1.Ginsenoside CK mediates the regulation of the AMPK/mTOR pathway in gastric cancer cell proliferation, apoptosis and autophagy

Objective:To investigate the effects of ginsenoside Compound K (CK) on proliferation, apoptosis, and autophagy in gastric cancer cells, and to further explore whether it exerts its effects by regulating the AMPK/mTOR signaling pathway.Methods:Human gastric cancer AGS cells were cultured in vitro to the logarithmic growth phase and randomly divided into control group (treated with 0.9% sodium chloride solution), ginsenoside CK group (treated with 50 μM CK) and ginsenoside CK+ Dorsomorphin group (treated with 50 μM CK combined with 10 μM Dorsomorphin). AGS cells were treated with ginsenoside CK at concentrations of 0, 5, 10, 20, 50, 100, and 200 μM, and cell viability was measured by the CCK-8 assay to determine the optimal concentration (50 μM). After treatment according to the above groups, cell viability was assessed by the CCK-8 assay; colony formation was evaluated by the plate clone formation assay; apoptosis was detected by the Tunel assay; autophagy was assessed by MDC fluorescence staining; and the expression levels and phosphorylation status of autophagy-related proteins (Beclin-1, LC3-Ⅱ), apoptosis-related proteins (Caspase-3, Bax, Bcl-2), and AMPK/mTOR signaling pathway-related proteins were analyzed by Western Blot. Statistical analysis of the data was performed using GraphPad Prism 9.1.2 software.Results:Compared with the control group, the apoptosis rate, autophagy degree and the expression levels of Beclin-1, LC3-Ⅱ, caspase-3, Bax and p-AMPK proteins were significantly increased in the ginsenoside CK group ( P<0.05), while the cell viability, clone formation rate, and the expression levels of Bcl-2 and p-mTOR proteins were decreased ( P<0.05). After the addition of AMPK pathway inhibitor Dorsomorphin based on 50 μM ginsenoside CK treatment, the apoptosis rate, the degree of autophagy, and the expression levels of Beclin-1, LC3-Ⅱ caspase-3, Bax, and p-AMPK proteins showed a decreasing tendency ( P<0.05). In contrast, the cell viability, the clone formation rate, and the expression levels of Bcl-2 and p -mTOR protein expression levels showed a certain rebound trend ( P<0.05). The anti-tumor effect of ginsenoside CK was reversed by the combined intervention of ginsenoside CK and dorsomorphin in AGS cells. Conclusion:Ginsenoside CK suppresses gastric cancer cell proliferation while enhancing apoptosis and autophagy through modulation of the AMPK/mTOR signaling pathway, offering a promising novel approach for gastric cancer therapy.
2.Validity and reliability test of the Chinese version of Nurse Delirium Care Competency Scale

Objective:To translate the Nurse Delirium Care Competency Scale (NDCCS) into Chinese and test the validity and reliability of Chinese version of NDCCS among ICU nurses.Methods:According to Brislin translation model, the Chinese version of NDCCS was developed by forward translation, back translation, cross-cultural adaptation and pre-survey. From October to December of 2023, 338 ICU nurses from Sir Run Run Shaw Hospital, Zhejiang University School of Medicine were selected by cross-sectional survey to test the reliability and validity of Chinese version of NDCCS.Results:A total of 329 valid questionnaires were collected. There were 50 males and 279 females, aged 20-53 years old. The Chinese version of NDCCS included 6 dimensions and 27 items. The scale-level content validity value was 0.985, and the item-level content validity value was 0.800-1.000. Exploratory factor analysis showed a six-factor structure, the cumulative variance contribution rate was 75.700%, the Cronbach′s α coefficients of the scale amount table were 0.952, and the Cronbach′s α coefficients of the dimensions were 0.811- 0.936,and the retest reliability was 0.932.Conclusions:The Chinese version of the NDCCS has good reliability and validity, and it is suitable for assessing the delirium care competence of ICU nurses in China.

4.Combination of online teaching and BOPPPS model to promote sticky learning

To enhance the learning stickiness, improve low completion rate of online teaching, and promote teaching quality has become the key to solve the contradiction in online teaching. In this paper, taking the teaching of biochemistry as example, based on the trigger mechanism, maintenance mechanism and migration mechanism of sticky learning, guided by the three-dimensional goal of "knowledge and skills, process and method, emotional attitude and values", the BOPPPS (bridge-in, objective, pre-assessment, participatory-learning, post-assessment, summary) teaching model was combined with online teaching. According to the interactive behavior in the course learning space, the Spearman rank correlation analysis was performed by SPSS 18.0 software to comprehensively evaluate the learning stickiness degree. The research has found that, due to its "micro but refined, compact structure and student-centered" characteristics, BOPPPS combining with online teaching can effectively make up for the time and space limitations of offline teaching and the excessively broad online teaching, bring benefits from the perspectives of "inclusion, attraction and production", promote students' active learning, and practically improve learning stickiness. The research provides a new idea for creating online "golden" courses.
5.Research progress of nursing succession in nursing management

Objective:In order to analyze the current research status of handover shift in nursing management, summarize, analyze and judge the existing literature, in order to provide reference for clinical nursing practice.Methods:Through literature review, it is planned to review the current situation, shortcomings and future development of nursing handover classes.Results:The handover process was generally divided into four stages, of which SBAR was the best practice tool for handing over key information. For the performance of handover shifts, NASR, PVNC-BR, HES and Handoff CEX were often used to evaluate the performance of shifts, and for the results of shifts, evaluations were mostly conducted at the levels of patient safety, process elements, and organizational management. At present, the use of electronic information systems, benign organizational culture and patient and family-centered clinical practice could effectively improve the efficiency and effectiveness of handover.Conclusions:The process and elements of the current shift mode are relatively complete, and the communication strategy is reasonable, but there are still many shortcomings and defects. This suggests that nursing managers should adopt scientific intervention methods and evaluation tools when paying attention to and reforming nursing handover in the future to continuously improve the quality of handover.
6.Translation, revision and assessment of reliability and validity of the Assessment of Interprofessional Team Collaboration Scale

0bjective To translate and revise the Assessment of Interprofessional Team Collaboration Scale (AITCS) into Chinese, then to evaluate the reliability and validity of the Chinese version of AITCS. Methods The Chinese version of AITCS was translated from the original one, back-translated and adjusted for cultural adaptation. The reliability and validity were tested among 288 nurses, 81 physicians, 25 respiratory therapists, 10 physical therapists and 3 nutritionists from a tertiary hospital in Hangzhou using convenience sampling. Results The average of scale-level content validity index was 0.98, unanimity of scale-level content validity index was 0.84 and item-level content validity index was 0.89-1.00. Exploratory factor analysis extracted three common factors,which explained 61.427% of the total variance,and each item had high factor loading quality (>0.4). The correlations coefficients between each dimension score and the total score ranged from 0.801 to 0.898 (P<0.05),and the correlations coefficients between each dimension score ranged from 0.607 to 0.698 (P<0.05). The Cronbach α of AITCS was 0.909, split-half reliability was 0.835 and test-retest reliability was 0.763. Conclusion The Chinese version of the AITCS has been proved to be reliable and valid. It is a valuable tool for evaluating interprofessional team collaboration among the health professional providers in mainland China.

The "student-centered" concept highlights the dominant position of students in the teaching process and makes up for the deficiency of traditional teaching mode.Based on the above idea,the teaching team of the author's department tried to improve teaching effect through effective teaching design in biochemistry.Firstly,the teaching goal (the goal of knowledge,ability and emotion) and the important situational factors the learning situation analysis,the characteristics of the course and the class,etc)should be identified.suitable teaching strategies he inspiration strategy,exploration strategy and problem solution strategy,etc.),teaching method (discussion,case study,problem based learning,etc.) and proper assessment (such as the summative assessment with formative evaluation) should be determined to organize classroom teaching.At the end of the class the timely feedback and the teaching reflection should be carried outto extend classroom teaching to extra-curricular class.The results showed that effective teaching designismore likely to stimulate students' learning enthusiasm,promote students' deep learning and achieve better teaching effects.
8.Intervention effect of chlorhexidine gargle on respiratory tract infection following general anesthesia

Objective To evaluate the effect of chlorhexidine gargle on respiratory tract infection (RTI)in patients following general anesthesia.Methods From January 2012 to June 2013,94 patients who received general anesthe-sia for removal of vertebral disc were randomly divided into two groups,with 47 cases in each group,control group gargled with normal saline,observation group accepted 2% chlorhexidine gargle.The occurrence of RTI and detec-tion of pharynx pathogens of two groups were compared.Results The incidence of RTI in observation group and control group was 8.51%(n=4)and 23.40%(n=11)respectively(χ2 = 12.95,P <0.05).4 patients in observation group were detected 4 isolates of pathogens,11 patients in control group were detected 26 isolates of pathogens,the detection rate of pharynx pathogens of observation group was lower than control group (χ2 =3.89,P <0.05).The main isolated pathogens were Pseudomonas aeruginosa ,Staphylococcus aureus ,Klebsiella pneumoniae and Esche-richia coli .Conclusion Chlorhexidine gargling can effectively reduce RTI following general anesthesia,it is worthy of clinical application.
9.Effect of depsides salts from Salvia miltiorrhiza on human hepatoma cell line SMMC-7721 subcutaneous xenografts in nude mice.

OBJECTIVE:
To exlpore the eff ect of depsides salts from Salvia miltiorrhiza on human hepatoma cell line SMMC-7721 xenograft tumors and the possible mechanisms.
METHODS:
A total of 36 nude mice were divided into 6 groups: A model group, a negative control group, a positive control group, and 3 treatment groups at low, middle or high dose (n=6). The tumor model of nude mice was given depsides salts at a dose of 10, 20 or 50 mg/kg every 3 day for 16 days. Then samples of subcutaneous tumors in nude mice were collected. The morphological changes of tumor samples were observed by HE staining and the expression of vascular endothelial growth factor (VEGF) and the tumor antigen Ki67 was detected by immunohistochemical method.
RESULTS:
The tumor growth was inhibited by all doses of depsides salts. The morphology of tumors was shrinkage, broken and irregularly arranged compared with the tumors in the model group and the negative control group. Morphological changes were more obvious in tumors with treatment at high dose. Expression of VEGF and Ki67 in treatment groups and the positive control group were lower than that in the model group and the negative control group, with a significant difference (P<0.05).
CONCLUSION
Depsides salts from Salvia miltiorrhiza can inhibit the growth of human hepatoma cell line SMMC-7721 tumor in nude mice, which is related to the inhibition of Ki67 and VEGF.

10.Change of serum myeloperoxidase and lipoxin A4 level in coronary heart disease patients with anxiety and/or depression.

OBJECTIVE:
To investigate the change of serum myeloperoxidase (MPO) and lipoxin A4 (LXA4) in coronary heart disease (CHD) patients with anxiety and depression and its clinical significance.
METHODS:
From December 2010 to February 2011, 143 CHD patients and 44 non-CHD patients (the control group) hospitalized in the Department of Cardiology at the Second Xiangya Hospital were enrolled. The hospital anxiety and depression scale (HADS) was used to evaluate the psychological state of all patients and the CHD patients were assigned to an anxiety and depression group (n=57) or a non-depression and anxiety group (n=86). The serum levels of high sensitive C-reactive protein (Hs-CRP), MPO, and LXA4 were examined, and the ratio of MPO and LXA4 (M/L) was calculated.
RESULTS:
The levels of Hs-CRP, MPO, and LXA4 as well as M/L ratios in both CHD groups were significantly higher than those in the control group (all P<0.01). Compared with the non-anxiety and depression group, the levels of MPO and LXA4, and M/L ratios in the anxiety and depression group increased (all P<0.05). Correlation analysis showed that MPO was positively correlated with the score of HADS-total (HADS-t), HADS-anxiety (HADS-a), or HADS-depression (HADS-d), while LXA4 was negatively correlated with HADS-t or HADS-d. Multiple ordinal logistic regression analysis revealed that higher HADS-t score, stable angina, unstable angina, and acute myocardial infarction were the independent impact factors for the elevation of M/L ratio.
CONCLUSION
Anxiety and depression may aggravate the inflammatory response in CHD patients. The imbalance between inflammation and anti-inflammation may be part of the mechanism.
