1.Study on influence of Daxx on cell cycle and chemotherapy in human ovarian cancer cells

Objective To explore the influence of down-regulating Daxx on cell cycle and chemotherapeutic drug resistance in human ovarian cancer cells.Methods SiRNA and NC negative control(NC) RNA of Daxx were constructed and divided into the NC group and silencing Daxx(siDaxx) group after transfecting to ovarian cancer cell line C13k.The doxorubicin concentration gradient of 0,0.15,0.30,0.60 μmol/L was set.The cellular cycle changes and apoptosis changes of these two groups were detected by using the flow cytometry.Western blot was used to detect the expression changes of apoptosis related protein cyclinB1 and cleavedparp.Results 0.30 μmol/L doxorubicin down-regulated Daxx to result in significant G2/M arrest(P<0.05).Down-regulating Daxx resulted in doxorubicin resistance in C13k cells.Conclusion The effect of Daxx on ovarian cancer chemotherapy might be related to its regulation on cell cycle.
2.Effect of domestic wire-reinforced epidural catheter on occurrence of adverse events during epidural block

Objective To evaluate the effect of domestic wire-reinforced epidural catheter on the occurrence of adverse events during epidural block.Methods Three hundred ASA Ⅰ or Ⅱ patients of both sexes,aged 18-64 yr,weighing 41-78 kg,scheduled for elective operations under combined spinal-epidural anesthesia,were randomly divided into 3 groups (n =100 each):polyvinyl chloride epidural catheter group (group A),imported wire-reinforced epidural catheter group (group B) and domestic wire-reinforced epidural catheter group (group C).Combined spinal-epidural anesthesia was performed routinely.The corresponding epidural catheter was inserted in each group.The catheterization without difficulty,paresthesia during catheterization,the number of patients in whom blood or cerebrospinal fluid was withdrawn from the epidural catheter,intravascular catheter insertion,injection obstruction,easiness during removal of the catheter,bleeding after removal,postoperative paresthesia and epidural hematoma within 1 week after operation were recorded.Results Compared with A group,the incidences of paresthesia during catheterization,the number of patients in whom blood or cerebrospinal fluid was withdrawn from the epidural catheter,injection obstruction and postoperative paresthesia were significantly decreased (P < 0.05),and no significant change was found in the other parameters in B and C groups (P > 0.05).There was no significant difference in all the parameters between B group and C group (P > 0.05).Conclusion Domestic wire-reinforced epidural catheter can decrease the occurrence of catheterization-induced damage to the nerve and blood vessels and the efficacy is comparable with that of imported wire-reinforced epidural catheter.
3.Clinical Effect of Acupuncture Combined with Rehabilitation on Dysphagia after Stroke at Different Stage

Objective To observe the therapeutic effect of acupuncture combined with rehabilitation on dysphagia after stroke at differentstage. Methods According to the randomized trial principle, 155 cases were divided into two groups: control group (n=80) and observationgroup (n=75). The control group was treated with rehabilitation training, and the observation group was treated with acupuncture and rehabilitationtraining, 5 times every week for 4 weeks. The two groups were assessed by TCM swallowing assessment scores and Kubota testbefore and after treatment. Results According to Kubota test, the total rate was 66.67% in the control group and 89.33% in the observationgroup with a significant difference between the groups (P<0.001). In the observation group, the total rate was 90.48% at acute stage and88.89% at the convalescence stage with a significant difference (P<0.01). According to TCM swallowing assessment, the total rate was64.00% in the control group and 74.67% in the observation group with no significant difference between the groups (P<0.05). In the observationgroup, the total rate was 90.48% at acute stage and 68.52% at the convalescence stage with a significant difference (P<0.001). ConclusionAcupuncture combined with rehabilitation facilitates to improve the swallowing function in stroke patients following dysphagia especiallyat acute stage.
