Objective:To investigate the role of platelet-derived zyxin in promoting tumor migration by platelets. Methods:The gene expression profile of platelets was analyzed from cancer patients by using the GEO database.Isolated platelets from wild-type(WT)and Zyx-/-mice were co-cultured with B16F10 cells labeled with green fluorescence to investigate the influence of zyxin deficiency on tumor cell migration, invasion, and wound healing.Optical microscopy was employed to evaluate the impact of zyxin deficiency on epithelial-mesenchymal transition(EMT)in B16F10 cells induced by platelets.Employing specific markers to label platelets, fluorescence confocal microscopy was utilized to investigate the impact of platelet-derived zyxin on the binding between tumor cells and platelets. And an aggregometer was employed to observe the influence of zyxin deficiency on tumor cell-induced platelet aggregation. Results:Compared to platelets from healthy volunteers, zyxin was upregulated in platelets from cancer patients.Zyx-/-mouse platelets exhibited a significant reduction in tumor cell invasion and migration, impaired wound healing, and delayed tumor cell EMT compared to WT mouse platelets.Additionally, zyxin deficiency attenuated the interaction between platelets and tumor cells, and diminished the capacity for tumor cell-induced platelet aggregation.Conclusion:Platelet-derived zyxin deficiency diminishes platelet-tumor cell interactions and weakens the ability of tumor cell-induced platelet aggregation, ultimately suppressing tumor cell migration.

Objective:To observe the accuracy of artificial intelligence-assisted design and 3D printed personalized osteotomy guide plate-assisted total knee arthroplasty (TKA).Methods:A total of 112 patients with knee osteoarthritis scheduled for TKA admitted to the Second People′s Hospital of Guangxi Zhuang Autonomous Region from February 2022 to February 2024 were prospectively included. They were divided into the intelligent group and the traditional group by lottery, with 56 cases in each group. The traditional group used conventional osteotomy guides, while the intelligent group used artificial intelligence-assisted design and 3D printed personalized osteotomy guides. The surgical conditions, postoperative imaging indicators, deviation range of hip-knee-ankle angle (HKA) and clinical efficacy of the two groups were observed. Results:There was no statistically significant difference in incision length between the two groups ( P>0.05). The operation time and blood loss in the intelligent group were both less than those in the traditional group (all P<0.05). The differences in postoperative HKA, sagittal tibial prosthesis angle (LTC), and sagittal femoral prosthesis angle (LFC) in the intelligent group were all smaller than those in the traditional group (all P<0.05), but there were no statistically significant differences in coronal femoral prosthesis angle (FFC) and coronal tibial prosthesis angle (FTC) between the intelligent group and the traditional group ( P>0.05). There was a statistically significant difference in the deviation range grade of HKA between the two groups ( P<0.05), and the proportion of HKA deviation ≤1° in the intelligent group was higher than that in the traditional group ( P<0.05). After the operation, the Visual Analogue Scale (VAS) scores for pain in both groups were lower than those before the operation (all P<0.05), and the range of motion (ROM) of the knee joint and the American Knee Society Score (KSS) were both increased compared with those before the operation (all P<0.05). However, there were no statistically significant differences in the VAS scores, ROM, and KSS of pain after the operation between the two groups (all P>0.05). Conclusions:Artificial intelligence-assisted design and 3D printed personalized osteotomy guide plates can improve the accuracy of TKA surgery, shorten the operation time and reduce the amount of blood loss.

Albendazole-glucan particles(ABZ-GPS)and abendazole(ABZ)were used to treat secondary alveolar echinococ-cosis in mice. The therapeutic effects of ABZ-GPS on alveolar echinococcosis in vivo were evaluated, and the feasibility of using glucan particles as anti-hydatid drug carriers was further verified.Mice with echinococcosis were randomly divided into an ABZ group, glucan nanoparticle(GP)group,ABZ-GPS group, and control group. After four courses of administration with a final concentration of 50 mg/mL,the therapeutic effects of ABZ-GPS were evaluated on the basis of imaging, histopathological changes, ultrastructure, and immunology. After ABZ-GPS and ABZ administration, clear liver lesion tissue necrosis and large numbers of infiltrating lymphocytes were observed.Significant differences in the average cyst wet weight(t=7.83,P<0.05),were observed between the ABZ-GPS group and ABZ group.Imaging revealed that ABZ-GPs were targeted to liver tissue.Pa-thology and ultrastructure analyses demonstrated that the alveolar hydatid cells in the liver in the control group and GP group grew well, and the vesicles were large, filled with cystic fluid, and translucent or transparent;the cyst wall tension was high;no calcification was observed;the stratum corneum and germinal layer were clear;and more fertile capsules and different num-bers of protocephalospora were present in the liver. In the ABZ group, the capsular cavity collapsed, and showed partial necro-sis and lymphocyte infiltration. In the ABZ-GP group, the corneum and germinal layer of echinococcus vesicles were difficult to identify, and we observed bulbous necrosis, central calcification, fibrous tissue hyperplasia, inflammatory cell infiltration, coarser, shorter or absent microvilli of the germinal layer, nuclear shrinkage, dissolution or disappearance, clear expansion of cytoplasmic microtubules, and myelin-like or vacuole-like changes.Therefore,ABZ-GPs showed good targeting and killing ac-tivity in vivo in mice with secondary alveolar coccosis.

Objective To develop a fused model based on combined intratumor-peritumor radiomics features and clinical risk factors to accurately predict the efficacy of radiotherapy for hepatocellular carcinoma(HCC).Methods A retrospective study was conducted on 124 HCC patients(95 males and 29 females)undergoing IMRT at the radiotherapy center of some hospital, who were divided into a training set and a validation set in a 7∶3 ratio.According to relevant standards for evaluating solid tumor treatment efficacy the patients at the progressive disease phase were enrolled into a non-response group, and the ones at the phases of complete response, partial response and stable disease were included into a response group.Firstly, radiomics features were extracted from pre-radiotherapy contrast-enhanced CT images, and were screened with the least absolute shrinkage and selection operator(LASSO)regression to obtain valuable radiomics features;secondly, the scores of intratumor radiomics features, peritumor radiomics features and combined intratumor-peritumor radiomics features underwent binary Logistic regression(LR)analysis by using the glmnet package in R software(version 4.2.0),so as to construct three models for intratumor radiomics features, peritumor radiomics features and combined intratumor-peritumor radiomics features;finally,a fused model was established based on the scores of combined intratumor-peritumor radiomics features and clinical risk factors, and the corresponding Nomogram, calibration curve and clinical decision curve were plotted.R software(version 4.4.1)was used for image analysis and statistical analysis. Results The fused model based on the scores of combined intratumor-peritumor radiomics features and clinical risk factors achieved an AUC of 0.87 on both the training and validation sets, outperforming the models based solely on intratumor radiomics features, peritumor radiomics features or combined intratumor-peritumor radiomics features. The Nomogram plot showed the prediction probability of the fused model for the short-term treatment response rate was 10%-90%within the total score range of 42-93 points, and the calibration and clinical decision curves indicated that the fused model gained advantages in prediction and clinical application. Conclusion The fused model based on the scores of combined intratumor-peritumor radiomics features and clinical risk factors predicts the short-term efficacy of HCC radiotherapy effectively, providing references for formulating radiotherapy plans and guiding subsequent treatment decisions.
