1.Optimization Study of Rat Models for Sequelae of Pelvic Inflammatory Disease

Objective:To establish a stable rat model of sequelae of pelvic inflammatory disease(SPID)with clinical characteristics,and to provide a reliable experimental model for the study of the pharmcological effect and mechanism of SPID.Methods:Twenty-four 7-week-old SD rats were divided into sham operation group,model-A(108 cfu/mL mixed bacterial solution,0.2 mL),model-B(109 cfu/mL mixed bacterial solution 0.2 mL),and model-C(108 cfu/mL E.coli 0.2 mL).The weight of the rat's uterine was weighed and the uterine index was calculated.The automatic hematology analyzer was used to detect the blood routine;hematoxylin-eosin staining(HE)and masson staining were used to detect uterine pathlogical changes in rats.Enzyme-linked immunosorbent assay(ELISA)was used to detect interleukin-1β(IL-1β),interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)in rat uterine tissue homogenates.Western blot was used to detect the expression of proteins related to NF-κB signaling pathway.Results:Compared with the sham operation group,the uterine index of model-A,model-B,and model-C were significantly increased(P<0.05,P<0.01).The levels of WBC and NE in the model-A increased significantly(P<0.01).The level of LY in model-B decreased significantly(P<0.01).The levels of IL-1β,TNF-α in model-A,model-B,and model-C were significantly increased(P<0.01).The levels of IL-6 in model-A and model-B were significantly increased(P<0.05,P<0.01).The collagen volume fraction of model-A and model-B were significantly increased(P<0.01).Mechanism study indicates that the expression levels of p-IKKβ/IKKβ,p-IκBα/IκBα and p-p65/p65 in model-A were significantly increased(P<0.01),and the expression levels of IκBα/β-actin were significantly decreased(P<0.01).The expression level of p-IKKβ/IKKβ in model-B was significantly increased(P<0.01).Conclusions:A stable rat model of SPID that conforms to clinical characteristics can be successfully constructed by combining 0.2 mL of mixed bacterial solution with a concentration of 108 cfu/mL and mechanical injury.This modeling method intervened in the expression of the NF-κB inflammatory signaling pathway.
2.The influence of cardiac rehabilitation therapy on the postoperative quality of life, cardiac function and prognosis of patients undergoing transcatheter aortic valve replacement

Objective:To explore the influence of cardiac rehabilitation therapy on the postoperative quality of life, cardiac function and prognosis of patients undergoing transcatheter aortic value replacement (TAVR).Methods:A total of 182 patients with aortic valve stenosis who underwent TAVR in the Union Hospital, Tongji Medical College, Huazhong University of Science and Technology from January 2021 to May 2023 were selected as the research subjects. The patients were divided into the observation group and the control group by the random number table method, with 91 cases in each group. The control group was given conventional drug treatment after the operation, while the observation group was given conventional drug treatment + cardiac rehabilitation treatment after the operation. The baseline clinical data, quality of life before and after intervention (Minnesota Quality of Life Scale score), cardiac function [6-minute walking distance (6MWT), B-type brain natriuretic peptide (BNP)] of the two groups of patients were compared. The left ventricular ejection fraction (LVEF), left ventricular end-systolic diameter (LVESD), left ventricular end-diastolic diameter (LVEDD), and the incidence of adverse events one year after TAVR were evaluated using the Cox regression model to assess the effect of cardiac rehabilitation therapy on the occurrence of adverse events one year after TAVR. Kaplan-Meier survival analysis was used to compare the cumulative rate of no adverse events one year after TAVR in the two groups of patients.Results:After the intervention in both groups, the Minnesota Quality of Life Scale score, 6MWT, BNP, and LVEF were significantly improved compared with those before the intervention (all P<0.05), and the Minnesota Quality of Life Scale score, 6MWT, BNP, and LVEF in the observation group after the intervention were better than those in the control group (all P<0.05). The incidences of stroke and heart failure in the observation group one year after TAVR were significantly lower than those in the control group (all P<0.05). Cox regression analysis showed that cardiac rehabilitation significantly reduced the risk of adverse events one year after TAVR in patients ( HR: 0.72; 95% CI: 0.46-0.97; P=0.035). Kaplan-Meier survival analysis showed that the cumulative rate of no adverse events at 3, 6, and 12 months after TAVR in the observation group was significantly higher than that in the control group (94.3%, 89.5%, 76.8% vs 83.4%, 74.6%, 57.8%, all P<0.05). Conclusions:Cardiac rehabilitation therapy can significantly improve the postoperative quality of life and cardiac function of patients with TAVR, bringing obvious survival benefits to such patients.
3.Analysis of factors influencing right atrial strain in patients with pulmonary arterial hypertension

Objective To analyze the correlation between right atrial strain at various stages and various influencing factors in patients with pulmonary hypertension,and to explore the role of right atrial strain in the assessment of pulmonary hypertension.Methods A total of 239 cases diagnosed with pulmonary hypertension who underwent echocardiography and complete right heart catheterization at hospital from October 2021 to December 2023 were included.Conventional ultrasound parameters such as right heart strain,right atrial area(RA area),inferior vena cava diameter(IVC diameter),and collapse rate of the inferior vena cava(IVC diameter changes)were measured.The heart rate(HR)corresponding to the ultrasound images were recorded.General information such as age and gender,as well as catheter data including mean right atrial pressure(mRAP),mean pulmonary artery pressure(mPAP),and pulmonary vascular resistance(PVR),were collected.The relationship between right atrial strain and its influencing factors was analyzed,and further analysis was conducted by dividing into shunt group and non-shunt group based on the presence or absence of left-to-right shunt disease.Results The correlation with RA reservoir strain(RASr)from high to low is RV global strain(RV4CSL),RV free wall strain(RVFWSL),RA area,IVC diameter,mRAP,age,HR,and PVR;the correlation with RAconduit strain(RAScd)from high to low is RV4CSL,RVFWSL,RA area,IVC diameter,mRAP,age,PVR,and HR;the correlation with RA contraction strain(RASct)from high to low is RA area,RV4CSL,RVFWSL,mRAP,IVC diameter,and HR.The collapse rate of the inferior vena cava is correlated with strain at various stages of the right atrium;gender is correlated with RASr and RASct.Conclusions Right atrial strain can reflect changes in right atrial function,with the highest correlation to right ventricular strain and right atrial area.Right atrial strain can indicate the severity of right ventricular function and right atrial remodeling,serving as an evaluative index for the condition and treatment outcomes of pulmonary arterial hypertension.

7.The effect of salidroside derivative pOBz on angiogenesis after ischemic stroke by regulating Notch signaling pathway

Aim To study the effect of p-benzoyl sali-droside(pOBz)on angiogenesis after ischemic stroke and to explore the underlying mechanism.Methods The MCAO model was prepared by suture method.Rats were divided into four groups:sham,MCAO,pOBz administration,and edaravone positive control,treated for seven days.The mNSS was used to assess the neurological impairment.Western blotting was em-ployed to detect CD31,NICD,and Hes-1 protein ex-pression,while immunofluorescence staining was ap-plies to quantify CD31-positive cells in ischemic brain tissue.In vitro an OGD/R model was established in HUVECs.Following treatment with varying pOBz con-centrations(0.01,0.1,1 μmol·L-1),the CCK-8 as-say was uses to measure cell viability,and in vitro tube formation assay was utilized to evaluate angiogenesis.Western blotting was employed again to assess CD31,NICD and Hes-1 protein levels.To further elucidate the mechanism,HUVEC were treated with the Notch inhibitor DAPT prior to grouping and pOBz administra-tion,and the same parameters were evaluated.Results pOBz significantly reduced the mNSS score of MCAO rats,increased CD31-positive cell counts,and upregu-lated CD31,NICD,and Hes-1 protein expression(P<0.01).In vitro results further showed that pOBz could dose-dependently increase the survival rate and angio-genesis ability of HUVEC induced by OGD/R,and promote CD31,NICD and Hes-1 proteins(P<0.01),and Notch inhibitor DAPT could reverse the above effects of pOBz.Conclusion pOBz promotes angio-genesis in HUVEC,and its mechanism involves activa-tion of the Notch signaling pathway.

Objective:To observe the effect of WAFF abdominal and pelvic mechanical exercise combined with pelvic floor muscle training during pregnancy on the recovery of pelvic floor muscle function in primiparas after childbirth.Methods:This study was a prospective case-control study,a total of 100 primiparas with a pregnancy of more than 16 weeks who were admitted to Longyan People's Hospital of Fujian from October 2021 to November 2023 were recruited and and randomly divided into control group(n=50)and observation group(n=50).The control group received pelvic floor muscle training,while the observation group received WAFF abdominal and pelvic mechanical exercise on the basis of the control group.The weight gain during pregnancy,mode of delivery,newborn birth weight,duration of second stage of labor,pelvic floor muscle strength,incidence of postpartum pelvic floor organ prolapse,incidence of stress urinary incontinence,pelvic floor electrophysiological indicators,and psychological states[Self rating Anxiety Scale(SAS),Self rating Depression Scale(SDS)]between two groups were compared.Results:The newborn birth weight in the observation group was lighter than that in the control group,the weight gain during pregnancy was less than that in the control group,the duration of second stage of labor was shorter than that in the control group,the number of cases of cesarean section was less than that in the control group,and incidence of postpartum pelvic floor organ prolapse,incidence of stress urinary incontinence were lower than those in the control group(P<0.05).The pelvic floor muscle strength in the observation group was better than that in the control group at 42 d postpartum(P<0.05).The fatigue levels of Class Ⅰ and Class Ⅱ muscle fibers in the observation group were lower than those in the control group,while the vaginal dynamic pressure,Class Ⅰ muscle fiber strength,and Class Ⅱ muscle fiber strength were higher than those in the control group(P<0.05).The SAS and SDS scores in the observation group after intervention were lower than those in the control group(P<0.05).Conclusion:WAFF abdominal and pelvic mechanical exercise combined with pelvic floor muscle training during pregnancy can effectively improve the pregnancy outcomes of primiparous women,improve pelvic floor electrophysiological indicators and psychological states,and reduce the incidence of postpartum pelvic floor organ prolapse and stress urinary incontinence.

10.2-Fluorofucose Increases Chemosensitivity of Human Breast Cancer Cells to Paclitaxel

Paclitaxel(PTX)is a first-line chemotherapy drug for breast cancer,but its resistance issues significantly impact clinical treatment efficacy.Fucosylation,especially core fucosylation,is closely related to tumor chemoresistance,resulting in poor chemotherapy responses and poor prognosis in patients.In this study,we investigated the effect and mechanism of the fucosylation inhibitor 2-fluorofucose(2-F-Fuc)on the chemosensitivity of paclitaxel-resistant breast cancer MCF-7/PTX cells.The drug resistanceindex(RI)of MCF-7/PTX cells was 8.49 by MTT assays.Western blotting,real-time PCR,enzyme-linked immu-nosorbent assay(ELISA)and Lens Culinaris Agglutinin(LCA)lectin imprinting showed that compared with MCF-7 cells,the expression of FUT8,MDR1and core fucosylation in MCF-7/PTX cells was high.Western blotting showed that 2-F-Fuc had a significant inhibitory effect on the growth of MCF-7/PTX cells,and the expression levels of FUT8 and MDR1 were significantly down-regulated after 2-F-Fuc treatment,and the down-regulation was more pronounced in the PTX and 2-F-Fuc combination group(P<0.05).Compared to the control,expression of PCNA in MCF-7/PTX cells in the PTX and the 2-F-Fuc group were down-regulated,and the apoptosis-related proteins,such as cleaved caspase-3 and Bax/Bcl-2 were in-creased.The level of p-PI3K and p-AKT were down-regulated,and the changes in the combination of 2-F-Fuc and PTX were more robust(P<0.05).The above results showed that the core fucosylation level of MCF-7/PTX cells was significantly increased,and 2-F-Fuc could reduce the core fucosylation level of MCF-7/PTX cells by inhibiting the expression of FUT8,and enhance the sensitivity of drug-resistant cells to PTX,which may correlate with the downregulation of PI3K/AKT signaling pathway proteins.
