1.The relationship between serum ferritin and metabolic associated fatty liver disease in adolescents

Objective To investigate the relationship between serum ferritin(SF)and metabolic associated fatty liver disease(MAFLD)in adolescents.Methods This study utilized data from 2017 to 2020 national health and nutrition examination survey database,ultimately including 559 adolescents aged 12~18 years,divided into MAFLD group(119 cases)and non-MAFLD group(440 cases).Data on demographic characteristics,serum iron metabolism indicators,liver function,lipid profiles,glucose levels,and other relevant parameters were collected.Liver steatosis and fibrosis were assessed using vibration-controlled transient elastography.Pearson correlation and Logistic regression analyses were performed to evaluate the relationship between SF and MAFLD.Results SF levels were significantly higher in MAFLD group compared to non-MAFLD group(P<0.05).Pearson correlation analysis indicated that SF was positively correlated with the controlled attenuation parameter,liver stiffness measurement,body mass index(BMI),alanine aminotransferase(ALT),γ-glutamyl transferase,triglycerides,and uric acid levels(P<0.05),but negatively correlated with high-density lipoprotein cholesterol levels(P<0.05).Adolescents were divided into 4 groups according to the quartiles of SF levels(Q1 goup:SF 1.9-25.6ng/ml;Q2 goup:SF 25.7-41.7ng/ml;Q3 goup:SF 41.8-70.4ng/ml;Q4 goup:SF 70.5-445.0ng/ml).The results of Logistic regression analysis showed that when the variables were not adjusted,the risk of MAFLD in Q4 group adolescents was significantly increased,this association disappeared after adjusting for gender,age,race,BMI,and other factors.BMI(OR=1.250,95%CI:1.176-1.329),globulin(OR=2.991,95%CI:1.218-7.343),ALT(OR=1.052,95%CI:1.003-1.104),and white blood cell count(OR=1.225,95%CI:1.041-1.441)were identified as independent risk factors for MAFLD.Conclusion SF levels are significantly elevated in adolescents with MAFLD but are not independently associated with the occurrence of MAFLD.BMI,globulin,ALT,and white blood cell count are important risk factors for adolescent MAFLD.

3.Experiences in construction of infection disease outpatient service

The infectious disease outpatient service as a frontier is an important fulcrum of public health service. Its standardized construction is an important support for ensuring medical safety, reducing nosocomial infections, and controlling the epidemic of infectious diseases. The sub-specialty outpatient service of infection diseases includes fever outpatient service, intestinal outpatient service, tuberculosis outpatient service, AIDS outpatient service, liver disease outpatient service, etc. According to the characteristics of each subspecialty outpatient service and combining with clinical practice, we elaborated the setting norms of subspecialty outpatient service for common infectious diseases from the perspective of planning and design, building layout, equipment and facilities configuration, staffing, daily management and demonstration.
4.Isolation and characterization in vitro and in vivo of pancreatic islets from NOD mice

Objective This study was conducted to establish a stable and highly efficient method for isolation and purification of pancreatic islets from NOD mice and to evaluate their characteristics in vitro and in vivo. Methods The is-lets were isolated from mouse pancreas using modified collagenase digestion and Ficoll density gradient centrifugation. The endocrine secretory function was assessed by insulin secretion in either low or high dose glucose stimulation. To evaluate the function of the graft,body weight and blood glucose were monitored,and IVGTT was performed. In addition,to assess sur-vival of the implanted islets,Pathology using HE staining and insulin immunostaining of the graft were performed. Results The average islet yield was 116 ± 12 islets/pancreas and purity was higher than 90%. Compared with islets from Kunming mice,the islets isolated from NOD mice were poorly responsive to glucose challenge. Blood glucose levels and body weight changes of the islet-transplanted diabetic mice were significantly improved compared with the sham-operated mice. In addi-tion,blood glucose levels in vivo after an IVGTT also significantly improved. However,these improvements were only main-tained for 2 weeks. Furthermore,HE staining and immunostaining assays demonstrated that there were insulin-positive cell clusters and lymphocyte infiltration in the graft-bearing kidney. Conclusions A large number of quality islets can be isola-ted and purified from NOD mice by using the modified mouse islet isolation method, which can be used to develop thera-peutic strategies to protect transplanted islets from rejection and autoimmune attack.
5.Association between gene polymorphism of plasminogen activator inhibitor and ischemic cerebrovascular disease in aged people of Henan Han nationality

Objective To investigate the association between gene polymorphism of plasminogen activator inhibitor (PAI-1)and ischemic cerebrovascular disease in aged people of Henan Han nationality.Methods A case control method was used,including 408 patients with ischemic cerebrovascular disease and 418 age and gender matched healthy controls.Polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) method was used to determine the distribution of allele and genotype frequencies of PAI-1 844G/A.Results In patient group,the gene frequencies of AA,AG,GG were 18.1%,41.4%,40.4%,and the A and G allele frequencies were 38.8% and 61.2%,respectively.In the control group,the gene frequencies of AA,AG,GG were 15.8%,47.4%,36.8%,and the A and G allele frequencies were 39.5% and 60.5%.There were no significant differences in the frequencies of genotypes and alleles between the two groups (P>0.05).The different genotypes of 844G/A of PAI-1 gene were not associated with hypertension,hyperglycemia,plasma homocysteic acid levels (P > 0.05).Conclusions PAI-1 gene-844G/A polymorphism may not be independent risk factor of ischemic cerebrovascular disease in aged people of Henan Han nationality.
