the incidence and mortality of lung cancer in tumor registration areas of Yunnan province in 2020,as well as the changing trends from 2012 to 2020,in order to provide the data support for lung cancer prevention and control.Methods The reported cases of lung cancer from 2012 to 2020 in tumor registration areas of Yunnan province were collected,and SAS 9.4 software was used to calculate the incidence,mortality and other indicators to describe the cur-rent situation of lung cancer incidence and mortality.JoinPoint 4.9.0.0 software was used to calculate the annual percentage change(APC)to analyze the trend of lung cancer incidence and mortality.Results In 2020,the crude incidence of lung cancer in Yunnan province was 46.68/100,000(male 58.56/100,000 and female 34.21/100,000),age-standardized incidence rate by Chinese stand-ard population(ASIRC)was 30.83/100,000,age-standardized incidence rate by World standard population(ASIRW)was 30.61/100,000;The crude mortality was 33.88/100,000(male 45.94/100,000 and female 21.21/100,000),age-standardized mortality rate by Chinese standard population(ASMRC)was 21.70/100,000,age-standardized mortality rate by World standard population(ASMRW)was 21.61/100,000.In 2020,the incidence and mortality of lung cancer in Yunnan province showed an upward trend with the increase of age,and was at a low level before the age of 39,and increased rapidly after the age of 40.It reached its peak in the age groups of 75-79 and 80-84,respectively.The crude incidence of lung cancer in urban and rural areas was 47.21/100,000 and 46.52/100,000,respectively;The crude mortality of lung cancer was 32.66/100,000 and 34.27/100,000,respectively.From 2012 to 2020,the crude incidence of lung cancer in Yunnan province increased with the year(APC=2.94%,95%CI=0.29%-5.67%,P<0.05);The crude incidence of female lung cancer and ASIRW increased with the year(crude incidence rate:APC=6.30%,95%CI=2.52%-10.21%,P<0.05;ASIRW:APC=5.91%,95%CI=1.21%-10.82%,P<0.05).Conclusion In 2020,the incidence and mortality of lung cancer in Yunnan province increase significantly after the age of 40.The overall incidence and mortality of lung cancer in men are higher than those in women,and higher in rural areas than that in urban areas.At the same time,the incidence of lung cancer in women is increasing year by year.Therefore,males 40 years old and over above in rural areas should be identified as key populations for lung cancer prevention and control.Simultaneously,women lung cancer also pay attention to the prevention and control.

6.Anesthesia management in cesarean section for patient with COVID-19: a case report.

Since the coronavirus disease 2019 (COVID-19) affects the cardio-pulmonary function of pregnant women, the anesthetic management and protection of medical staff in the cesarean section is significantly different from that in ordinary surgical operation. This paper reports a case of cesarean section for a woman with COVID-19, which was successfully performed in the First Affiliated Hospital of Zhejiang University School of Medicine on February 8, 2020. Anesthetic management, protection of medical staff and psychological intervention for the pregnant woman during the operation were discussed. Importance has been attached to the preoperative evaluation of pregnant women with COVID-19 and the implementation of anesthesia plan. For moderate patients, intraspinal anesthesia is preferred in cesarean section, and try to reduce its influence in respiration and circulation in both maternal and infant; general anesthesia with endotracheal intubation should be adopted for severe or critically ill patients. Ensure the safety of medical environment, and anesthetists should carry out level-Ⅲ standard protection. Special attention and support should be paid to maternal psychology: fully explanation before operation to reduce anxiety; relieve the discomfort during operation, so as to reduce tension; avoid the bad mood due to pain after operation.

Since the corona virus disease 2019 (COVID-19) affects the cardio-pulmonary function of pregnant women, the anesthetic management in the cesarean section for the patients, as well as the protection for medical staff is significantly different from that in ordinary surgical operation. 8.A new quantitative 16S rRNA amplicon sequencing method.

In recent years, 16S rRNA amplicon sequencing technology has been widely used to study human gut microbiota and to detect unknown pathogens in clinical samples. However, its resolution to bacterial population can only reach the relative abundance of genus level, and different factors affect the final bacterial profile, such as sample concentrations, PCR cycle numbers and amplification primers. In order to solve these problems, we developed a quantitative 16S rRNA amplicon sequencing method by combining random tag and internal marker method. The new methods improved the accuracy of human gut microbiota, reduced the impact of experimental operation on the results, and improved the comparability between sequencing and other molecular biological methods.

9.Itraconazole against invasive fungal infection in patients of multiple organ dysfunction syndrome

Objective To evaluate the efficacy and safety of itraconazole injection in treatment of invasive fungal infection in the patients with multiple organ dysfunction syndrome.Methods Invasive fungal infection in 15 patients with multiple organ dysfunction syndrome was treated with 200 mg itraconazole injection,twice a day on the first two days,then once a day,for 8 to 14 days.During the treatment,the symptoms and signs,the results of chemical detection were recorded.Everyday fungus,sputum,stool smear or culture were performed.Results The recovery rate,effective rate,and fungal clearance in this trial with itraconazole injection were 80%(12/15),86.7%(13/15),and 86.7%(13/15) respectively.The side effect was not found.Conclusion Itraconazole injection is an effective and safe drug against invasive fungal infection in patients with multiple organ dysfunction syndrome.
