Objective To explore the effect of SGLT2 inhibitor(SGLT2i)Dagaglipzin(Dap)on mitochondrial autophagy in cardiomyocytes in diabetic cardiomyopathy(DCM)by regulating mitochondrial membrane transporter TOM20.Methods H9C2 cardiomyocytes cultured in vitro were divided into normal control(Con)group,HG+PA group, knockout TOM20(HG+PA+siTOM20)group,knockout control(HG+PA+siNC)group,Dap intervention(HG+PA+Dap)group, and solvent control(HG+PA+DMSO)group. The expression of TOM20 was predicted by RNA-seq second-generation sequencing in HG+PA group. The expression of TOM20 and mitochondrial autophagy related proteins was detected by Western blot, the mitochondrial membrane potential was measured by immunofluorescence staining, and the mitochondrial autophagy level was evaluated by transmission electron microscopy. Results Compared with Con group, protein expressions of TOM20 and P62 were increased(P<0.05),while protein expressions of ULK1,Parkin,Beclin1 and LC3-B were decreased(P<0.05),mitochondrial membrane potential was destroyed, the number of mitochondrial autophagosomes was reduced, and autophagy activity was inhibited in HG+PA group.Compared with HG+PA+siNC group and HG+PA+DMSO group, the protein expressions of TOM20 and P62 were decreased(P<0.05),while the protein expressions of ULK1,Parkin,Beclin1 and LC3-B were increased(P<0.05),the damage of mitochondrial membrane potential decreased, the number of mitochondrial autophagosomes increased, and the inhibition of autophagy activity reversed in HG+PA+siTOM20 and HG+PA+DAPA groups. Conclusions SGLT2 inhibitors appear to partially restore mitochondrial function in diabetic cardiomyopathy by suppressing TOM20 expression, potentially through activation of mitochondrial autophagy.

Objective To conduct Rh blood group serological testing and third-generation sequencing(TGS)on 22 RhD variant voluntary blood donors in Chongqing and explore the phenotypic distribution and genotyping of RhD variants in Chongqing. Methods From January to August 2023,individuals who participated in blood donation in our blood center were selected as the study objects.RhD variant phenotype identification was performed using routine serological methods.Once the RhD variants were identified, tests on different antigenic epitopes of RhD were conducted using a D-screen assay kit.Furthermore, after the genomic DNA from 22 RhD variant blood samples was extracted, imbraided primers design and multi-segment amplification and splicing were used to sequence the full-length RHD gene for TGS.The RHD gene sequence was analyzed using SnapGene software. Results Among the 22 RhD variants,8 were DVI type 3(36.36%),with the main mutation of RHD-CE(3-6)-D hybrid allele.Six cases(27.27%)showed partial weak D15 type, with the main mutation of c.845G>A.There were 6 cases of Asia type Del(27.27%),with the main mutation of c.1227G>A.One case was weak D17 type with a mutation of c.340C>T and 1 case speculated to be partial D(c.491A>T,p.Asp164Val,missense mutation). Conclusion The most common RhD variant phenotype among blood donors in Chongqing is DVI type 3,and the full-length haplotype sequence of RHD variant alleles can be obtained by Pacific Bioscience single-molecule real-time sequencing(SMRT).

Objective To preliminarily understand the current status of medication safety management of medical institutions in China. Methods Medication Safety Panel in China Core Group of International Network for the Rational Use of Drugs (INRUD) and Chinese Pharmacological Society Professional Committee of Drug-induced Diseases jointly established a research group. Basing on the voluntary principle, members (medical institutions)of the group did medication safety self-assessment using the questionnaires of "2011 ISMP Medication Safety Self Assessment? for Hospitals (Chinese version)", which included 10 key elements,20 core indicators, and 270 assessment projects. The questionnaires were handed out on August 17,2018 and needed to be completed and submitted within 2 months. Results As of October 19,2018,67 hospitals of 16 provincial administrative regions in total had submitted their questionnaires, including 61 (91. 0%)3A hospitals and 6 (9%)2A hospitals. The average value of total scores of medication safety self-assessment in the 67 hospitals was 58. 9% (7. 6% -90. 0%). None of the 67 hospitals evaluated the key element Ⅵ(medication device acquisition, use, and monitoring). The scores of the other 9 key elements from high to low were 67. 6%,66. 2%,65. 1%,64. 8%,64. 1%,58. 2%, 54. 5%,54. 4%,and 52. 5% respectively for element Ⅶ (environmental factors, workflow and staffing patterns),element Ⅳ(drug labeling, packaging and nomenclature),element Ⅸ (patient education), element Ⅲ(communication of drug orders and other drug information),element Ⅷ (staff competency and education),element Ⅴ(drug standardization, storage and distribution),element Ⅹ (quality processes and risk management),element Ⅰ (patient information),and element Ⅱ (drug information). Conclusion The data of medication safety from 67 hospitals of 16 provincial administrative regions were obtained through the first national self-assessment questionnaire survey in medical institutions, which initially reflected the current status of medication safety in medical institutions in China.

Objective To study the changes of neutrophil surface molecule CD64 and inflammatory factor levels in patients with burn infection. Methods 46 cases of patients with burn infection who were treated in the department of burn in our hospital between March 2010 and October 2015 were selected as the observation group. Meanwhile, 46 cases of healthy people who underwent physical examination in our hospital during the same period were included in the control group. The levels of neutrophil surface molecule CD64, C-reactive protein (CRP), white blood cell count (WBC) and inflammatory factor [interleukin IL-6 (IL-6), interleukin (IL-8), tumor necro-sis factor (TNF-α) in the two groups of subjects were determined by flow cytometry and were compared. Results In the observation group, the levels of CD64, CRP, WBC and inflammatory factors were significantly higher than those in the control group (P < 0.05). Conclusion The levels of CD64, CRP, WBC and inflammatory factors in patients with burn infection are significantly higher than those in healthy people , which indicates that neutrophil surface molecule CD64, inflammatory factors and burn infection are closely correlated.

OBJECTIVE:To establish an HPLC method for the detection of Prednisone acetate, Hydrocortisone, Dexamethasone acetate, Piroxicam, Diclofenac sodium, Indometacin and Ibuprofen, the 7 chemical constituents in antirheumatic traditional Chinese medicine. METHODS: The chromatographic separation was performed on a Hypersil BDS C18 column (200 mm?4.6 mm,5 ?m) using methanol-1 g?L-1KH2PO4(58∶42) (adjusted to pH 4.0?0.05 with H3PO4) as the mobile phase at a flow rate of 1.0 mL?min-1. The detective wavelength was set at 220 nm. RESULTS: Seven chemical constituents were all well-separated with good linearity within respective range of sample size. CONCLUSION: The method is rapid and convenient, and can be used for the quantitative and qualitative analyses of 7 chemical constituents in antirheumatic traditional Chinese medicine and to detect whether there is addition of chemical constituents in antirheumatic traditional Chinese medicines.
