OBJECTIVES: To investigate the antioxidative mechanism of snake venom-derived protein C activator (PCA) in mitigating vascular endothelial cell injury. METHODS: Human umbilical vein endothelial cells (HUVECs) were cultured in DMEM containing 1.0 g/L D-glucose and exposed to hypoxia (1% O₂) for 6 h followed by reoxygenation for 2 h to establish a cell model of oxygen-glucose deprivation/reoxygenation (OGD/R). The cell model was treated with 2 μg/mL PCA alone or in combination with 2-ME2 (a HIF-1α inhibitor) or DMOG (a HIF-1α stabilizer), and intracellular production of reactive oxygen species (ROS) and protein expression levels of HIF-1α, BNIP3, and Beclin-1 were detected using DCFH-DA fluorescence probe, flow cytometry, and Western blotting. The OGD/R cell model was transfected with a BNIP3-specific siRNA or a scrambled control sequence prior to PCA treatment, and the changes in protein expressions of HIF-1α, BNIP3 and Beclin-1 and intracellular ROS production were examined. RESULTS: In the OGD/R cell model, PCA treatment significantly upregulated HIF-1α, BNIP3 and Beclin-1 expressions and reduced ROS production. The effects of PCA were obviously attenuated by co-treatment with 2-ME2 but augmented by treatment with DMOG (a HIF-1α stabilizer). In the cell model with BNIP3 knockdown, PCA treatment increased BNIP3 expression and decreased ROS production without causing significant changes in HIF-1α expression. Compared with HUVECs with PCA treatment only, the cells with BNIP3 knockdown prior to PCA treatment showed significantly lower Beclin-1 expression and higher ROS levels. CONCLUSIONS Snake venom PCA alleviates OGD/R-induced endothelial cell injury by upregulating HIF-1α/BNIP3 signaling to suppress ROS generation, suggesting its potential as a therapeutic agent against oxidative stress in vascular pathologies.

Objective To explore the clinical efficacy of different ligation methods in laparoscopic radi-cal resection of rectal cancer and sigmoid colon cancer and the influencing factors of anastomotic leakage.Methods A total of 82 patients who underwent laparoscopic radical resection of rectal cancer and sigmoid co-lon cancer in this hospital from December 2022 to December 2023 were selected and divided into the low-liga-tion group(n=42)and the high-ligation group(n=40)according to different surgical methods.The inferior mesenteric artery(IMA)classification,IMA length,operation time,intraoperative blood loss,total number of lymph node dissections,number of lymph node dissections in group 253,postoperative hospital stay,exhaust time,incidence of urinary retention,incidence of anastomotic leakage,and incidence of preventive stoma were compared between the two groups.Univariate and multivariate logistic regression analyses were conducted to analyze the influencing factors of anastomotic leakage in patients undergoing radical resection of rectal cancer and sigmoid colon cancer.Results There was no statistically significant difference in IMA length,IMA classi-fication proportion,operation time,intraoperative blood loss,the total number of lymph node dissections,the number of lymph node dissections in group 253,and the postoperative hospital stay between the two groups(P>0.05).Compared with the high-ligation group,the low-ligation group had a shorter exhaust time,lower incidence of anastomotic leakage and preventive ostomy,the differences were statistically significant(P<0.05).Anastomotic leakage occurred in 7 patients.Univariate analysis showed that whether left colic artery(LCA)was preserved,tumor location,presence of underlying diseases,and preventive stoma were associated with anastomotic leakage in patients undergoing radical resection of rectal cancer and sigmoid colon cancer(P<0.05).Multivariate logistics analysis showed that whether LCA was preserved,tumor location,presence of underlying diseases,and presence of preventive stoma were not influencing factors for anastomotic leakage in patients undergoing radical resection of rectal cancer or sigmoid colon cancer(P>0.05).Conclusion In laparoscopic radical resection of rectal and sigmoid colon cancer,the clinical efficacy of preserving LCA is com-parable to that of not preserving LCA,and it can reduce the risk of anastomotic leakage and the probability of preventive stoma,promoting the early recovery of intestinal function.

Objective: To understand the current status of health education faculty in Yuzhong District of Chongqing, so as to explore the implementation path to improve the construction of health education faculty. Methods: In August 2023, stratified random sampling was used to directly select a total of 555 teachers and school doctors from 17 schools in 11 streets of Yuzhong District, who were invited to conduct a questionnaire survey. A total of 409 fulltime or parttime health education teachers were included in the analysis of the results. Both χ2 test and Fisher exact probability method were analyzed for compare the differences in the frequency of conducting health education courses and specializing in specializing areas among different plant health education teachers in Yuzhong District. Results: Health education course faculty were mainly female teachers, accounting for 81.66%. The proportions of physical education (PE) teachers, school doctors, head teachers and teachers from other disciplines were 14.67%, 8.31%, 62.84%, 14.18%, and the class teachers (81.33%) and school doctor (80.95%) were found of the highest proportion of providing health education courses, and the teaching section was mainly elementary school (first to third grade: 42.30%, fourth to sixth grade: 44.74%). Among the teachers, school doctors had the highest proportion (88.24%) of receiving training related to health education courses. And there was a significant stastical difference in the frequency of conducting health education classes among PE teachers, school doctor, head teachers and the teachers of other courses (χ2=54.46, P<0.01). The results of Fisher exact probability method showed that the differences in selfassessment of the most and least skilled areas among PE teachers, school doctor, head teachers and teachers of other courses were statistically significant (P<0.01). Conclusions Health education faculty in Yuzhong District is mainly composed of PE teachers, school doctor, head teachers and teachers of other courses. The standardized training and assessment of teaching for school doctor could promote the school nurse group developing into specialized health education faculty.

Objective To explore the value of serum leucine-rich-alpha-2-glycoprotein-1(LRG1)and CC motif chemokine ligand 19(CCL19)detecting levels in children with henoch-schonlein purpura nephritis(HSPN)for early clinical diagnosis and prognosis.Methods A sum of 108 children with HSPN who were treated at Handan Central Hospital from May 2021 to May 2023 were selected as the study subjects,meantime,72 healthy children were as the control group.The levels of LRG1,CCL19,IgM,IgA,and IgG in serum were detected.Logistic regression method was applied to analyze the influencing factors of HSPN,receiver operating characteristic was applied to evaluate the clinical value of LRG1 and CCL19 levels for early diagnosis of HSPN in children.The differences in serum LRG1 and CCL19 levels among children with HSPN in the acute,chronic,and recovery stages were analyzed,Spearman correlation was applied to analyze and explore the correlation between serum LRG1,CCL19 levels and the duration of HSPN in children.Results The levels of LRG1(184.36±23.64 ng/L)and CCL19(463.19±89.46 ng/L)in the HSPN group were obviously higher than those in the control group(149.42±18.29 ng/L,208.83±52.97 ng/L),and the differences were significant(t=10.600,21.710,all P＜0.05).The Logistic regression results showed that LRG1[OR(95％CI):1.429(1.057～1.933)]and CCL19[OR(95％CI):1.842(1.216～2.791)]were both influencing factors for the occurrence of HSPN in children(P＜0.05).According to the analysis of receiver operating characteristic(ROC),the areas under the curve(AUC)of serum LRG1,CCL19 and their combined diagnosis of HSPN in children were 0.868,0.881 and 0.952,respectively,and their combined application in clinical diagnosis was better than that of serum LRG1 and CCL19 in their separate diagnosis(Z=3.147,3.487,all P=0.001).The levels of LRG1(203.49±24.89 ng/L,177.56±23.19 ng/L)and CCL19(591.13±98.32 ng/L,415.61±89.82 ng/L)in the serum of HSPN patients in the acute and chronic stages were obviously higher than those in the rehabilitation stage(158.53±21.96 ng/L,295.17±69.61 ng/L),and the differences were significant(t=6.917,12.101;5.320,3.102,all P＜0.05),while the levels of LRG1 and CCL19 in the serum of HSPN patients in the acute stage were obviously higher than those in the chronic stage,and the differences were significant(t=5.059,8.750,all P＜0.05).Spearman correlation analysis showed that the serum levels of LRG1 and CCL19 in children with HSPN were positively correlated with their course of disease(r=0.506,0.689,all P＜0.001).Conclusion The levels of serum LRG1 and CCL19 are elevated in HSPN children,and combined detection of serum LRG1 and CCL19 can improve the early clinical diagnostic value of HSPN and evaluate the prognosis of children with HSPN.

Objective:To study the methods of hormone-related teaching in standardized residency training of endocrinology.Methods:From February 2022 to June 2022, 25 residents in the Department of Endocrinology, Peking University Third Hospital were given a standardized residency training. The teaching mode combined with a hundred years of insulin development was adopted to practice insulin teaching, and questionnaires were evaluated before and after the teaching. Chi-square test was performed by SPSS 20.0.Results:A total of 31 physicians participated in the training, and 25 residents who were included in the first-stage standardized training were analyzed. Before the training, only 3 people (12%) knew the development history of insulin, and 10 people (40%) answered all the questions about clinical use of insulin correctly. After the training, 23 residents (92%) said they knew or were familiar with the history of insulin development, and 20 residents (80%) answered the questions about the clinical use of insulin correctly, all of which were higher than those before the training, and the difference was statistically significant.Conclusion:The integration of medical history in standardized residency training in endocrinology department can effectively improve the teaching effect, and enhance the humanity quality of young doctors, which can become an effective vehicle in standardized residency training.

Objective To explore the expression of zinc finger protein 217 (ZNF217) in non-small cell lung cancer (NSCLC) and its correlation with prognosis of patients. Methods A total of 120 NSCLC patients in Chongqing Three Gorges Central Hospital from January 2012 to October 2013 were selected. Immunohistochemical method was used to test the expression of ZNF217 in NSCLC tissues and paracancerous tissues. The correlation of ZNF217 expression with patient's clinicopathological features was analyzed. At the same time, the Kaplan-Meier method and Cox regression model multiple factor analysis method were used to explore the factors affecting the prognosis of patients after NSCLC radical operations. Results ZNF217 mainly existed in cell nucleus of NSCLC. The positive expression rate of ZNF217 in the cancer tissues was higher than that in the paracancerous tissues [52.5％ (63/120) vs. 20.1％ (25/120), χ 2 = 25.909, P < 0.05]. The positive expression rate of ZNF217 increased with the increase of tumor T stage (χ 2 = 7.333, P = 0.026), N stage (χ 2 = 7.782, P = 0.020) and TNM stage (χ 2 = 11.557, P = 0.003). The overall survival (P = 0.007) and progression-free survival (P = 0.004) of patients with positive ZNF217 were poorer than those of patients with negative ZNF217. Cox multiple factor analysis showed that ZNF217 was an independent risk factor affecting the prognosis of NSCLC. Conclusion ZNF217 is an independent risk factor affecting the prognosis of NSCLC, and it may be a potential target for accurate treatment of NSCLC.

Objective To explore the effect of multidisciplinary team nursing model in patients with lung cancer.Methods Totally 104 patients with lung cancer admitted in Chongqing Three Gorges Central Hospital from October 2013 to September 2016 were selected and divided into the observation group (multidisciplinary team) and the control group (routine care) according to random number table method. The quality of life,the incidence of complications and nursing satisfaction between two groups were compared.Results In the observation group,the scores of neuritis,catarrh,cough,dyspnea,chest pain,shoulder pain,hemoptysis and pain in other parts were significantly lower than those of the control group (P<0.05). Fifty-two cases in the observation group,there was one case of respiratory failure,one case of arrhythmia and one case of thrombosis, so the total incidence rate of complications was 5.77%;fifty-two cases in the control group,there was two cases of respiratory failure,one case of arrhythmia,one case of pulmonary infection and four cases of thrombosis,so the total incidence rate of complications was 15.38%. The satisfaction rate in the observation group was 98.08%, while in the control group,it was 80.77%.Conclusions Multidisciplinary team has obvious advantages in the nursing care of patients with lung cancer,and it can improve the quality of life of patients and reduce the incidence of adverse complications.

Objective To investigate the adiponectin(ADIPOQ)gene polymorphisms and which association with colorectal cancer(CRC).Methods Genotyping of blood samples were performed for 250 case-control pairs.Taqman real-time PCR was used to test the three single nucleotide polymorphisms(SNPs),namely rs266729,rs2441 766,rs1 501299.Logistic regression was applied to assess the effects of three SNPs,the gene-gene,and gene-environment interactions on CRC risk.Results The ADIPOQ rs266729 GG and CG+GG genotype had a higher CRC risk than those carrying the CC genotype[OR (95%CI ):1.87 (1.01 - 3.47),1.63 (1.14-2.32),respectively].The same results was observed in cases who carried TG+GG genotype vs .TT genotype in rs2441 766 [OR(95%CI ):1.45(1.02-2.06)].The rs1 501299 GT+TT genotype had a lower CRC risk than those carrying the GG genotype [OR(95%CI ):0.61(0.43-0.88)].Furthermore,in two-factor gene-environment interaction analyses,rs266729 presented signifi-cant interactions with Body mass index (BMI),with OR of 1.1 6 (95%CI :1.03-1.30).Conclusion The results suggest that vari-ants in ADIPOQ may contribute to increased colorectal cancer risk and this contribution may be modified by BMI.
