OBJECTIVE To investigate the potential mechanism of epigallocatechin gallate (EGCG) enhancing the sensitivity of hepatocellular carcinoma (HCC) cells to lenvatinib based on the phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway. METHODS Five human HCC cell lines (HepG2, Huh-7, SMMC-7721, SNU-368 and SNU-739) were used to evaluate the effects of lenvatinib alone and in combination with EGCG on survival rates, clone number, proliferation rate, invasion number and the expressions of mRNAs and proteins related to the PI3K/Akt signaling pathway. The PI3K activator insulin-like growth factor-1 (IGF-1) was introduced to investigate the effect of activating the PI3K/Akt signaling pathway on the sensitization effect of EGCG. RESULTS Compared with the control group, lenvatinib (10 μmol/L) and different concentrations of EGCG+ lenvatinib (1, 5 and 10 μg/mL EGCG+10 μmol/L lenvatinib) significantly reduced the survival rates and clone numbers of all five HCC cell lines in a dose-dependent manner (P<0.05). Lenvatinib (10 μmol/L) and EGCG+lenvatinib (10 μg/mL EGCG+10 μmol/L lenvatinib) also markedly inhibited the proliferation rate and invasion numbers of these cells, and decreased the mRNA expressions of PI3K, Akt, mammalian target of rapamycin (mTOR), P70S6K and 4EBP, and the phosphorylation levels of PI3K and Akt, as well as the protein expressions of mTOR and B cell lymphoma-2 (Bcl-2) in HepG2 cells or all five HCC cells; conversely, the mRNA and protein expressions of phosphatase and tensin homologue deleted on chromosome 10(PTEN), and the protein expressions of caspase-3 and cleaved caspase-3 were significantly upregulated, with more pronounced effects observed in the EGCG+lenvatinib group than in the lenvatinib group (P<0.05). Compared with the lenvatinib group and the EGCG+lenvatinib group, the clone number, proliferation rate and invasion number of HepG2 cells in the EGCG+lenvatinib+IGF-1 group (10 μg/mL EGCG+10 μmol/L lenvatinib+50 ng/mL IGF-1) were significantly increased (P<0.05). CONCLUSIONS EGCG can enhance the sensitivity of HCC cells to lenvatinib, and its underlying mechanism may be related to the inhibition of the activation of PI3K/Akt signaling pathway activation.

Patients with severe trauma require an extremely timely treatment and transfusion plays an irreplaceable role in the emergency treatment of such patients. An increasing number of evidence-based medicinal evidences and clinical practices suggest that patients with severe traumatic bleeding benefit from early transfusion of low-titer group O whole blood or hemostatic resuscitation with red blood cells, plasma and platelet of a balanced ratio. However, the current domestic mode of blood supply cannot fully meet the requirements of timely and effective blood transfusion for emergency treatment of patients with severe trauma in clinical practice. In order to solve the key problems in blood supply and blood transfusion strategies for emergency treatment of severe trauma, Branch of Clinical Transfusion Medicine of Chinese Medical Association, Group for Trauma Emergency Care and Multiple Injuries of Trauma Branch of Chinese Medical Association, Young Scholar Group of Disaster Medicine Branch of Chinese Medical Association organized domestic experts of blood transfusion medicine and trauma treatment to jointly formulate Chinese expert consensus on blood support mode and blood transfusion strategies for emergency treatment of severe trauma patients ( version 2024). Based on the evidence-based medical evidence and Delphi method of expert consultation and voting, 10 recommendations were put forward from two aspects of blood support mode and transfusion strategies, aiming to provide a reference for transfusion resuscitation in the emergency treatment of severe trauma and further improve the success rate of treatment of patients with severe trauma.

Aim To investigate the molecular mechanism by which quercetin inhibits the malignant behavior of breast cancer cells. Methods Breast cancer cell lines MCF-7 and MB231 were used as the research models. Lentiviral transfection was employed to establish tumor cells with high expression of ERa and MAL-AT-1. The expression of MALAT-1 was assessed using RT-qPCR,and ERa expression was determined through Western blot. Subsequently, CCK-8 assay and colony formation assay were conducted to evaluate cell proliferation. PI staining and adenovirus transfection were performed to observe the inhibitory effects of quercetin on breast cancer cell proliferation. Results 17|3-es-tradiol ( E2 ) promoted the proliferation of MCF-7 breast cancer cells, while 5 jjunol L quercetin reversed the promoting effect of E2 on proliferation ( P 0. 05 ) . Quercetin had no effect on MB231 breast cancer cells. Overexpression of ERa significantly inhibited the pro-proliferative effect of E2 on MB231-ERa cells, and quercetin further suppressed this effect. Additionally , quercetin inhibited the expression of MALAT-1. However, this inhibitory effect was reversed by overexpression of MALAT-1, leading to enhanced cell proliferation , cell cycle progression, and clonal formation a-bility. Conclusions Quercetin exerts its anti-tumor effects on breast cancer cells by regulating MALAT-1, dependent on the presence of estrogen receptor. Quercetin shows potential as a therapeutic drug for breast cancer targeting the estrogen receptor.

Chinese Journal of School Health 2024;45(7):1031-1035
Objective:
To analyze the acute effects of exposure to fine particulate matter (PM2.5) components on primary school students lung function, so as to provide a scientific basis for protecting childrens respiratory health.
Methods:
From 2019 to 2021, the study selected a total of 2 120 primary school students from grades 3 to 5 in Tianjin using a stratified random sampling method to undergo lung function tests. At the same time, the shortterm exposure levels were simulated by combining PM2.5 components and student addresses, and the acute impact of PM2.5 exposure on primary school students lung function was analyzed by generalized linear models (GLM) and weighted quantile sum (WQS) regression models.
Results:
The average daily concentration of PM2.5 in the air of Tianjin from 2019 to 2021 was 81.14 μg/m3, which was higher than the national standard. The results of lung function testing showed that there was no statistically significant difference in lung function measurement indicators such as forced vital capacity (FVC), forced expiratory volume at 1 second (FEV1), peak expiratory flow (PEF), 75% forced expiratory volume in 75 (FEF75), and 25% forced expiratory volume in 25 (FEF25) among primary school students in different regions of Tianjin (F=1.23, 0.87, 2.34, 1.56, 0.98, P>0.05). But the GLM analysis results showed that all components of PM2.5 in the air had adverse effects on the lung function indicators of primary school students. When the concentrations of fluorene (FLU) and pyrene (PYR) increased by 10 ng/m3, the FVC of primary school students decreased by 166.44 and 61.94 L respectively. The WQS regression model analysis results showed that the mixed exposure of PM2.5 components particularly significant damaging effects on lung function indicators in primary school students, especially the FLU and PYR components in polycyclic aromatic hydrocarbons, as well as the heavy metal lead.
Conclusions
Both single and mixed exposure to various PM2.5 components in the air have adverse effects on the lung function of primary school students. Among them, the influences of FLU and PYR in polycyclic aromatic hydrocarbons, as well as heavy metal Pb, are particularly significant.Potential pollution sources should be controlled to protect the respiratory health of primary school students by comprehensive prevention and control measures.
