1.Compound Danshen Tablets ameliorate myocardial ischemia/reperfusion injury-induced ventricular remodeling by regulating autophagy via AMPK/mTOR signaling pathway.

OBJECTIVE:
Left ventricular remodeling induced by myocardial ischemia/reperfusion injury (MI/RI) is a common cardiac dysfunction. Accumulating evidence has demonstrated that autophagy plays a vital role in protecting against ventricular remodeling. This study aims to investigate the performance of Compound Danshen Tablets (CDT) in rescuing ventricular remodeling and whether autophagy as the potential mechanism.
METHODS:
The left anterior descending arteries of rats were temporarily ligated for 30 min to construct the MI/RI model. Ventricular remodeling was induced by reperfusion for 28 d, during which the MI/RI rats were administered CDT (300 mg/kg and 600 mg/kg), atorvastatin (2 mg/kg), and diltiazem (16 mg/kg). Cardiac function and structure were examined by echocardiography. Immunohistochemistry, Masson's trichrome staining, and hematoxylin-eosin (HE) staining were utilized to assess the fibrosis and histological alterations in the heart tissue. The expression of autophagy-related proteins was detected using Western blotting.
RESULTS:
CDT attenuated the cardiac dysfunction, structural changes, histopathological changes and fibrosis induced by MI/RI. CDT significantly enhanced the level of Beclin1 and microtubule-associated protein 1 light chain 3 beta (LC3β), and reduced p62 levels in MI/RI rats. Moreover, CDT significantly increased the phosphorylation of adenosine monophosphate-activated protein kinase (AMPK) and inhibited mammalian target of rapamycin (mTOR) phosphorylation.
CONCLUSION
CDT ameliorated MI/RI-induced ventricular remodeling by activating autophagy and improving autophagic flux via the AMPK/mTOR signaling pathway.
2.Analysis of supply satisfaction with community-embedded elderly care services in Nanchang city

ObjectiveTo investigate the supply satisfaction with community-embedded elderly care services in Nanchang city, which can assist in optimizing its supply system of elderly care services and build a policy system for high-quality development of community-embedded elderly care services for reference. MethodsA total of 15 indicators from three dimensions: resource embeddedness, functional embeddedness, and operational-mode embeddedness, were selected to design the Elderly Care Service Supply Satisfaction Scale. A simple random sampling method was used to survey elderly residents in 25 community-embedded elderly care institutions in Nanchang city, and the binary logistic regression model and structural equation model were used for analysis. ResultsFirst, resource embeddedness and operational-mode embeddedness had a significantly positive impact on supply satisfaction. Second, the weight of resource embeddedness, functional embeddedness, and operational-mode embeddedness on supply satisfaction decreased gradually. Third, the “number of service personnel” and “adequacy of service types” in the resource embeddedness dimension, the “entertainment services” and “medical services” in the functional embeddedness dimension, and the “charging standard” and “service response degree” in the operational-mode embeddedness dimension had a greater impact on supply satisfaction in their corresponding dimensions. ConclusionTo develop community-embedded elderly care, it is essential to strengthen the construction of the team of elderly care professionals, enrich the supply content of elderly care services, and increase the development support with the help of resource embeddedness; establish a unified charging system for elderly care, improve the responsiveness of elderly care services, and enhance the development internal driving force based on operational-mode embeddedness; improve the level of integrated medical and elderly care, emphasize the return of the family as the main body, and perfect the development system by using functional embeddedness as a link.
3.Advances in the role of STAT3 in tumor immunology

Signal transducer and activator of transcription 3(STAT3)are nuclear transcription factors that regulates expression of genes that are involved in cell cycle,cell survival,and cancer-related immune response.Activated STAT3 may participate in oncogenesis by stimulating cell proliferation,resisting apoptosis and promoting tumor angiogenesis and metastasis.Furthermore,abnormal activation of STAT3 is associated with poor tumor prognosis.Recent advances have revealed a crucial role of persistent activation of STAT3 in cancer progression and immune escape.STAT3 is also an important driver of mediated immune escape in tumor immunity,so it has become an effective target in immunotherapy.Therefore,STAT3 became frequently studied targets for more specific cancer immunotherapies.This article elaborated the multiple functions of STAT3 in innate immunity,adaptive immunity and tumor immunity.Further,it put forward to the prospects and issues that urgently need to be addressed in the future.
4.Correlation between lead exposure and clinical heterogeneity in patients with amyotrophic lateral sclerosis

Objective To investigate the relationship between lead exposure and the clinical heterogeneity of amyotrophic lateral sclerosis(ALS).Methods Baseline data were collected from 49 ALS patients hospitalized at the PLA General Hospital.Blood metal levels(copper,zinc,calcium,magnesium,iron,lead)were measured by graphite furnace atomic absorption spectrometry.The Revised ALS Functional Rating Scale(ALSFRS-R)was used to assess the severity of motor function impairment.Patients were grouped by site of onset and ALSFRS-R score,then blood metal levels were compared between two groups.General linear regression,ROC curves and Logistic regression models were applied to analyze the correlation between blood lead levels and disease severity to identify risk factors affecting motor function in ALS patients,and analyze its predictive value.Results The differences of blood metal levels between bulbar onset patients and non-bulbar onset patients were no statistical significance(all P>0.05).Blood lead levels were significantly lower in the high ALSFRS-R score group than in the low ALSFRS-R score group,and the difference was significant(P=0.02).Blood lead levels were negatively correlated with ALSFRS-R scores in ALS patients(Y=-0.1267X+45.86,R2=0.08937,P=0.04).The area under the ROC curve was 0.70,indicating that blood lead had predictive value for ALS severity.The Logistic regression model demonstrated that elevated blood lead level was an independent risk factor for the severity of ALS disease(OR=1.107,95%CI:1.006-1.219,P=0.04).Conclusions Blood lead levels in ALS patients correlated with disease severity and exhibited predictive utility.Elevated blood lead was an independent risk factor for the severity of motor function impairment in ALS.
5.Relationship between social media use and internalized homophobia in homosexual and bisexual persons

Objective:To explore the relationship between social media use and internalized homophobia in homosexual and bisexual persons,and the role of self-esteem and perceived social support.Methods:A total of 626 homosexual and bisexual persons were selected through online social media platforms,and the Social Media Use In-tensity Scale(SNSIS),Self-Esteem Scale(SES),Perceived Social Support Scale(PSSS)and Internalized Homo-phobia Scale(IHS)were used for measurement.Use the SPSS macro program PROCESS to test moderated media-tion effects.Results:The average SNSIS score was(3.0±1.0).The SES score played a partial mediating role in the relationship between SNSIS scores and IHS scores,and the value of mediating effect was 21.43%.The PSSS scores moderated the relationship between the SNSIS scores and the SES scores(β=-0.10,P<0.01),and the re-lationship between the SES scores and the IHS scores(β=-0.11,P<0.01).Conclusion:Social media use by ho-mosexual and bisexual persons is associated with internalized homophobia,self-esteem mediated the relationships,and perceived social support plays a moderating role.

9.Protective Effect and Mechanism of Salvia Polyphenolic Acid for Injection on Cerebral Ischemia-reperfusion Injury in Rats

Objective To investigate the neuroprotective effect and mechanism of salvia polyphenolic acid for injection (SAFI) on cerebral ischemia-reperfusion injury in rats. Methods A total of 100 SD rats were randomly divided into sham surgery group,model group,low-,medium-and high-dose (5,10,20 mg·kg-1) salvia polyphenolic acid groups,with 20 rats in each group. After being continuously administrated by intraperitoneal injection of SAFI once daily for three days,the rat model of middle cerebral artery occlusion/reperfusion (MCAO/R) was established using the thread embolization method at 1 hour after the last administration. The neurological deficit of rats was evaluated by Zea Longa score. The cerebral infarction volume was detected by 2,3,5-triphenyltetrazolium chloride(TTC) staining. The levels of serum NADPH oxidase(NOX),4-hydroxynonanal(4-HNE),8-hydroxydeoxyguanosine (8-OHdG),monocyte chemoattractant protein-1 (MCP-1),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-18(IL-18),interleukin-6(IL-6),and intercellular adhesion molecule-1 (ICAM-1) were detected by ELISA kits. Hematoxylin-eosin (HE) staining and Nissl staining were used to observe the pathological changes of brain tissue and the morphology of neurons. The apoptosis of neuronal cells in brain tissue was detected by TUNEL. Immunofluorescence staining was used to detect the expression level of glial fibrillary acidic protein (GFAP) in brain tissue. Western Blot was used to detect the protein expression of NLRP3 and Caspase1 in brain tissue. Results Compared with the sham surgery group,neurological deficit scores in model group increased remarkably (P<0.01). The cerebral infarction volume increased significantly (P<0.01). Serious pathological damage of brain was observed,and neuronal density decreased significantly(P<0.01). The apoptosis rate of cortical cells increased obviously (P<0.01). The levels of serum NOX,4-HNE,8-OHdG,MCP-1,TNF-α,IL-1β,IL-18,IL-6 and ICAM-1 increased significantly (P<0.05,P<0.01). The protein expression of GFAP,NLRP3 and Caspase1 in brain significantly upregulated (P<0.01). Compared with the model group,neurological deficit scores in medium-and high-dose SAFI groups decreased remarkably (P<0.01). The cerebral infarction volume decreased significantly (P<0.01). Neuronal damage was ameliorated to varying degrees,and neuronal density increased significantly(P<0.05,P<0.01). The apoptosis rate of cortical cells decreased obviously (P<0.01). The levels of serum NOX,4-HNE,8-OHdG,MCP-1,TNF-α,IL-1β,IL-18,IL-6 and ICAM-1 decreased significantly (P<0.05,P<0.01). The protein expression of GFAP,NLRP3 and Caspase1 in brain significantly downregulated(P<0.01). Conclusion SAFI has a protective effect on MCAO/R rats,which can significantly reduce oxidative stress and inflammatory responses,thereby reducing pathological damage and apoptosis of brain tissue. Its mechanism may be related to the inhibition of NLRP3/Caspase-1 signaling pathway and astrocyte activation.
10.Establishment of primary breast cancer cell line as new model for drug screening and basic research

Background and purpose:In 2016 the National Cancer Institute(NCI)decided stopping to use NCI-60 cell lines for drug screening,suggesting that tumor cell lines were losing their value as a tool for drug discovery and basic research.The reason for NCI-60 cells'retirement'was that the preclinical studies based on traditional cellular and animal models did not obtain the corresponding expected efficacy in clinical trials.Since the major cancer behaviors,such as proliferation and metastasis,are fundamentally altered with long-term culture,the tumor cell lines are not representative of the characteristics of cancer in patients.Currently,scientists hope to create a new cancer model that are derived from fresh patient samples and tagged with details about their clinical past.Our purpose was to create patient-derived breast cancer primary cell lines as new cancer model for drug screening and basic research.Methods:Breast cancer tissues were collected in the Department of Breast Surgery,Affiliated Hospital of Guizhou Medical University.The collection of tumor tissue samples was approved by the Ethics Committee of the Affiliated Hospital of Guizhou Medical University(approval number:2022 ethics No.313),and the collection and use of tumor tissues complied with the Declaration of Helsinki.The primary breast cancer cell lines were isolated from the patient's breast cancer tissues and cultured in BCMI medium.After the cells proliferated,the media were replaced with DEME medium.Cell line STR genotyping was done to determine cell-specific genetic markers and identification.Clone formation assay and transplantation assay were done to analyze the ability of breast cancer primary cell lines to form tumors.Results:We created 6 primary breast cancer cell lines.The 6 primary breast cancer cell lines from the patients were tagged with the definitively clinicopathological features,clinical diagnosis,therapeutic regimens,clinical effectiveness and prognostic outcomes.The STR genotyping assays identified the genetic markers and determined the identities of the 6 primary breast cancer cell lines.Clone formation assays and transplantation assay showed that the proliferative capacities of the patient-derived primary breast cancer cell lines were significantly greater compared with the conventional breast cancer cell lines.Conclusion:We created a panel of 6 patient-derived primary breast cancer cell lines as new cancer model for drug screening and basic research in breast cancer.
