1.Role of liver cancer stem cells in hepatocellular carcinoma and related strategies for targeted therapy

Hepatocellular carcinoma (HCC) is a malignant tumor with relatively high incidence and mortality rates worldwide, and its therapeutic resistance and recurrence mechanism are closely associated with liver cancer stem cells (LCSC). This article systematically introduces the biological characteristics of LCSC and their key role in the progression of HCC, reviews the functional characteristics of the specific surface markers (such as EpCAM and CD133) and related signaling pathways (such as Wnt/β-catenin, TGF-β, and STAT3), elaborates on the interaction between LCSC and tumor microenvironment, and summarizes the latest clinical treatment strategies targeting LCSC and the countermeasure for existing resistance mechanisms. The article points out that LCSC promote tumor development and progression through metabolic reprogramming and immune microenvironment remodeling, and it is proposed to establish a standardized detection system for LCSC specific markers and promote a triple synergistic therapeutic paradigm combining targeted therapy, immune regulation, and traditional chemotherapy, in order to provide new ideas for the clinical intervention of HCC.
2.Novel pathogenesis and intervention strategies for liver cirrhosis based on the gut microbiota-bile acid axis

Liver cirrhosis is the final stage of the progression of various chronic liver diseases, often accompanied by serious complications and high mortality rates. Recent studies have shown that the interaction between gut microbiota and bile acid metabolism (the gut microbiota-bile acid axis) is closely associated with liver cirrhosis. This article systematically reviews the mechanism of action of the gut microbiota-bile acid axis in the progression of liver cirrhosis, elaborates on the pathological features of liver cirrhosis and its harm to the body, and summarizes the association of the gut microbiota-bile acid axis with the development and progression of liver cirrhosis. It also analyzes the key regulatory role of this axis in the progression of liver cirrhosis and explores its potential application value as a therapeutic target for liver cirrhosis, in order to provide a theoretical basis for exploring more effective clinical intervention methods.
3.Analysis of the prevalence and clinical characteristics of pertussis in children with different cough durations

Objective:To analyze the prevalence and clinical characteristics of pertussis in children with different cough durations.Methods:From January 2021 to October 2022, information on children aged 0-18 years who visited eight hospitals in Shandong Province due to cough was enrolled. Pertussis serological antibody testing and/or nucleic acid testing were performed. The prevalence and clinical characteristics of pertussis were compared among the acute cough group, protracted cough group, and chronic cough group using the χ2 test or Fisher′s exact test. Results:A total of 1 565 children with cough were included in the study, of which 348 (22.24%) were laboratory-confirmed pertussis. There was a significant difference in the laboratory-confirmed rate of pertussis among different cough groups ( χ2=83.424, P<0.001). The confirmation rate of pertussis in the protracted cough group (42.21%) was significantly higher than that in the acute cough group (16.49%, P<0.05) and chronic cough group (19.50%, P<0.05). In each cough group, the age of children was significantly associated with the confirmed rate of pertussis, and the confirmed rate was relatively high in children aged 3 months to <2 years. Pertussis vaccination was significantly associated with the confirmed rate in all groups, and the confirmed rate was higher in unvaccinated children. Among laboratory-confirmed pertussis cases, the incidence of typical symptoms such as paroxysmal cough, whoop, and post-tussive emesis or sleep disturbance was significantly higher than that in the non-confirmed cases. In the protracted and chronic cough groups, the proportion of non-confirmed cases complicated with asthma/cough variant asthma (CVA) was significantly higher than that in pertussis-confirmed cases. Conclusion:There are differences in the confirmation rate of pertussis among children with different cough durations. The confirmation rate is significantly associated with age, vaccination status, and clinical symptoms. Enhancing clinical vigilance against pertussis, conducting early diagnosis, and getting timely and standardized vaccination are crucial for effectively controlling pertussis and preventing outbreaks.
4.Efficacy of nucleos(t)ide analogues combined with peginterferon α-2b on antiviral therapy in children with chronic hepatitis B

Objective:To analyze the efficacy of combination of peginterferon α-2b(Peg-IFN α-2b)with nucleos(t)ide analogues(NAs)on antiviral therapy in children with chronic hepatitis B(CHB)and to provide an optimized clinical treatment strategies for CHB children.Methods:A retrospective analysis was conducted on 30 CHB children treated in The First Affiliated Hospital of the Army Medical University(Southwest Hospital)from January 2022 to January 2025 with treatment duration at least 48 weeks. The enrolled children were aged between 2 and 17 years and divided into the NAs combined with Peg-IFN α-2b(NPI)group(n=13)and NAs group(n=17)by their therapy regimens. The characteristics of baseline,week 12,week 24,week 48 and week 96 were compared between groups,as well as the differences in response to biochemical,immune and viral indicators at each observation point. Logistic regression analysis and receiver operating characteristic(ROC)curve analysis were performed to identify factors influencing the HBsAg seroclearance. Results:At baseline of treatment,the proportion of HBeAg positivity in the NPI group and the NAs group was high(76.9% vs 86.6%, χ2=0.679, P=0.628),and the alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in the NPI group were significantly lower than those in the NAs group( P<0.001). At 24 weeks,the decrease in HBsAg in the NPI group was also significantly higher than that in the NAs group( Z=-3.161, P=0.002). Finally,the cumulative seroclearance rate of HBsAg at 96 weeks in the NPI group was significantly higher than that in the NAs group(46.15% vs 5.88%, χ2=0.679, P=0.025). Mulitivariate Logistic regression analysis showed that treatment regimen and gender were risk factors affecting the outcome of HBsAg( P<0.05). ROC curve analysis showed that the increase in ALT at 12 weeks compared with baseline(AUC=0.857,Cutoff value=3.615 IU/L),the decrease in ALT at 24 weeks(AUC=0.870,Cutoff value=47.85 IU/L),and the decrease in HBsAg at 12 weeks and especially at 24 weeks(AUC=0.885,Cutoff value=0.97log IU/ml)were effective predictors of HBsAg prognosis at 96 weeks. Conclusion:In CHB children,antiviral regimen Peg-IFN α-2b combined with NAs was more effective than NAs alone in improving the HBsAg seroclearance rate of CHB,and the effects in female were better than in male. The decline of HBsAg and the fluctuation of ALT in the early treatment period are valid predictors of HBsAg clearance.
5.Rapid enrichment and SERS differentiation of various bacteria in skin interstitial fluid by 4-MPBA-AuNPs-functionalized hydrogel microneedles

Bacterial infection is a major threat to global public health,and can cause serious diseases such as bacterial skin infection and foodborne diseases.It is essential to develop a new method to rapidly di-agnose clinical multiple bacterial infections and monitor food microbial contamination in production sites in real-time.In this work,we developed a 4-mercaptophenylboronic acid gold nanoparticles(4-MPBA-AuNPs)-functionalized hydrogel microneedle(MPBA-H-MN)for bacteria detection in skin inter-stitial fluid.MPBA-H-MN could conveniently capture and enrich a variety of bacteria within 5 min.Surface enhanced Raman spectroscopy(SERS)detection was then performed and combined with ma-chine learning technology to distinguish and identify a variety of bacteria.Overall,the capture efficiency of this method exceeded 50%.In the concentration range of 1 × 10 7 to 1 × 10 10 colony-forming units/mL(CFU/mL),the corresponding SERS intensity showed a certain linear relationship with the bacterial concentration.Using random forest(RF)-based machine learning,bacteria were effectively distinguished with an accuracy of 97.87%.In addition,the harmless disposal of used MNs by photothermal ablation was convenient,environmentally friendly,and inexpensive.This technique provided a potential method for rapid and real-time diagnosis of multiple clinical bacterial infections and for monitoring microbial contamination of food in production sites.
6.Deciphering the Role of Shank3 in Dendritic Morphology and Synaptic Function Across Postnatal Developmental Stages in the Shank3B KO Mouse.

Autism Spectrum Disorder (ASD) is marked by early-onset neurodevelopmental anomalies, yet the temporal dynamics of genetic contributions to these processes remain insufficiently understood. This study aimed to elucidate the role of the Shank3 gene, known to be associated with monogenic causes of autism, in early developmental processes to inform the timing and mechanisms for potential interventions for ASD. Utilizing the Shank3B knockout (KO) mouse model, we examined Shank3 expression and its impact on neuronal maturation through Golgi staining for dendritic morphology and electrophysiological recordings to measure synaptic function in the anterior cingulate cortex (ACC) across different postnatal stages. Our longitudinal analysis revealed that, while Shank3B KO mice displayed normal neuronal morphology at one week postnatal, significant impairments in dendritic growth and synaptic activity emerged by two to three weeks. These findings highlight the critical developmental window during which Shank3 is essential for neuronal and synaptic maturation in the ACC.

9.Current status of construction and monitoring of healthcare-associated in-fection system in Guangxi area

Objective To investigate the current status of construction and monitoring of healthcare-associated in-fection(HAI)system in maternal and child health care institutions in Guangxi Zhuang Autonomous Region,and provide reference for improving the quality of HAI management.Methods After discussed by members of expert group of HAI Prevention and Control Professional Committee of Guangxi Maternal and Child Health Care Associa-tion,the"Questionnaire on the construction and monitoring of HAI system in maternal and child health care institu-tions in Guangxi Zhuang Autonomous Region"was designed.The questionnaires were distributed to all maternal and child health care institutions in the region via"Questionnaire Star",and a WeChat group was established to ad-dress any questions in questionnaires.Results A total of 104 maternal and child health care institutions were in-cluded in the study,including 6 primary,74 secondary,and 24 tertiary level institutions.All maternal and child health care institutions have established HAI management departments,95.19%(n=99)of institutions held HAI management committee meeting at least once every six months.Among 201 HAI management professionals,most were female(90.55%),with nursing specialty(72.64%),with a bachelor's degree(66.17%),and aged 31-50 years old(73.13%).Only 30.77%of the maternal and child health care institutions had HAI monitoring system.Seventy institutions(67.30%)have carried out the special action of improving the pathogen detection rate before antimicrobial therapy for hospitalized patients,out of which 55(78.57%)were led by the HAI management depart-ment.Ninety-seven(92.27%)maternal and child health care institutions conducted surgery-related monitoring,with 50.96%of institutions relying on manual monitoring;only 30.77%of institutions have carried out the special action of"Strengthening perioperative infection prevention and control to ensure surgical quality and safety".Conclusion HAI management system in maternal and child health care institutions in Guangxi has been basically es-tablished and can carry out monitoring work,but personnel compostion is limited and talent reserve is insufficient.The information construction of HAI monitoring is underdeveloped,with a lagged implement of special action.Ad-ministrative support is needed to enhance infection prevention and control at the grassroots level.All levels of ma-ternal and child health care institutions should communicate and exchange work experience.Hospitals should in-crease their support for the construction of HAI information technology,improve level of HAI monitoring as well as prevention and control capabilities.
10.Preliminary evaluation of the application of digital smile design in clinical internships for undergraduate students

Objective To investigate the application value of digital smile design in clinical internships for undergraduate students in oral restoration,and to provide a method for clinical teaching of aesthetic restoration of anterior teeth.Methods A total of 18 under-graduate students who participated in clinical internships in the prosthotontics department at the Stomatological Hospital of Sun Yat-sen University from 2021 to 2023 were selected as the research subjects.Traditional oral photography and analysis of the red and white aes-thetics of anterior teeth were taught,and digital smile design training was conducted using the anterior tooth aesthetic design module of 3-Shape oral scanning software.Digital smile design would be applied to patients with anterior tooth aesthetic problems.The students wererequired to fill out the evaluation questionnaire.Results The survey evaluation results indicated that digital smile design training could enhance students'understanding and recognition of the aesthetics of anterior teeth(100%),improve their confidence in anterior teeth cases(88.89%),and also help cultivate students'systematic thinking in diagnosis and treatment(88.89%),improve clinical photography skills(100%),and enhance their ability to communicate with patients(83.33%)and produce case reports(88.89%).All 18 students completed at least 1 case of anterior teeth aesthetic restoration guided by DSD during clinical practice.Conclusion Digital smile design can improve students'thinking and abilities of digital system,improve their awareness and understanding of aes-thetic restoration of anterior teeth,and has application prospects in clinical practice training of oral restoration.
