Journal of Southern Medical University 2023;43(4):577-584
OBJECTIVE:
To explore the mechanism underlying the inhibitory effect of quercetin against testicular oxidative damage induced by a mixture of 3 commonly used phthalates (MPEs) in rats.
METHODS:
Forty male Sprague-Dawley rats were randomly divided into control group, MPEs exposure group, and MPEs with low-, median- and high-dose quercetin treatment groups. For MPEs exposure, the rats were subjected to intragastric administration of MPEs at the daily dose of 900 mg/kg for 30 consecutive days; Quercetin treatments were administered in the same manner at the daily dose of 10, 30, and 90 mg/kg. After the treatments, serum levels of testosterone, luteinizing hormone (LH), follicle stimulating hormone (FSH), and testicular malondialdeyhde (MDA), catalase (CAT) and superoxide dismutase (SOD) were detected, and testicular pathologies of the rats were observed with HE staining. The expressions of nuclear factor-E2-related factor 2 (Nrf2), Kelch-like ECH2 associated protein 1 (Keap1) and heme oxygenase 1 (HO-1) in the testis were detected using immunofluorescence assay and Western blotting.
RESULTS:
Compared with the control group, the rats with MPEs exposure showed significant reductions of the anogenital distance, weight of the testis and epididymis, and the coefficients of the testis and epididymis with lowered serum testosterone, LH and FSH levels (P < 0.05). Testicular histological examination revealed atrophy of the seminiferous tubules, spermatogenic arrest, and hyperplasia of the Leydig cells in MPEs-exposed rats. MPEs exposure also caused significant increments of testicular Nrf2, MDA, SOD, CAT and HO-1 expressions and lowered testicular Keap1 expression (P < 0.05). Treatment with quercetin at the median and high doses significantly ameliorated the pathological changes induced by MPEs exposure (P < 0.05).
CONCLUSION
Quercetin treatment inhibits MPEs-induced oxidative testicular damage in rats possibly by direct scavenging of free radicals to lower testicular oxidative stress and restore the regulation of the Nrf2 signaling pathway.

ObjectiveTo establish a method for the analysis of oligosaccharides in Atractylodes lancea rhizome based on ultra-performance liquid chromatography tandem quadrupole-time-of-flight mass spectrometry (UPLC-Q-TOF-MS) and a method for the quantification of oligosaccharides in A. lancea rhizome based on UPLC-evaporative light scattering detector (ELSD), and to investigate the oligosaccharide characteristics of A. lancea rhizome from different habitats. MethodUPLC-Q-TOF-MS was used to identify the oligosaccharides in A. lancea rhizome with the mobile phase of 0.1% ammonia acetonitrile solution (A)-0.1% ammonia solution (B) for gradient elution (0-0.5 min, 98%A; 0.5-2.0 min, 98%-89%A; 2.0-2.5 min, 89%-86%A; 2.5-5.5 min, 86%-80%A; 5.5-6.5 min, 80%-72%A; 6.5-9.5 min, 72%-63%A; 9.5-14.0 min, 63%-50%A; 14.0-16.0 min, 50%A; 16.0-16.5 min, 50%-98%A; 16.5-20 min, 98%A), the column temperature of 60 ℃ and the flow rate of 0.2 mL·min-1. Electrospray ionization (ESI) was used to collect data in negative ion mode and the detection range was m/z 50-1 500. The qualitative analysis of oligosaccharides was accomplished by retention time, relative molecular weight, primary and secondary MS information of characteristic fragment ions in combination with reference substance information. UPLC-ELSD was employed to determine the contents of nine oligosaccharides in A. lancea rhizome with the mobile phase of 0.1% ammonia acetonitrile solution (A)-0.1% ammonia solution (B) for gradient elution (0-1 min, 98%-75%A; 1-7 min, 75%-70%A; 7-18 min, 70%-55%A; 18-23 min, 55%A; 23-23.5 min, 55%-98%A; 23.5-28 min, 98%A), the drift tube temperature of ELSD was set at 50 ℃. Principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA) were used to analyze the classification and differential components between A. lancea rhizome from different habitats. ResultA total of 24 oligosaccharides, containing 11 pairs of isomers, were identified from A. lancea rhizome. Among them, compared with samples from Anhui, Chongqing, Nanjing and Shaanxi, the contents of kestose(GF2), 1F-fructofuranosylnystose (GF4), kestohexose (GF5), fructo-oligosaccharide DP10 (GF9) in samples from Maoshan were statistically significant (P<0.05), and the total mass fraction of sucrose (GF1)-GF9 reached 16.47%. The peak area ratio of fructose-fructose oligosaccharide to its isomer sucrose-fructose oligosaccharide was greater than 1 in samples from Maoshan. ConclusionThe types and contents of oligosaccharides in A. lancea rhizome vary greatly among different habitats, and the peak area ratio of fructose-fructose oligosaccharide to sucrose-fructose oligosaccharide >1 may be one of the geoherb characteristics of A. lancea rhizome, which can provide a reference for the development, utilization and quality control of this herb.

Journal of Medical Biomechanics 2021;36(2):E238-E244
Objective Taking pig kidney as an example, through a series of comparative and analogical experiments, the influencing factors of compressive stress at relaxation stage of biological tissues were analyzed, and a more accurate and widely applicable biomechanical model at relaxation stage was established. Methods The compressive stress relaxation experiments of pig kidney under different conditions were carried out by using the self-built mechanical experiment platform. The collected data were analyzed and mapped, and various factors affecting the relaxation force changes were summarized. Based on the conclusion, the neural network learning algorithm was used to model the force change process at relaxation stage of pig kidney. Results The pre-extrusion pressure and relaxation time were the main influencing factors for compressive stress changes of biological tissues at relaxation stage. The average error of test sample validation experiment was 6.4 mN, and the average prediction error of generalization sample validation experiment was 34.9 mN, so the modeling effect was good. Conclusions Neural network modeling algorithm has the advantages of strong generalization ability and good fault tolerance, which contributes to providing more realistic force tactile feedback prediction for virtual surgery system. It is also a new idea for mechanical modeling of nonlinear biological tissues.

Objective:To use a self-developed, prefabricated cap for making interocclusal records in implant-supported fixed prosthetic treatment, and to evaluate its effect of clinical application and accuracy of transferring intra-oral intercuspal position. Methods:Series of prefabricated caps for occlusal recording of implant-supported fixed prostheses were designed based on the healing abutments, and fabricated with three-dimensional (3D) printing. According to the inclusion and exclusion criteria, 12 partially edentulous patients who visited the Department of Prosthodontics, Peking University School and Hospital of Stomatology between November 2020 and September 2021, had lost no fewer than 2 contiguous teeth in distal extension (Kennedy Class Ⅰ or Ⅱ), and had received implant placement 3 months ago were enrolled. Self-control study design was used. Two occlusal records of each included case were obtained by the following 2methods:for the test group, appropriate prefabricated caps were used; and for the control group, polyvinyl siloxane occlusal record was directly set on the healing abutments. The working casts were mounted on the mechanical articulator using the 2 groups of occlusal records successively. Accuracy of occlusal relationship of the mounted casts was evaluated. Diagnostics test was performed to obtain the sensitivity and positive predictive value, which were determined in photographs by comparing the intra-oral occlusal contact points with those in the mounted casts, point-by-point. Virtual casts were taken by intraoral and extraoral scans in intercuspal position and imported to Geomagic Studio 2014. Then the root mean square values of occlusal clearance space between the upper and lower occlusal surfaces of remaining posterior teeth, and the deviations of the interocclusal position of the occlusal surfaces of the remaining teeth in the mandibular arches when superimposing the maxillary arches of the intraoral and extraoral scans were calculated. As a self-control design, paired t test was used. Results:Twelve participants were enrolled, including 3 men and 9 women aged (52.6±12.1) years, and 36 missing teeth. The prefabricated caps could fit on the healing abutments with good retention and stability. The sensitivity of the test group (0.73±0.14) was significantly better than that of control group (0.63±0.12, P<0.01), with no significant difference in positive predictive value between the 2 groups ( P>0.05). The deviations of the interocclusal position of the occlusal surfaces of the remaining teeth were (357.0±140.2) μm for the test group, and (399.4±206.3) μm for the control group, with no significant difference between them ( P>0.05). Conclusions:Interocclusal position record based on prefabricated cap in this study for implant-supported fixed prosthetic treatment can improve the consistency between the intra-oral occlusion and the occlusion in dental casts. This technique has good accuracy, clinical convenience and usability.
