Objective To explore the effects of prohibitin 2(PHB2)on sensitivity of non-small cell lung cancer cell line A549 to erlotinib(Erl)and its potential mechanisms.Methods RACK1-specific small interfering RNA was transfected in A549 cells for knocking-down of PHB2.The effects of PHB2 inhibition on cell proliferation and apop-tosis induced by Erl were observed.The colocalization of microtuble-associated protein light chain 3 alpha(LC3)and mitochondria was visualized by MitoTracker staining and green fluorescent protein-microtuble-associated protein light chain 3 alpha(GFP-LC3)transfection.Cell proliferation was detected by 5-ethynyl-2′-deoxyuridine(EdU)staining.Cell colony formation was evaluated by colony forming assay.Apoptotic index of A549 cells was evaluated by TUNEL.Western blot was used to measure the expressions of PHB2 and LC3Ⅱ.Mitochondrial transmembrane potential,cytochrome c and respiratory chain complexⅠ/Ⅱ/Ⅴactivity were analyzed by the commercially availa-ble kits.Results Compared with the siPHB2 and siCtrl+Erl group,the EdU-positive A549 cells and the number of cell colonies decreased significantly(P＜0.05),while the TUNEL-positive A549 cells increased significantly(P＜0.05)in the siPHB2+Erl group.In addition,compared with the siPHB2 and siCtrl+Erl group,mitochondrial transmembrane potential and respiratory chain complexⅠ/Ⅱ/Ⅴactivity decreased significantly(all P＜0.05)and the levels of cytochrome c increased in mitochondrial fractions(P＜0.05)and decreased in cytosolic fractions(P＜0.05)in the siPHB2+Erl group.Conclusions PHB2 inhibition significantly improves sensitivity of A549 cells to Erl,which may be explained by inhibition of PHB2-mediated mitochondrial autophagy.

Objective:To study the clinical efficacy of nanocrystalline needles combined with 5% minoxidil tincture in the treatment of androgenetic alopecia of male.Methods:A total of 60 patients with androgenetic alopecia were randomly divided into treatment group and control group, with 30 patients in each group. The control group was treated with 5% minoxidil tincture 2 times a day. On the basis of the control group, the treatment group was treated with nanocrystalline needles combined with 5% minoxidil tincture for 10 minutes 2 times a week. The clinical effects of different treatments were compared after six months.Results:There was a significant difference in overall response rate between the treatment group and the control group (80.00% vs. 53.30%, P<0.05). The improvement of the each clinical features of image by dermoscopy in patients with hair loss disease included more than 20% of hair diameter diminish, vellus hair increased, partly acomia and yellow dot sign had a significant difference between the treatment group and the control group ( P<0.05). Conclusion:Nanocrystalline needles combined with 5% minoxidil tincture has a good clinical effect in the treatment of androgenetic alopecia.

Objective:To investigate the risk factors on androgenetic alopecia (AGA) of primary healthcare staffs.Methods:Primary healthcare staffs were investigated by a questionnaire.Results:A total of 934 healthcare staffs (550 males and 384 females) were surveyed, of which 238 cases were diagnosed as AGA. 68.8% of AGA patients had poor mental health self-assessment; 52.94% of AGA patients had a family history; the prevalence in the people who usually had the night shift was 26.03%, which was higher than that in the people who had no night shift.Conclusions:The prevalence of AGA in healthcare staffs is higher than the national average, which might be related to working pressure, mental status, night shifts, genetic and alcohol drinking.

Objective To evaluate the relationship of serum thrombospondin?1(TSP?1)with the micro?inflammation in maintenance hemodialysis(MHD),and to explore its clinical prognosis value in the MHD patients. Methods A total of 84 MHD patients in our hospital were enrolled and prospectively followed for 2 years. The serum levels of TSP?1 and clinical inflammatory markers were detected. Patients were divided into groups according to different serum TSP?1 levels. The clinical inflammatory markers were detected by using ELISA analysis. Pearson simple correlation analysis method was applied to analyze the correlation between TSP?1 levels and inflammation related indicators. At the same time the prognosis and turnover of MHD patients was analyzed by using Kaplan Meier survival curve and survival rate was compared by Deleted:compared log?rank test. Cox regression analysis was used to calculate the hazard ratio (HR) and Deleted:using 95% confidence interval (CI). Results The indexes of blood lipid and inflammatory factors in the TSP?1 high?level groups were higher than that in TSP?1 low?level groups (P<0.05). Correlation analysis showed that the serum TSP?1 level was positively correlated with the serum lipid and inflammatory factors. Survival curve analysis showed that the mortality rate of TSP?1 high?level group was higher than that of TSP?1 low?level groups. Cox hazards analysis revealed that the patients with high?level TSP?1 had a higher risk for mortality than these TSP?1 low?level patients. This predictive value still existed after multivariate adjustment for age,blood lipid,serum albumin and other factors (P < 0.001). Conclusion The serum TSP?1 levels were associated with micro?inflammation and had a significant value in predicating the prognosis of MHD patients.

Objective To analyze the effect of Ziniliangxue compound in treating psoriasis vulgaris patients with clinical efficacy and serum IL-8, IL-17, IL-23.MethodsIn the control group were treated with routine treatment, in the experimental group were treated with Ziniliangxue compound treatment, effect of serum IL-8, IL-17, IL-23 and in the clinical effect of the two groups were compared.ResultsIn the observation group of patients with clinical curative effect total have efficiency (88.57%) was significantly higher than that of control group in patients with clinical efficacy in patients with total efficiency (65.71%), the difference has statistical significance (P<0.05);after treatment, the observe of serum IL-8(14.211±2.314), IL-17(12.643±1.232) and IL-23(12.856±1.321)were significantly better than the control of serum IL-8(17.653±2.512), IL-17(19.432±1.685) and IL-23(19.786±1.754), the difference has statistical significance (P< 0.05).ConclusionZiniliangxue compound on psoriasis vulgaris clinical treatment effect and can effectively reduce the patients serum IL-8, IL-17, IL-23 levels.It is worthy of clinical application.

OBJECTIVETo evaluate the published data on association between present/null polymorphism of glutathione S-transferase M1 (GSTM1) and breast cancer risk in Chinese population in order to abttain a more precise and comprehensive estimation of the relationship.

METHODSA meta-analysis was performed to investigate the association between GSTM1 polymorphism and susceptibility to breast cancer in Chinese population by searching Pubmed, Embase, Cochrane library, CNKI, VIP, Wanfang and CBD database. The data were screened according to the inclusion and exclusion criteria, and extracted, and the quality of included studies was evaluated. The pooled odds ratios (OR) with 95% confidence intervals (95%CI) were calculated using RevMan 5.2 and Stata 12.0 software. Publication bias and sensitivity analysis were also assessed.

RESULTSA total of 15 case-control studies involving 5,176 cases and 5 890 controls were included in the meta-analysis. The results showed that individuals with GSTM1 null genotype harbored a significantly increased risk of breast cancer compared to that with GSTM1 non-null genotype in Chinese population (OR=1.34, 95%CI=1.12-1.60, P=0.002). The subgroup analysis by region revealed that the individuals with GSTM1 null genotype were significantly associated with an increased risk of breast cancer in southern and northern China populations (southern: OR=1.14, 95%CI=1.01-1.28, P=0.03; northern: OR=2.65, 95%CI=2.04-3.34, P<0.01).

CONCLUSIONThe current meta-analysis demonstrates that the GSTM1 polymorphism is significantly associated with susceptibility to breast cancer in Chinese population, and the GSTM1-deficit may increase the risk of breast cancer.

Objective To establish the capability level classification system and capability level staffing standard of ICUs in tertiary hospitals, in an effort to raise the efficiency of nursing resources while ensuring quality of care of patients. Methods Such methods as experts meetings and Delphi Method were called into play to establish the nurses capability level system in ICUs with the convenient sampling method, we made clinical observation and research in 5 ICUs of 3 tertiary hospitals in Shanghai. Results Nurses in ICUs were classified into four levels, so were their responsibilities and job description. This way the system of nursing staff capability levels takes shape, and the staffing standards for nurses in ICUs of tertiary hospitals come into being. Conclusion Such tiered staffing standards in tertiary hospitals enable clinical nursing managers to deploy and efficiently use nursing staff of all levels.

Objective To calculate and analyze the grading nursing costs in a grade Ⅲ-A general hospital, and give suggestions to the government for rational pricing. Methods 6 wards were selected as the subjects, we calculated the total expenses by the ratio of income method, meanwhile, by sharing the cost we calculated the actual costs of grading nursing and let them underwent variance analysis. Results The actual cost of grade 1 nursing was (64.06±14.58) yuan, the actual cost of grade 2 nursing was (40.06±13.49) yuan, and the actual coat of grade 3 nursing was (31.11±5.81) yuan, by statistic analysis, the actual grad-ing nursing costs were significantly different from the current pricing system. Conclusions The grouping of nursing expense ia a difficult point in nursing cost study, and by calculating the actual grading nursing cost, we could develop measures to control the cost effectively. And the grading nursing total coat calculat-ing is the foundation of rational prices setting of grading nursing.

Objective To classify the nursing faculty in a certain third-class and first-grade hospi-tal.Methods We carried out 2 times of expert consultations about the classification of 14 nursing man-agement faculty in a certain third-class and first-grade hospital by Delphi method.Besides we had further interviews with 5 nursing experts and 12 nurses by means of qualitative interview.Results We got weight of rationality and harmony degree of expert attitude with a authoritative coefficient of 0.7867.The results of qualitative interview included 4 thematic concep4 understanding degree,meaning,basis of classification system.Conclusion Scientific and operational classification system for clinical nurses could set basis for reasonable settlement of nurses resource and design of nurses'career.

BACKGROUNDTo evaluate the relationship between estrogen receptor (ER) expression and level of serum sexual hormones in male patients with lung cancer.

METHODSThe levels of serum estradiol (E 2), testosterone (T), follicle stimulating hormone (FSH) and lutenising hormone (LH) were measured in 25 patients with lung cancer and 30 healthy men by enzyme immunoassay magnetic solid phase (IEMA), the ER expression was detected in 25 cancer tissues, 25 paracancerous tissues, and 11 benign pulmonary tissues by immunocytochemistry (ICC).

RESULTSThe level of plasma E₂ in male patients with lung cancer was significantly higher than that in normal controls [(22.4±15.7) ng/L [WTBX]vs ( 12.6±4.8) ng/L, P=0.001 9] while the level of T was significantly lower while the level of T was significantly lower [(2.9±1.3) μg/L [WTBX]vs (4.1±1.5) μg/L, P= 0.003 0]. The ratio of E₂/T of male patients with lung cancer was also remarkably higher than that of control . The ratio of E₂/T of male patients with lung cancer was also remarkably higher than that of control [(9.7±10.0) ×10⁻³[WTBX]vs ( 3.4±1.6)×10⁻³, P=0.004 6]. The expression rate of ER in lung cancer tissue samples was 60.0% (15/25), but no ER expression was found in the paracancerous tissues and benign pulmonary tissues. The level of E₂ had positive correlation with the expression of ER in male patients with lung cancer (. CONCLUSIONSThere are disorders and imbalances of sexual hormone metabolism in male patients with lung cancer, and these imbalances relate to the expression of ER. The elevation of E₂ level in peripheral blood might be related to the overexpression of ER.
