1.Analysis of factors influencing the operation time and complications of endoscopic unilateral thyroidectomy by a gasless unilateral axillary approach

Objective To explore the factors that may influence the operation time and complications of endoscopic unilateral thyroidectomy by a gasless unilateral axillary approach.Methods Clinical data of 203 consecutive patients who underwent endoscopic unilateral thyroidectomy by a gasless unilateral axillary approach from December 2022 to November 2023 were collected.Results Gender,BMI,TG,glandular lobe volume,number of lesions,and benign and malignant tumors affected the length of surgery;Among them,male,BMI>19.45 kg/m2 and glandular lobe volume>9.25 mL were independent influences on the length of surgery(P<0.05).TG and tumor maximal diameter affected the occurrence of complications;There was no independent influence factor.Conclusion Female patients with BMI≤19.45 kg/m2 and glandular lobe volume ≤ 9.25 mL(which can be measured preoperatively by 3D slicer)are relatively less difficult to perform the procedure and may be more suitable for beginners to learn.

3.Status of cognition and needs of family integrated care among parents of premature infants in a children 's hospital in Chongqing

Objective:To investigate the cognition and demands of parents of premature infants on family integrated care (FICare) and to provide reference for formulating appropriate FICare programs.Methods:Using the convenient sampling method, a total of 160 parents of premature infants who were followed up in Neonatal Integrated Clinic of Children 's Hospital of Chongqing Medical University from March to July 2021 were selected as the research objects. The parents of premature infants were surveyed using a self-designed Questionnaire on the Cognitive Needs of Parents of Premature Infants on FICare. A total of 160 questionnaires were distributed and 140 valid questionnaires were recovered, with the effective recovery rate of 87.5%. Results:Among 140 parents of premature infants, 135 parents thought FICare was beneficial. 71.9% (97/135) parents chose to participate in 1 to 4 hours of care every day, but 31.1% (42/135) parents lacked confidence in care. Among the FICare providers selected by the parents, 94.1% (127/135) were neonatologists, 88.9% (120/135) were neonatal nurses, 62.2% (84/135) were breast-feeding counselors and 62.2% (84/135) were dietitians. 66.7% (90/135) parents and 58.5% (79/135) parents chose simulation teaching and online course as the ideal training mode. Discharge guidance was the service that parents urgently needed, with a score of 74.5%.Conclusions:Parents of premature infants have multi-level and multi-dimensional demands for FICare, so it is necessary to develop multi-level and all-round FICare services based on parents ' demands in the future to improve the outcomes of families of premature infants.
4.Research progress on parental coping ability of premature infants after discharge

5.Secondary sex ratio analysis of singleton babies born following assisted reproductive technology

Objective:To explore the influencing factors of the secondary sex ratio (SSR) of singleton babies born following assisted reproductive technology.Methods:A total of 7020 single babies delivered by assisted reproductive technology at the Reproductive Medicine Center of the First Affiliated Hospital of USTC (Anhui Provincial Hospital) from January 2000 to December 2018 were collected in a retrospective cohort study. The effect of influencing factors, such as maternal age, fertilization method, embryo transfer type and embryo transfer period on the sex of singleton babies were analyzed.Results:Among 7020 babies, 3730 were boys and 3290 were girls. The total SSR was 113∶100. The SSR of in vitro fertilization (IVF) was significantly higher than that of intracytoplasmic sperm injection (ICSI) fertilization (121∶100 vs. 95∶100, P<0.001), while other factors had no significant effect on the SSR of infants born. In the stratified analysis, there was no difference in SSR between IVF and ICSI in the blastocyst embryo transfer group with mother's age 35 years or older ( P<0.05), while the SSR of IVF in the other groups was higher than that of ICSI (<35 years old group: P<0.001; fresh embryo group: P=0.001; frozen-thawed embryo group: P=0.003; cleavage embryo transfer group: P<0.001). Univariate logistic regression analysis showed that the fertilization method in singleton pregnancy had a statistically significant effect on baby SSR ( OR=0.792, 95% CI=0.712-0.881, P<0.001). After removing confounding factors, logistic regression analysis showed that the fertilization method in singleton pregnancy had a statistically significant effect on neonatal SSR (a OR=0.793, 95% CI=0.713-0.883, P<0.001). Conclusion:The fertilization method of assisted reproductive technology can affect the SSR of singleton newborns. The SSR of IVF is significantly higher than that of ICSI and the difference was significant.

7.Effects of Shikonin on Autophagy and Apoptosis of Human Colon Cancer HCT 116 Cells

OBJECTIVE:To study the effects of shikonin on autophagy and apoptosis of human colon cancer HCT 116 cells. METHODS:After treating HCT 116 cells for 48 h with shikonin at 0(blank control )10,20,40 μg/mL,MTT method was used to detect inhibitory rate of cell proliferation. Flow cytometry was used to detect cell apoptosis rate. RT-qPCR assay and Western blotting were respectively used to detect mRNA and protein expressions of microtubule associated protein light chain 3(LC3)and autophagy-related protein Beclin- 1 and p 62. RESULTS :Compared with blank control ,after treated with 10,20,40 μ g/mL shikonin for 48 h,proliferation inhibitory rate and apoptosis rate of HCT 116 cells were increased significantly (P<0.05 or P< 0.01). After treated with 10,20,40 μg/mL shikonin for 48 h,mRNA and protein expressions of LC 3,Beclin-1 and p 62 in HCT116 cells were increased to different extents ;except that mRNA expression of LC 3 was not increased significantly after treated with 10 μg/mL shikonin,the difference were statistically significant in other indexes ,compared with blank control (P<0.05 or P<0.01). CONCLUSIONS :Shikonin can induce the apoptosis of human colon cancer HCT 116 cells and activate its autophagy pathway.
8.Application of integrated medical-nursing management model for clinical nursing teaching in neonatology department

Objective:To explore the application of integrated medical-nursing management model for clinical nursing teaching in neonatology department.Methods:A total of 28 undergraduate nursing students having internship in neonatology department from August 2018 to February 2020 were enrolled in the study, among which 15 nursing students from August 2018 to May 2019 were selected as the control group, and 13 nursing students from August 2019 to February 2020 were selected as the observation group. The control group was given traditional teaching, and the observation group was given integrated teaching of medical care. The theoretical scores, nursing operation, comprehensive quality, total scores and teaching satisfaction of the two groups were compared. SPSS 19.0 was used for t test. Results:The theoretical scores of the observation group and the control group were (96.31±0.85) points and (88.47±1.92) points, respectively. Nursing operation scores of observation group and control group were (95.18±2.06) points and (93.15±2.13) points. Comprehensive quality of observation group and control group were (96.23±1.48) points and (90.47±1.88) points. The total scores of the observation group and the control group were (96.83±1.27) points and (91.20±1.31) points, respectively. All the differences were statistically significant ( P<0.05). Ther were significant differences between the two groups in terms of teaching satisfaction including teaching methods, contents, teachers' satisfaction and total satisfaction( P<0.05). Conclusion:In the clinical nursing teaching of neonatology department, the integrated medical-nursing management model is helpful to improve the clinical comprehensive quality and teaching satisfaction of undergraduate nursing students.

10.Effects of Benzoyl Aconitine on Autophagy and Apoptosis of Human Lung Cancer Cell Line A 549

OBJECTIVE: To study the effects of benzoyl aconitine (BAC) on autophagy and apoptosis of human lung cancer A549 cells, and to investigate its mechanism in anti-non-small cell lung cancer. METHODS: A549 cells were treated with different doses of BAC (10, 50, 100, 200, 400 μmol/L), and then cell morphology was obtained; the proliferation inhibition rate of the cell was determined by CCK-8 assay. The cells were divided into control group (without drug), BAC low-dose and high-dose groups (200, 400 μmol/L). After treated with relevant drugs, the apoptosis rate of cells was determined by flow cytometry. The gene and protein expression of apoptosis-related factors Bcl-2, Bax, Caspase-3 as well as autophagy-related factors Beclin1, LC3, P62 were determined by RT-PCR and Western blotting assay. RESULTS: After treated with different doses of BAC, the cells were shrunken and sparsely arranged; inhibitory rate of cell proliferation was increased significantly in BAC 100, 200, 400 μmol/L groups (P<0.05 or P<0.01). Results of flow cytometry showed that the apoptotic rates of cells were increased to different extents in BAC low-dose and high-dose groups after treated for 24 and 48 h, in a concentration and time-dependent manner. Compared with control group, mRNA and protein expression of Bcl-2 and P62 were decreased to different extents in BAC groups; mRNA expression of Bax, Caspase-3, Beclin1 and LC3 as well as protein expression of Bax, Active caspase-3, P62, Beclin1, LC3Ⅱ/Ⅰ were increased to different extent; there was statistical significance in mRNA expression of Caspase-3, and protein expression of Bcl-2, Active Caspase-3, Beclin1, LC3Ⅱ/Ⅰ and P62 in BAC low-dose group as well as all target mRNA and protein expression in BAC high-dose group (P<0.05 or P<0.01), in dose-dependent manner. CONCLUSIONS: BAC can inhibit the proliferation and promote the apoptosis of A549 cells, promote Beclin1, LC3(LC3Ⅱ/Ⅰ),Bax and Caspase-3 (Active Caspase-3) gene and their protein expression, but inhibit P62 and Bcl-2 gene and their protein expression. The mechanism may be related to BAC inducing apoptosis by promoting excessive autophagy of cells.
