1.Iodine nutritional status of children aged 8-10 in Wuhan from 2019 to 2023

Objective:
To explore the iodine nutrition status of children in Wuhan from 2019 to 2023, and to evaluate the effect of iodine deficiency disorders control in focus groups in Wuhan, so as to provide a basis for consolidating elimination of iodine deficiency disorders.
Methods:
A total of 13 000 non boarding primary school students aged 8-10 were selected from 13 districts of Wuhan by stratified random sampling method.Household salt samples were collected to measure salt iodine content, random urine samples were analyzed for urinary iodine concentration. And B ultrasound was used to measure thyroid volume in students. The median of salt iodine, coverage rate of iodized salt, consumption rate of qualified iodized salt, median of urinary iodine, and the goiter rate were calculated. And Mann-Whitney U- test, Kruskal-Wallis H- test and Chi-square test were applied to compare between groups. Chi-square trend test was used to analyze the changing trends of coverage rate of iodized salt, consumption rate of qualified iodized salt and goiter rate among children in Wuhan.
Results:
The median of iodine content of children s household salt was 23.8 (21.7, 26.1) mg/kg, and the coverage rate of iodized salt was 98.7%, and the consumption rate of qualified iodized salt was 94.5 %. The consumption rates of qualified iodized salt showed an overall upward trend from 2019 to 2023 ( χ 2 trend =5.57, P <0.05). The median of urinary iodine of children was 220.1 (136.7, 326.0) μg/L,and boys had higher median of urinary iodine than girls [228.3(143.2, 336.0),210.2(129.1, 315.7) μg/L] ( Z =6.60, P <0.01). The median of urinary iodine of children in suburbs was higher than those in urban areas [236.3 (150.7, 342.2) , 207.1 (124.5, 309.8) μg/L]( Z =11.00, P <0.01). A total of 4 600 children were examined for thyroid volume, and the range of goiter rates were 1.1% to 3.4%, with an average goiter rate of 2.5%, which showed an overall downward trend from 2019 to 2023 ( χ 2 trend =5.11, P <0.05).
Conclusions
The iodine nutrition is sufficient and iodine nutrition status is good among children in Wuhan. It should continue to carry out monitoring and evaluation of children s iodine nutrition, guide the public to supplement iodine scientifically,so as to maintain the appropriate level of iodine for children.
2.Compound Danshen Tablets ameliorate myocardial ischemia/reperfusion injury-induced ventricular remodeling by regulating autophagy via AMPK/mTOR signaling pathway.

OBJECTIVE:
Left ventricular remodeling induced by myocardial ischemia/reperfusion injury (MI/RI) is a common cardiac dysfunction. Accumulating evidence has demonstrated that autophagy plays a vital role in protecting against ventricular remodeling. This study aims to investigate the performance of Compound Danshen Tablets (CDT) in rescuing ventricular remodeling and whether autophagy as the potential mechanism.
METHODS:
The left anterior descending arteries of rats were temporarily ligated for 30 min to construct the MI/RI model. Ventricular remodeling was induced by reperfusion for 28 d, during which the MI/RI rats were administered CDT (300 mg/kg and 600 mg/kg), atorvastatin (2 mg/kg), and diltiazem (16 mg/kg). Cardiac function and structure were examined by echocardiography. Immunohistochemistry, Masson's trichrome staining, and hematoxylin-eosin (HE) staining were utilized to assess the fibrosis and histological alterations in the heart tissue. The expression of autophagy-related proteins was detected using Western blotting.
RESULTS:
CDT attenuated the cardiac dysfunction, structural changes, histopathological changes and fibrosis induced by MI/RI. CDT significantly enhanced the level of Beclin1 and microtubule-associated protein 1 light chain 3 beta (LC3β), and reduced p62 levels in MI/RI rats. Moreover, CDT significantly increased the phosphorylation of adenosine monophosphate-activated protein kinase (AMPK) and inhibited mammalian target of rapamycin (mTOR) phosphorylation.
CONCLUSION
CDT ameliorated MI/RI-induced ventricular remodeling by activating autophagy and improving autophagic flux via the AMPK/mTOR signaling pathway.
3.Establishment and preliminary study of four patient-derived primary breast cancer cell lines

Objective To establish primary breast cancer cell lines from patient tissues and offer a new cancer cell model for basic research.Methods Breast cancer biopsy tissues were digested with typeⅡcollagenase and cultured in BCMI medium.When the cells proliferated rapidly,the medium was switched to DMEM.STR genotyping was per-formed to identify specific genetic markers of the four primary breast cancer cell lines.Colony expansion assays and sphere formation assays were conducted to analyze its tumorigenicity.Real-time PCR and Western blot experiments were used to analyze the expression of the epithelial-mesenchymal transition(EMT)molecule markers.Migration and invasion assays were performed to assess the metastatic potential of the primary breast cancer cells.Results We es?tablished four primary breast cancer cell lines:BC25#,BC51#,BC56#,and BC57#.These cell lines were cultured in DMEM medium,passaged multiple times and tagged with details about their clinical past.STR genotyping identified specific genetic markers for each of the four primary breast cancer cell lines.Clonogenic and sphere formation assays revealed that the four lines have a stronger tumor?forming capability compared to the classic breast cancer cell line T?47D.Real?time PCR and Western blot experiments showed that,compared to T?47D,the four primary breast cancer cell lines have decreased E?cadherin expression and increased vimentin expression.Migration and invasion assays indicated that BC25#had a higher metastatic potential than the traditional breast cancer cell line T?47D.Conclusions Four primary breast cancer cell lines,BC25#,BC51#,BC56#and BC57#are successfully estab?lished,which may act as new cancer cell model for laboratory research of breast cancer.
4.Effects and mechanism of astragalus mongholicus extract on lung injury in rats with rheumatoid arthritis induced interstitial lung disease

Objective :
To investigate the effects of astragalus mongholicus extract ( AME) on lung injury and the myeloid differentiation factor 88 ( MyD88 ) / Toll-like receptor 4 ( TLR4 ) / nuclear factor kappa B ( NF-κB) p65 pathway in rheumatoid arthritis induced interstitial lung disease (RA-ILD) rats.
Methods :
SD rats were randomly divided into a control group,RA-ILD group,low-dose AME group (5 g / L) ,high-dose AME group ( 10 g / L) ,and high-dose AME + lipopolysaccharide (LPS) group ( 10 g / L AME + 1 mg / L TLR4 activator LPS) .Except for the control group,rats in all other groups were injected with bovine type Ⅱ collagen,Freund ’s complete adjuvant, and bleomycin to establish the RA-ILD model.The arthritis index and lung tissue wet-dry weight ratio of rats were tested.ELISA was applied to detect the levels of inflammatory factors interleukin (IL) -1 β , IL-6 and tumor necrosis factor-α ( TNF-α) in bronchoalveolar lavage fluid. Hematoxylin eosin staining was used to observe pathological changes of rat knee joint tissue and lung tissue.Western blot was applied to detect the expression of autophagy fac- tors Beclin 1,microtubule-associated protein 1A /1B-light chain 3 (LC3) Ⅱ / Ⅰ , and MyD88 /TLR4 /NF-κB p65 pathway related proteins in lung tissue.
Results :
Compared with control group,knee joint tissue and lung tissue of rats in RA-ILD group were damaged,the arthritis index,lung tissue wet-dry weight ratio,levels of IL-1 β , IL-6, and TNF-α , the expression levels of MyD88 and TLR4 proteins ,and p-NF-κB p65 /NF-κB p65 ratio increased (P<0. 01) ,the expression of Beclin 1 and LC3 Ⅱ / Ⅰ proteins decreased (P<0. 01) .Compared with RA-ILD group,the low-dose and high-dose AME groups showed reduced tissue damage in rats,the arthritis index,lung tis- sue wet-dry weight ratio,levels of IL-1 β , IL-6,and TNF-α , the expression levels of MyD88 and TLR4 proteins, and p-NF-κB p65 /NF-κB p65 ratio showed a dose-dependent decrease (P<0. 05 or P<0. 01) ,the expression of Beclin 1 and LC3 Ⅱ / Ⅰ proteins showed a dose-dependent increase (P<0. 05 or P<0. 01) .Compared with high- dose AME group,the tissue damage of rats in the high-dose AME + LPS group was worsened,the arthritis index, lung tissue wet-dry weight ratio,levels of IL-1 β , IL-6,and TNF-α , the expression levels of MyD88 and TLR4 pro- teins,and p-NF-κB p65 /NF-κB p65 ratio were higher (P<0. 01) ,the expression of Beclin 1 and LC3 Ⅱ / Ⅰ pro- teins was lower (P <0. 01 ) .
Conclusion
AME inhibits the MyD88 /TLR4 /NF-κB p65 pathway and alleviates lung injury in RA-ILD rats.
5.The expression of miR-133a and miR-424 in serum and their relationship with T lymphocyte subsets in patients with connective tissue disease combined with interstitial lung disease

To investigate the expression of miR-133a and miR-424 in the serum of patients with connective tissue disease(CTD)and interstitial lung disease(ILD)and their relationship with T lymphocyte subpopulations,total of 96 CTD-ILD patients treated in our hospital from December 2019 to December 2022 were selected as CTD-ILD group,while 96 CTD patients without ILD were as the control group.The real-time fluorescence quantitative PCR(qRT-PCR)method was applied to detect serum levels of miR-133a and miR-424;flow cytometry was applied to detect the levels of T lymphocyte subpopulations.Pearson method was applied to analyze the relationship of miR-133a and miR-424 with T lymphocyte subpopulations.Compared with the control group,the level of serum miR-133a in the CTD-ILD group was obviously reduced,while the expression level of miR-424 was obviously increased(P<0.05).Under different degrees of pulmonary ventilation disorders,the expression level of miR-133a in the serum of mild,moderate,and severe patients decreased obviously,while the expression level of miR-424 increased obviously(P<0.05).Under different grades of pulmonary diffusion dysfunction,the expression level of miR-133a in the serum of mild,moderate,and severe patients reduced obviously,while the expression level of miR-424 increased obviously(P<0.05).Furthermore,the levels of CD4+and CD4+/CD8+in the CTD-ILD group were obviously increased,as compared to the control group,while the levels of CD8+and CD3+were obviously reduced(P<0.05).In addition,miR-133a was negatively correlated with CD4+,and positively correlated with CD8+and CD3+;miR-424 was positively correlated with CD4+,and negatively correlated with CD8+and CD3+(P<0.05).In conclusion,the expression level of miR-133a in serum of CTD-ILD patients is decreased,while the expression level of miR-424 is increased,and both of them are related to the T lymphocyte subpopulations.
6.A survey of adult thyroid volume and related influencing factors in Wuhan

Objective:To learn about the thyroid volume and goiter status of adults in Wuhan, analyze related influencing factors, and provide a basis for revising the diagnostic criteria for adult goiter.Methods:From December 2019 to November 2021, a multi-stage cluster random sampling method was used in 13 districts of Wuhan. Two communities or townships were selected from each district, 100 permanent residents over 16 years old were selected from each community or township according to the inclusion and exclusion criteria for general information collection, thyroid ultrasound examination, urinary iodine detection and physical examination were performed, and the influencing factors of thyroid volume were analyzed.Results:A total of 2 578 adults aged 16 and older in Wuhan were selected, including 1 168 males and 1 410 females, with an average age of (41.79 ± 13.01) years old. The median thyroid volume (interquartile range) in adults was 6.36 (4.88, 8.11) ml, the rate of goiter was 0.70% (18/2 578), and the detection rate of thyroid nodules was 35.49% (915/2 578). The median urinary iodine (interquartile range) was 185.32 (107.93, 277.08) μg/L. According to Spearman correlation analysis, the thyroid volume of adults in Wuhan was positively correlated with their height, weight, body mass index (BMI), and body surface area (BSA, r = 0.12, 0.22, 0.21, 0.20, P < 0.001). Through multiple linear regression analysis, the regression equation was obtained: thyroid volume = 7.72 + 1.19 × thyroid nodules - 1.04 × gender + 0.15 × age group. Patients with thyroid nodules had a thyroid volume greater than those without, males greater than females, and the elderly group greater than the younger group. Conclusion:The goiter rate in Wuhan is relatively low, and factors such as gender, age, presence of thyroid nodules, height, weight, BMI, BSA and other body measurement are the influencing factors of thyroid volume.
7.Establishment of primary breast cancer cell line as new model for drug screening and basic research

Background and purpose:In 2016 the National Cancer Institute(NCI)decided stopping to use NCI-60 cell lines for drug screening,suggesting that tumor cell lines were losing their value as a tool for drug discovery and basic research.The reason for NCI-60 cells'retirement'was that the preclinical studies based on traditional cellular and animal models did not obtain the corresponding expected efficacy in clinical trials.Since the major cancer behaviors,such as proliferation and metastasis,are fundamentally altered with long-term culture,the tumor cell lines are not representative of the characteristics of cancer in patients.Currently,scientists hope to create a new cancer model that are derived from fresh patient samples and tagged with details about their clinical past.Our purpose was to create patient-derived breast cancer primary cell lines as new cancer model for drug screening and basic research.Methods:Breast cancer tissues were collected in the Department of Breast Surgery,Affiliated Hospital of Guizhou Medical University.The collection of tumor tissue samples was approved by the Ethics Committee of the Affiliated Hospital of Guizhou Medical University(approval number:2022 ethics No.313),and the collection and use of tumor tissues complied with the Declaration of Helsinki.The primary breast cancer cell lines were isolated from the patient's breast cancer tissues and cultured in BCMI medium.After the cells proliferated,the media were replaced with DEME medium.Cell line STR genotyping was done to determine cell-specific genetic markers and identification.Clone formation assay and transplantation assay were done to analyze the ability of breast cancer primary cell lines to form tumors.Results:We created 6 primary breast cancer cell lines.The 6 primary breast cancer cell lines from the patients were tagged with the definitively clinicopathological features,clinical diagnosis,therapeutic regimens,clinical effectiveness and prognostic outcomes.The STR genotyping assays identified the genetic markers and determined the identities of the 6 primary breast cancer cell lines.Clone formation assays and transplantation assay showed that the proliferative capacities of the patient-derived primary breast cancer cell lines were significantly greater compared with the conventional breast cancer cell lines.Conclusion:We created a panel of 6 patient-derived primary breast cancer cell lines as new cancer model for drug screening and basic research in breast cancer.
8.Impact of stressors and stress responses on the daily life of college students with migraine

BackgroundStress is closely related to migraine attacks, however, previous studies on stressors, stress responses and their impact on the daily life of college students with migraine remain unclear. ObjectiveTo explore the impact of stressors and stress responses on the daily life of college students with migraine, in order to provide references for improving daily life of this population. MethodsFrom August 2018 to August 2019, 458 students from North Sichuan Medical College who met the International Classification of Headache Diseases third edition (ICHD-3), were selected using random sampling method. General data and headache characteristics were collected. The 6-item Headache Impact Test Questionnaire (HIT-6) was used to assess the impact of migraine on daily life. Student-Life Stress Inventory (SLSI) was used to assess the stressors and stress responses. Hamilton Anxiety Scale (HAMA) and Hamilton Depression Scale-24 item (HAMD-24) were used to assess anxiety and depressive symptoms. Pearson correlation analysis were used to examine the relationship between HIT-6 score and scores on various scales. Multiple linear regression was used to analyze the influencing factors on daily life in college students with migraine. ResultsIn the SLSI, stressors in the dimensions of frustration (r=0.138, P<0.01), conflict (r=0.168, P<0.01), pressure (r=0.157, P<0.01), change (r=0.148, P<0.01), self-imposed (r=0.158, P<0.01) , as well as physiological response (r=0.256, P<0.01), emotional response (r=0.241, P<0.01), behavioral response (r=0.164, P<0.01), HAMA total score (r=0.192, P<0.01), dHAMD-24 total score (r=0.250, P<0.01), and SLSI total score (r=0.250, P<0.01), were positively correlated with HIT-6 score. Cognitive response (r=-0.104, P<0.05) was negatively correlated with HIT-6 score. Stepwise multiple linear regression revealed that physiological response (β=0.140, P<0.05), anxiety (β=0.159, P<0.05), and cognitive response (β=-0.091, P<0.05) could predict the impact on daily life of college students with migraine. ConclusionPhysiological response, cognitive response and anxiety may be the independent influencing factors on the daily life of college students with migraine. Expanded services such as clinical comprehensive evaluation of anti-tumor drugs and management of off-label drug use are an important development trend of oncology pharmacy. The column named “Oncology Pharmacy Management and Services” is introduced, which discussed in depth from the two parts of “frontier progress of anti-tumor drugs” and “comprehensive clinical evaluation of anti-tumor drugs”, aiming to provide new ideas for the exploration of oncology pharmaceutical services, so as to further accelerate the transformation of oncology pharmaceutical care and promote the high-quality development of oncology pharmacy.
10.Research Progress of Pharmacokinetics Studies of Anti-tumor Drugs in Patients with Third Space Fluid

Third space fluid(TSF) is a common complication of advanced malignancies, including malignant pleural effusion, malignant ascites, intracranial effusion, and pelvic effusion, etc. The pharmacokinetics(PK) of anti-tumor drugs in vivo are influenced by various factors, and TSF is one of the potential factors that contributes to PK variations, which may consequently affect the efficacy and safety of anti-tumor drugs. This paper aimed to comprehensively investigate PK studies related to anti-tumor drugs in patients with malignant tumors accompanied by TSF. The paper summarized the PK characteristics of common cytotoxic drugs, small molecule targeted drugs, and monoclonal antibodies in both blood and TSF.
